Effects of Malloti Cortex Water Extract, Bergenin, and Acetylbergenin on Liver Fibrosis Induced by Bile Duct Ligation in Rats

  • The effects of Malloti Cortex Water Extract (MCWE), bergenin (isolated as an active component from MCWE), and acetylbergenin (synthesized from acetylation of bergenin) on the liver fibrosis induced by bile duct ligation (BDL) in rats. We studied hydroxypro1ine (HYP) as a marker of collagen accumulation in the liver, alanine aminotransferase (s-ALT), aspartate aminotransferase (s-AST), and alkaline phosphatase (s-ALP) as serum markers of liver cell damage induced by BDL, MCWE, bergenin, and acetylbergenin decreased towards normal the accumulated levels of HYP in the liver and the elevated serum levels of s-ALT, s-AST and 5-ALP. The results indicate that MCWE, bergenin, and acetylbergenin ameliorated the liver damage induced by BDL in rats.

Protective Effect of Sachungwhan against CCl4-induced Hepatotoxicity

  • Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine(CRE), total cholesterol(TCHO), triglyceride(TG), low density lipoprotein cholesterol(LDL-CHO), high density lipoprotein cholesterol(HDL-CHO), total protein(TP), albumin(ALB) and total bilirubin(BIL) in serum. Hepatic parameters monitored were levels of cholesterol(CHO), triglyceride(TG), malondialdehyde(MDA), content of cytochrome P450(CYP), level of glutathione(GSH), and activities of NADPH-CYP reductase, superoxide dismutase(SOD), catalase(CAT), glutathione S-transferase(GST), glutathione reductase(GR), glutathione peroxidase(GPx). The histopathological examination showed that the treatment of Sachungwhan relieved the ballooning degeneration of hepatocytes which had been generated by $CCl_4$. The results suggested that hepatoprotective effects of Sachungwhan possibly are due to their promising antioxidative activity.

  • CCl4 has been known as typical chemical which induce acute hepatitis accompanying increase in the levels of acute phase proteins in serum. In this study, after acute liver damage was induced by CCl4 in Sprague-Dawley rats, the levels of serum acute phase proteins were examined using 2-dimensional electrophoresis and lactic dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels in blood were also examined at 1, 2, 3 days after the induction to confirm acute hepatitis. Effect of Silk Fibroin on the Protection of Alcoholic Hepatotoxicity in the Liver of Alcohol Preference Mouse

  • Silk fibroin (SF) derided from the domestic silk worm, bombyx mori, is the natural protein and widely used as bio-functional materials as well as apparels. We studied the livers protective effect of SF from alcohol-induced hepatotoxicity in the alcohol preference mouse. To increase more absorption of SF in experimental animals, molecular weight of SF was lowered by 2N of HCI aqueous solution at 10$0^{\circ}C$ for 48 hrs. SF was added to liquid diet with alcohol and fed to the alcohol preference mice for 4 weeks. To assess the liver function, the concentration of alanine aminotransferase (AlT), aspartate aminotransferase (AST) and cholesterol present in either blood or liver tissue were measured. As compared with non-SF treated groups the SF-treated showed significantly low concentrations of ALT, AST, cholesterol and triacylglycerol values, respectively. Histopathological examination revealed that the extent of hepatocyte injury in the SF-treated group was reduced when it was compared with non SF-treated group. These results suggest that SF may have liver protective effects against alcohol-induced hepatotoxicity.

장기간의 Pyridoxine 부족이 새끼쥐 뇌의 지방조성에 미치는 영향 (Effect of Long-Term Pyridoxine Depletion on Lipid Composition in the Developing Rat Brain)

