Serotype and antimicrobial susceptibility of Actinobacillus pleuropneumoniae isolates from pigs in Korea (돼지에서 분리한 Actinobacillus pleuropneumoniae의 혈청형 분포 및 항생제 감수성)

  • Actinobacillus (A.) pleuropneumoniae is the causative agent of pleuropneumonia which is one of the most important respiratory diseases in pigs worldwide. A total of 32 A. pleuropneumoniae isolates from diseased pigs during 2008 to 2010 were serotyped by polymerase chain reaction method. The susceptibility of the isolates to 13 antimicrobial agents were determined by disk diffusion test. In all the 32 isolates examined in this study, serotype 5 (16 isolates: 50%), 1 (7 isolates: 21.9%), 2 (5 isolates: 15.6%) and 12 (1 isolate: 3.1%) were found. Of all tested antimicrobial agents, resistance to oxytetracycline was found in 96.9% of isolates, followed by resistance to amikacin (81.2%), neomycin (68.7%), kanamycin (53.1%), penicillin (50.0%), gentamicin (43.7%), florfenicol (25.0%), ampicillin (18.7%), colistin (9.4%), trimethoprim/sulfamethoxazole, ceftiofur (8.3%), amoxicillin/clavulanic acid (3.1%) and enrofloxacin (0%). Oxytetracycline or florfenicol-resistant isolates were examined for the presence of resistance gene. Among the 31 oxytetracycline-resistant isolates, tetB, tetH and tetO genes were detected in 22 (71%), 8 (26%) and 1 (3%) isolates, respectively. The floR genes were detected in 8 (100%) of the 8 florfenicol-resistant A. pleuropneumoniae isolates.

  • A 50-kb chromosome DNA region was isolated from Streptomyces kanamyceticus by screening the fosmid genomic library, using the 16S rRNA methylase gene (kmr) as a probe. Sequence analysis of this region revealed 42 putative open reading frames (ORFs), which included biosynthetic genes such as genes responsible for 2-deoxystreptamine (2­DOS) biosynthesis as well as genes for resistance and regulatory function. Also, the kanamycin acetyltransferase gene (kac) was characterized by in vitro enzyme assay, which conferred E. coli BL21 (DE3) with 10, 50, and 80-times higher resistance to kanamycin A, tobramycin, and amikacin, respectively, than the control strain had, thus strongly indicating that the isolated gene cluster is very likely involved in kanamycin biosynthesis. This work provides a solid basis for further elucidation of the kanamycin biosynthesis pathway as well as the productivity improvement and construction of new hybrid antibiotics.

Antimicrobial Drug Resistance in Shigella cultures Isolated in Korea (1983) (이질균속의 항균제 내성)

  • One hundred and forty strains of Shigella cultures isolated from the twelve hygiene laboratories of cities and provincial level and general hospital laboratories in 1983, and were tested for their resistance to 13 antimicrobial drugs and their R-Plasmid transfer. One hundred and forty (100%) of isolates were susceptible to amikacin, gentamicin, tobramycin, a total of 94.3% of all shigella isolates were resistant to 1 or more of the 13 antimicrobial agents tested. The most commonly found resistance was to chloramphenicol (94%), followed by streptomycin (93%), tetracycline (92%), piperacillin (90%), ampicillin (83%), cefoperazone (42%), nalidixic acid (14%), cephalothin (17%), rifampicin (22%), and kanamycin (6%). Sixty percent of strains among 140 were resistant to ampicillin, chloramphenicol, streptomycin, tetracycline at same time. The transfer of drug resistance by conjugation was tested and 94 strains (94.3%) which were resistant to one or more drugs were found to transfer their drug resistance to E. coli.

Serological Distribution and Antibiotic Susceptibility of Salmonella sp. Isolated from Pusan Area in 2000 (2000년도 부산지역에서 분리된 살모넬라속균의 혈청학적 분포 및 항균제 감수성)

