Development and Validation of an Analytical Method for β-Agonists in Livestock and Fishery Products Using LC-MS/MS (LC-MS/MS를 이용한 축·수산물 중 β-agonist계 시험법 개발 및 검증)

  • BACKGROUND: The β-agonists known as phenyl ethanolamine derivatives have a conjugated aromatic ring with amino group. They are used as tocolytic agents and bronchodilator to human and animal generally, and some of them are used as growth promoters to livestock. METHODS AND RESULTS: β-agonists in samples were extracted by 0.4 N perchloric acid and ethyl acetate. The target compounds were analyzed by liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS). Validation of method was performed according to CODEX guidelines (CAC/GL-71). The matrix matched calibration gave correlation coefficients>0.98, and the obtained recoveries were in the range of 62.0-109.8%, with relative standard deviation ≤ 20.1%. In addition, a survey was performed to inspect any residual β-agonist from 100 samples of livestock and fishery products and ractopamine was detected in one of the 100 samples. CONCLUSION(S): In this study, we established the analytical method for β-agonists through using the expanded target compounds and samples. And we anticipate that the established method would be used for analysis to determine veterinary drug residues in livestock and fishery products.

Current Status and Perspectives of Quality Improvement in Sesame (참깨 품질 연구의 현황과 문제점 및 전망)

  • Sesame(Sesamum indicum L.) is probably the most ancient oilseed crop known in the world. The seed of sesame is used in a variety of ways as food. The whole seed may be eaten raw, either roasted or parched, or fed to birds and stock. Sesame oil is used as a salad or cooking oil, in shortening, margarine and in the manufacture of soap. Minor uses are as a fixative in the perfume industry and formerly as a carrier for fatsoluble substances in pharmaceuticals such as penicillin. One of the minor constituents of sesame oil, sesamin, is used for its synergistic effect in pyrethrin insecticides, in addition of a small quantity of this substance markedly increases the effectiveness of fly sprays. The meal remaining after oil extraction can be used as and animal feed-stuff or as manure. In general sesame meal is considered to be equal to cottonseed or soybean meal as a protein supplement for livestock and poultry. It is especially high in certain amino-acids such as methionine, which is low in soybean meal, and thus can be combined with it or similar meal to form a more balanced ration. An attempt to summarize the literature review on quality improvement of sesame was made to discuss the accomplishments of the past and perspectives in the future. The reviews on quality improvement of sesame were mainly discussed in connection with the cultural practices and genetic informations in current status. The emphasis focussed on environmental variation of quality in cultural practices, such as harvest time, variety by location, climatic condition, fertilizer application, and growth regulator treatment. On the genetic variation of quality, it was discussed on variety background, mutation breeding, correlations, and inheritance of quality related characteristics. It also was discussed on relationship between quality and plant traits, storage condition or period, and seed coat color. Moreover, current research status were reviewed on some minor elements such as sesamin, oxalic acid, and trypsin inhibitor. As a results of the review, the lack of an effort to quality improvement in each utilization area was indicated as a problem area. More active efforts for the improvement of quality were also insufficient to incorporate the available genes for quality in breeding method or collection and analysis of breeding materials. Therefore, researches in the future would be recommended to emphasize on these problem areas.

Convergence study on the quality evaluation of ginseng sprout produced smart farm according to organic acid treatment and packing containers during storage (스마트팜 생산 새싹인삼의 유기산 처리 및 포장 용기에 따른 품질 평가에 대한 융합연구)

  • In this study, the physical quality and microbial changes of ginseng sprout according to the pretreatment process and packaging container were evaluated to improve the storage properties of ginseng sprout produced in smart farm. Quality change during storage (10 days) according to pretreatment method (ascorbic acid, citric acid, peroxyacetic acid) and packaging container (expanded polystyrene (EP), polypropylene (PP), polyethylene (PE), polypropylene + polyethylene + cast polypropylene (PP+PE+CPP)) was evaluated in terms of texture, viable cell count, water content, and color. As a result of comparison according to the type of pretreatment, the citric acid treatment group showed the lowest texture change and the effect on inhibition of bacterial growth. On the other hand, citric acid, which was most effective among pretreatments, was treated in all samples and then stored in 4 types of containers. Specially, the ginseng sprout in PP packaging container was not observed significant softening or color changes after 10 days storage, and the lowest changes in viable cell number. Therefore, this study was shown that citric acid treatment and use of PP packaging container are effective in increasing the shelf life of ginseng sprout.

