Antioxidant, Liver Protective and Angiotensin I-converting Enzyme Inhibitory Activities of Old Laying Hen Hydrolysate in Crab Meat Analogue

  • The purpose of this study was to evaluate the antioxidative activities of Crab meat analogue prepared with protein hydrolysates obtained from mechanically deboned chicken meat (MDCM) from spent laying hens. 2,2-diphenyl-1-picrylhydrazyl hydrate (DPPH) radical-scavenging activity was increased by adding MDCM hydrolysates during storage, and activity correlated with the concentration of DPPH added up to 6 weeks of storage. Hydroxyl radical-scavenging activity was increased in all analogues containing MDCM hydrolysates. At 0 days of storage, angiotensin I-converting enzyme (ACE)-inhibitory activity was increased by the addition of MDCM hydrolysates. Activity did not correlate after 6 weeks of storage, in which ACE-inhibitory activity was increased with low concentrations of MDCM hydrolysates, but no ACE-inhibitory activity was observed at higher concentrations. The liver-protecting activity of crab meat analogue was shown to be around 60% of the positive control; however, it was not significantly different among the samples during storage. These results support the use of MDCM as a source of health-promoting constituents in crab meat analogue.

각종 버섯류로부터 안지오텐신 전환효소 저해제의 탐색 (Screening of Mushrooms Having Angiotensin I-Converting Enzyme Inhibitor)

  • 항고혈압 효능이 우수하며 부작용이 없는 고혈압 예방 제품을 개발하고자 농업과학기술원에서 분양 받은 52종의 버섯을 대상으로 일반성분, 추출 수율, 그리고 ACE 저해활성을 조사하여 활성이 우수한 버섯을 선정한 다음 ACE 저해물질의 추출 최적조건을 검토하였다. 시료 버섯은 $7.1{\sim}56.5%$의 조단백질과 $0.2{\sim}4.4%$의 조지방 및 $30.3{\sim}86.6%$의 탄수화물을 각각 함유하고 있었으며 비늘버섯 ASI 24027 자실체의 물 추출물 수율이 68%로 제일 높았다. 그러나 ACE 저해활성은 비늘 버섯 ASI 24012 균주의 자실체를 물 추출로부터 얻은 추출물($IC_{50}$: 0.45 mg)에서 가장 높았다. 그리고 이 버섯의 ACE 저해물질 최적추출조건은 자실체 분말을 물로 $30^{\circ}C$에서 1시간 추출했을때 가장 많이 용출 되었으며 이때 ACE 저해활성도도 67.6%($IC_{50}$: 0.20 mg)로 가장 높았다.

열수 및 효소적 가수분해로 제조된 틸라피아 비늘 젤라틴 가수분해물의 ACE 저해 활성 (Angiotensin I Converting Enzyme Inhibitory Effects of Gelatin Hydrolysates Prepared from Tilapia mossambica Scales by Hot Water and Enzymatic Extraction)

  • Fish scales have potential in functional food preparation due to their antioxidant and antihypertensive properties. We investigated the angiotensin I converting enzyme (ACE) inhibitory activity of Tilapia mossambica scale extracts. Hydrolysates of tilapia scales were prepared by enzymatic extraction using five proteases (${\alpha}$-chymotrypsin, Alcalase, Kojizyme, Protamex and trypsin) after scales were treated with hot water for 3 hr. Scale enzymatic hydrolysates prepared using both hot water and enzyme treatments exhibited elevated hydrolysis (about 25%-55%) compared to only enzyme treatment (about 15%-45%). Enzymatic hydrolysates (1 mg/mL) prepared by both hot water and enzyme treatments also showed significantly increased ACE inhibitory activities from about 20%-75%. The pattern of ACE inhibitory activities was similar to the degree of hydrolysis. Alcalase and ${\alpha}$-chymotrypsin hydrolysates displayed the highest ACE inhibitory activities ($IC_{50}$ = 0.83 mg/mL and 0.68 mg/mL, respectively). In addition, the ACE inhibitory effects of $IC_{50}$-chymotrypsin hydrolysates increased with decreasing molecular weight (5 kDa>, 10 kDa> and 30 kDa>), with the 5 kDa> fraction displaying the highest ACE inhibitory activity (about 89.9% and $IC_{50}$ = 0.1 mg/mL). We suggest that the peptide compounds of enzymatic hydrolysates prepared from tilapia scale enhances ACE inhibitory activity and might be useful as an antihypertensive material.

