개불(Urechis unicinctus) 당단백질의 성분조성과 특성 (Chemical Composition of Glycoprotein from Urechis unicinctus)

  • To confirm the food quality of Urechis unicinctus which have been favored as a special raw seafood in southern area in Korea, the chemical composition of Urechis unicinctus and those glycoprotein were studied. Freeze dried Urechis unicinctus was composed of more than 70% of crude protein and 15% of total carbohydrate. The amino acids such as glycine(18.69%), glutamic acid(12.50%) and aspartic acid(9.37%) were noted as major components of total amino acids. The predominant free amino acids were alanine(32.98%), glycine(27.50%), asparagine(19.65%) and taurine(8.29%), and the sum of them were more than 88% to total free amino acids, so they may cause unique taste of Urechis unicinctus with sweetness. As the basis of sugar composition analysis, 56.35% of glucose and 30.49% of N acetylglucosamine were contained respectively, and they might also play an important role in a sweet taste. The leading carbohydrate moiety of glycoprotein from Urechis unicinctus was identified as glucose and N acetylglucosamine similarly to raw muscle, and they occupied more than 50% of total sugar content. Fucose(18.32%) and D glucuronic acid(12.31%) also detected in higher levels com pared to raw muscle. The total amino acid profile of glycoprotein showed a similar trend to raw muscle protein but glycine was detected a significantly lower than that in raw muscle. The glycoprotein from Urechis unicinctus was composed of 4 kinds of subunits, i.e. 25kDa, 20kDa, 18kDa and 12.5kDa of molecular weights through the SDS polyacrylamide gel electrophoresis. On the basis of the IR spectrum of absorptions appeared in 1,240cm-1 and 850cm-1, the glycoprotein had sulfate groups.

Chemical and Textural Properties in Commercial Fermented Soybean Curds of Sufu

  • A survey aiming to find out the chemical and textural properties of commercial fermented soy bean curd called sufu was conducted. Sixteen brands of plain sufu produced in the Northern or the Southern part of China were collected and examined for their crude protein, crude fat, texture profiles, free amino acids, and free fatty acid contents. Twenty-one free amino acids were extracted and derivatized using a commercial kit followed by separation and analyzed by the gas chromatography-mass spectrometry (GC-MS). Similarly, ten free fatty acids were extracted using alumina, eluted, separated and analyzed. The content ranges of crude fat and protein were 22~36% and 31~38%, respectively. In texture profile analysis, ranges of the texture parameters were 131~493 g (hardness), 0.4~0.5 (cohesiveness), -137 to -50 gs (adhesiveness), 0.6~1 (springiness), 47~220 g (gumminess) and 32~177 g (chewiness). Twenty-one different free amino acids, especially alanine, glycine, $\alpha$-aminobutyric acid, valine, leucine, allo-isoleucine, aspartic acid, glutamic acid and lysine in large amount, as well as ten fatty acids in total, notably linoleic acid (9-octadecanoic acid), oleic acid (9,12-octadecadienoic acid), linolenic acid (9,12,15-octadecadienoic acid), hexadecanoic acid and octadecanoic acid were found. This information provides important quality reference ranges for product developers and manufacturers to optimize and produce the plain sufu.

Physicochemical attributes, oxidative stability, and microbial profile of boneless sirloin and bone-in T-bone steaks from Hanwoo steer with reference to dry-aging

  • We investigated the comparative physicochemical attributes, oxidative stability, and microbial characteristics of 28 days dry-aged meat in between boneless sirloin (gluteus medius) and bone-in T-bone steaks (infraspinatus) muscles from Korean Native Hanwoo Steer (KNHS). Results reveal that regardless of the muscles, dry-aging increased protein content and water-holding capacity (WHC) (p < 0.05). Meat from infraspinatus-aged muscle led to darker meat with higher pH values than un-aged meat (p < 0.05). However, fat content, CIE a*, and CIE b* remained unchanged in both muscles at aging. At aged meat, thiobarbituric acid reactive substances (TBARS) values from bone-in infraspinatus muscle was 2.5-fold higher than boneless gluteus medius muscle (p < 0.05). Dry-aging led to an increase in the contents of total unsaturated fatty acids (UFAs), monounsaturated fatty acids (MUFAs), and UFA/saturated fatty acids (SFA) in both muscles (p < 0.05). Furthermore, gluteus medius aged muscle concentrated with olic acid (C18:1) compared to infraspinatus aged muscle. Irrespective of the muscles, dry-aging enhanced the total free amino acids (FAAs) as well as tasty, and bitter amino acid contents whereas decreased the tasty/bitter amino acids (p < 0.05). Aromatic amino acid, tryptophan that converted to serotonin was 2-fold higher in boneless gluteus medius muscle than bone-in infraspinatus muscle at pre and post aging processes (p < 0.05). Aged Infraspinatus muscle increased total bacteria (p < 0.05) while no salmonella spp. was detected in both muscles. Taken together, our study confirms that 28 days dry-aging profiling the quality characteristics of boneless sirloin (gluteus medius) and bone-in T-bone steaks (infraspinatus) distinctly while gluteus medius aged steak performs better owing to oxidative stability and functional compounds than infraspinatus aged steak.