Liver Protective Effect of the Co-treatment of Rhei Radix et Rhizoma and Silymarin on TAA-induced Liver Injury (대황과 실리마린의 병용투여의 간섬유화 보호 효과)

  • Objective: Liver fibrosis is a highly conserved wound-healing response and the final common pathway of chronic inflammatory injury. This study aimed to evaluate the potential anti-fibrotic effect of the combination of Rhei Radix et Rhizoma water extract (RW) and silymarin in a thioacetamide (TAA)-induced liver fibrosis model. Methods: The liver fibrosis mouse model was established through the intraperitoneal injection of TAA (1 week 100 mg/kg, 2-3 weeks 200 mg/kg, 4-8 weeks 400 mg/kg) three times per week for eight weeks. Animal experiments were conducted in five groups; Normal, Control (TAA-induced liver fibrosis mice), Sily (silymarin 50 mg/kg), RSL (RW 50 mg/kg+silymarin 50 mg/kg), and RSH (RW 100 mg/kg+silymarin 50 mg/kg). Biochemical analyses were measured in serum, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), malondialdehyde (MDA), and ammonia levels. Liver inflammatory cytokines and fibrous biomarkers were measured by Western blot analysis, and liver histopathology was evaluated through tissue staining. Results: A significant decrease in the liver function markers AST and ALT and a reduction in ammonia and total bilirubin were observed in the group treated with RSL and RSH. Measurement of reactive oxygen species and MDA revealed a significant decrease in the RSL and RSH administration group compared to the TAA induction group. The expression of extracellular matrix-related proteins, such as transforming growth factor β1, α-smooth muscle actin, and collagen type I alpha 1, was likewise significantly decreased. All drug-administered groups had increased matrix metalloproteinase-9 but a decreasing tissue inhibitor of matrix metalloproteinase-1. RSL and RSH exerted a significant upregulation of NADPH oxidase 2, p22phox, and p47phox, which are oxidative stress-related factors. Furthermore, pro-inflammatory proteins such as cyclooxygenase 2 and interleukin-1β were markedly suppressed through the inhibition of nuclear factor kappa B activation. Conclusions: The administration of RW and silymarin suppressed the NADPH oxidase factor protein level and showed a tendency to reduce inflammation-related enzymes. These results suggest that the combined administration of RW and silymarin improves acute liver injury induced by TAA.

The Status of Maize Production and Distribution in Cambodia (캄보디아 옥수수 생산 및 유통현황 연구)

  • In recent, the cultivation of maize in the world is being increased as a feed for the animal due to the increase of the large amount of meat consumption. However, reduction of supply for the feeding maize by the natural disaster such as drought, flooding, and demanding materials for bioenergy are attributed to increase the cereal price of the world that caused for the fluctuation of prices. The research has been done to provide useful information to the Korean agricultural investor regarding the current status of maize production and distribution in Cambodia. The maize cultivation of Cambodia in 2009 presented as a second major crop at the area of 6.8% in the total crop production area, and it continuously increased maize production yearly basis for the internal uses as well as for the exportation. The total amount of production showed by amount of 0.92 million tons due to the increased production of unit area. The yield production of unit area in current shows about 4 tones per hectare. About 90% of total production produced during August to September at wet season, and area of Batambang province produced about 70% of the total production. The production of feeding maize cultivated about 70% of the total cultivation area, and the exportation have done around 60% and remains were consumed as a domestic uses. The prospects of maize production in Cambodia will be increased due to the acceleration of livestock industry as 8.7% of annual growth rate.

Pig Image Learning for Improving Weight Measurement Accuracy

  • The live weight of livestock is important information for managing their health and housing conditions, and it can be used to determine the optimal amount of feed and the timing of shipment. In general, it takes a lot of human resources and time to weigh livestock using a scale, and it is not easy to measure each stage of growth, which prevents effective breeding methods such as feeding amount control from being applied. In this paper, we aims to improve the accuracy of weight measurement of piglets, weaned pigs, nursery pigs, and fattening pigs by collecting, analyzing, learning, and predicting video and image data in animal husbandry and pig farming. For this purpose, we trained using Pytorch, YOLO(you only look once) 5 model, and Scikit Learn library and found that the actual and prediction graphs showed a similar flow with a of RMSE(root mean square error) 0.4%. and MAPE(mean absolute percentage error) 0.2%. It can be utilized in the mammalian pig, weaning pig, nursery pig, and fattening pig sections. The accuracy is expected to be continuously improved based on variously trained image and video data and actual measured weight data. It is expected that efficient breeding management will be possible by predicting the production of pigs by part through video reading in the future.

