Chemical compositions and antioxidant activities of Cheonnyuncho (Opuntia humifusa) stems and fruit (천년초의 성분특성과 항산화 활성)

  • The purpose of this study was to investigate the chemical composition of the freeze-dried stems and fruit of the cactus Cheonnyuncho. The analysis showed that powdered stems have the highest fat content (1.91%) and the powdered fruits have he highest protein content (2.62%). The K content of the fruits higher than that of the stems, while the Ca, Mg, Na and P contents of the stems were higher than those of the fruits. Both the stems and fruits powders contained high levels of the amino acids glutamic acid and aspartic acid. The free sugars such as sucrose, fructose, and glucose were detected in both the stems and fruits. The 75% ethanol (EtOH) extract showed a relatively high antioxidative activity compared to those of the water and 75% methanol (MeOH) extracts. Furthermore, the 75% EtOH extract of the stem powder exhibited a total polyphenol content of 3.60 g/100 g, and a total flavonoid content of 2.00 g/100 g. The antioxidant activities of the stem and fruit powder extracts, measured in DPPH radical scavenging experiments, were higher than that of the control group.

Changes of Constituent Components in Chestnut during Storage (밤의 저장 중 성분변화)

  • Studies were carried out to investigate the changes of constituent components in chestnuts (Castanea crenata Sieb. et Zucc) during storage at $20^{\circ}C$ for 9 weeks and $1^{\circ}C$ for 15 weeks. Ascorbic acid content of chestnut was 4.08 mg% in oxidized form and 17.7 mg% in reduced form which was 81.3% of total ascorbic acid. The reduced form gradually decreased during storage, while the oxidized forms increased during 5 weeks at $20^{\circ}C$ and 7 weeks at $1^{\circ}C$. The major organic acids in chestnut were malic acid, citric acid and quinic acid. Citric acid significantly decreased during storage, while malic acid increased during storage at $20^{\circ}C$ and decreased at $1^{\circ}C$. Free sugars in chestnuts were identified as glucose, fructose, sucrose and maltose. Glucose and fructose decreased after 5 weeks storage at $20^{\circ}C$ followed by an increase thereafter. Sucrose and maltose also increased. The major free amino acids in the chestnut were glutamic acid, aspartic acid, arginine, alanine, proline and serine. Glutamic acid, arginine, alanine, threonine, phenylalanine, valine and lysine increased during storage at $20^{\circ}C$, while proline, serine, isoleucine. leucine and ${\gamma}-aminobutyric$ acid decreased.

Chemical Characteristics of Citron(Citrus junos) Juices (유자 착즙액의 화학적 특성)

  • It was aimed to study the chemical characteristics and volatile compounds of juices extracted by press (PE) and centrifugal extractors (CE). Citrons cut up cross direction were used as samples in PE. Proportion ratio of peel, flesh, and seed of citron were 43.9, 39.4 and 16.7%, respectively. Yield of juice increased with weight to 120g of citron fruit. pH, brix, pulp, oily materials, amino nitrogen and yield of juice extracted by CE were higher than those of juice by PE. Acidity and transmittance of juice extracted by PE were higher than those of juice by CE. Free sugars of citron juice were consisted of fructose, glucose and sucrose. Free sugar contents of juice by CE were twice as much as those of PE. Major volatile compounds of total steam distillation matters of citron juice extracted by PE were dl-limonene, ${\gamma}-terpinene\;{\beta}-farnesene$, sabinene, linalool, ${\beta}-myrcene$ and terpinolene, while those by CE were dl-limonene, ${\gamma}-terpinene$, sabinene, ${\beta}-myrcene,\;{\alpha}-pinene$, linalool and terpinolene. dl-Limonene and ${\gamma}-terpinene$ consisted of $73{\sim}83%$ of total volatile compounds. dl-Limonene of juice by CE increased about 6% that of juice by PE. ${\beta}-Farnesene$ were observed trace in juice by CE but presented in 7% in juice by PE. The results would suggested that chemical characteristics and volatile compounds of citron juice seemed to depend on extraction methods.