천연 및 양식산 넙치의 함질소엑스분과 아미노산조성 (Comparison of Nitrogenous Extractives, Amino Acids in Wild and Cultured Bastard)

  • 천연산 및 양식산 어류의 식품성분에 관한 일련의 연구로서, 횟감으로 즐겨 먹는 고급어종인 넙치를 천연산과 양식산으로 구분하고 아울러 유사어종인 도다리의 함질소엑스분 및 아미노산 조성을 분석, 비교하였다. 양식산 넙치는 천연산에 비해 수분함량이 다소 않은 반면 조단백질, 조지방 함량은 약간 적었으나 대제로 성분조성이 비슷하였다. 총 유리아미노산 함량은 천연산 넙치가 305.03mg/100g, 양식 산이 253.42mg/100g, 도다리가 340.10mg/100g이었고, taurine의 함량이 월등히 많고 다음이 alanine, glycine, lysine순이었다. 유리아미노산 조성은 모두 유사하였다. 핵산관련물질 중 IMP 함량은 천연산 넙치가 408.31mg/100g, 양식산이 356.26mg/100g으로 도다리의 178.61mg/100g에 비해 월등히 높았고. TMAO, total creatinine 역시 천연산이 양식산에 비해 함량이 많았다. 시료어의 주요 구성아미노산은 glutamic acid, aspartic acid, leucine, isoleucine, valine 등으로 서로 비슷하였고 전체함량은 천연산 넙치가 21.94g/100g, 양식산이 20.12g/100g, 도다리가 17.63g/100g이었다. 사료의 아미노산 조성과 양식어의 조성 사이에 뚜렷한 상관관계가 없었다.

마늘의 alcohol 침지 중 휘발성 향기성분과 침출유리아미노산 함량 (Elution Profiles of Volatile Compounds and Free Amino Acids during Alcohol Soaking of Garlic(Allum sativum L.))

  • 마늘주의 산업화에 앞서 기초 자료로써 활용하고자, 마늘을 소주 등의 술에 침지하였을 때 마늘의 휘발성 기능성분과 유리아미노산이 술로 침출되는 정도 및 형태에 관하여 조사하고자, 생마늘을 20% 주정에 5주간 침지하고 매주 휘발성 향기성분과 유리아미노산들이 침출되는 양적 및 형태적 변화를 조사하였다. 마늘의 주요 유리 아미노산은 L-aspartic acid, L-glutamic acid, L-arginine, L-alanine, L- proline, L-asparagine 및 L-serine이었으며, 유리 아미노산들 모두 침지 기간의경과에 따라 침출량이 증가하였다. 또한, L-threonine, L-proline, L-valine 및 L-leucine 등의 중성아미노산들과 방향족아미노산인 tyrosine과 phenylalanine은 3주 후부터 80% 이상 침출된 반면에, 산성, 염기성 및 함황아미노산들은 5주 후에도 80% 미만이었다. 생마늘의 휘발성 향기성분은 diallyl trisulfide, diallyl disulfide, methyl allyl disulfide, 2-vinyl-4H-1,3-dithiin, 3-vinyl-3,4-dihydro-1,2-dithiin, 3,5-diethyl-1,24-trithiolane, isobutyl isothiocyanate 및 diallyl sulfide 등의 황화합물들이 주요 성분들이었다. 생마늘로부터 주정으로 침출된 휘발성 향기성분은 allyl alcohol, diallyl disulfide, 3,5-diethyl-1,2,4-trithiolane, diallyl trisulfide, 3,4-dimethoxyfuran의 5종이었으며, 침지기간의 경과에 따라 3,5-diethyl-1,2,4-trithiolane는 계속 침출량이 증가하였지만 그 외 4종은 감소하였다. 침 지기간 중 furfural, 5-methylfurfural, 5-hydroxymethylfurfural, dimethyl pyrazine, furfuryl alcohol, 3-hydroxy-2-bytanone과 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one이 새로이 생성되었으며, 침지기간의 경과에 따라 침출주에 대부분 축적되는 양상을 보였다.

