Cytotoxicity of Angelicae Radix from Korea, China, and Japan on HepG2 Cells (한국 ${\cdot}$ 중국 ${\cdot}$ 일본 당귀(當歸)의 HepG2 세포 독성 비교 연구)

  • The purpose of this study is to examine the cytotoxicity of species of Angelica (Angelicas Radix; the root of Angelica gigas Nakai, A. sinensis (Oliv.) Diels, and A. acutiloba Kitag.) on HepG2 cells. The water extracts of roots of Angelica gigas (WAG), A. sinensis (WAS), and A. acutiloba (WAA) were studied for HepG2 cell viability by a modified MTT assay in the concentrations of 5, 10, 50, 100, 250, 500 ug/ml for 24, 48, 72 h. WAG and WAS did not reduced the cell viability significantly. But WAA reduced the cell viability in the concentration of 500 ug/ml for 24 h (85.45%), 48 h (75.01%). In conclusion, WAG and WAS have not the significant cytotoxicity on HepG2 cells in the suitable dose.

Validations of Analysis Methods for Decursin and Decursinol Angelate of Angelicae gigantis Radix by Reversed-Phase Liquid Chromatography

  • A reversed-phase liquid chromatographic method for decursin and decursinol angelate of Angelicae gigantis Radix, an important crude drug in Korean traditional medicine, was developed and validated. Decursin and decursinol angelate, the structure isomer (pyranocoumarin) each other, are the main organic constituents in Angelicae gigantis Radix. This method was developed using a RP-18 column, UV detector at 280 nm and 50% acetonitrile solution containing 0.01 M sodium dodecyl sulfate and 25 mM sodium dihydrogen phosphate (pH 5.0) as the mobile phase. Various validation parameters were included and evaluated satisfactorily. Linearity was established in range 2-75 mg/ml of decursin and decursinol angelate (correlation coefficient = 0.9997 and 0.9995, respectively). This analytical method showed good accuracy (98.1% and 99.5%, respectively). Precision (repeatability) revealed a relative standard deviation value of 1.71% (decursin) and 3.19% (decursinol angelate). For intermediate precision measure the considered variables were equipment and days. A robustness test showing the influence of deferent counter-ion concentration in mobile phase was also performed.

  • Essential oil of the root of Angelica koreana Max. (Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain nine monoterpenes such as ${\alpha}-pinene$(7.0%), camphene, ${\beta}-pinene$, myrcene, ${\alpha}-phellandrene$, ${\Delta}-3-carene$, p-cymene, limonene(2.8%), terpinolene and also found to contain m-cresol(11.6%), citronellol, citral b, methylcinnamate, eudesmol and osthol. 2-Hydroxy-5-methylacetophenone and twelve compounds were tentatively identified.

  • Essential oil of the root of Angelica dahurica Benth et Hook(Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain eleven monoterpenes such as ${\alpha}-pinene(4.74%),\;campben, {\beta}-pinene,\;myrcene,\;{\alpha}-phellandrene,\;{\delta}-3-carene(39.4%),\;{\alpha}-terpinene,\;{\rho}-cymene,\;{\beta}-phellandrene,\;{\alpha}-terpinene,\;terpinolene\;and\;also\;found\;to\;contain\;4-vinylguaiacol,\;iso-elemicin,\;{\beta}-elemene$, caryophyllene, ligustilide, osthol and seven tentatively identified sesquiterpenes.

Four sesquiterpenes and five sesquiterpene alcohols were tentatively identified by comparison of their mass spectra.

The Domestic Trends of Osteoarthritis-related Research on Each Constituent Herb of ChondroT - Focused on the Journal of Korean Medicine (ChondroT 구성약물의 골관절염 관련 국내 연구 동향 - 한의학 학술지를 중심으로)

  • Objectives The purpose of this study is to analyze the osteoarthritis-related effect of each constituent herb of ChondroT, the herbal medicine that used for osteoarthritis. Methods We searched osteoarthritis-related studies on each constituent herb of ChondroT via five Korean Medicine web databases. We classified these studies by each constituent herb (Ostericii Radix, Angelica Gigas Nakai, Clematidis Radix, Lonicerae Flos, Phellodendri Cortex) and researched the osteoarthritis-related effect. Results We collected 18 studies (two studies of Ostericii Radix, five studies of Angelica Gigas Nakai, three studies of Clematidis Radix, six studies of Lonicerae Flos, and two studies of Phellodendri Cortex). All of these studies were composed of in vivo and in vitro, and there were six pharmacopuncture experimental study. All experiments showed the significant effects on specific inflammatory mediators. Conclusions These results suggest that ChondroT will be able to apply to the treatment of osteoarthritis through continuous research and development.

