Effects of SNP in TSH-β Gene of Chicken on Economic Traits (닭의 경제 형질에 미치는 TSH-β 유전자 변이 효과 분석)

  • Thyroid hormone (TH) plays a role in growth of the poultry. Thyroid-stimulating hormone (TSH) stimulates production and distribution of TH, and is a heterodimer which is formed by ${\alpha}$- and ${\beta}$-subunits. Most of TSH activity is known to rely on ${\beta}$-subunit. TSH-${\beta}$ gene is located on chicken chromosome 26 and associated with growth performances. Therefore, this study aimed to investigate the association of TSH- ${\beta}$ SNP (G1031C) with economic traits (layday, layw, layno, bw150, bw270, layw270) in Korean Native Black chicken, Rhode Island Red and Cornish. Allele frequency of GG genotype in Rhode Island Red (RIR) was found to be 1.00 in this study. A significant effect was only observed on body weight at day 150 in Cornish. In Cornish, body weights of chicken with the CC genotype ($302.15{\pm}6.336$) were significantly higher than that of the GG genotype ($294.56{\pm}4.537$) (p<0.05). These findings suggest that the G1031C SNP of TSH-${\beta}$ gene can be used for improvement of growth-related traits in Cornish.

Lodging Related Traits and Yield of Two Rice Varieties as Affected by Paclobutrazol at Different N Levels and Split Rates (질소(窒素) 시비량(施肥量)과 분시비율(分施比率)이 다른 조건(條件)에서 Paclobutrazol 처리(處理)가 출수후(出穗後) 수도품종(水稻品種)의 도복(倒伏) 관련형질(關聯形質)과 수량(收量)에 미치는 영향(影響))

  • An experiment was carried out to know the effects of paclobutrazol on the lodging related traits and yield of a japonia (Seomjinbyeo) and an indica (Samgangbyeo) rice variety grown at 3 N levels (10, 20, 30 kg/10a) and 3 N split rates [basal + top dressing 15 days after transplanting (DAT) : top dressing 25 days before heading (DBH) was 100 : 0, 80 : 20, and 60 : 40]. Paclobutrazol (3kg/10a of 0.6% G) was applied 15 DBH and lodging related traits were observed 3, 13, 23, and 33 days after heading (DAH). The results obtained summarized as follows. 1. Culm length was increased as N level increased, but decreased by application of paclobutrazol. Culm length was not affected by N split rates in both varieties. 2. Lodging was not occurred at all plots, but lodging index of Seomingbyeo was much greater than that of Samgangbyeo. In both varieties loding index increased up to 23 DAH due to an increase in fresh weight and a decrease in breaking strength and levelled off thereafter due to a decrease in both fresh weight and breaking strength. 3. Culm length of Seomjinbyeo and fresh weight of shoot and culm length of Samgangbyeo were positively correlated with lodging index, but breaking strength was not correlated with lodging index in both varieties during the most of ripening stages. Direct effect of culm length contributing to lodging index was positive and much greater than that of fresh weight and breaking strength in Seomjinbyeo, but in Samganbyeo direct effect of culm length and fresh weight was positive, but that of breaking strength was negative with similar magnitudes. 4. Yield in brown rice of Seomjinbyeo was not affected by N level, but split application of N increased yield. In contrast, yield of Samgangbyeo was increased as N level increased, but yield was not affected by N split rates. Paclobutrazol did not affect yield of rice in Seomjinbyeo, but decreased yield by 5.2% in Samgangbyeo due to a decrease in the number of spikelets per panicle.

