대금음자(對金飮子)가 흰쥐의 만성 알콜성 근위축에 미치는 영향 (The Effects of Daekumeumja on Alcohol-induced Muscle Atrophy in Rats)

  • Chronic alcoholic myopathy is one of the most common skeletal muscle disorders. It is characterized by a reduction in the entire skeletal musculature, skeletal muscle weakness, and difficulties in gait. Patients with alcoholic hepatitis and cirrhosis have severe muscle loss that contributes to worsening outcome. Although the myopathy selectively affects Type II (fast twitch, glycolytic, anaerobic) skeletal muscle fibers, total skeletal musculature is reduced. The severity of the muscle atrophy is proportional to the duration and amount of alcohol consumed and leads to decreased muscle strength. The mechanisms for the myopathy are generally unknown but it is not due to overt nutritional deficiency, nor due to either neuropathy or severe liver disease. Skeletal muscle mass and protein content are maintained by a balance between protein synthesis and breakdown and in vivo animal models studies have shown that ethanol inhibits skeletal muscle protein synthesis. Daekumeumja is a traditional Korean medicine that is widely employed to treat various alcohol-induced diseases. Muscle diseases are often related to liver diseases and conditions. The main objective of this study was to assess that Daekumeumja extract could have protective effect against alcoholic myopathy in a Sprague-Dawley rat model. Rats were orally given 25% ethanol (5ml/kg, body weight) for 8 weeks. After 30 minutes, rats were administrated with Daekumeumja extract. Controls were similarly administrated with the vehicle alone. The weights of gastrocnemius, soleus and plantaris muscles were assessed and the morphologic changes of gastrocnemius and plantaris muscles were also assessed by hematoxylin and eosin staining. In results, The muscles from ethanol treated rats displayed a significant reduction in muscle weight and average cross section area compared to Normal group. Daekumeumja extract treated group showed increased muscle weight and muscle fiber compared to the ethanol treated group. It was concluded that Daekumeumja extract showed ameliorating effects on chronic alcohol myopathy in skeletal muscle.

Ginseng Extract Regulates the Alterations of Sleep Architecture and EEG Power Spectra in Restraint Stressed Rats

  • The present investigation was conducted to evaluate the regulation of sleep architecture by the red ginseng water extract (RGE) in acutely and chronically restraint stressed rats. Adult rats were fitted with sleep.wake recording electrodes. Following post-surgical recovery, rats were extensively habituated for freely moving polygraphic recording conditions. Polygraphic signs of sleep-wake activities were recorded for 24 h after RGE administration and induction of stress and were analyzed to understand the regulation of sleep architecture. Acute stress decreased wakefulness and increased total sleep, non-rapid eye movement (NREM) sleep, and rapid eye movement (REM) sleep in both the daytime and nighttime recording. RGE shortened the daytime NREM and REM sleep, without changing the wakefulness and total sleep. RGE increased nighttime wakefulness, and decreased total, NREM and REM sleep. Chronic stress increased wakefulness and decreased total sleep in the daytime recording, and increased REM and decreased NREM sleep in both the day and night time recording. RGE ameliorated chronic stress and induced alterations of REM and NREM sleep in the day and night time sleep architecture. Acute and chronic stress could also induce alternations in cortex electroencephalogram (EEG) recording during NREM, REM sleep and wakefulness. These findings suggest that RGE may modulate the sleep behavior in acutely and chronically stressed rats and the ameliorating effect of RGE on the sleep architecture may involve in modulation of $\alpha$-, $\theta$- and $\delta$- wave activities of the cortical EEG.