Correlation analysis of muscle amino acid deposition and gut microbiota profile of broilers reared at different ambient temperatures

  • Objective: Temperature could influence protein and amino acid deposition as well as gut microbiota profile and composition. However, the specific effects of ambient temperature on amino acids deposition and gut microbiota composition remain insufficiently understood. Methods: A total of 300 one-day-old Avian broilers were randomly divided into three groups and reared at high, medium, and low temperature (HT, MT, and LT), respectively. Breast muscle and fecal samples were collected for amino acid composition analysis and 16S rRNA gene sequence analysis. Results: Our data showed that compared to the MT group, there was a decrease of muscle leucine and tyrosine (p<0.05), as well as an increase of methionine in the HT group (p<0.05) and a decrease of serine in the LT group. Examination of microbiota shift revealed that at genus level, the relative abundance of Turicibacter and Parabacteroides was increased in the HT group (p<0.05) and that the relative abundances of Pandoraea, Achromobacter, Prevotella, Brevundimonas, and Stenotrophomonas in the LT group were higher than those in the MT group (p<0.05). In addition, there were substantial correlations between microbes and amino acids. In the HT group. Turicibacter was negatively correlated with aspartic acid and tyrosine, whereas Parabacteroides was positively correlated with methionine (p<0.05). In the LT group, there were multiple positive correlations between Achromobacter and arginine, isoleucine or tyrosine; between Prevotella and cysteine or phenylalanine; between Brevundimonas and cysteine; and between Stenotrophomonas and cysteine as well as a negative correlation between Stenotrophomonas and serine. Conclusion: Our findings demonstrated that amino acid content of breast muscle and intestinal microbiota profile was affected by different ambient temperatures. Under heat exposure, augmented abundance of Parabacteroides was correlated with elevated methionine. Low temperature treatment may affect muscle tyrosine content through the regulation of Achromobacter.

  • This experiment was planned to develop a functional supplements by food resources to prevent and lessen the bad effects caused by the environmental pollution such as bad food, air, water and heavy metals exposed to people these days. As for Its primary stage, the nutrient profile and sensory characteristics of the duck-extract were evaluated in this study. The duck-extract was formulated by the mixture of bone and internal organs of ducks and 6 medicinal herbs which were able to be used as foods and known to help the excretion and the repression of the poison inside the body for a long time. As we compared the six medicinal herbs mentioned above to the vegetable herbs, the nutrient profile of the medicinal herbs were superior to the vegetable herbs. The duck-extract was composed of protein 49.92%, carbohydrate 37.02%, dietary fiber 20.99%, lipid 7.60% and ash 5.46%. The ratio of Ca to P was 1 : 1.4, which was a suitable ratio for absorption, the contents of Na and K were low and those of micro-elements such as Fe, Mn, Zn and Cu high compared to other meat extracts. The essential amino acids accounted for 30.91% of total amino acids. The result of sensory evaluation was better in overall preferences than the ones which are already in the market. From these results, the nutrient profile of the duck-extract was estimated to be able to supply enough nutrients to the people whose nutritional balance was destroyed these days. This study also showed the effective way of using duck-extract and its application to the oriental medicine.

고-Lysine 보리 돌연변이 계통 M98 종실의 아미노산 조성 및 Proteome Profile 특성 (Characterization of Grain Amino Acid Composition and Proteome Profile of a High-lysine Barley Mutant Line M98)

