This study, including two in vitro experiments and an in vivo experiment were conducted to evaluate effects of Passtein$^{(R)}$ on crude protein degradability, ruminal fermentation characteristics and nutrient digestibility. In in vitro experiment protein degradability was examined using borate-phosphate buffer and neutral detergent, and using protease from Stroptomyces griseus at 39$^{\circ}C$ for 0, 2, 4, 8, 12, and 48 h. In addition, an in vivo experiment was conducted in a switch back design and ruminal fermentation and nutrient digestibility were determined. Four ruminal-fistulated Holstein cows weighing 300kg in mean body weight randomly allotted to 2 treatments (control and Passtein$^{(R)}$ supplementation). Although there was no significant difference on protein fraction between treatments, it appears that Passtein$^{(R)}$ supplementation decreased buffer soluble protein fraction compared to control. Protein degradability was not affected by Passtein$^{(R)}$ from 0 h to 4 h, but decreased at 12 h and 48 h compared to control. Degradation of immediately degradable fraction was higher in Passtein$^{(R)}$ treatment, but degradation of fermentable fraction was lower in Passtein$^{(R)}$ treatment compared to control. The pH and $NH_3$-N concentration tended to increase in Passtein$^{(R)}$ treatment, but VFA production, microbial counts and enzyme activity tended to decrease in Passtein$^{(R)}$ treatment compared to control. In addition, nutrient digestibility in the total tract tended to increase in Passtein$^{(R)}$ treatment compared to control.
We conducted two experiments to evaluate the effects of chromium picolinate(CrP) on growth performance, carcass characteristics and plasma components in Holstein bulls. In trial Ⅰ, eight finishing Holstein bulls(300${\pm}$6.99Kg) were allocated to 2 treatments(control and 0.05% CrP) with 4 replication for 10-months. In results, growth performance was not affected by CrP addition. The plasma insulin concentration in 0.05% CrP group was about 2 times higher than the control group of Holstein bulls. The levels of plasma NEFA were significantly decreased to 59.00 mEq/dl with 0.05% CrP treatment(P<0.05), but the levels of plasma glucose and PUN were not altered by 0.05% CrP treatment. The grade of carcass was not different between control and 0.05% CrP group, but back fat thickness in 0.05% CrP group was increased in 22.33% compared with control group. In trial 2, fifteen growing- finishing Holstein bulls(160${\pm}$4.63Kg) were allocated to 3 treatments(control, 0.025% CrP and 0.05% CrP) with 5 replication for 14-months. During the overall experimental period, growth performance was not affected by CrP levels. The levels of hormone and metabolites were not affected by CrP supplementa- tion. The carcass characteristics were not different between control and treatment. These results show that the CrP may have no effects for beef cattle production because of degradation of CrP conjugation in the rumen. However treatment of short term provide a possibility the effects of development for lipogenesis.
This study was conducted to evaluate the feeding value of ascidian tunic shell the effects of its dietary supplementation on laying performance, egg-yolk pigmentation, egg-shell strength and egg taurine content. A total of 168 brown layers at the age of 29wks in commercial cage were fed for 4 wks with 7 different diets containing ascidian tunic shel1(AST) at varying levels of 0$\sim$5% Dm or 0% AST with 100ppm carophyll red. No differences were found in egg production and weight among the treatments indicating that ascidian tunic shell did not adversely affect the laying performances. Adding the ascidian tunic shell to the diets increased egg-yolk pigmentation compared to the control and resulted in simillar or better effect on egg-yolk pigmentation compared to 100ppm carophyll red. The data suggest that ascidian tunic shell may be used as feed ingredients in layer diet enrichment of egg-yolk pigmentation in the place of carophyll red(chemical pigment). Specific gravity and breaking strength of egg shell were significantly increased by the adding ascidian tunic shell to the diet, suggesting that ascidian tunic shell may be used as feed ingredients for increasing egg shell strength. Also taurine content of egg was significantly increased with increasing supplementation of ascidian tunic shell to the diet(p<0.05). Therefore, ascidian tunic shell may be used as feed ingredients in laying hen diet to improve egg quality such as egg-yolk pigmentation, egg-shell strength and egg taurine enrichment.
