Antinociceptive and anti-inflammatory activities of Pandanus fascicularis Lamk. leaves in animal models

  • The present study was carried out to elucidate the potential of, chloroform extract of Pandanus (P.) fascicularis Lamk (Family-Pandanaceae) leaves on antinociceptive, behavioral study and anti-inflammatory effects using various animal models The dried, powdered leaves of, P. fascicularis were extracted successively with petroleum ether ($60\;-\;80^{\circ}C$) and chloroform in soxhlet apparatus. The chloroform extract (yield 21.6% w/w with respected to dry powdered plant material) was selected for all experimental procedure. Two models were employed to investigate the effects on nociception, the tail immersion and hot plate method in Swiss albino mice and anti inflammatory effect were investigated by employing the carrageenan induced rat paw edema test in. adult Wister albino rats. Behavioral study was investigated by elevated plus maze method in Swiss albino mice. Results were revealed that the PFCE was found significant antinociceptive effect (P < 0.001) at the dose levels of 100, 200 and 400 mg/kg, orally in mice and produced remarkable antiinflammatory effect (P < 0.001) at the same dose levels used in the rats. Behavioral study of the PFCE has no significant anxiolysis effect when used orally. It concludes that, PFCE possessed remarkable antinociceptive effect and anti-inflammatory effect but no anxiolytic effect on animal models.

Improving the Quality of Response Surface Analysis of an Experiment for Coffee-supplemented Milk Beverage: II. Heterogeneous Third-order Models and Multi-response Optimization

  • This research was motivated by our encounter with the situation where an optimization was done based on statistically non-significant models having poor fits. Such a situation took place in a research to optimize manufacturing conditions for improving storage stability of coffee-supplemented milk beverage by using response surface methodology, where two responses are $Y_1$=particle size and $Y_2$=zeta-potential, two factors are $F_1$=speed of primary homogenization (rpm) and $F_2$=concentration of emulsifier (%), and the optimization objective is to simultaneously minimize $Y_1$ and maximize $Y_2$. For response surface analysis, practically, the second-order polynomial model is almost solely used. But, there exists the cases in which the second-order model fails to provide a good fit, to which remedies are seldom known to researchers. Thus, as an alternative to a failed second-order model, we present the heterogeneous third-order model, which can be used when the experimental plan is a two-factor central composite design having -1, 0, and 1 as the coded levels of factors. And, for multi-response optimization, we suggest a modified desirability function technique. Using these two methods, we have obtained statistical models with improved fits and multi-response optimization results with the predictions better than those in the previous research. Our predicted optimum combination of conditions is ($F_1$, $F_2$)=(5,000, 0.295), which is different from the previous combination. This research is expected to help improve the quality of response surface analysis in experimental sciences including food science of animal resources.

Histopathological Comparison of Animal Models of Skin Inflammation and Inhibition of the Inflammatory Responses by Plant Flavonoid, Wogonin

  • Wogonin(5,7-dihydroxy-8-methoxyflavone), an anti-inflammatory plant flavonoid, was previously demonstrated to modulate the several parameters of animal skin inflammation. This compound inhibited edematic response as well as proinflammatory gene expression. In this investigation, the histopathological changes of the lesions from different types of experimental skin inflammation were compared and the potential therapeutic effect of topically applied wogonin was evaluated. From the results, it was found that multiple TPA treatment drastically increased ear edema accompanied with epidermal hyperplasia and inflammatory cell infiltration, while phenol treatment provoked only edematic response in the dermal area. Wogonin somewhat differently inhibited these animal models of skin inflammation.

