To decrease the bolting rate of Angelica gigas, The growth inhibitors such as MH, Pp333 (paclobutrazol) and CCC(chloroniequat or Cycocel) were treated twice with the interval of 20 days atthe forming stage of flower bud.Growth and bolting rate in plot treated inhibitors were retarded and decreased, and the yield ofroots was also decreased compared with non - treatment. The treatments of MH and Pp333 amongthree inhibitors showed the better effects in decreasing the bolting than that of CCC, but they did notshow the significant in the yield and contents of decursin among each of them.In order to improve the decreasing effects by chemicals, selection of suitable reagents, concentrationand number of treatment should be investigated.
Angelicae gigantis Radix (dried root of Angelica gigas) including major bioactives such as decursin and decursinol angelate provides rich flavors and several healthy benefits. Recent studies have shown that ultrafine powders of herbal medicines provide better physical properties and biological activities. Thus, ultrafine Angelica powder was added into the oligosaccharide syrup to provide flavors and healthy benefits in this study. Angelicae gigantis Radix was pulverized into d(0.1) = 3.220, d(0.5) = 7.822, and d(0.9) = 7.817 $\mu$m respectively using an air-flow mill. The ultrafine Angelica powder was added into the oligosaccharide syrup process with different ratios of water to oligosaccharide syrup at 1:5, 1:8, 1:11, and 1:14. The physicochemical properties such as viscosity and bulk density were measured. The Stokes' law was applied to predict the sedimentation velocity of the added Angelica powder in the syrup. The Angelica syrup prepared in this experiment showed good stability since the Angelica particles precipitated down slowly. The ratio of water to oligosaccharide syrup at 1:11 showed the optimal preparation in terms of the stability and the viscosity. The ultrafine-sized herbal powders such as Angelicae gigantis Radix have potentials for various food and pharmaceutical applications.

This study was carried out to investigate characteristics of growth and active compounds of Angelica gigas population distributed in Mt. Jeombong. Quadrates were established in native area to harvest root part of Angelica gigas and analyze soil properties in September and October 2017. It was found that Angelica gigas populations were collected above the altitude 758 m near the valley covered with Fraxinus mandshurica and Acer pictum, and so on. In case of soil nutrient, soil organic matter, available phosphorous, and total nitrogen ranged 5.8-25.2%, 23.0-67.9 mg/kg and 0.3-1.3% respectively. Also, the Fresh weight of root ranged 28.8-65.3 g in September and 22.3-75.6 g in October. The content of active compounds ranged 2.7-4.7% in decursin and 2.9-4.5% decursinol angelate in October that was increased by 1% and 2.4% respectively compared with that in September. In the final analysis, there was positive correlation between fresh weight of root and exchangeable cation (natrium, calcium, magnesium). Also, there was positive correlation between active compounds and the content of water in root part of Angelica gigas.

This study were carried out to find bolting response of cultivation in different regions and to isolate FLC (FLOWERING LOCUS C) homologs in Angelica gigas Nakai. The mean temperature of different regions, ordering in altitude, were as follows: 100 m > 350 m > 530 m > 700 m. The largest amount of rainfall was occurred in the region of 350 m while the longest time of sunshine was occurred in the region of 100 m. The content of soil chemical properties in regions showed pH 6.2 ~ 7.4, T-N 0.17 ~ 26, organic mater $1{\sim}32gkg^{-1}$, $P_2O_5$${151{\sim}664_{mgkg}}^{-1}$, exchangeable potassium and calcium and magnesium were 0.78 ~ 1.15, 3.9 ~ 10.0, ${0.7{\sim}3.2_{cmol}}^{+kg-1}$. L5 line of A. gigas was occurred in bolting at all regions, but the bolting ratio was 60.0% in 700 m region with non-mulching treatment. Manchu of A. gigas was not occurred in bolting at all regions. The accumulation bolting ratio of L5 line by non-mulching was higher than that of mulching as 90.4% and 72.8% in 100 m region. The MADS-box transcription factor FLC is one of the well-known examples as a strong floral repressor. We decided to isolate FLC homologs from A. gigas as a starting point of flowering mechanism research of this plant. We have isolated two RT-PCR products which showed very high amino acid sequence homology to Arabidopsis FLC.
The bioactive materials (phenolic compounds, flavonoids, minerals, decursin and decursinol angelate) and biological activities (DPPH [$\alpha,\alpha$'-diphenyl-$\beta$-picrylhydrazyl] free radical scavenging capability, reducing power, and tyrosinase activity) in the extracts of leaf, stem mixture (AGLS), and root (AGR) from Angelica gigas Nakai were examined by using water, hot water and ethanol solvent. The highest extract yield (21.89%) was found in the water extract of AGR. The highest concentrations of phenolic compounds and flavonoids in the ethanol extracts of AGLS and AGR were 14.99% and 14.79%. Major minerals of AGLS and AGR were K, Mg, Fe, Na and Ca. Decursin and decursinol angelate were the major ingredients of Angelica gigas, detected at 18.71 and 18.89 min of retention time by HPLC analysis, respectively. The highest concentrations of decursin and decursinol angelate in the Angelica gigas ethanol extract were found in root ($41.7\;{\mu}g/g$) and leaf ($34.04\;{\mu}g/g$). The highest free radical scavenging activity was found in the hot water extracts of AGLS and AGR, and its activity was stronger in all extracts of AGLS than AGR. The highest reducing power was found in the ethanol extracts of AGLS and AGR and this was dependent on the sample concentration. The hot water extracts of AGLS and AGR revealed the highest inhibition activity on tyrosinase. Overall, these results may provide the basic data needed to understand the biological activities of bioactive materials derived from Angelica gigas.