기혼여성의 취업여부에 따른 시간압박감과 관련요인의 차이 : 아내의 시간, 남편의 시간, 그리고 가사노동 사회화의 효과 (Differential in Married Women's Perceived Time Pressure by Employment : Testing the Effects of Couple Time Use and Housework Outsourcing)

  • The purpose of this study is to examine the differences in perceived time pressure between employed and non-employed married women in Korea. The sample of married couple from the Korean Time Use Survey 2009 (n = 6,948 couple diaries) was used to examined the difference in the level of perceived time pressure over the life cycle of two group. This study also investigated the married women's time use, husband's time use as well as hour of domestic outsourcing and whether such factors are associated with lowering perceived time pressure of married women. Results showed employed wives felt almost 3 times more time pressed than non-employed wives, and the gap mostly remained over different life cycles. Total work hours were associated with increasing the time pressure of both groups of women, while regenerating time was associated with ameliorating time pressure. The result suggests that the time pressure gap between employed and non-employed wives can be partly due to the relative length of total labor hours and regenerating time among the two groups. Ordered logit analysis revealed that husband's paid work hours were not associated with wive's feeling rushed, but husband' unpaid work hours were positively linked with time pressure of their wives. Results indicated if employed or non-employed wives are feeling very busy, there is a high chance that husband might give them some help. We found employed wives spend more hours on housework outsourcing, and purchasing goods for housework was not associated with lowering the feeling of pressure of employed wives. There were both similarity and difference in factors associated with women's feeling pressed among employed and non-employed wives. It implies that social process and it's strategies to alleviate the time pressure can be different by women's employment status.

흑삼농축액을 이용한 흑삼두부 제조 및 품질 특성 (Preparation and Characterization of Black Ginseng Tofu)

  • This study was performed to process Tofu as an hypolipidemic food using black ginseng. Before processing we performed the ameliorating effect of black ginseng on the serum levels of triglyceride and total cholesterol in diabetic mice (db/db mice). As a result, the diabetic mice, whose diet was supplemented with black ginseng has a reduced level of serum lipid total cholesterol ($124.58{\pm}10.59mg/dL$) compared to non-supplemented diabetic mice, The supplemented mice exhibited a significant decrease (p<0.05) in serum lipid triglycerides ($80.32{\pm}35.40mg/dL$), which provided the efficacy of black ginseng in reducing hyperlipemia, thus indirectly proving the prevention and treatment of obesity. Then we processed Tofu as a hypolipidemic food using 0~8% black ginseng extract. We evaluated the quality characteristic after producing black ginseng Tofu. For color value, as the addition level of color increases, the value of L (lightness) decreased and a (redness), b (yellowness) increased. With increase in black ginseng concentrate additional level increases, the hardness, gumminess, and chewiness values increased (p<0.05), but the springiness and cohesiveness showed no significant differences. In case of sensory evaluation, Tofu with the addition of 2% black ginseng concentrate (BGT2) showed the highest preference overall. To sum up, black ginseng demonstrated pharmacological effects in treating diabetic complications like hyperlipidemia and reducing body deposit fat.

Retrospective Review of Effectiveness of Various Pharmacological Agents in Treating Burning Mouth Syndrome

  • Purpose: Burning mouth syndrome (BMS) is a chronic pain condition involving the oral and perioral regions, often characterized by a burning sensation and pain in elderly patients. In this study, we investigated the effectiveness of pharmacological agents for the treatment of BMS patients through a retrospective chart review. Methods: We enrolled 61 BMS subjects (57 females, 4 males; $66.4{\pm}10.9$ years of age) from among consecutive patients treated pharmacologically from January 2014 to June 2015 at Chonnam National University Dental Hospital. Patients with secondary BMS associated with local factors were excluded. The treatment period, number of pharmacological agents tried, and effectiveness of the drugs administered to each subject were analyzed. Results: The mean treatment period for the management of BMS was 2.5 months. More than three agents were tried to control BMS symptoms in 17 subjects (27.9%); two agents were used in 10 subjects (16.4%), and a single agent in 24 subjects (39.3%). Clonazepam was prescribed most frequently and was effective at relieving symptoms in 30 of 39 subjects (76.9%). Paroxetine was moderately effective, relieving symptoms in 7 of 17 subjects (41.2%). Some of the subjects benefited from tricyclic antidepressants, gabapentin, and lipoic acid. A topical local anesthetic used to supplement other systemic agents had ameliorating effects in four of six subjects. Conclusions: Within the study limitations, clonazepam was the most effective drug and antidepressants were efficacious in some subjects for relieving the symptoms of BMS. These pharmacological agents could be considered as first-line drugs for the management of BMS.