  • The study was conducted to find out the gene effects on body weights at weaning and at the age of 60 days in mice, with 343 progenies from full-dialell crosses of four lines of BALB/c, CBA, C3H and C57BL. The progenies were reared at the Experimental Animal Farm, College of Agriculture, Kyungpook National University from November, 1984 to February, 1985, and data collected from the progenies were analyzed into general combining ability, maternal effects, specific combining ability and reciprocal effects with Harvey's model. General combining ability effects estimated in line-crosses were -0.6033~0.5298 for weaning weights and -0.5086~1.0012 for body weights at the age of 60 days. General combining ability for BALB/c and C57BL were significantly better than general combining ability for CBA and C3H for both traits (P<0.05). Maternal effects for C3H were significantly larger than the maternal effects of BALB/c for both traits (P<0.05). The estimates of maternal effects were -0.9678~0.4609 for weaning weights and -1.1886~0.0729 for body weights at the age of 60 days. Specific combining ability effects were estimated to be significant (P<0.05), and the estimates were -0.1999~0.3380 for weaning weights and -0.4056~0.3317 for body weights at the age of 60 days. Reciprocal effects were found to be largest in BALB/c${\times}$C57BL and BALB/c${\times}$C3H. The estimates were -0.5049 from BALB/c${\times}$C57BL and 0.4972 from BALB/c${\times}$C3H form weaning weights, and -1.0336 from BALB/c${\times}$C57BL and 1.2793 from BALB/c${\times}$C3H for body weights at the age of 60 days.

The Effects of Various Factors on Milk Yield and Variation in Milk Yield Between Milking, Milk Components, Milking Duration, and Milking Flow Rate in Holstein Dairy Cattle (착유우의 연속유량, 유량변이, 유성분, 체세포수, 비유지속시간, 비유속도에 대한 산차, 착유시간, 유기 및 착유간격의 효과)

  • This study was carried out to estimate the effects of parity, milking time, milking interval and days in milk(DIM) on variation in milk yield between consecutive milkings(am to pm to am), morning and evening milk yield and its components, somatic cell counts(SCS), milking duration, milk flow rate and peak milk flow in Holstein dairy cattle. Records from one hundred and twenty two heads of Holstein cattle at National Livestock Research Institute, Korea were used for this study from July 1 to August 8, 2005. The experimental herd had average 1.6$\pm$0.9 parities, 199.8$\pm$109.1 DIM and 12.26$\pm$4.06kg milk yields at each milking. Milking yield, percent milk fat and SNF, milking duration and average milk flow were significantly varied by parity, milking time and DIM. Percent milk protein and lactose were varied by parity and DIM, however SCS and average milk flow were affected by parity and milking time. Milking interval significantly affected the consecutive, morning and evening milk yield and average milk flow. However, MUN was not affected by parity, milking time, DIM and milking interval. Milk yield was decreased with increasing parity. Milk yield in the morning was higher than that of in the evening. Milk yield between consecutive milking was not affected by parity, however, affected by milking time. Percent milk Fat, SNF and SCS were higher at in evening milk than those of in morning milk. Milk protein, lactose, SNF, SCS, milking duration and peak milk flow rate were influenced by parity. This study suggested that milk yield variation between consecutive milking, milking flow rate, and milking duration could be important traits for enhancing Holstein cattle productivity however, and more study is needed to estimate genetic parameters for such traits.

Performance Comparison of Local and Imported Dairy Semen Used in Rep. of Korea (젖소의 주요경제형질에 대한 국내생산 및 수입 정액의 능력비교)

  • The data used in the study were taken from the national dairy genetic evaluation of bulls from 1986 up to 2001. It was conducted to compare the phenotypic performance and genetic merits in terms of production traits, linear traits and selection indexes of four types of dairy semen (TypeⅠ:semen from Korean proven bull, Type Ⅱ:semen from proven bull imported by National Agricultural Cooperative Federation(NACF), Type Ⅲ:semen imported from USA, Type Ⅳ:semen imported from Canada) used in Korea. The result of national dairy genetic evaluation was used to compare genetic merits. Type Ⅲ was superior in the phenotypic performance of milk yield, milk fat and milk protein and was significantly different from Type Ⅰ. Types of semen were not significantly different in fat when semen from bull born after 1991 were compared. Likewise, types of semen were not significantly different in the genetic merit for milk yield, milk fat, milk protein and milk protein %. Moreso, for profit index (MFP) and Korean type production index (KTPI) it was not statistically different. However type Ⅰ was superior in milk fat % and was significantly different from other types. Type Ⅳ was superior in Final Score of conformation test (FS) and Udder Composite Index (UDC) and significantly different from Type Ⅰ. When semen from bull born after 1991 were compared, types of semen were not significantly different in milk yield and milk protein, although type I was superior in milk fat, milk fat %, milk protein % and MFP and was significantly different from others. Moreover type Ⅲ and type Ⅳ was superior in UDC and were significantly different from others.