  • Physicochemical components of dried green algae, Seaweed fulvescens (Capsosiphon fulvescens) with $96.02{\pm}0.02%$ moisture content, were investigated. Crude protein, crude lipid, ash and carbohydrate content of C. fulvescens were $31.76{\pm}0.26,\;1.01{\pm}0.21,\;13.58{\pm}0.62\;and\;53.65{\pm}0.73$, respectively. Predominant minerals were Na, Mg, Ca, K and P. Fe $(122.83{\pm}11.19mg/%)\;and\;Se\;(0.90{\pm}0.07mg%)$ contents in were higher than those of other seaweeds. Major free sugars were identified as fructose, glucose, sucrose, maltose and galactose. Organic acid including L-malic acid, citric acid, tartaric acid and d-malic acid were detected. Most abundant amino acids were glutamic acid, aspartic acid and alanine. Main fatty acids were palmitic acid, linolenic acid and stearidonic acid, with 58.37% polyenes.

  • Chemical composition and antimicrobial activity of Houttuynia cordata Thunb. were investigated to develop a natural food preservative from it. Aspartic acid, glutamic acid, glycine and arginine were major amino acids of Houttuynia cordata Thunb., but were present in a trace amount. Free sugars were composed of glucose, fructose, sucrose and maltose and major fatty acids were linolenic acid, linoleic acid, oleic acid, and palmitic acid. Mineral elements were potassium, calcium, magnesium, iron, zinc, and copper. Antimicrobial activities were shown in acidic, neutral and phenolic fraction of Houttuynia cordata Thunb., but not in basic fraction. Among the four fractions, neutral fraction showed the strongest antimicrobial activities against microorganisms tested, such as Pseudomnas aeruginosa, Micrococcus luteus, Bacillus subtilis, Aspergillus parasiticus and Aspergillus flavus. Minimum inhibitory concentration (MIC) and minimum lethal concentration (MLC) of the neutral fraction varied according to microoganisms tested. The lowest values of MIC (0.0075 g eq./mL) and MLC (0.10 g eq./mL) were obtained from Pseudomonas aeruginosa.

Prevention of Swelling and Quality Improvement of Sunchang Traditional Kochujang by Natural Additives (천연첨가물을 이용한 전통고추장의 유통중 팽창억제 및 품질개선)

  • To suppress the gas forming caused by some yeast in the pack of traditional kochujang prepared at Sunchang area, which is the most severe problem during distribution of commercial products, mustard or horseradish powder as natural preservatives was mixed to the ingredients of kochujang and left for fermentation in clay pot as commercial scale for 180 day. The composition changes including quality and gas formaing in the kochujang containing those additives were monitored physicochemically and organoleptically. The fermented kochujang containing those additives were monitored physicohemically and organoleptically. The fermented kochujang containing 0.6% of horseradish showed lower organic acid content than that of the control but amino type nitrogen content, which is one of the important quality reference of kochujang, was higher in the one containing 0.6% natural preservatives. No color changed in the kochujang containing mustard but L and a value were increased in the kochujang containing horseradish. No difference of free reducing sugars was showed by the addition of those natural preservatives and the addition of 0.6% horseradish to kochujang had completely stopped gas forming during fermentation. The overall quality of the kochujang containing 0.6% horseradish was superior than that of the other treatments.

  • 미역의 뿌리부분인 미역귀에는 알긴산과 후코이단 성분이 풍부하고 고혈압과 당뇨등과 같은 성인병을 예방하는데 도움이 된다. 후코이단은 미역과 다시마 같은 갈조류에서 추출된 천연물질로 특히 면역력을 활성화하고 항암작용으로 잘 알려져 있다. 알긴산 등 유용성분이 함유된 것으로 알려진 미역귀의 유용성분을 분석하여, 향후 미역귀를 활용한 제품개발의 기초자료를 제공하고자 본 연구를 수행하였다. 본 연구에 사용된 미역귀는 2015년 12월 완도 금일수산에서 제공받아 연구재료로 활용하였다. AOAC방법에 따라 분석한 미역귀의 일반성분은 수분이 대부분을 차지하였으며, 조섬유와 가용성 무질소물 함량이 높게 나타났다. 이는 시료로 사용한 미역귀가 수확 후 바로 냉동보관 되어서 수분함량이 높은 것으로 보여진다. 유리당은 미역귀 1g에 증류수를 가하여 $60^{\circ}C$에서 4시간 추출 한 후, 원심분리(3,000 rpm, 30 min)하여 $0.45{\mu}m$ membrane filter (Millipore Co., USA)로 여과한 여액을 HPLC (High Performance Liquid Chromatography)를 이용하여 분석한 결과, 미역귀의 유리당은 Galactose 1종만이 검출되었다. 미역귀의 구성아미노산과 유리아미노산은 $0.45{\mu}m$ membrane filter로 여과하여 얻은 여액을 일정량 취하여 AccQ-Tag 시약을 사용하여 유도체화 시킨 후 HPLC로 분석하였다. 미역귀의 구성아미노산은 glutamic acid, aspartic acid, alanine, lysine, threonine, valine, arginine 순으로 높은 함량을 나타내었으며, 유리아미노산은 glutamic acid, alanine, threonine, arginine, lysine 순으로 높은 함량을 나타내었다. 미역귀의 유리아미노산함량은 표고에 비하여 현저히 낮은 함량이 나타났으며, 주요아미노산은 해조류의 풍미를 대표하는 아미노산인 glutamic acid와 alanine으로 나타났다.