  • 최근 비만이 전 세계적인 문제로 대두되면서 임상 연구의 해석에 필요한 중요 자료를 제시해 줄 수 있는 동물 모델을 이용한 생리학적 기전 연구에 대한 관심이 높아지고 있다. 인삼은 많은 동물 실험에서 항비만 또는 항당뇨 효과가 보고되었으나 인체의 임상에서 비만을 연구한 논문은 거의 없는 실정이다. 이 연구에서는 신체계측치수와 대사지표를 활용한 동물 모델에서의 인삼의 항비만 효과의 근거수준을 평가하고자 한다. 전임상 단계에서 비만에 대한 인삼의 효과를 연구한 대조군 연구, 비교 연구를 포함시키고자 한다. 실험적으로 비만을 유도하는 도중 혹은 이후에 인삼을 투여하고, 일차평가변수는 신체계측치수, 이차평가변수는 지방조직의 무게, 섭취음식의 총량, 대사지표 등을 포함한다. 언어, 출판일 등 특별한 제한 없이 MEDLINE, Embase, PubMed, Web of Science, Scopus를 통해 논문 검색을 시행한다. 본 연구에서의 자료 수집은 개인 정보를 포함하지 않으며, 사생활 침해의 우려가 없으므로 윤리적 승인 대상에서 제외된다. 연구의 전체과정을 수행한 후 연구결과는 연관 저널에 출간하거나 관련 학회에 발표할 예정이다. 본 연구 프로토콜은 the Collaborative Approach to Meta-Analysis and Review of Animal Data from Experimental Studies (CAMARADES) website (http://www.camarades.info)에 등록되었다.

알레르기성 비염 동물모델에서 한약제재 경구투여에 대한 국내 연구보고 고찰 (Oral administration of the Herbal Medicines in animal models for treatment of Allergic Rhinitis: a Review of Animal Study Reports Published in Korea)

  • Objective : This study is to review the effect of herbal medicines on treatment of Allergic Rhinitis in animal models reported in Korean domestic journals.Methods : The Databases(NDSL, Koreantk, OASIS, KISS) are searched with terms as Allergic Rhinitis. among them, only animal study reports were reviewed. Animal model, intervention, and observation methods of measuring indicators were extracted.Results : 27 articles were reviewed. All studies used OVA to induce Allergic Rhinitis. 7 studies used simple herbal medicines, 20 studies used complex herbal medicines, To evaluate the effect of the Herbal medicines, 23 studies used immunological examination, 18 studies used histological examination, 16 studies used hematologic examination, 3 studies used macroscopy The evidences of 5 studies is insufficient to show that Herbal medicines are effective for treatment of Allergic Rhinitis.

Optimization of the Growth Rate of Probiotics in Fermented Milk Using Genetic Algorithms and Sequential Quadratic Programming Techniques

  • Prebiotics (peptides, N-acetyglucoamine, fructo-oligosaccharides, isomalto-oligosaccharides and galactooligosaccharides) were added to skim milk in order to improve the growth rate of contained Lactobacillus acidophilus, Lactobacillus casei, Bifidobacterium longum and Bifidobacterium bifidum. The purpose of this research was to study the potential synergy between probiotics and prebiotics when present in milk, and to apply modern optimization techniques to obtain optimal design and performance for the growth rate of the probiotics using a response surface-modeling technique. To carry out response surface modeling, the regression method was performed on experimental results to build mathematical models. The models were then formulated as an objective function in an optimization problem that was consequently optimized using a genetic algorithm and sequential quadratic programming approach to obtain the maximum growth rate of the probiotics. The results showed that the quadratic models appeared to have the most accurate response surface fit. Both SQP and GA were able to identify the optimal combination of prebiotics to stimulate the growth of probiotics in milk. Comparing both methods, SQP appeared to be more efficient than GA at such a task.

실험적 자가면역성 말초신경염에서의 조직병리적 병변 및 CD5+ B-림프구의 발현 (Histopathologic Features and CD5+ B-lymphocyte Expression in the Experimental Allergic Neuritis)

