Effects of Sprinkler and Fan Cooling System in Loose Barn on the Physiological Parameter and Milk Productivity in Holstein Cows During Summer (개방식우사내 Fan과 Sprinkler설치가 착유우의 생리적 변화 및 유생산성에 미치는 영향)

  • This experiment was carried out to study the effect of sprinkler and fan cooling system on the physiological parameter, milk production and milk composition for Holstein cows in hot, humid climates. Thirty cows were assigned to one of two sections of open-sided loose barn. Water nozzles of sprinkler system were spaced in line at 1.2m intervals. Forced air was provided by 85cm diameter fans at rate of 3.4㎥/sec. The results obtained from these experiments are as follows: 1. There was no significant difference in meteorological data between control and fan + sprinkler cooling system(treatment group). 2. Skin temperature and rectal temperature of the treatment group were significantly lower than those of the control group (32.96 : 39.53$^{\circ}C$ vs 34.02 : 41.21$^{\circ}C$ respectively) (P<0.05). 3. Serum cortisol concentration of the treatment group(0.90$\mu\textrm{g}$/dL) was lower than that of control group(1.44$\mu\textrm{g}$/dL)(P<0.05). 4. Milk production of cows cooled with a sprinkler and fan cooling system was significantly higher than that of no cooling system (P<0.0l). 5. Lactose, protein and solid-not-fat content of milk were not changed by the treatments. Milk fat content of the control(3.23%) was low compared with the treatment group(3.38%). Somatic cell count was reduced by 26.63% in the treatment. The results indicate that a sprinkler and fan cooling systems can provide an effective means to relieve heat stress and enhance productivity of lactating Holstein cows during hot and humid summer season.

Study on the Development of Measuring System for Fermentation Degree of Liquid Swine Manure Using Visible Ray (가시광선을 이용한 돈분뇨 액비 부숙도 측정장치 개발에 관한 연구)

  • This study was conducted to develop an measuring system and method for fermentation degree of liquid swine manure by visible ray. The constituent changes of liquid swine manure were examined. pH gradually increased with time, but EC gradually decreased. Malodor strength decreased gradually with aeration treatment with time. Control needed more time to decrease malodor strength than aeration treatment. In aeration treatment, there was no germination of seeds (radish, chinese cabbage) up to 6 weeks and germination rate at 15th week was over 50%. However, in control, there was no germination up to end of experiment. Circular chromatography method showed that there was change after 10th week in aeration treatment but there was no change up to end of experiment in control. As a result, the fermentation degree of liquid swine manure would have relations among pH, EC, germination rate, malodor concentration, and reaction of circular chromatography. The simple analytical instrument for liquid swine manure consisted of a tungsten halogen and deuterium lamp for light source, a sample holder, a quartz cell, spectrometer for spectrum analyzer, a malodour measuring device, a software, etc. Results showed that the simple analytical instrument that was developed can approximately predict the fermentation degree of liquid swine manure by visible ray. Generally, the experiment proved that the simple analytical instrument was reliable, feasible and practical for analyzing the fermentation degree of liquid swine manure.

A Study on Livestock Odor Reduction Using Water Washing System (수세탈취시스템을 이용한 축산악취저감에 관한 연구)

  • The odor problem in the livestock is increasing by 7% annually. Most importantly, the livestock odor problem in swinery accounts for the maximum ratio (54%). In this study, we reviewed the possibility of deodorizing swinery using an odor reduction device that can be used with the water washing system. First, the study confirmed that the solubility of odor gas, which was hydrogen sulfide, was very low regardless of the contact time with solvent, but the solubility of methyl mercaptan was found to increase along with the increase in time. The solubility of other odor gases, such as dimethyl sulfide, dimethyl disulfide and ammonia, was considerably high. Consequently, it is considered that if the odor reduction device for the water washing system deodorization is used in a swinery, the time during which the exhaust gas is in contact with usable water must be extended, or solvent quantity must be expanded. However, it is predicted that although hydrogen sulfide is easily generated in the anaerobic condition, it is difficult to expect high odor reduction efficiency because this gas has low solubility in water, especially in case it is used in the deodorization of the water washing system. The result of the solubility experiment using the bench-scale device practically manufactured represented the higher odor reduction ratio than expected. This result was possible because the removal efficiency of dust particles could be reached up to 93%. Therefore, it is judged that also the odor gas absorbed on dust particles could be removed by removal of dust. Consequently, it is expected that the higher order reduction ratio will be possible by structural improvement for increasing contact with water and odor gas.

