The objective of this study was to assess parameters affecting nitrite accumulation, which offers advantages in terms of less aeration energy and carbon consumption for denitrification. The influence of the alkalinity to $NH_4{^+}-N$ concentration ratio, pH, FA(free ammonia) concentration and temperature on nitrite accumulation was investigated. The experiment was performed with supernatant from dewatering process of anaerobic digested sludge using a submerged biofilm reactor. The influent contains high strength of ammonium nitrogen and the alkalinity was insufficient for complete nitrification. An increased nitrite accumulation was observed with increase in alkalinity to $NH_4{^+}-N$ concentration ratio. The increase in alkalinity to $NH_4{^+}-N$ concentration ratio has been a maior reason for the high pH value and FA concentration in the reactor. It can be considered that selective inhibition of Nitrobacter can be causes of nitrite accumulation. The nitrite accumulation increased with increment of temperature at fixed alkalinity to $NH_4{^+}-N$ concentration ratio.
The effects of temperature on nitrification of enriched nitrifiers were investigated by using kinetics and thermodynamics method through the batch test. Aquaculture recirculating water, which was sampled at Chung Cheong Buk-Do Inland Fisheries Research Institute, was analized to observe the characteristics of nitrification. Temporal variation of ammonium, nitrite and nitrate concentration was measured at batch experiments. Activation energy was calculated using Arrhenius equation with the oxidation rates of specific ammonium or nitrite ion. These oxidation rates were measured at temperature range of $6-35^{\circ}C$ and ammonium concentration range of 0.2-1.8 mg/L. Two distinct activation energy of Nitrosomonas sp. at temperature $6-15^{\circ}C\;and\;15-35^{\circ}C$ was 93.1 and 25.0 KJ/mol, respectively. Nitrate accumulation was observed at temperature over $15^{\circ}C.$
A sequencing batch reactor was operated under different pH conditions to see the influence of free ammonia (FA) and free nitrous acid (FNA) to microbial community on ammonium partial nitrification. Long-term influences of FA and FNA were evaluated by polymerase chain reaction-denaturing gradient gel electrophoresis and fluorescence in situ hybridization. Nitrite accumulation was successfully achieved at pH 8.2 and 6.3. The shifts in the microbial community were observed when influent ammonia concentration increased to 1 g $NH_4$-N/L at pH 8.2, and then when pH was dropped to 6.3. Both Nitrosomonas and Nitrosospira were selected during the startup of the reactor, and eventually became dominant members as ammonia-oxidizing bacteria. The results of molecular microbiological analysis strongly suggested that the composition of microbial community was changed according to the method used to control nitrite-oxidizing bacteria.

Recently, the interests on economical nitrogen removal from wastewater are growing. As a method of the novel nitrogen removal technology, nitrogen removal via nitrite pathway by selective inhibition of free ammonia and free nitrous acid on nitrite oxidizing bacteria have been intensively studied. The inhibition effects of free ammonia and free nitrous acid are low when domestic wastewater is used, however, because of its relatively lower nitrogen concentration than the wastewater from industry and landfill, etc. In this study, a sequencing batch reactor (SBR) is proposed for nitrogen removal to investigate the effect of the low nitrogen concentration on nitrite accumulation. Nitrification efficiency reached almost 100% during the aerobic cycle and the maximum specific nitrification rate ($V_{max,nit}$) reached $17.8mg\;NH_4{^+}-N/g\;MLVSS{\bullet}h$. During the anoxic cycle, average denitrification efficiency reached 87% and the maximum specific denitrification rate ($V_{max,den}$) reached $9.8mg\;NO_3{^-}-N/g\;MLVSS{\bullet}h$. From the analysis the main reason of nitrite accumulation in the SBR was free nitrous acid rather than free ammonia. Nitrite accumulation increased with the decrease of organic content in the wastewater and the mechanism is not well understood yet. From the result of fluorescent in situ hybridization, the distribution of nitrite oxidizing bacteria was in equilibrium with ammonium oxidizing bacteria when nitrite accumulation did not occur.
Some ammonium oxalate soluble pectic fragments prepared from cultured cell wall of Ephycla distrahya elicited the accumulation of p-coumarocylamino acids (p-CAA) in E. distachya cultures while water soluble and alkali soluble fractions had no activity. Partial purification of the pectic fragments fraction using DEAE-cellulose chromatography afforded two active fractions (PS-I and PS-II) which were composed of mainly uronic acids (98-99 w/w %). They elicited the accumulation of p-CAA in an amount of 52-60 nmol per gram fresh weight of cultures. The acidic sugar compositions of PS-I and PS-II were found to be galacturonic acid and glucuronic acid by TLC analysis. They were supposed to act as endogenous elicitors of p-CAA accumulation. In order to investigate the effect of ethylene on p-CAA accumulation, Ethrel, which is known as ethylene generator, and ACC(1-aminocyclopropane-1-carboxylic acid), a direct precusor of ethylene biosynthesis, were added to the culture. However, they did not glycopeptide elicitor [(Con A-II)], either. Consequently, no relationships between ethylene and p-CAA accumulation were recognized. Several tentative elicitors were teted for their activity. Commercial yeast glucan, $CuCl_2$, laminarin and laminariheptaose had slight activity whereas ${\alpha}$-methylmannopyranoside and commercial yeast mannan had no elicitor activity. ${\alpha}$-methylmannopyranoside which has been known as a tentative inhibitor of glucan elicitor in Glycine max did not affect on the elicitor activity of Con A-II.