  • Background : The pathogenesis of acute inflammatory demyelinating polyradiculoneuropathy (AIDP), Guillain Barre syndrome (GBS) is not clear, but it has been known that the immune mechanisms play an important role. Authors performed this study to establish an animal model of experimental allergic neuritis (EAN) by immunizing the myelin components of peripheral nerves and to understand the electrophysiological and histopathological features as well as the ${CD_5}^+$ B-lymphocyte changes in peripheral bloods in the EAN models. Methods : Lewis rats weighing 150-200 gm were injected subcutaneously in soles two times with total myelin, P0, P1, or P2 proteins purified from the bovine cauda eguina. The EAN induction was assessed by evaluating clinical manifestations. The electrophysiological and histopathological features were studied as routine methods. The ${CD_5}^+$ Blymphocytes were double stained using monoclonal FITC conjugated anti-rat CD45RA and R-PE conjugated anti-rat ${CD_5}^+$ antibodies and calculated using a fluorescence activated cell sorter (FACS). Results : The EAN animal models were established. In two out of five, in one out of two, in none out of three, and in none out of one Lewis rats injected with purified total myelin, P0, P1, P2 proteins respectively, They showed slow spontaneous motor activity and weak resistance against pulling back by tails. The typical electrophysiological and histologic findings in total protein and P0 induced EAN animal models were the decreased conduction velocity, the decreased compound muscle action potential (CMAP) amplitude and the dispersion phenomenon. The perivascular infiltrates of lymphocytes with focal demyelinating process were found in light microscopy. The ${CD_5}^+$ B-lymphocyte expression in three EANs were 2.38%, 3.50% 2.50%, which were not significantly increased, compared with those in normal controls. Conclusion : The EAN animal models were successfully established by injecting the total myelin and P0 myelin and they showed electrophysiological and histological features typical of demyelinating process. However they did not show an increased expression of ${CD_5}^+$ B-lymphocyte in peripheral bloods which could be indirect evidence of humoral autoimmunity.

Use of Germ-Free Animal Models in Microbiota-Related Research

  • The large intestine is a home for trillions of microbiota, which confer many benefits on the host, including production of vitamins, absorption of nutrients, pathogen displacement, and development of the immune system. For several decades, germ-free animals have been used to study the interaction between the host and its microbiota. This minireview describes the technical aspects for establishing and maintaining germ-free animals and highlights the advantages and disadvantages for germ-free animals as experimental models.

Use of Transgenic and Mutant Animal Models in the Study of Heterocyclic Amine-induced Mutagenesis and Carcinogenesis

  • Heterocyclic amines (HCAs) are potent mutagens generated during the cooking of meat and fish, and several of these compounds produce tumors in conventional experimental animals. During the past 5 years or so, HCAs have been tested in a number of novel in vivo murine models, including the following: lacZ, lacI, cII, c-myc/lacZ, rpsL, and $gpt{\Delta}$ transgenics, $XPA^{-/-}$, $XPC^{-/-}$, $Msh2^{+/-}$, $Msh2^{-/-}$ and $p53^{+/-}$ knock-outs, Apc mutant mice ($Apc^{{\Delta}716}$, $Apc^{1638N}$, $Apc^{min}$), and $A33^{{\Delta}N{\beta}-cat}$ knock-in mice. Several of these models have provided insights into the mutation spectra induced in vivo by HCAs in target and non-target organs for tumorigenesis, as well as demonstrating enhanced susceptibility to HCA-induced tumors and preneoplastic lesions. This review describes several of the more recent reports in which novel animal models were used to examine HCA-induced mutagenesis and carcinogenesis in vivo, including a number of studies which assessed the inhibitory activities of chemopreventive agents such as 1,2-dithiole-3-thione, conjugated linoleic acids, tea, curcumin, chlorophyllin-chitosan, and sulindac.

  • Small animal models are extensively utilized in the study of biomedical sciences. Current animal experiments and analysis are largely restricted to in vitro measurements and need to sacrifice animals to perform tissue or molecular analysis. This prevents researchers from observing in vivo the natural evolution of the process under study. Imaging techniques can provide repeatedly in vivo anatomic and molecular information noninvasively. Small animal imaging systems have been developed to assess biological process in experimental animals and increasingly employed in the field of molecular imaging studies. This review outlines the current developments in nuclear medicine imaging instrumentations including fused multi-modality imaging systems for small animal imaging.