Comparison of Weekly and Batch Management System for Sows (모돈의 주간관리와 그룹관리 비교)

  • Conventionally, many pig producers have utilized a continual sow managements system that the matings, farrowings and weanings are done weekly basis. But this transitional method is not able to cut the cycle of diseases and fully apply all-in/all-out system because of the continuous flow of sows and pigs. Conventional weekly management system is currently limiting in small farm to work efficiently both for workers and pigs. Therefore, pig producers have found novel management methods for applying all-in/all-out system, improving pig health, leading to better growth, lowering mortality and reducing medication costs nowadays. Moreover, all-in/all-out pig management system has known as a strategy for improving productivity in swine farm. The batch system is one of the best management methods to adopt all-in/all-out pig management system that prevent spreading diseases in pig and remove cycle of diseases. Batch farrowing system is a concept for providing a group of sows that delivery within a specific farrowing interval and inducing a large enough scale of piglets to fill the weaner facilities. There are different types of batch farrowing system with batch size and interval of farrowing when several factors at the swine farm are considered such as total number of sows, available facilities in the farm, and the efficiency of workforce. Sow managements such as farrowing, weaning and breeding, every 3 weeks rather than weekly, 2 or 5-week interval have advantages for workers and reproductive cycle of sows as well as pig flow. Because there are several pros and cons both in weekly and batch management system, various factors should be considered to apply the most suitable management system in each individual farm. To improve poor swine productivity in Korea compared to ED, batch system for sows will be an alternative choice which is able to prevent high incidence of diseases in swine farm such as PMWS, PRRS, PRDC and PED because all-in/all-out pig management can be also applied automatically by using this management system.

Evaluation on Heating Effects of Geothermal Heat Pump System in Farrowing House (지열 난방시스템을 이용한 분만돈사의 난방효과 분석)

  • Geothermal heat pump system (GHPS) is an energy-efficient technology that use the relatively constant and renewable energy stored in the earth to provide heating and cooling. With the aim of using GHPS as a heating source, it's possibilities of application in farrowing house were examined by measuring environmental assessment and sow's performance. A total of 96 sows were assigned to 2 pig housings (GHPS and conventional housing) with 48 for four weeks in winter season. During the experimental period, indoor maximum temperature in GHPS-housing was measured up to $26.7^{\circ}C$, average temperature could maintain $21.2^{\circ}C$. The mean value of dust levels and $CO_2$, $NH_3$ and $H_2S$ gas emissions were decreased in GHPS-housing compare with those of conventional housing. Litter size, birth weight, parity and weaning weight did not differ between housings. However, feed intake of sow in GHPS-housing was lower than that of conventional housing. In energy consumption for heating, electric power consumption increased in GHPS-housing than the conventional housing, a 2,250 kwh increase, whereas there is no fuel usage for heater in GHPS-housing. Amount of ground water circulated for heating in cold weather for earth heat exchanger was 8.4-12.9 ton per day. In conclusion, GHPS may have environmental benefits and effectiveness of heating in farrowing housing and affect the performance in sows.