Isolation and Culture of Protoplasts from Embryonic Cotyledons of Pinus densiflora S. et Z. (소나무 자엽(子葉)을 이용(利用)한 원형질체(原形質體) 나출(裸出) 및 배양(培養))

  • Protoplasts were isolated from cotyledons of germinating Pinus densiflora S. et Z. seeds. The seeds were germinated for nine days, and excised embryonic cotyledons about 3-4mm long were incubated with Cellulase Onozuka R-10 and Macerozyme. After 8 hours of incubation, large number of viable protoplasts were isolated. Isolated protoplasts were cultured in a medium containing basal salts of $B_5$ medium, vitamines, amino acids, organic acid, sugars, and growth hormones. The first evidence of protoplast budding was observed after twelve hours in culture, and it suggested that high potential of the embryonic cotyledons for rapid cell division affected the early budding, rather than effect of culture medium was shown in twelve hours. The three- to four-cell stage was reached after three to four days of culture. Most cell divisions were achieved by additional buddings rather than equal binary cell division. No further cell division was observed beyond the four-cell stage. Protoplasts isolated from fully expanded cotyledons (germinated for 17 to 24 days) seldom initiated or failed to initiate cell division.

Physicochemical Characteristics of Cold-Air Dried Persimmons and Traditional Dried Persimmons (냉풍건조 곶감과 천일건조 곶감의 이화학적 품질특성)

  • This study was performed to investigate physicochemical properties of cold-air dried persimmon and traditional dried persimmons. Moisture contents and water activity were lower at cold-air dried persimmon than traditional persimmons. Crude protein and crude fat were not significant differences, Crude ash and N-free extracts of cold-air dried persimmon were, relatively, high as comparing to the traditional dried persimmons. Brix in cold-air dried persimmon was the highest. In the hunter's color values, L, a and b values of cold-air dried persimmon were lowed than traditional dried persimmons. There was not significant differences at the texture of semi-dried persimmons, but cold -air dried persimmon were relatively high than the traditional dried persimmons in hardness, gumminess, chewiness. Free sugars contents were only glucose and fructose in an sample. Free amino acids contents were seventeen sorts, glutamic acid and alanine were high in order of contents.

Quality Characteristics of Various Beans in Distribution (시중에 유통되는 콩의 종류에 따른 품질 특성)

  • The goal of this study was to evaluate the quality characteristics of various beans in distribution. The quality characteristics investigated were proximate composition, color, free sugars, organic acids, amino acids, and minerals. Bean samples analyzed were white soybeans (Glycine max. (L.) Merrill), kidney beans (Phaseolus vulgaris var. humilis), black soybeans (Glycine max (L.) Merr.), black small soybeans (Rhynchosia nulubilis), sword beans (Canavalia gladiata), and green beans (Phaseolus vulgaris). The highest contents of crude fat and crude protein were 17.60${\pm}$0.14% for white soybeans, and 42.38${\pm}$0.15% for black soybeans, respectively. Higher color values compared to the other samples were $L^*$ (64.07${\pm}$0.97) for sword beans, $a^*$ (15.64${\pm}$0.48) for kidney beans, and $b^*$ (22.92${\pm}$0.09) for white soybeans. The highest contents of sucrose, oxalic acid, and malic acid in black small soybeans were 54.23 mg/g, 23.26 mg/100 g and 18.24 mg/100 g, respectively. Xylose, galactose, lactose, malonic acid, succinic acid, and lactic acid were not detected in the soybeans studied, whereas the glutamic acid content of soybeans ranged from 2.68 to 6.18 g/100 g. Levels of K and Mg contents in soybean were higher than those of the other minerals.
