Protective Effects of Chalcone Derivatives for Acute Liver Injury in Mice

  • The hepatoprotective effects of chalcone derivatives were evaluated in D-galactosamine/lipopolysaccharide (D-GaIN/LPS)-induced fulminant hepatic failure in mouse. Thirteen chalcone derivatives were synthesized for study and their hepatoprotective effects were evaluated by assessing aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in serum. Chalcone preparations were injected into mice at 12 hand 1 h before intraperitoneal injection of D-GaIN/LPS. After abdominal administration, changes in AST and ALT between the control and treated groups were observed. Ten of the synthesized chalcone derivatives exhibited inhibitory effects on D-GaIN/LPS-induced levels of AST and ALT in mice. Compounds 2, 3, 8, 9, and 12 markedly reduced serum AST and ALT at 8 h, inhibited hepatocyte necrosis and showed significant hepatoprotective activities. The activity of compound 3 was compared with the bifendate (DDB) through oral administration. Compound 3 showed much higher inhibitory effects than bifendate for decreasing AST and ALT activity. The results indicate that compound 3 has strong hepatoprotective activity through suppression of tumor necrosis factor­alpha (TNF-alpha) preduction, reduction of the histological change in the liver, and attenuated of hepatocyte apoptosis confirmed by DNA fragmentation assay.

  • Biliary atresia (BA) is the result of fibrosing destructive inflammatory process affecting intrahepatic and extrahepatic bile ducts, which lead to cirrhosis and portal hypertension. Kasai portoenterostomy has been the standard operative procedure in biliary atresia. Recently, there has been remarkable increase in the survival rate in cases of BA. However, long-term survivors are not clearly evaluated in Korea. To define long-term prognosis factors of patients who underwent surgery for BA, a retrospective study was undertaken of 10 (37 %) patients surviving more than 10 years among 27 patients who underwent one of Kasai procedures between 1981 and 1995. Hepatomegaly was present in 4 and splenomegaly in 7 patients. Serum bilirubin was normalized at 1 year after operation. Aspartate aminotransferase (AST, GOT), Alanine aminotransferase(ALT, GPT) were normalized at 12 years and alkaline phosphatase(ALP) was normalized at 13.5 years. Cholangitis developed mainly within 5 years after operation so close follow up is needed. Life long follow-up is needed because of progressive deterioration of liver function even after 10 years.

고들빼기가 흰쥐의 성장률, 단백질 및 지질농도에 미치는 영향 (Effect of Godulbaegi(Ixeris sonchifolia H.) Powder on Growth, Protein and Lipid Concentration in Rats)

  • The effects of godulbaegi(Ixeris sonchifolia H.) powder on body weight gain, lipid and protein concentrations, and enzyme activities were studied in growing male rats for 4 and 8 week. The fats (Sprague-Dawley) were fed diets containing 5% godulbaegi or without. The body weght gain, food efficiency ratio, protein efficiency ratio, hemoglobin concentration and hematocrit value of rats fed godulbaegi diet were similar to the control diet. The ratio of spleen weight to body weight in godulbaegi group for 8 weeks was greater than in that of control group, but they were similar both feeding groups in liver and kidney. The concentrations of the total lipid and triglyceride in serum were increased in rats fed godulbaegi diet for 4 week than in those fed control diet, but those were decreased in rats fed godulbaegi diet for 8 week than in those fed the control diet. The concentrations of the phospholipid, total cholestrol and HDL-cholestrol in serum were decreased in rats fed godulbaegi diet for 8 week than in those fed control diet. The HDL-cholestrol/total cholestrol ratio and athergenic index were similar to both diet groups. There were no differences in growth, serum total protein, albumin concentrations, aspartate aminotransferase and alanine aminotransferase activities between rats fed godulbaegi diet and those fed the control diet.

키토산과 세리신 및 콜라겐 펩타이드 추출 혼합물이 이상지질혈증의 지질대사 개선에 미치는 영향 (Improvements Caused by Chitosan, Sericin and Collagen Peptide Extract Complexes on Lipid Metabolism in Dyslipidemia)

  • The objective of this study was to assess improvements caused by chitosan, sericin and collagen peptide extract complexes (1:1:1, w/w/w, CSC-F-005) in lipid concentrations in the sera of dyslipidemic rats (SD strain) fed on experimental diets for 5 weeks. Serum concentrations of total cholesterol, HDL-cholesterol, ratio of HDL-cholesterol concentration to total cholesterol, atherosclerotic index, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid and blood glucose were effective on the metabolic regulation of dyslipidemic rats. The activities of alkaline phosphatase, aspartate aminotransferase and alanine aminotransferase in serum were remarkably lower in the extract complexes (CSC-F-005) than in the dyslipidemic model. From the above results shows that CSC-F-005 extract complexes were effective on the improvement of the lipid metabolism in sera of dyslipidemic rats.