  • 본 연구는 찰쌀보리와 M98의 종실에 함유된 아미노산 조성 및 단백질 profile의 특성 차이를 분석하여 M98의 고-Lysine 및 함몰배유 형질에 관련된 생화학적 정보를 얻고자 수행하였으며, 그 결과를 요약하면 다음과 같다. 1. 찰쌀보리와 M98 종실의 총 16개 아미노산 조성을 분석한 결과, lysine을 비롯하여 총 6개 아미노산의 조성비가 M98에서 찰쌀보리에 비해 1.2~1.8배 높았다. Proline과 그것의 전구물질인 glutamic acid의 조성비는 M98에서 찰쌀보리에 비해 절반수준으로 낮았는데, glutamic acid를 전구물질로 하여 합성되는 arginine의 조성비는 M98에서 찰쌀보리보다 1.8배 높았다. 2. 찰쌀보리와 M98간의 종실 단백질을 이차원전기영동으로 분리한 결과, 총 70개 단백질 spot에서 발현양상의 차이가 확인되었다. 찰쌀보리에 비해 M98에서 발현양이 높은 단백질 spot은 44개였으며, 발현양이 낮은 단백질 spot은 18개였다. 나머지 8개 단백질 spot에서 7개의 발현은 찰쌀보리에서만 확인되었고, 1개의 발현은 M98에서만 확인되었다. 3. 찰쌀보리와 M98의 종실에서 발현양상 차이를 나타낸 총 53개의 단백질 spot을 선택하여 nESI-LC-MS/MS 방식으로 동정하였으며, 대부분의 단백질 spot은 종실의 다양한 생물학적 과정에 관여하는 단백질이었다. 특히, 28개 단백질 spot은 M98의 함몰배유 및 고-Lysine 형질과 연관된 단백질이었다. 4. 본 연구에서 밝혀진 M98 종실의 아미노산 조성 및 단백질 발현특성 정보는 향후 M98 유래 계통을 이용한 유전학적 검토 등을 통하여 고-Lysine 형질 연관 바이오마커 및 보리 신품종 개발에 유용한 정보로 활용될 수 있을 것으로 판단되었다.

Isolation and characterization of LHT-type plant amino acid transporter gene from Panax ginseng Meyer

  • A lysine histidine transporter (LHT) cDNA was isolated and characterized from the roots of Panax ginseng, designated PgLHT. The cDNA is 1,865 bp with an open reading frame that codes for a protein with 449 amino acids and a calculated molecular mass of 50.6 kDa with a predicted isoelectric point of 8.87. Hydropathy analysis shows that PgLHT is an integral membrane protein with 9 putative membrane-spanning domains. Multiple sequence alignments show that PgLHT shares a high homology with other plant LHTs. The expression profile of the gene was investigated by real-time quantitative polymerase chain reaction during various chemical treatments. PgLHT was up-regulated in the presence of abscisic acid, salicylic acid, methyl jasmonate, NaCl, and amino acids. To further explore the function of PgLHT gene, full-length cDNA of PgLHT was introduced into P. ginseng by Agrobacterium rhizogenes A4. The overexpression of PgLHT in the hairy roots led to an obviously increase of biomass compared to the controls, and after addition of the amino acids, the overexpressed-PgLHT hairy roots grew more rapidly than untreated controls during early stage of the culture cycle. The results suggested that the PgLHT isolated from ginseng might have role in the environmental stresses and growth response.

채취시기별 방사무늬김(Porphyra yezoensis)의 식이섬유, 아미노산 및 지방산 함량 변화 (Seasonal Variation in the Dietary Fiber, Amino Acid and Fatty Acid Contents of Porphyra yezoensis)

  • Porphyra yezoensis is potentially an excellent source of dietary fiber, amino acids, and eicosapentaenoic acid (EPA) because this red seaweed is available in large quantities and is rich in polysaccharides, proteins, and n-3 fatty acids. This study determined the insoluble dietary fiber (IDF), soluble dietary fiber (SDF), neutral detergent fiber (NDF), acid detergent fiber (ADF), amino acid, and fatty acid contents of P. yezoensis harvested monthly from November 2011 to March 2012. The total dietary fiber (TDF) and IDF contents ranged from 27.2-34.9% and 18.5-26.9%, respectively, and were greater in March than November. The SDF content ranged from 4.9-8.4% and did not differ significantly during growth. Galactose and 3,6-anhydro galactose were the major sugars in IDF and SDF. The higher levels of galactose and 3,6-anhydro galactose in IDF might be due to associated porphyran-type polysaccharides. Mannose and xylose were also major sugars in IDF. The total amino acid contents decreased gradually from November to March. The total amino acid composition of Porphyra was dominated by alanine, glutamic acid, arginine, and aspartic acid. No significant changes in the fatty acid profile were observed throughout the study period. The dominant fatty acid during all seasons was EPA, which comprised as much as 50% of the total fatty acid content.