  • A total of 79 Salmonella spp. were isolated from Pusan area in 2000. The serotypes of 79 Salmonella isolates were classified as 42 strains of S. typhi(53.1%), 24 strains of S. enteritidis(30.4%), 9 strains of S. montevideo(11.4%), 2 strains of S. typhimurium(2.5%), 1 strain of S. infantis(1.3%) and 1 strain of S. indiana(1.3%) strains(16.5%) of Salmonella sp. were isolated at May July, respectively. The isolates of S. typhi were sensitive to most sntibiotics except streptomycin. All isolates of S. typhi were especially sensitive to tobramycin, gentamicin, colistin, kanamycin, samikacin, sulfamethozazole/ trimethoprim, cefriaxone, ceftazdime, cifrofloxacin, cefoxitin and cefotaxime. Isolates of S. enteritidis wer presented higher resistance than isolates of S. typhi. Twenty-four strains of S. enteritidis were sensitive to kanamycin, amikacin cifrofloxacin, cefoxitin and cefotaxime, however 13 strains(54.2%) of S. enteritidis were resistant to carbenicillin, ampicillin and ticarcillin. Nine strains of S. montevideo were sensitive to most antibiotics except carbenicillin and streptomycin. Each 1 stain of S. indiana and S. infantis was sensitive to most antibiotics used in this study except streptomycin. Three kinds of resistant pattern (CB, SM, TE, AM, TC). In the case of S. enteritidis isolates, 9 kinds resistant pattern were detected. Most frequent resistant pattern of S. enteritidis isolates was CB, AM, TC type(16.7%)

Species distribution and antimicrobial susceptibility of staphylococci isolated from canine clinical specimens (개의 임상재료로부터 분리된 포도상구균의 균종분포 및 항균제 감수성)

  • Specimens collected from various pyogenic lesions of dogs were culturally examined for staphylococci and all staphylococcal isolates obtained from the specimens were also tested for susceptibility to 14 antimicrobial agents. A total of 123 isolates of staphylococci were identified. Of these, 120 were Staphylococcus intermedius and 3 were S aureus. All isolates were susceptible to oxacillin, cefazolin, cephalothin and amikacin, whereas more than 85% of the isolates were resistant to ampicillin, penicillin G and tetracycline. S intermedius isolates could be divided into 8 different biotypes by biotyping with the most common type accounting for 66.7% of the isolates. One hundred and seventeen(97.5%) isolates could be also divided into 26 different antibiogram patterns. The predominant antibiogram type accounted for 34.2% of the isolates. Antibiogram typing was found to be effective in distinguishing epidemiologically related isolates of S intermedius.

Biotype, serotype and antibiotic susceptibility of Yersinia enterocolitica isolated from swine (돼지에서 분리한 Yersinia enterocolitica의 생물형, 혈청형 및 항균제 감수성)

  • A study on the isolation of Yersinia from the feces of healthy pigs and the biotype and serotype and susceptibility to 16 antimicrobials was carried. Out of 853 pigs, Yersiniae were isolated from 349 pigs(40.9%). Of 349 isolates, 289 isolates(82.8%) were Yersinia enterocolitica and 54(15.5%) were Y kristensenii, 3(0.9%) were Y pesudotuberculosis and the rest 3(0.9%) were Y prederiksenii. Out of 289 isolates of Y enterocolitica, the predominants biotype was 3B comprising of 165 isolates(57.1%) and followed by biotype 2, comprising of 49 isolates(17.0%), bioptype 3A, comprising of 41 isolates(14.2%) and biotype 4 comprising of 23 isolates(8.0%). And the predominant serotype was 0 : 3 comprising of 231 isolates(79.9%) and followed by serotype 0 : 9 comprising of 42 isolates(14.5%) and 0 : 21 comprising of 10 isolates(3.5%). Y. enterocolitica were resistant to cephalothin(99%), novobiocin(99%), erythromycine(83%), ampicillin(83%) and carbenicillin(81%) and susceptible to amikacin(100%), colistin(100%), gentamicin(100%), kanamycin(100%), polymyxin B(100%), tobramycin(100%), chloramphenicol(99%), nalidixic acid(99%), neomycin(99%), streptomycin(99%) and tetracycline(99%). Most strains of biotype 2/serotype 0 : 9 were susceptible to carbenicillin(100%) and ampicillin(61%) but the other biotype/serotypes were resistant to these antibiotic.

Antimicrobial Susceptibility Patterns of Staphylococcus intermedius Isolated from Skin Lesions of Thoroughbred Horse. (더러브렛 말의 피부병변에서 분리한 Staphylococcus intermedius의 약제 감수성 양상)

  • Staphylococcus intermedius (S. intermedius) has been rarely isolated from horse. In this study, we investigated the antimicrobial susceptibility patterns of S. intermedius isolated from skin lesions of Thoroughbred horse in Daegu Equestrian Association. The skin lesions were showed with dehairing and slight purulent inflammation. Bacteria were isolated from skin lesions and identified as S. intermedius by biochemical tests and MicroLog (BIOLOG, California, USA). Antimicrobial susceptibility test of S. intemedius isolates was performed with 33 antimicrobial agents (BBL, Maryland, USA) by using the agar disk diffusion method. It showed a high sensibility in the amikacin, amoxycillin/clavuramic acid, ampicillin, cefoxitin, cephalothin, chloramphenicol, ciprofloxacin, enrofloxacin, florfenicol, kanamycin, neomycin, nitrofurantoin, norfloxacin, ofloxacin, oxacillin, penicillin G, tetracycline and vancomycin. The horse was treated with penicillin, and cured completely after two weeks. The present results showed valuable information for treatment and prevention of skin disease in horse.

