H$_2$-producing bacteria were isolated from anaerobic granular sludge. Out of 72 colonies (36 grown under aerobic conditions and 36 under anaerobic conditions) arbitrarily chosen from the agar plate cultures of a Suspended sludge, 34 colonies (15 under aerobic conditions and 19 under anaerobic conditions) produced H2 under anaerobic conditions. Based on various biochemical tests and microscopic observations, they were classified into 13 groups and tentatively identified as follows: From aerobic isolates, Aeromonar spp. (7 strains), Pseudomonas spp. (3 strains), and Vibrio spp. (5 strains); from anaerobic isolates, Actinomyces spp. (11 Strains), Clostridium 5pp. (7 strains). and Porphyromonas sp. When glucose was used as the carbon substrate, all isolates showed a similar cell density and a H$_2$ production yield in the batch cultivations after 12 h (2.24-2.74 OD at 600 nm and 1.02-1.22 mol H$_2$/mol glucose, respectively). The major fermentation by-products were ethanol and acetate for the aerobic isolates, and ethanol, acetate and propionate for the anaerobic isolates. This study demonstrated that several H$_2$ producers in an anaerobic granular sludge exist En large proportions and their performance in terms of H$_2$ production is quite similar.
The swine waste industry is growing rapidly along with the world human population. The trend is toward more concentrated piggeries with numbers of herds in the thousands. Associated with these increased herds are large quantities of wastes, including organic matter, inorganic nutrients, and gaseous emissions. The trend in swine waste management is toward treatment of these wastes to minimize negative impact on the health and comfort of workers and animals and the atmosphere, water, and soil environments. Treatment of these wastes has traditionally involved land application, lagoons, oxidation ditches, and conventional batch and continuously stirred reactor designs. More sophisticated treatment systems are being implemented, involving advanced anaerobic digester designs, integrated with solids separation, aerobic polishing of digester effluents, and biological nutrient removal. This review discusses the present and future role of anaerobic processes in piggery waste treatment with emphasis on reactor design, operating and performance parameters, and effluent processing.
This study is to investigate the kinetics of anaerobic process, the effect of mass transfer on process, and the characteristics of the conventional anaerobic bioreactor, and develop new high efficiency bioreactor. In the new bioreactor wastewater containing highly concentrated organic materials, was treated without diluting wastewater. In this experiment the high COD removal rate (about 88%) and gas production(about 200l/d) was showed with short residence time(1.5 day). This performance is about 10 times as large as the conventional reactor.
Laboratory experiments were conducted to investigate the performances of anaerobic filters packed with ceramic tube and pall-ring media treating a dairy waste. The media packing volume was 65% of effective volume of anaerobic filter. Organics removals of anaerobic filters were maintained above 80% even at an organics loading rate of $10kgCOD/m^3/d$, and this was comparable to aerobic treatment of organic wastes. Organics removals of the ceramic tube anaerobic filters were always lower than those of the pall-ring anaerobic filters due to intrinsic physical property of ceramic tube, especially lower void space which caused to clogging and entrapment of biogas, substrate transfer limitation, and irregular evolution of biogas leading to loss of solids and biomass. This was clearly observed in higher concentration of TSS in the effluent from the ceramic tube anaerobic filter despite of higher retention capacity of TSS compared with pall-ring media. Vertical distribution of organics and solids in the filters showed above 90% of organics and solids in influent were removed below 20% of reactor height, and 50% of remaining organics and solids were removed though media packing zone. Effluent quality from the anaerobic filter was heavily depended on media itself as well as suspended biomass formed below media. It is therefore concluded that the type of media played an important role in biomass accumulation arid gas-liquid-solid separation efficiency. Type of media did not affect the start-up behaviors of the anaerobic filter, and supernatant from anaerobic digested sludge showed a good performance as a seeding materials.
In order to assess the performance of co-digester using pig slurry and food waste at the farm scale biogas production facility, the anaerobic facility that adopts the one-stage CSTR of 5 $m^3\;day^{-1}$ input scale was designed and installed under the conditions of the OLR of 2.33 kg $m^3\;day^{-1}$ and HRT of 30 days in an pig farmhouse. Several operation parameters were monitored for assessment of the process performance. The anaerobic facility was operated in three stages to compare the performance of the anaerobic co-digester. In the Stage I, that was fed with a mix of pig slurry to food waste ratio of 7:3 in the input volume, where input TS content was 4.7 (${\pm}0.8$) %, and OLR was 0.837-1.668 kg-VS $m^3\;day^{-1}$. An average biogas yield observed was 252 $Nm^3\;day^{-1}$ with methane content 67.9%. This facility was capable of producing an electricity of 626 kWh $day^{-1}$ and a heat recovery of 689 Mcal day-1. In Stage II, that was fed with a mixture of pig slurry and food waste at the ratio of 6:4 in the input volume, where input TS content was 6.9 (${\pm}1.9$) %, and OLR was 1.220-3.524 kg-VS $m^3\;day^{-1}$. The TS content of digestate was increased to 3.0 (${\pm}0.3$) %. In Stage III, that was fed with only pig slurry, input TS content was 3.6 (${\pm}2.0$) %, and OLR was 0.182-2.187 kg-VS $m^3\;day^{-1}$. In stage III, TS and volatile solid contents in the input pig slurry were highly variable, and input VFAs and alkalinity values that affect the performance of anaerobic digester were also more variable and sensitive to the variation of input organic loading during the digester operation. The biogas produced in the stage III, ranged from 11.3 to 170.0 $m^3\;day^{-1}$, which was lower than 222.5-330.2 $m^3\;day^{-1}$ produced in the stage II.