  • 각종 미강 추출물들의 항고혈압성 엔지오텐신 전환효소 저해 활성을 측정하고 이들의 추출 최적조건을 검토하였다. 각종 미강 추출물들중에서 일품벼 미강의 물 추출물이 77%의 가장 높은 안지오텐신 전환효소 저해활성 보였다. 일품벼 미강중의 안지오텐신 전환효소 저해물질은 물을 1:10(w/v)으로 미강 분말에 첨가한 후 $30^{\circ}C$에서 12시간 추출하였을 때 가장 많이 용출되었다.

ACE-inhibitory Effect and Physicochemical Characteristics of Yogurt Beverage Fortified with Whey Protein Hydrolysates

  • This study investigated the ACE-inhibitory effect of yogurt beverage fortified with hydrolysates as well as the suitability of hydrolysates as a nutraceutical additive to yogurt beverage. Three whey protein hydrolysates hydrolyzed by alcalase, protamex, and trypsin were each added to yogurt beverage at concentrations of 1.25, 2.5, and 5 mg/mL. Yogurt beverage fortified with 2.5 mg/mL of hydrolysates had 61-69% ACE-inhibitory activity, whereas yogurt beverage fortified with 5 mg/mL of hydrolysates showed 74% ACE-inhibitory activity. There were no significant differences in ACE-inhibitory activity between the alcalase or protamex hydrolysates during storage; however, trypsin hydrolysate exhibited significant differences. On the other hand, physicochemical characteristics such as pH (3.47-3.77), titratable acidity (0.81-0.84%), colority, viable cell count, and sensory qualities were not significantly different among the tested yogurt beverage samples during storage. These results showed that yogurt beverage fortified with whey protein hydrolysates maintained antihypertensive activity and underwent no unfavorable changes in physicochemical characteristics regardless of enzyme type.

  • Angiotensin I converting enzyme (ACE) in renin-angiotensin system is a cause of essential hypertension, which covers most hypertension, one of the major adult diseases. Thus, the inhibition of ACE would be indispensable for the prevention and cure of hypertension. Therefore, a lot of studies on the ACE inhibitor have been conducted. Peptides from the protein hydrolysate have been reported as an remarkable inhibitor. Especially, various ACE inhibitory peptides were isolated and identified from marine products for their utilization as value added products. (omitted)

약용 식물 발효액(들빛)의 항산화, Angiotensin Converting Enzyme 저해 및 Nitric Oxide 생성 조절 효과 (Antioxidative activity and Angiotensin Converting Enzyme Inhibitory activity of Fermented Medical Plants (DeulBit) and Its Modulatory Effects of Nitric Oxide Production)