  • 이유한 Sprague Dawley 암컷 쥐에게 성장, 임신, 수유기간 동안 Pyrridoxine이 충분한 식이와 Pyridoxine 이 부족된 식이를 주어 사육했으며, 또는 다른 군은 성장, 임신기간은 pyridoxine이 부족된 식이를 공급받아가 생후 5, 10, 21일에 각각 pyridoxine 이 풍부한 식이로 바꾸어 주어회복정도를 살펴보았다. 출생후, 5, 10, 15, 21,35, 50 일에 체중을 재고 새끼를 희생시켜 뇌와 적혈구를 실험에 사용하였다. 적혈구에서는 alanine aminotransferase 활성을 측정하였고, 뇌에서는 cholesterol, proteolipid protein, cerebroside를 측정하였다. Pyridoxine이 부족된 쥐는 실험기간동안 유의적으로 체중과 뇌무게가 감소했으며, erythrocyte alanine aminotransferase 활성의 증가정도가 훨씬 높은 것으로 보아 체내 비타민 B6 의 영양상태가 나쁨을 알 수 있었다. 할편, 뇌의 ceregroside의 gakfid은 유의적인 차리를 보였으며 5, 10, 21일에 supplementation 시킨 쥐의 이유후의 cerbroside 의 생성속도의 증가로 보아 supplementation 시킨 쥐의 myelinatino 이 저해되었음을 알수 있다. 한편, cholesterol, proteolipid protein 도 부족식이에 의해 영향을 받았는데 생후 5 일에 supplementation 시킨 쥐는 정상수준으로 회복되나 10 일 이후에 supplementation 시켰을대는 정상수준에 미치지 못하였다.

The efficacy of aspartate aminotransferase-to-platelet ratio index for assessing hepatic fibrosis in childhood nonalcoholic steatohepatitis for medical practice

  • Purpose: Childhood obesity is associated with nonalcoholic fatty liver disease (NAFLD), and it has become one of the most common causes of childhood chronic liver diseases which significant as a cause of liver related mortality and morbidity in children in the United States. The development of simpler and easier clinical indices for medical practice is needed to identify advanced hepatic fibrosis in childhood NAFLD instead of invasive method like liver biopsy. FibroScan and aspartate aminotransferase (AST)-to-platelet ratio index (APRI) have been proposed as a simple and noninvasive predictor to evaluate hepatic fibrosis in several liver diseases. APRI could be a good alternative to detect pathologic change in childhood NAFLD. The purpose of this study is to validate the efficacy of APRI for assessing hepatic fibrosis in childhood NAFLD based on FibroScan. Methods: This study included 23 children with NAFLD who underwent FibroScan. Clinical, laboratory and radiological evaluation including APRI was performed. To confirm the result of this study, 6 patients received liver biopsy. Results: Factors associated with hepatic fibrosis (stiffness measurement >5.9 kPa Fibroscan) were triglyceride, AST, alanine aminotransferase, platelet count, APRI and collagen IV. In multivariate analysis, APRI were correlated with hepatic fibrosis (>5.9 kPa). In receiver operating characteristics curve, APRI of meaningful fibrosis (cutoff value, 0.4669; area under the receiver operating characteristics, 0.875) presented sensitivity of 94%, specificity of 66%, positive predictive value of 94%, and negative predictive value of 64%. Conclusion: APRI might be a noninvasive, simple, and readily available method for medical practice to predict hepatic fibrosis of childhood NAFLD.

The Relationships between Respiratory Virus Infection and Aminotransferase in Children

  • Purpose: We sought to examine the relationship between the clinical manifestations of nonspecific reactive hepatitis and respiratory virus infection in pediatric patients. Methods: Patients admitted to the pediatric unit of Konyang University Hospital for lower respiratory tract disease between January 1, 2014 and December 31, 2014 and who underwent reverse transcriptase polymerase chain reaction tests were examined. The patients were divided into those with increased levels of alanine aminotransferase (ALT) or aspartate aminotransferase (AST) and those with normal ALT or AST levels. Further, patients with increased ALT and AST levels were individually compared with patients in the normal group, and the blood test results were compared according to the type of respiratory virus. Results: Patients with increased ALT or AST levels had one more day of hospital stay, on average, compared with patients in the normal group ($5.3{\pm}3.1$ days vs. $4.4{\pm}3.0$ days, p=0.019). Patients in the increased ALT level group were younger and had a longer mean hospital stay, compared with patients in the normal group (p=0.022 and 0.003, respectively). The incidences of increased ALT or AST were the highest in adenovirus infections (6/24, 25.0%), followed by enterovirus (2/11, 18.2%) and respiratory syncytial virus A (21/131, 16.0%) infections. Conclusion: Nonspecific reactive hepatitis is more common among patients with adenovirus, enterovirus and respiratory syncytial virus infection, as well as among those infected at a younger age. Compared with AST levels, ALT levels are better indicators of the severity of nonspecific reactive hepatitis.