The Use of the Internal Transcribed Spacer Region for Phylogenetic Analysis of the Microsporidian Parasite Enterocytozoon hepatopenaei Infecting Whiteleg Shrimp (Penaeus vannamei) and for the Development of a Nested PCR as Its Diagnostic Tool

  • The increasing economic losses associated with growth retardation caused by Enterocytozoon hepatopenaei (EHP), a microsporidian parasite infecting penaeid shrimp, require effective monitoring. The internal transcribed spacer (ITS)-1 region, the non-coding region of ribosomal clusters between 18S and 5.8S rRNA genes, is widely used in phylogenetic studies due to its high variability. In this study, the ITS-1 region sequence (~600-bp) of EHP was first identified, and primers for a polymerase chain reaction (PCR) assay targeting that sequence were designed. A newly developed nested-PCR method successfully detected the EHP in various shrimp (Penaeus vannamei and P. monodon) and related samples, including water and feces collected from Indonesia, Thailand, South Korea, India, and Malaysia. The primers did not cross-react with other hosts and pathogens, and this PCR assay is more sensitive than existing PCR detection methods targeting the small subunit ribosomal RNA (SSU rRNA) and spore wall protein (SWP) genes. Phylogenetic analysis based on the ITS-1 sequences indicated that the Indonesian strain was distinct (86.2% nucleotide sequence identity) from other strains collected from Thailand and South Korea, and also showed the internal diversity among Thailand (N = 7, divided into four branches) and South Korean (N = 5, divided into two branches) samples. The results revealed the ability of the ITS-1 region to determine the genetic diversity of EHP from different geographical origins.

Effects of the Feeding Methods on Body Growth and Sexual Maturity in Broiler Breed Male (육용종웅계(肉用種雄鷄)의 사양방법(飼養方法)이 체성장(體成長) 및 성성숙(性成熟)에 미치는 영향(影響))

  • This experiment was conducted to find out the effects of the six different feeding methods on the development of body weight, testis, comb and pituitary gland, and the sexual maturity of White Plymouth Rock cockerels. From hatching to 22 weeks of age, the weights of whole body, testis, comb and pituitary gland, and the histological changes of testis and the semen characteristics were checked every other week. The results obtained in this expeniment were as follows: 1. The growth rates of the self-feeding groups were faster than those of the limited feeding groups (70 percent of the self-feeding) by about 2 weeks. The weights of testis and comb showed the most marked increase at 20 weeks of age in the self-feeding groups and at 22 weeks of age in the limited feeding groups, respectively. 2. The weights of pituitary gland from hatching to 22 weeks of age at all observation weeks were not recognized significantly among the compared groups except 4, 14 and 16 weeks of age. 3. Correlations between week of age, body weight, testis, comb and pituitary gland, in the course of 22 weeks, were highly significant. 4. The diameters of lumina and tubules in the seminiferous tubules increased very slowly until 10 weeks of age. They showed the most marked increase at 12 weeks of age in the self-feeding groups and at 14 weeks of age in the limited feeding groups, and then continueously increased until 22 weeks of age. 5. Primary spermatocytes appeared at first at 8 weeks in the all treatment groups. Secondary spermatocytes appeared at first at 10 weeks in the self-feeding groups and at 12 weeks in the limited feeding groups. At 14 weeks of age spermatids and spermatozoa were found at first in the self-feeding groups but spermatids were found in the limited feeding groups. 6. Age of the first ejaculation was between 14 and 16 weeks of age in the all treatment groups. The average semen volume and sperm concentration ranged from $0.1{\sim}0.2m{\ell}/ejaculate$ and $5.6{\sim}9.8{\times}10^8sperm/m{\ell}$ at the age of the first ejaculation but $0.30{\sim}0.35m{\ell}/ejaculate$ and $22.4{\sim}42.7{\times}10^8sperm/m{\ell}$ at the 20 weeks of age in the all treatment groups.