  • 본 연구에서는 결명자, 오미자, 구기자, 박하, 국화, 산마, 생강, 둥글레, 감초, 오가피, 녹차, 다시마를 발효시킨 들빛 발효액으로 여러 가지 생리활성에 대하여 분석하였다. 우선, 들빛 발효액의 플라보노이드 함량과 DPPH 활성을 측정하였다. 그 결과 들빛 발효액의 총 플라보이드 함량은 $3.4{\pm}0.5\;mg/g$으로, DPPH법을 통해 측정한 들빛 발효액의 radical 소거능은 96%로 나타났다. 들빛 발효액의 SOD 유사활성은 29%로 나타내었고, XOase에 의해 생성된 superoxide radical 소거능은 88%로 높게 나타났다. 들빛 발효액의 항고혈압 측정실험에서는 66%의 저해률을 나타내어 ACE 저해 활성이 뛰어난 것으로 나타났다. 혈전분해능에 대한 들빛 발효액의 분석 결과는 혈전용해제로 알려져 있는 plasmin과 유사한 활성을 보이고 있다. 들빛 발효액과 면역활성과의 연관성은 NO 생성 증가율과 LPS에 의해 유도되는 NO 생성 억제율 분석으로 조사되었다. 그 결과, 들빛 발효액은 무려 15배의 높은 NO 생성 증가율을 보였다. 또한 LPS에 의해 유도된 NO 생성 억제율은 들빛 발효액 100%에서 42%로 나타나 높은 면역조절능을 나타내었다. 이상의 결과는 들빛 발효액의 우수한 생리활성을 증명하고 있고, 항산화력, 항고혈압 효과, 혈전분해능 및 면역 조절활성이 높은 것으로 나타나 기능성 음료의 소재로서 그 활용도가 높을 것으로 판단된다.

  • The epoxyalkanoyl derivatives were designed and synthesized as ACE inhibitors. Coupling of unsaturated carboxylic acids with amino acids and following epoxidation with dimethyldioxirane gave the epoxyalkanoyls with high yield. The inhibitory activity of synthesized compounds on angiotensin converting enzyme was $IC_{50}$ values of 0.06~5.5 ${\mu}M$.

발아 벼 부위별 추출물의 항산화활성과 Angiotensin Converting Enzyme I 저해활성 (Antioxidant and Angiotensin Converting Enzyme I Inhibitory Activity on Different Parts of Germinated Rough Rice)

  • 한국산 벼의 발아 전과 후의 부위별 및 추출용매별 항산화성분 및 활성변화를 살펴보았다. 발아 전과 후의 벼를 왕겨, 현미 및 싹으로 분리하고 각각 열수 및 70% 에탄올을 이용하여 추출하였다. 총 폴리페놀 함량은 현미 물추출물에서 발아 전 1.67에서 발아 후 5.84 mg/g으로 증가하였으며, DPPH 라디칼소거능은 벼 에탄올추출물에서 발아 전 22.95 %에서 발아 후 31.32%로 증가하였지만 왕겨와 현미 부분에서는 발아 후 감소하였다. 총 항산화력은 에탄올추출물에서 발아 전과 후에 큰 차이를 보이지 않았으나 싹 부분에서는 4.41 mg AA eq/g으로 높게 나타났다. 환원력은 에탄올 추출물 현미에서 발아 전 0.32에서 발아 후 0.45로 증가하였다. ACE 저해활성은 왕겨에서 가장 높았으며 발아 후 증가하였다. 본 연구결과 발아 시 항산화활성이 증가하는 원인에 대한 추후 연구가 필요하다 판단된다.

Antioxidant and ACE Inhibitory Activity of Cultivated and Wild Angelica gigas Nakai Extracts Prepared Using Different Extraction Conditions

  • The purpose of this study was to investigate the biological activities of cultivated Angelica gigas Nakai (CAG) and wild Angelica gigas Nakai (WAG) extracts prepared by extraction with water, 30% ethanol, 60% ethanol, or 90% ethanol. The electron donating ability of the WAG extracts was higher than that of the CAG extracts and 0.1% and 1.0% solutions of the comparative substance, L-ascorbic acid. The superoxide dismutase-like activity of the CAG extracts was higher than that of WAG extracts. Superoxide dismutase-like activity was highest (33.95%) in the CAG water extract. The total polyphenol content was highest in the 60% ethanol extracts of WAG. The nitrite scavenging ability of the CAG and WAG extracts was highest at a pH of 1.2. The tyrosinase inhibitory effect was highest (43.72%) in the water extract of WAG. The angiotensin converting enzyme inhibitory activity was highest (83.84%) in the 60% ethanol extract of WAG. The results of the present study will be useful for understanding the antioxidant and angiotensin-converting enzyme inhibitory activities of Angelica gigas Nakai extracts.