Comparison of Hemoglobin Activity of Angelica gigas, Angelica sinensis and Angelica acutiloba roots (참당귀, 중국당귀, 일당귀의 헤모글로빈 활성도 비교)

  • Objectives : Angelicae gigas, A. sinensis and A. acutiloba are three types of plants used as Angelicae Radix (Dang-Gui). Many doctors of Korean medicine want to know the difference in clinical use of these three species. This study aimed to compare the hemoglobin-related activity of the extracts of Angelicae gigas, A. sinensis and A. acutiloba roots by measuring the intensity of binding oxygen to hemoglobin using Raman spectroscopy. Methods : Hemoglobin activity was measured by chemical analysis and Raman spectroscopy to compare the pharmaceutical efficacy of three Angelica root extracts. The oxygenated hemoglobin intensity, blood decursinol and acetylcholinestrase(AChE) concentration in mice were measured. In addition, the effects of three Angelica root extracts on oxygenated hemoglobin intensity, decursinol and AChE concentration in red blood cells (RBC) from human were also investigated. Results : The contents of decursin, decursinol and decursinol angelate, which affected physiological activity and RBC properties, were higher in the extract of A. gigas root than in those of A. sinensis and A. acutiloba roots. Moreover, oxygenated hemoglobin intensity in the A. gigas extract was higher than that of other two species in the blood of mice and human RBCs. Also, the blood decursinol and AChE concentrations of A. gigas root extract were higher than that of A. sinensis and A. acutiloba roots. Conclusions : These results suggest that A. gigas is more effective in treating disease related oxygen deficiency in RBC deformation under oxidative stress.

Nodakenin and Decursin Contents of Fermented Angelicae Gigantis Radix by 4 Species Strain (4종의 발효균주가 당귀의 nodakenin과 decursin의 함량에 미치는 변화)

  • Objectives : The purpose of this study was to investigate the nodakenin and decursin contents in each fermented Angelicae gigantis Radix by 4 species of ferment strains. Methods : The strains of fermented Angelicae gigantis Radix were Lactobacillus plantarum(SE1), L. acidophilus(AC), Bacillus subtilis(B2) and B. licheniformis(BL2). The fermentative changes of nodakenin and decursin were analyzed using HPLC. Results : All of 4 species strains reduced nodakenin and decursin concentration in Angelicae gigantis Radix by fermentation. But fermentability were difference between strains. The nodakenin concentration of AC was lower than B2. The decursin concentrations of SE1 and AC were lower than B2 and BL2. Conclusions : 4 species of fermental strain were difference decomposable rate with nodakenin and decursin in fermented Angelicae gigantis Radix.

  • A mouse whole animal bioassay was employed to screen for potential mutagenicity of ethanol/water extracts of 16 Chinese herbal drugs that are commonly prescribed in Korea. Specific cytogenetic toxicity was measrured by recording evidence of clastogenesis toxicity was measured by recording evidence of clastogenesis via the mouse bone marrow micronucleus test. Male ICR mice administered ethanol extract of Pinelliae tuber (Pinellia eternata Breitenbach, ARACEAE, 양복) and ddY female mice administered extract of Angelica Koreanae radix(Angelica Koreana Maximowicz, UMBELLIFERAE, ) (both by oral administration, at a dose of 600 mg/kg), in a short-term dosing schedule, demonstrated significant increase in micronucleated polychromatophilic erythrocytes, indicating the increase of clastogenicity.

Determination of Byakangelicin in Angelicae dahuricae Radix by High Performance Liquid Chromatography (고속액체크로마토그라피에 의한 백지근 중 Byakangelicin의 정량)

  • A new method for quantitative determination of byakangelicin in Angelicae dahuricae Radix by high performance liquid chromatography was established. A reversed-phase system with a ${\mu}Bondapak$ $C_{18}$ column using THF : dioxane : MeOH : HAc : 5% $H_3PO_4$ : $H_2O$=72.5 : 62.5 : 25 : 10 : 1 : 329 as a mobile phase was developed. Byakangelicin together with ter-O-byakangelicin and oxypeucedanin methanolate, and isooxypeucedanin as an internal reference were detected at 350 nm and the analysis was successfully carried out within 30 min.