Effect of ω3-Fatty Acid Desaturase Gene Expression on Invasion and Tumorigenicity in Human Tongue Squamous Cell Carcinoma Cells (인체 혀의 편평세포암 세포에서 ω3-fatty acid desaturase 유전자 발현이 침윤 및 종양형성에 미치는 영향)

  • Omega-3 polyunsaturated fatty acids (${\omega}3$-fatty acid) have been found to possess anticancer properties in a variety of cancer cell lines and animal models, but their effects in human tongue squamous cell carcinomas (SCCs) remain unclear. This study was designed to examine the effect of ${\omega}3$-fatty acid desaturase (fat-1) gene expression on invasion and tumorigenicity in human tongue SCC cells and the molecular mechanism of its action. Docosahexaenoic acid (DHA) treatment inhibited in vitro invasion in a dose-dependent manner. In zymography, matrix metalloproteinase-9 (MMP-9) and Matrix metallopeptidase-2 (MMP-2) activities were reduced, and MMP-9 and MMP-2 promoter activities were inhibited by the DHA treatment. In addition, cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) promoter reporter activities were inhibited in SCC-4 and SCC-9 cells after the DHA treatment. To investigate the effect of a high level of endogenous ${\omega}3$ fatty acids, a stable SCC-9 cell line expressing the ${\omega}3$-desaturase gene (fSCC-9sc) was generated. The growth rate and colony-forming capacity of fSCC-9sc were remarkably decreased as compared with those of fSCC-9cc. Likewise, the tumor size and volume of fSCC-9sc implanted into nude mice were significantly inhibited, with increases in the cell death index. Furthermore, a transwell chamber invasion assay showed a reduction in cell invasion of the fSCC-9sc lines when compared with that of the fSCC-9cc line. These findings suggested that fat-1 gene expression inhibited tumorigenicity, as well as invasion in human tongue SCC cells. Thus, utilization of ${\omega}3$ fatty acids may represent a promising therapeutic approach for chemoprevention and the treatment of human tongue SCCs.

Effects of Supplemental Lighting on Growth and Yield of Sweet Pepper (Capsicum annuum L.) in Hydroponic Culture under Low Levels of Natural Light in Winter (동계시설내 보광이 수경재배 착색단고추(Capsicum annum L.)의 생육에 미치는 영향)

  • This study was conducted to examine the effect of supplemental lighting on the growth and yield of hydroponically grown sweet pepper (Capsicum annuum L. cv. sprit) under low levels of natural light in winter. The plants were treated with natural light only (control), 3-hour supplemental lighting before sunrise, after sunrise and after sunset with high pressure sodium (HPS, 400W). As the result of these three treatments, the supplemental lighting promoted photosynthesis in the low light intensity condition and particularly photosynthesis was more active right after sun rise in the morning, 1.5-$3.0{\mu}molCO_2{\cdot}m^{-2}{\cdot}s^{-1}$ comparing to those of supplemental lighting after sunset, 0.5-$1.5{\mu}molCO_2{\cdot}m^{-2}{\cdot}s^{-1}$. Transpiration rate and stomatal conductance sharply increased with supplemental lighting after sunrise then they decreased again after turning the lights off. Stomatal size was observed $32.2{\mu}m^2$ after supplemental lighting, whereas the size of the natural light was almost closed at $7.7{\mu}m^2$. The average plant height of sweet papper cv. spirit was 185 cm before sunrise, 188 cm after sunrise and 208 cm after sunset with supplemental lighting for 3hours while the control was 171 cm. With supplemental lighting a better number of fruit set per plant was measured 4.3 before and after sunrise, 3.7 after sunset but 2.6 in the control. Interestingly, there were no significant differences in the sugar content ($^{\circ}Brix$) degree between treatment of supplemental lighting, whereas slight differences between seasons were seen. The marketable fruit yield of sweet pepper (cv. spirit) was $116.0kg{\cdot}ha$ with supplemental lighting, whereas the control (natural light only) was $75.8kg{\cdot}ha$. Despite of spending electricity and depreciation cost, the economic analysis showed net income with supplemental lighting after sunrise was 51% higher than control treatment in cv. spirit.