Preventive Effect of the Korean Traditional Health Drink (Taemyeongcheong) on Acetaminophen-Induced Hepatic Damage in ICR Mice

  • This study was to investigate the preventive effect of taemyeongcheong (TMC, a Korean traditional health drink) on acetaminophen (APAP, 800 mg/kg BW)-induced hepatic damage in ICR mice. TMC is prepared from Saururus chinensis, Taraxacum officinale, Zingiber officinale, Cirsium setidens, Salicornia herbacea, and Glycyrrhizae. A high dose of TMC (500 mg/kg BW) was found to decrease APAP-induced increases in serum levels of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and lactate dehydrogenase. TMC pretreatment also increased the hepatic levels of hepatic catalase, superoxide dismutase, glutathione peroxidase, and glutathione, and reduced serum levels of the inflammatory cytokines tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-6 in mice administered APAP (P<0.05). TMC (500 mg/kg BW) reduced hepatic mRNA levels of TNF-${\alpha}$, IL-$1{\beta}$, IL-6, COX-2, and iNOS by 87%, 84%, 89%, 85%, and 88%, respectively, in mice treated with APAP (P<0.05). Furthermore, histological observations suggested TMC pretreatment dose-dependently prevented APAP-induced hepatocyte damage. These results suggest that TMC could be used as a functional health drink to prevent hepatic damage.

Hypoglycemic Effect and Hepatic Detoxification Activity of Extracts from Crataegus fructus and Morus alba L. in Alcohol-treated Rats

  • To consider potentially new sources which have hypoglycemic effect and accelerating alcohol detoxification, this study was designed to investigate the effect of Crataegus fructus and Morns alba L. in alcohol-treated rats. I compared the body weight, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alcohol dehydrogenase (ADH), and aldehyde dehydrogenase (ALDH) of rats administered both alcohol and extract of experimental plants to rats treated with alcohol alone. Administration of extracts of C. fructus and M alba, respectively, resulted in a significant reduction in the blood glucose level and the activities of ADH of liver compared to the control rats, and administration of extract of M. alba showed significantly lower on bodyweight gain in the rats than in other treated rats. In contrast, the activities of ALDH of liver were increased. The activities of AST and ALT between the only alcohol-treated rats and the alcohol and experimental plants-treated rats were no significant difference. The results suggest that C. fructus and M alba have a hypoglycemic effect, and reduce liver damage by accelerating acetaldehyde metabolism in alcoholic rats, so the combined effect of C. fructus and M alba may be considered as an alternative remedy for hangovers, alcohol-induced overweight and alcohol-induced diabetes.

Proline accumulation and transcriptional regulation of proline biothesynthesis and degradation in Brassica napus

  • To understand the molecular mechanism underlying proline accumulation in Brassica napus, cDNAs encoding ${\Delta}^1$-pyrroline-5-carboxylate synthetase (BnP5CS), ornithine $\delta$-aminotransferase (BnOAT) and proline dehydrogenase (BnPDH) were isolated and characterized. Southern blot analysis of BnP5CSs in B. napus and its diploid ancestors suggested a gene loss may have occurred during evolution. The expression of BnP5CS1 and BnP5CS2 was induced, while the expression of BnPDH was inhibited under salt stress, ABA treatment and dehydration, prior to proline accumulation. The upregulation of BnOAT expression was only detected during prolonged severe osmotic stress. Our results indicate that stress-induced proline accumulation in B. napus results from the reciprocal action of activated biosynthesis and inhibited proline degradation. Whether the ornithine pathway is activated depends on the severity of stress. During development, proline content was high in reproductive organs and was accompanied by markedly high expression of BnP5CS and BnPDH, suggesting possible roles of proline during flower development.

Extracts of Codonopsis lanceolata Decrease $CCl_4$-Induced Hepatotoxicity on High Fat Diet in Rats

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)­induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.

Enzyme Activities and Histochemical Changes in the Hind Limb Muscle of the Mouse Treated with 6-Aminonicotinamide

  • We investigated enzyme activity and histochemical changes in hind limb of mouse treated with 6-aminonicotinamide (6-AN). The activity of aspartate aminotransferase, alanine aminotransferase and creatine phosphokinase in 6-AN treated group were significantly higher than those of the control and pair-fed groups. Also, the activity of lactic dehydrogenase in 6-AN treated group was the highest among the three groups, whereas that of the pair-fed group were higher than that of the control group. In the 6-AN treated group, oxidative histochemical stains, nicotinamide adenine dinucleotide reductase (NADH), succinyl dehydrogenase (SDH) showed increased scattered fibers in 6-AN treated subsarcolemma. Cytochrome c oxidase (COX) stain showed decreased up to 85% in 6-AN treated fibers. These results demonstrate that 6-AN antagonizes cell metabolism and induces the morphological deformity like the other mitochondrial muscle diseases. Therefore, we suppose that these data would be useful indexes for disclosing the mechanism of mitochondrial muscle disease.

Effects of Fruit Extract Drink on Alcohol Metabolic Enzymes in Ethanol-treated Rats

  • Alcoholism is a significant global health problem. Alcohol dehydrogenase and aldehyde dehydrogenase play important roles in the metabolism of alcohol and aldehyde. In this study, we aimed to investigate the eliminatory effects of a fruit extract drink on alcohol metabolism in drunken Sprague-Dawley (SD) rats. Male SD rats were given a fruit extract drink or a commercial product (10 mL/kg) 30 min prior to 40% (5 g/kg) ethanol ingestion. To assay the effect of the fruit extract drink on blood ethanol concentration, blood samples were taken from the saphenous vein at 3 and 5 h after ethanol ingestion. The blood concentrations of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase were significantly lower in the fruit extract drink group than in the control group, in a time-dependent manner. However, the alanine aminotransferase and aspartate aminotransferase activities of all experimental groups were unaltered compared to those of the control group. These results suggested that fruit extract drink intake can have a positive effect on the reduction of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase concentrations in the blood and may alleviate acute ethanol-induced hepatotoxicity by altering alcohol metabolic enzyme activities.