The objective of this study was to evaluate the effects of dietary cation-anion difference (DCAD) with or without ruminally protected fat and niacin on the thermoregulatory ability, milk yield and milk composition of lactating dairy cows during summer in Korea. Thirty mid-lactating Holstein cows (134±12.4 DIM and 23.4±2.3kg/d of milk yield) were divided into three groups (10 animals/group). Cows were housed in a free-stall barn and were provided with forced- air ventilation (wind velocity = 4 m/s) using 41 cm diameter fans. Diet one was formulated to contain low DCAD (+15 DCAD) while the remaining two diets were higher in DCAD (+30 DCAD). One higher DCAD diet was formulated to contain by-pass fat and the second higher DCAD diet contained the niacin along with by-pass fat. The maximum ambient temperature during July was 28.5℃ which could be seen as a period of mild heat stress. As summer progressed, August was characterized as a severe heat stress condition with maximum ambient temperature (32.4℃) and THI (74.0). Dry matter, crude protein and total digestible nutrients intake was not affected by the DCAD level and supplementation of ruminally protected fat or niacin. Milk production was higher in cows fed diets supplemented with fat and niacin than those fed un-supplemented diet. No difference in milk yield was observed in cows fed diets supplemented with fat or niacin plus fat. Milk fat and rectal temperature were not affected by the DCAD level and supplementation of ruminally protected fat or niacin. However, respiration rate was decreased in cows fed diets supplemented with either fat or fat and niacin compared to those fed. The results of the present study indicated that higher DCAD (+30) and supplementation of fat along with niacin can somehow mitigate the negative effects of heat stress on milk yield and physiology of lactating Holsteins during July and August in Korea. In present study reduced respiration rate and increased milk yield in lactating cows may be attributed to the cooling effect of supplemented fat along with vasodilatory functions of niacin. Arthrospira platensis is the oldest marine microalgae on the planet, is said to contain most of the nutrients needed by the human body. It's components are reported to contain a large amount of various substances such as phycocyanin, chlorophyll and β-carotene, and are known to have an aging and whitening effect. In this study, UVB-induced reactive oxygen species reduction efficacy and antioxidant activity of spirulina purified water extract were investigated. effect was confirmed by measuring DPPH radical scavenging activity, FRAP reducing power and ABTS+ radical scavenging activity of 0.05, 0.10, 0.50 1.0 mg/mL of spirulina purified water extract. The coagulation rate, hatching rate and heart rate toxicity were measured by treating spirulina purified water extract with 0.05, 0.10, 0.50 mg/mL concentration using Zebrafish, an alternative experimental animal model. UVB-induced ROS measurement was treated with spirulina extract at 0.05, 0.10, 0.50 mg/mL concentration, and then stained with DCFH-DA to confirm the inhibitory effect of ROS. As a result of measuring antioxidant effect, DPPH, FRAP and ABTS+ showed concentration-dependent antioxidant effects in comparison with ascorbic acid. and measuring the coagulation rate, hatching rate, and heart rate using Zebrafish, an alternative experimental animal, it was confirmed that there was no toxicity in 0.05 and 0.10 mg/mL except 0.5 mg/mL compared to the control group. The ROS scavenging activity of UVB-induced zebrafish showed higher ROS reduction than the positive control. The results of this study suggest that spirulina and purified water extracts are valuable for UV and skin protection cosmetics.