Transcriptional Activation and Repression of Cell Cycle Regulatory Molecules by Trichostatin A (Trichostatin A 처리에 의하 세포주기 조절인자들의 전사활성화 및 불활성화)

  • The dihydrofolate reductase (dhfr) promoter contains cis-acting element for the transcription factors Spl and E2F. Transcription of dhfr gene shows maximal activity during the Gl/S phase of cell cycle. The member of the Spl transcriptional factor family can act as both negative and positive regulators of gene expression. There was a report that Spl-Rb and E2F4-pl30 complexes cooperate to establish stable repression of dhfr gene expression in CHOC400 cells. Here, we examined the role of HDAC in dhfr, cyclin E, and cyclin A gene regulation using the histone deacetylation inhibitor, trichostatin A (TSA) in U2OS and C33A cells, a Rb-positive human osteosarcoma cell line, and a Rb-negative cervical carcinoma cell line, respectively. When the dhfr promoter constructs were applied in U2OS cells, TSA markedly stimulated over 14-fold of dhfr promoter activity through dhfr-Spl sites by the deletion of an E2F element. In contrast, the deletion of dhfr-Spl binding sites completely abolished promoter stimulation by TSA. The dhfr promoter activity including dhfr-Spl sites increased only 2-fold in C33A cells. Promoter activity containing only dhfr-E2F site did not have much effect by the treatment of TSA in both U2OS and C33A cells. On the other hand, treatment with TSA induced significantly mRNA expression of dhfr and cyclin E, whereas levels of cyclin A decreased in U2OS cells, but had no effect in C33A cells. These results indicate that TSA have contradictory effect, activation of dhfr and cyclin E genes on Gl phase, and down-regulation of cyclin A on G2 phase through transcriptional regulation in U2OS cells.

Stability of the Fat Ingredients of Deep Fried Instant Noodles, Biscuits, and Cookies (라면, 비스킷, 및 쿠키속의 유지성분(油脂成分)의 안전성(安定性)에 관한 연구(硏究))

  • Deep fried instant noodles, biscuits, and cookies were prepared, using the same beef tallow as their fat ingredient. In addition to wheat flour and beef tallow, the common and major ingredients, the deep fried instant noodle contained 1.5% salt before frying, the biscuits 20.0% sucrose and 10.0% nonfat milk solid before baking, and the cookies 20.0% sucrose before baking. The three products and a portion of beef tallow, which was to be used as control, were stored in an incubator at $47.0{\pm}1.5^{\circ}C$. The peroxide value and the free fatty acid value of the control and the extracted fat were determined regularly during the storage period. The fat incorporated in the biscuits exhibited far greater stability to rancidity development than that of the control with regard to both peroxide value and free fatty acid value development. However, the fat incorporated in the deep fried instant noodles and the cookies showed much poorer stability than that of the control. Factors like a deep frying process and/or the presence of a significant amount of salt in the deep fried instant noodles appeared to promote the rancidity development of the fat incorporated in the product. On the other hand, Maillard type browning reaction products in the biscuits seemed to retard effectively the rancidity development of the fat incorporated in the product.

Physico-chemical and Microbiological Changes of Traditional Meju during Fermentation in Kangweondo Area (강원도 지방의 재래식 메주 발효중 이화학적 특성 및 미생물의 변화)