Microbial community shifts, associated with performance data, were investigated in an anaerobic batch digester treating high-solid food waste under mesophilic conditions using, a combination of molecular techniques and chemical analysis methods. The batch process was successfully operated with an organic removal efficiency of 44.5% associated with a biogas yield of 0.82 L/g $VS_{removal}$. Microbial community structures were examined by denaturing gel gradient electrophoresis. Clostridium and Symbiobacterium organisms were suggested to be mainly responsible for the organic matter catabolism in hydrolysis and acidogenesis reactions. The dynamics of archaeal and methanogenic populations were monitored using real-time PCR targeting 16S rRNA genes. Methanosarcina was the predominant methanogen, suggesting that the methanogenesis took place mainly via an aceticlastic pathway. Hydrogenotrophic methanogens were also supported in high-solid anaerobic digestion of food waste through syntrophism with syntrophic bacterium. Microbial community shifts showed good agreement with the performance parameters in anaerobic digestion, implying the possibility of diagnosing a high-solid anaerobic digestion process by monitoring microbial community shifts. On the other hand, the batch results could be relevant to the start-up period of a continuous system and could also provide useful information to set up a continuous operation.
Purpose: In the process of anaerobic digestion, stirring of the digester and feeding of organic waste into the digester have been considered important factors for digestive efficiency. The objective of this study was to determine the most appropriate conditions for both stirring interval of the digester and organic feeding frequency in order to improve anaerobic digestion performance. Methods: A 5-L anaerobic digester was used to conduct continuous batch tests to process swine manure and food waste. Four different stirring intervals of the digester were used: 5 min/h, 10 min/2 h, 15 min/3 h, and 20 min/4 h. Results: The application of swine manure to the digester every 5 min/h resulted in the highest production of biogas as well as the highest removal rates of volatile solids (VS) and total chemical oxygen demand. Stirring the digester with a mixture of swine manure and food waste at intervals of 5min/h and 10min/2 h produced the highest biogas yields of 515.3 mL/gVS and 521.1 mL/gVS, respectively. To test different supply frequencies, organic waste was added to the digester in either a 12-hor 24-h cycle. The 24-h cycle produced 1.5-fold greater biogas production than that during the 12-h cycle. Conclusions: Thus, from the above results, to optimize anaerobic digestion performance, the ideal stirring condition must be 5min/h for swine manure feeding and 10min/2h for co-digestion of food waste and swine manure in a 24-h cycle.

Purpose: Since 2013, marine dumping of wastewater has been banned, and research on eco-friendly and efficient land treatment has emerged. This study compared and tested changes in biogas production and anaerobic process efficiency depending on whether or not enzyme pretreatment was performed during anaerobic digestion from single-phase and two-phase to medium-temperature. Research design, data and methodology: The total sugar, direct sugar, pH, and acidity before and after fermentation were analyzed by G/C by anaerobic fermentation of the liquor wastewater, food wastewater 1, and food wastewater 2 at 30℃ for 67 hours, and the amount of methane gas generated was analyzed by balloon volume. Results: It was found that stable organic acid concentration and pH were found in the enzyme-treated food wastewater 2, and the amount of methane gas generated was also increased. Conclusions: When anaerobic digestion of the liquor wastewater and the food wastewater together, the performance of enzyme pretreatment resulted in increased digestive efficiency. It will be the basic data that can contribute to carbon neutrality and greenhouse gas reduction by increasing the production of biogas.
A mathematical model was formulated to simulate the long-term performance of an anaerobic bioreactor designed to digest Korean food wastes. The system variables of various decomposition steps were built into the model, which predicts the temporal characters of Solid waste, and volatile fatty acid (VFA) in the reactor, and gas production in response to various input loadings and temperatures. The predicted values of VFA and gas production were found to be in good agreement with experimental observations in batch and repeated-input systems. Finally, long-term reactor performance was simulated with respect to the seasonal temperature changes from 5C in winter to 25C in Summer at different food waste input loadings. The simulation results provided us with information concerning the success or failure of a process during long-term operation .