The Hippo-YAP signaling pathway is critical for cell proliferation, survival, and self-renewal in both Drosophila and mammals. Disorder of Hippo-YAP pathway leads to tumor development, progression and poor prognosis in various cancers. YAP/TAZ are the key downstream effectors of the Hippo pathway and they can be inhibited through LATS1/2, core kinases in the Hippo pathway, mediated phosphorylation. In this study, we investigated the effect of Angelica gigas Nakai extract (AGNE) on Hippo-YAP/TAZ pathway. First, ANGE induced YAP/TAZ phosphorylation and dissociation of the YAP/TAZ-TEAD transcription complex. By qRT-PCR, we found that ANGE inhibits the expression of YAP/TAZ-TEAD target gene, CTGF and CYR61. In addition, the transcriptional activity of YAP/TAZ was not suppressed significantly in LATS1/2 double-knockout (DKO) cells by ANGE compared to LATS1/2 wild-type (WT) cells, which means AGNE inhibits YAP/TAZ signaling through direct action on LATS1/2. Further, it was confirmed that AGNE-induced activation of LATS1/2 inhibited the migration potential of the vector-expressing cells by suppressing YAP/TAZ activity. The reduced migration potential was restored in active YAP-TEAD expressing cells. Taken together, the results of this study indicate that ANGE downregulates YAP/TAZ signaling in cells through the activation of LATS1/2.
Korean angelica(Angelica gigas Nakai), a oriental medicinal plants is important and in great demand in Korea. This crop is a biennial which is to die after flower, so as could not harvest the root. A new variety 'Anpung' was developed from the selection of the local collecting population by medicinal crop team at NCES in 2001. This showed stable bolting during its cropping year and have high quality and yield potential. The color is greenish purple in stem and red purple in flower. It was longer in plant height and root length, thicker in root diameter, more leave and latheral roots comparing with the check variety 'Manchu'. The dry root was 72 g/plant showing heavier than check variety of 65 g/plant. It showed such low bolting rate as 0.5%, on the other hand 49.3% in landrace when it cultured by transplanting with heating nursery seedlings, and $0{\sim}15%$ with field nursery seedlings. The dry root yield of the variety was averaged 3,630 kg/ha out yielding the check variety by 11% at the three regional yield test from 1999 to 2001. Finally, 'Anpung' showed higher content of extracts up to 46.3% in 'Anpung' and 45% in check variety, decursin and decursinol contents was 3.48% and 2.79%, showing slightly higher than check variety.
The most important subject in cultivation of A. gigas NAKAI would be how to decrease the bolting rate. Belcause the decrease and poor of the yields and quality is imevitable in such field where the bolting rate is high. This study was carried out to investigate the time of flower-bud differentiation as a part of improvements of cultivation method. Shoot apex was fixed in Farmer's solution(ETOH 3:Acetic acid 1), thin paraffin ribbon was made, and the tissue was stained by Safranin solution. Differentiation time of flower-bud in A. gigas differed greatly from individuals by the seedling size, generally begun from middle of May to late of June, it was time that two leaves spread. In this results, Photoperiocid types of A. gigas NAKAI were regarded as LI type or LS type of dull-photosensitivity.

Two pyranocoumarin constituents have been isolated from Angelica gigas and were identified as decursinol angelate (1) and decursin (2) by means of NMR analysis, respectively. Human acyl-CoA:cholesterol acyltransferase (hACAT) inhibitory activity of decursinol angelate (1) and decursin (2) was evaluated. Decursin (2) showed significantly inhibitory activity against hACAT1 and hACAT2 with $IC_{50}$ value of 137 and $168\;{\mu}M$, respectively, whereas decursinol angelate (1) exhibited weak ACAT inhibitory activity. These results suggested that decusin from A. gigas might be effective for the prevention and the treatment of hypercholesterolemia or atherosclerosis by inhibitory effect on hACAT. The contents of decursinol angelate (1) and decursin (2) were analyzed in various parts of A. gigas including flower, seed, leaf and root using LC/MS/MS (ESI, positive ion mode, MRM mode). The content of decursinol angelate was increased in order of flower, seed, leaf, and root and decursin content was increased in order of flower, seed, leaf, and root. It was expected that unused parts including leaf and flower of A. gigas might be useful as new functional sources by their high contents of decursin and decursinol angelate.

The purpose of the present study is a comparison of the antioxidant effects of Angelica gigas Korea (AG), Angelica sinensis of China (AS), and Angelica acutiloba of Japan (AA), and comparison of the effects of AG, AS and AA on tight-junction related genes in human keratinocyte HaCaT cells. All species showed a strong antioxidant effect, and AA was higher than AG and AS in antioxidant effects. The cytotoxicity was confirmed to be higher in AS than AG and AA at a concentration of $1,600{\mu}g/ml$ using the MTS assay in HaCaT cells. We analyzed the effects of AG, AS, and AA on mRNA expression levels of various tight-junction related genes in HaCaT cells. We found that no obvious changes in expression of Claudin 1, 3, 4, 6, 7, 8, Occludin, JAM-A, ZO-1, ZO-2, and tricellulin by treatment of all species, suggesting that there is less possibility of side effects and skin moisturizing effects due to changes in tight-junction gene expression. Our results suggest that AG, AS, and AA are thought to be effective in reducing the oxidative stress of the skin and preventing the aging of the skin.