Antimicrobial Susceptibility of Coagulase-Negative Staphylococci Isolated from Bovine Mastitis Between 2003 and 2008 in Korea

  • A total of 1,444 coagulase-negative staphylococci (CNS) isolates from bovine mastitic milk samples collected during 2003-2008 in Korea were identified to the species level. Of 14 species identified, S. simulans, S. haemolyticus, and S. sciuri accounted for over 60% of the isolates. All the CNS isolates were tested for susceptibility to eight antimicrobials commonly used in dairy cattle. With a few exceptions, similar resistance patterns were observed among the CNS species: penicillin and ampicillin showed the lowest activity, whereas amikacin, cephalothin, and gentamicin were highly effective. About 39% (557/1,444) of the CNS isolates were pan-susceptible, whereas 12% (175/1,444) showed resistance to four or more antimicrobials tested.

Prevalence and factors associated with Blackleg outbreak in north western Gyeongbuk province (경북 문경지방에서 발생한 기종저의 역학적 특성)

  • Outbreaks from September 1999 to July 2000 of blackleg in 4 milking and fatting cattle farms in the vicinity of Munkyong(northwestern part of Gyeongbuk province) were investigated biochemical characteristics of isolates and were surveyed epidemiological works during the period of 2 months from August to September 2000. Clostridium chauvoei, the etiological agent of blackleg is a serious pathogen of cattle and sheep causing fever, depression and lameness with high mortality in growing phase of 6 to 18 months old mainly beef cattle. Biochemical and cultural properties of 4 isolated Cl chauvoei were identical to those of reported results and isolates were susceptible to ampicillin, bacitracin, chlorarnphenicol, cephalothin, ciprofloxacin, erythromycin, norfloxacin, penicillin, and tetracycline, while they were resistant to amikacin, colistin, gentamicin, kanamycin, neomycin, streptomycin and trimethoprim-sulfamethoxazole. In the results of epidemiological survey, outbreak farms that concentrated region of milking or fatting cattle in each sites were so close contact within a radius of about 1km that they are separated only by wire fence or shrub harrier and flies, rodents, wild cats and birds could have the chance to move around between those farms. The origin of the outbreak blackleg were uncertain, but the transmission between farms were mainly through the movement of farm personnel, vehicles which were struck the feces or manures from farm and other mechanical factors such as flies, rodents and birds furthermore occurred 4 farms were not vaccinated. In conclusion, a periodic disinfection, vaccination and quarantine prevention are ideal methods of damage reduction by blackleg.

An Investigation on Concentration of Airborne Microbes in a Hospital (병원내 공기중 미생물의 농도에 관한 조사연구)

  • A survey was conducted to measure concentration of airborne microbe in a hospital using RSC air sampler during October~November 1991.The result was as follows: 1) In an agar strip GK-A media for total counts of microbial particles. The highest count were 1384 CFU/m$^3$ in the main lobby, followed by 912 CFU/m$^3$, in the obstetric room, 688 CFU/m$^3$ in 1CU. By gram staining, the distribution for organisms in the air were shown 74.1% in gram possitive cocci followed by 16.8%, in gram possitive bacilli 6.7% in gram negative bacilli and 4.7% in yeast, but low organism was detected in recovery room with 194 CFU/m$^3$. 2) In agar strip S media for Staphylococci the count at the main lobby was detected in the recovery room with 92 CFU/m$^3$, Tests of coagulase were negative Staphylococci with 78%, and positive Staphylococci with 22%. The Staphylococci were highly resistance to penicillin, ampicillin and sensitive to amikacin, cefazolin, gentamycin and chloramphenicol. 3) In agar strip C media for coliform bacteria the colony counts at the main lobby was 139 CFU/m$^3$ and treatment room was 190 CFU/m$^3$, most frequently isolated microorganisms were non fermentative bacilli. 4) In agar strip HS media for yeast and molds. Most frequently colony counts 17~76 CFU/m$^3$, 0.5% lactophenol cotton blue stains were shown unidentified 77.2%, 8.1%, in Penicillium 8.1% in Aspergillus, and 3.8% in mucor.