Association Between the Polymorphism in the Promoter Region of Porcine A-FABP Gene and Growth Traits in Duroc Pigs (돼지 Duroc 품종에서 A-FABP promoter의 다형성과 성장형질의 연관성)

  • A polymorphism was found in the promoter region of porcine adipocyte fatty acid binding protein gene(A-FABP) gene which plays a key role in the binding and transportation of free fatty acid in adipocyte and deposition of intramuscular fat. Mutation was detected a substitution(T406C) using SSCP analysis and subsequently confirmed by sequencing the fragment in Duroc pigs. This T-406C mutation might change the binding activity for transcription factor nuclear factor 1(NF1). In this population, this mutation was genotyped using HinfⅠRFLP, and found three kinds of genotypes(TT, TC, and CC) showing their frequencies of 42.3, 44.3, and 13.4%, respectively. We statistically analyzed the association between the A-FABP genotypes and growth traits and found that the body weights of the pigs containing 406C/(TC or CC) were heavier for the body weight at the age of 20 weeks than those containing genotype TT(P<0.05), but not for those at the age of 0, 3, and 10 weeks. Pigs containing genotype CC had also a higher value for the average daily gain and lower values for the date for 90kg of body weight and food conversion ratio than those of 406T/- genotype. In addition, without the significant difference of back fat thickness, there was a significant association between the existence of allele CC and lean meat and eye muscle area(P<0.05). As a result of this study, we suggest that the allele T406C in the promoter region of A-FABP gene play an important role in deposition of intramuscular fat and weight in the later growth period. This polymorphism will be an useful molecular marker for breeding of Duroc pigs.

Association between Genotypes of the Isocitrate Dehydrogenase 3, beta subunit (IDH3B) Gene and Carcass Traits in an F2 Crossbred Population of Landrace × Jeju (Korea) Black Pigs (제주흑돼지와 랜드레이스 교배 2세대의 도체형질과 IDH3B 유전자형의 상관관계)

  • This study tested the association between genetic polymorphisms of the isocitrate dehydrogenase 3, beta subunit (IDH3B) gene and economic traits in an F2 crossbred population of Landrace × Jeju (South Korea) Black pigs. A 304-bp insertion/deletion mutation in promoter region was screened for determining genotypes of the IDH3B gene in a total of 1,105 F2 pigs. Three genotypes (AA, AB, and BB) were identified in the founder, F1, and F2 populations. Association analysis showed significant differences in carcass weights (CW), backfat thicknesses in three positions of the body (4th-5th ribs, BF5; 11th-12th ribs, BF12; 13th rib-1st lumbar, BFL), and carcass lengths (CL) (p<0.05), but not in meat color (MC), eye muscle area (EMA), or marbling scores (MARB) (p>0.05). The F2 IDH3B BB homozygotes showed heavier CW (80.790±0.725 kg) and shorter CL (101.875±0.336 cm) than the other genotypes (p<0.05). In addition, the BF levels between the 4th - 5th and 11th - 12th vertebrae were thicker in the carcasses of pigs with the IDH3B BB genotype than with the other genotypes (p<0.05). These results suggested that genetic variations in the IDH3B gene may serve as molecular genetic markers for improving the Landrace × Jeju Black pig crossbreeding systems.

Genetic Parameters for Milking Duration, Milk Flow and Milk Yield Per Milking in Holstein Dairy Cattle (홀스타인 착유우의 착유시간, 착유속도 및 착유량에 대한 유전모수 추정)

  • This study estimated characteristics and genetic parameters for milking traits in Holstein dairy cattle. Daily records (12,561) were available from a hundred fifty four lactating cows with sixty six sires at National Livestock Research Institute from November, 2005 to April, 2006. The cows were fed same rations, and were milked twice daily using milking parlour equipped with Alpro-system?? to collect milking records automatically. The experimental cows have averaged 1.9 parities, 139.7 days in milk, and 13kg milk yield per milking. Average milk flow was 64% of peak milk flow. Milking duration, peak milk flow, average milk flow, and milk yield per milking were 5.57±1.67 minutes, 3.58±0.79kg/minute, 2.28±0.51kg/minute, and 13.02±4.09kg, respectively. All traits in the morning milking were higher than those of in the evening milking, and were also increased with parity. Heritabilities for milking duration, peak and average milk flow, and milk yield per time were 0.49, 0.70 0.58 and 0.36, respectively. The genetic correlations of milking duration with peak milk flow, average milk flow, and milk yield per time were 0.48, 0.54, and 0.41, respectively. The correlations of milk yield per milking with peak milk flow and average milk flow were 0.23 and 0.30, respectively. In conclusion, milk flow was increased with milk yield; however milk flow was opposite relationship with milk duration. Further studies are not only necessary to warranty genetic parameters, but pre-installation of automatic recording system to collect daily milking record is also necessary.