Industrial wastes from pulp and food plants were treated with microorganisms to clarify organic waste-water and to produce cells as animal feed, and results were summarized as follows. (1) Waste-water from pulp, beer, bread yeast, and ethanol distillation plants contained $1.4{\sim}1.5%$ of total sugar, $0.25{\sim}0.35%$ nitrogen, and biological oxygen demand (BOD) was $400{\sim}25,000$, chemical oxygen demand (COD), $500{\sim}28,000$, and pH, $3.8{\sim}7.0$. The BOD and COD were highest in waste-water from ethanol distillation plants among others. (2) Bacterial and yeast counts were $4{\times}10^4-1{\times}10^9,\;2{\times}10^2-7{\times}10^4/ml$ in waste-water. (3) Bacteria grew better in pulp waste and yeasts in beer, bread yeast, and ethanol distillation waste. (4) Saccharomyces cerevisiae SAFM 1008 and Candida curvata SAFM 70 were the most suitable microorganisms for clarification of ethanol distillation waste. (5) When liquid and solid waste from ethanol distillation were treated with microbial cellulase, xylanase, and pectinase, solid waste was reduced by 36%, soluble waste was increased, and recuding sugar content was increased by 1.3 times which provided better medium than untreated waste for cultivation of yeasts. (6) Optimum growth conditions of the two species of yeast in ethanol distillation waste were pH 5.0, $30^{\circ}C$, and addition of 0.2% of urea, 0.1% of $KH_2PO_4$ and 0.02% of $MgSO_4$. (7) Minimum number of yeast for proper propagation was $1.8{\times}10^5/ml$. (8) C. curvata70 was better than cerevisae for the production of yeast cells from ethanol distillation waste treated with microbial enzymes. (9) S. cerevisiae produced 16 g of dried cell per 1,000ml of ethanol distillation waste and reduced BOD by 46%. C. curvata produced 17.6g of dried cell and reduced BOD by 52% at the same condition. (10) Yeast cells produced from the ethanol distillation waste contained 46-52% protein indicating suitability as a protein source for animal feed.
A computer simulation was carried out to investigate the optimum capacity of liquid manure tank spreader which is used as a tractor attachment. Soil physical properties, such as soil moisture content, bulk density, soil hardness and soil types were measured in the 10 major rice production area for computer simulation. Mathematical model which include soil physical properties and vehicle factor was used for computer simulation. Most of the soil type of the investigated area was sandy clay loam. Soil moisture content ranged between 30 and 40% mostly. Soil bulk density was in the range of 1,500 to 1,700 kg/$m^3$. Soil hardness ranged between 1 to 18 $cm^2$. Soil hardness incorporate the effects of many soil physical properties such as soil moisture content, soil type and soil bulk density, and so the range of soil hardness is greater than any other physical properties. The capacity of liquid manure tank spreader was above 3,000 kg$_{f}$ for the most of the investigated areas, and mostly in the range of 4,000 to 6,000 $kg_f$ depending upon the slip. But for the soft soil area such as Andong and Asan, the tractor itself has mobility problem and shows no pulling force for some places. For this area, the capacity of liquid manure tank spreader ranged between 1,000 and 2,000 $kg_f$ mostly, so the capacity of liquid manure tank spreader should be designed as a small capacity trailer compared to the other area.mpared to the other area.
In this study, current status of domestic deer farms and its feeding were surveyed. The information of supply and demand of feed f3r deers including its industry were also examined and then analyzed to make fundamental data for deer farms and government policy. The results are as follows. 1. Over 40 years old farmers were about 63% of total deer farms while 2030 years olds were less than 1% indicating that young people still evade agriculture. Moreover, considering education, over 52% of the farmers have bachelor degree showing much higher rates compared to the other agricultural fields. The reason can be assumed that the labor burden is not serious in deer farming while it is not dirty job compared to the other livestock management. Those higheducated people can be easily trained as experts of deer farming to improve its international competition. 2. Most of investigated farms raise Korean spotted deers and Elk showing that the percentage of Elk has greatly increased(However, many farmers have complained about purchasing methods and they insisted that the sales organization should be controlled by government). 3. 57% of total cost of production is for feed while most of feeds are imported from abroad. It indicated that it is urgent to make counterplan for saving feed cost. 4. It is necessary to develop feeds for deers in the near future while most of the examined farmers currently use normal assorted feed. Typical roughage sources feeds are rice straw, alfalfa hay, browses feed, and so on. Most of them are currently imported except the rice straw indicating urgently needed to develop domestic bulky feed. 5. The present questions are development of processed goods of velvet antler, establishment of reasonable management system, difficulty of velvet antler selling, feed supply, and so on. It is necessary for government and academic world to develop reasonable policy and scientific research program.