  • By using Korean native soybean, traditional meju was prepared in Chuncheon, Kangweondo according to the traditional process. Analysis of physico-chemical, enzymatic and microbiological changes during meju fermentation were carried out in order to obtain a basic information for industrial scale production of meju. The enviroments for natural meju fermentation were $10{\sim}15^{\circ}C$ and $60{\sim}70%{\;}RH$. Moisture content decreased from 59% to 11% (exterior section) and 19% (interior section). the pH of meju rapidly increased up to 8.5 at $33^{rd}{\;}day$ of fermentation and thereafter decreased down to 7.9 at $70^{th}{\;}day$ of fermentation. Souble protein content was 1.47% at initial stage and increased up to $6.31{\sim}7.34%$ at $33^{rd}{\;}day$ of fermentation. Amino nitrogen content was $460{\sim}770{\;}mg%$ at $70^{th}{\;}day$ of fermentation. the color of meju became gradually black and decreased in redness and yellowness. During the process, protease and lipase seemed to play an important role in the digestion of soy protein and fat. Acidic protease activity increased up to $135.9{\sim}152.4{\;}unit/g$ at $33^{rd}{\;}day$ of fermentation and were $181.3{\sim}272.6{\;}unit/g$ at $70^{th}{\;}day$ of fermentation. Lipase activity increased up to 6 unit/g (interior section) and 15 unit/g (exterior section) at $70^{th}{\;}day$ of fermentation. the viable cell count of meju was at the level of $10^8{\;}CFU/g$ during the overall fermentation period. Aerobic halophilic count was $1.51{\times}10^7{\;}CFU/g$ at initial stage and maintained $10^8{\;}CFU/g$ level during the process. Initial anaerobic cell count was $2.0^9{\times}10^4{\;}CFU/g$ and increased up to $10^5{\;}CFU/g$ level at 47 days. Yeast and mold counts were $10^4{\sim}10^5{\;}CFU/g$ for the fermentation period.

Levels of Plasma Glucose and Lipid in Rats Fed Bread Supplemented with Natural Extracts (천연추출물이 첨가된 식빵을 섭취한 흰쥐의 혈당 및 지질수준에 미치는 영향)

  • In this study, 4-week-old rats were fed bread supplemented with Terminalia chebula (TC), Plantago asiatica (PA), Linder obtusiloba (LO), and Capsosiphon fulvescens (CF) ethanol extracts, to determine the decrease in blood glucose levels, as well as the anti-inflammatory and lipid-enhancing effects. Previous studies have demonstrated the antioxidative effects of these ethanol extracts. After sacrifice, the liver tissue, whole blood, and serum samples were collected for biochemical analysis. The results showed a significant decrease in blood glucose level, lipid peroxidation, malondialdehyde (MDA) level, HbA1c level, total cholesterol, and low-density lipoprotein (LDL)-cholesterol (p<0.05) and an increase in high-density lipoprotein (HDL)-cholesterol level in rats fed bread supplemented with LO and CF ethanol extracts (p<0.05). Therefore, the results of this study demonstrate that bread supplemented with LO and CF ethanol extracts can potentially affect the blood glucose level and lead to lipid enhancement.

Changes in Feed Value, Forage Productivity, and Grazing Intensity at Mountainous Pasture Grazed by Growing Korean Native Goat (Capra hircus coreanae) (육성 흑염소 방목 산지초지에서 사료가치, 사초 건물 수량 및 방목 강도의 계절별 변화에 관한 연구)

  • This study was conducted to determine grazing intensity of growing Korean native goats (Capra hircus coreanae) on mountainous pasture. It was carried out to obtain basic information for improvement of mountainous pasture management and establishing feeding system of Korean native goats. Castrated, male goats (n=10) with average initial body weight (BW) of $23.33{\pm}2.15kg$ and an average age of 4 months were used in this study. Grazing goats were supplemented by concentrates with 1.0% of BW. The crude protein content of forage was the highest in October ($22.71{\pm}0.25%$) and there were significantly differences (p<0.05) in monthly comparison. The forage productivity of pasture was the highest from May to June ($1718.7{\pm}207.5{\sim}1672.0{\pm}422.8kg/ha$ but it was decreased in July ($1356.0{\pm}103.8kg/ha$) because of drought and summer depression. Average daily gains (ADG) of goats were the highest in June ($99.5{\pm}6.4d/g$). Grazing intensity was calculated by forage productivity and dry matter intake (DMI) and was the highest in May (65 head/ha). As shown in the results of this research, grazing intensity was suggested to average 39 head/ha from May to October. It is desirable that adequate grazing intensity was maintained by adjusting supplemental feed.