Identification of a SNP in Chicken CaSR Gene and Its Effect on Economic Traits (닭의 CaSR 유전자내 단일 염기 변이 탐색 및 경제 형질간의 연관성 분석)

  • The Function of the calcium sensing receptor (CaSR) is to control calcium levels by altering PTH (parathyroid hormone) secretion and renal calcium resorption. The influence of calcium on the basal and stimulated release of several hormones from chicken pituitary glands has been determined in vitro. The objective of this study was to identify SNP in chicken CaSR gene and to investigate the effect of the SNP on economic traits. The sequencing analysis method was used to identify nucleotide polymorphisms within chicken CaSR gene. This study identified SNP at position 1949 bp(Genebank accession No : XM_416491) in the exon 1. The SNP changed the amino acid to alanine(GCC) from serine(TCC). This SNP showed three genotypes, AA, AS and SS by digestion with the restriction enzyme NcoⅠ using the PCR-RFLP method. The A963S showed significant effect only on the first lay day (P<0.05) in Leghorn population. Leghorn with the genotype AA had significantly faster the first lay day(137.6) than the genotype AS(143.0, P<0.05). Also, the A963S showed significant effect only on the first lay day(P<0.05) and mean of egg weight(P<0.05) in KNC population. KNC with the genotypes AA ans AS had significantly faster the first lay day (151.0 and 152.6, respectively) than the genotype SS(159.4, P<0.05). And the genotypes SS had significantly heavier the mean of egg weight(50.4 kg, P<0.05) than the genotype AA ans AS (47.5 and 47.8 kg, respectively). According to result of this study, an a allele of the A963S was found to have a significant effect on the first lay day. It will be possible to use this SNP marker on selecting chicken to improve the first lay day.

Estimation of Breeding Values for Economic Traits in Korean Native Chicken (한국재래닭의 경제 형질에 대한 육종가 추정)

  • This study was carried out to evaluate the breeding values for economic traits in Korean native chicken on the basis of 11,583 birds of record along 7 generations collected from 1995 to 2001 at National Livestock Research Institute, Korea. The results obtained are summarized as follows : The breeding value estimates of the body weights at 150 and 270 days in Dark Brown, Light Brown, Gray Brown, Black and White strains were $-16.563\~0.474\;and\;-18.836\~21.700,\;-14.714\~9.272\;and\;-23.025\~11.297,\; -25.186\~12.953\;and\;-28.368\~18.156,\;-15.549\~6.329\;and\;-18.264\~4.963,\;-10.708\~20.284\; and\;-25.112\~10.132g$, respectively. The breeding value estimates of the ages at first egg in Dark Brown, Light Brown, Gray Brown, Black and White strains were $-1.236\~0.047,\;-1.509\~0.298,\;-1.362\~0.114,\;-1.436\~0.199\;and\;-1.385\~0.718$, respectively. The breeding value estimates of the egg weight at first egg and at 270 days were $-0.105\~0.075\;and\;-0.120\~0.398,\;-0.109\~0.042\;and\;-0.205\~0.173,\;-0.166\~0.084\;and\;-0.256\~0.091,\;-0.119\~0.066\;and\;-0.051\~0.132,\;-0.112\~0.027\;and\;-0.070\~0.101g$, respectively. The breeding value estimates of the egg Production to 270 days in Dark Brown, Light Brown, Gray Brown, Black and White strains were $0.298\~l.807,\;-0.049\~l.146,\;-0.396\~l.125,\;-1.206\~l.173\;and\;-0.474\~2.641$, respectively.