Partial nitirification and denitrification process has been reported to be technically feasible and economically favorable, especially for wastewater treatment with high ammonium concentration or low Carbon/Nitrogen ratio. This research was performed to survey nitrite accumulation by nitritation in treating ADEPT effluent of piggery wastewater, which contains highly concentrated ammonia. To estimate the possibility of nitrite accumulation, DO concentration and SRT were investigated as key operational parameters. This result proved that nitritation to nitrite was steadily obtained under short sludge retention time. Oxygen limitation was proved to be just a subsidiary parameter. Energy efficiency of nitritation-denitritation process was higher than complete nitrification-denitrification because external carbon requirement for denitritation could be saved. Though the influent contained significant nonbiodegradable organic substrate, total nitrogen removal efficiency was more than 51% in nitritation-denitritation system.
L-canavanine was added to GAs treated barley seeds, and induced amylase activity, soluble protein content, and arginine content were mesured. Canavanine, added at the beginning of the incubation period, inhibited amylase activity and protein accumulation. Amylase activity decreased markedly by addition of canavanine at 6 hr after incubation, where soluble protein content was not affected. The addition of canavanine after 12 hr incubation did not show serioud inhibited effect on the amylase activity and protein accumulation. GAs incubation caused decrement in arginine content per mg protein, but it was somewhat recovered by canavanine treatment. The longer the time between GAs and canavanine addition was, the less the recovery ration was. Arginine content in the $\alpha$-amylase fraction (ammonium sulfate 20~50% saturation) was lower than in 0~20% fraction, but higher than in 50~80% fraction. These results and control expreiments, using cordycepin and cycloheximide, support the idea that canavanine might incorporate into protein.
Determination of cyanide ion has been studied by adsorptive stripping voltammetry using hanging mercury electrode. Cyanide ion complexed with copper ion is adsorpbed on the electrode and oxidised at the positive potential scan. Optimal conditions of CN determination were found to be ; supporting electrolyte solution ; 0.1 M NaCl of ammonium buffer at pH 10, accumulation potential; -800 mV vs Ag/AgCl, accumulation time ; 300 s, scan rate ; 50mV/s. The linear concentration of cyanide ion was observed in the range $1{\times}10^{-8}$, $1{\times}10^{-7}M$. The detection limit(n/s=3) was $0.13{\mu}g/L$($5{\times}10^{-9}M$) with 3.5% RSD.
Nitrite accumulation is not unusual in batch processes such as sequencing batch reactor (SBR) with high-strength of ammonium or nitrate wastewaters. A possible mechanism of nitrite inhibition on Acinetobacter was depicted in a biochemical model, which the protonated species, nitrous acid form of nitrite, affects proton relating transport at the proton-pumping site crossing the cell membrane under unlimited carbon and phosphorus conditions. This effect exerts inhibition of phosphorylation under aerobic condition and yields low APT/ADP ratio, consequently decrease poly-P synthesis and phosphorus uptake from outside the cell in the model.
Searches for the nutrition requirements of three strains of Brevibacterium ammoniagenes reported in the previous paper were carried out with an aim of achieving the striking accumulation of L-glutamic acid and the large multipication of cells. It was recognized that all three strains required both biotin and thiamine, together with amino acids such as histidine or cysteine, for their good growth and extracellular L-glutamic acid accumulation. The quantity of biotin required for remarkable growth of these microorganisms was quite different from that for the maximum production of L-glutamic acid. This result, however, did not apply in the case of thiamine. It was also confirmed that, of 18 amino acids, histidine and cysteine were the msot effective organic nitrogen sources, while the most available inorganic ammonium salt resulting in a large amount of L-glutamic acid-production and considerable cell gorwth was found to be only urea. Maximum accumulation of extracellular L-glutamic acid, more than 50%(w/w) of the initial sugar content, could be obtained from fermentation in the medium containing wheat-bran extract(Brev. ammoniagenes T-1 and Brev.ammoniagenes Y-2) or rice-bran extract(Brev. ammoniagenes YR-2), which confirmed us a possibility that these bacteria might be employed for industrial fermentation of L-glutamic acid.