An energy metabolism study was conducted with two commercial strains of meat-type chickens, Hubbard and Cobb, and two egg strains, Hy Line and Korean-bred Hanhyup 325. The heat production of growing chickens from each strain were measured by the use of an open-circuit gravimetric respiration calorimeter. The data obtained from this study were summarized as fallows. 1. The average body weight of 9-wk-old Hubbard broilers reared in battery cages was 2,570g/bird. The average body weights of 9-wk-old Hy Line chicks and Hanhyup 325 were 777 and 748g/bird, respectively. 2. At 3 weeks of age, the Hubbard broiler chicks consumed two times the feed consumed by Hy Line chicks (54.6g VS. 26.7g/bird/day). These values increased to 151g and 57.2g/bird/day, respectively, at 8 weeks of age, indicating that the difference in feed intake between meat and egg-type chicks tends to increase as they grow older. In terms of water consumption, the 5-wk-old Hubbard broiler chicks drank $226m{\ell}/bird/day$ as compared with $58m{\ell}$ by Hy Line chicks. These values increased to 282 and $70m{\ell}$, respectively, at 8 weeks of age. 3. The excreta outputs of Hubbard broilers and Hy Line chicks were 18.7 and 6.1g DM/bird/day at 4 weeks of age, and 41.5 and 10.0g DM/bird/day at 8 weeks of age, respectively. 4. The energy metabolizability of broiler chicks were 75.4~77.1% compared to 75.0~83.5% by egg-type chicks. 5. The respiratory quotient (RQ) was between 0.78 and 0.97. There seems to be no difference in RQ between meat and egg-type chicks. The RQ tended to decrease when feed intake was low and vice versa. 6. Both meat and egg-type chicks produced $83.1{\sim}123.1Kcal/kg^{\frac{3}{4}}B.W./day$. The considerably low value of $83Kcal/kg^{\frac{3}{4}}B.W./day$ was obtained when the chicks were off the feed under the stressful conditions. The high value of 123.1Kcal was obtained when the chicken chamber temperature rose to $27{\sim}34^{\circ}C$.

Quail is a very useful animal model for studying vertebrate development because of its small body size and unique reproductive traits. This species is also ideal model for producing germline chimeras via transferring exogenous primordial germ cells (PGCs) into the recipient embryo. To increase the contribution efficiency of donor PGCs into recipients' tissues, decreasing the population of endogenous PGCs has been rate-limiting factor. We therefore conducted this study to investigate if gamma ($\gamma$)-irradiation depletes endogenous PGCs in developing quail embryo. Firstly, freshly laid stage X quail embryos were irradiated with various output of $\gamma$-irradiation and its teratogenic effect on the embryo was evaluated. Although a dose-dependent increase in the number of embryo showing malformation was found as the output increased (0, 250, 500, 750, and 1,000 rads), only a maximum of 10.1% of embryos were abnormal in 1,000 rads. Immunocytochemical analysis using the QCR1 antibody, which is specific marker for quail PGCs, was conducted to analyze the effect of sterilization. As results, $\gamma$-rays at a dose-rate of 500 rads/73 sec onto undeveloped stage X embryo significantly reduced the number of germ cells to an average of 75.55 % and 82.03 % in male and female embryos, respectively. We conclude that $\gamma$-ray selectively targets PGCs while affects minimally to the somatic development in quail embryo. Our results will not only provide important data for germline chimera production but can be used for analyzing the effect of ionized rays on the differentiating germ cells in various stages during animal development.