칠피(漆皮) 부탄올 분획물이 LPS로 유도된 RAW 264.7 대식세포에 미치는 영향 (Effects of Butanol extract from Rhois Vernicifluae Cortex (RVC) in lipopolysaccharides-induced macrophage RAW 264.7 cells)

  • Objectives : RVC has long been used for a useful natural agent ameliorating inflammation related symptoms in the folk medicine recipe. This study was performed to investigate effects of RVC on the inflammation and oxidation in RAW 264.7 cells. Methods : The RVC was extracted with 80% ethanol and sequentially partitioned with solvents in order to increase polarity. With the various fractions, we determined the activities on the inflammation and oxidation in RAW 264.7 cells. Results : 1. Among the various solvent extracts of RVC, the butanol fraction showed the most powerful inhibitory ability against nitric oxide (NO) production in lipopolysaccharide (LPS)-induced RAW 264.7 cells without affecting cell viability. 2. Butanol fraction showed a oxidation inhibition effect by decreasing the DPPH and OH radicals. 3. Butanol fraction exhibited the inhibitory avilities against iNOS and COX-2. 4. Reverse transcriptase polymerase chain reaction (RT-PCR) and Westem blotting analysis revealed that the BuOH fraction provided a primary inhibitor of the iNOS protein and mRNA expression in LPS-induced RAW 264.7 cells. Among the up-regulater molecules of iNOS and COX-2, the BuOh fraction of RVC was shown the inhibitory activity of phoshporylation of c-Jun N-terminal kinase (JNK) 1/2 and threonine protein kinase (AKT), the one of the MAPKs pathway. Conclusion : Thus, the present study suggests that the response of a component of the BuOH fraction to NO generation via iNOS expression provide a important clue to elucidate anti-inflammatory and anti-oxidation mechanism of RVC.

Land Cover Change and Urban Greenery Prediction in Jabotabek by using Remote Sensing

  • The tremendous growth of population and physical development in the largest urban agglomeration in Indonesia -the Jakarta Metropolitan Region, also known as Jabotabek (Jakarta, Bogor, Tanggerang, Bekasi)- has created many environmental problems, such as land use conversion, increasing urban temperature, water and air pollution, intrusion of seawater, and flooding. These problems have become more serious as the urban green space (trees, shrubs, and groundcovers) has decreased rapidly with the urbanization process. Urban green space directly benefits the urban environment through ameliorating air pollution, controlling temperature, contributing to the balance of the hydrological system, and providing space for recreation and relaxation. Because there is little hard data to support the claim of decreasing greenery in Jabotabek, it is necessary to measure the amount of urban green space. The paper describes the spatial analysis of urban green space within Jabotabek through the use of a geographical information system (GIS). We used GIS and remote sensing to determine land cover change and predicted greenery percentage. Interpretation of Landsat data for 1972, 1983, 1990, and 1997 showed that Jabotabek has experiences rapid development and associated depletion of green open space. The proportion of green open space fell by 23% from 1972 to 1997. We found a low percentage of urban green space in the center of Jakarta but a high percentage in fringe area. The amount of greenery is predicted by the Ratio Vegetation Index (RVI) model: predicted greenery (%) = [146.04] RVI - 134.96. We consider that our result will be useful for landscape planning to improve the environment of Jabotabek.

트리메틸틴 처리로 유도된 기억·학습 능력 손상 모델에 대한 계피와 금앵자 혼합추출물의 개선 효과 (Ameliorating Effects of Cinnamomum loureiroi and Rosa laevigata Extracts Mixture against Trimethyltin-induced Learning and Memory Impairment Model)

  • Background: A critical features of Alzheimer's disease (AD) is cognitive dysfunction, which partly arises from decreased in acetylcholine levels. AD afftected brains are characterized by extensive oxidative stress, which is thought to be primarily induced by the amyloid beta ($A{\beta}$) peptide. In a previous study, Cinnamomum loureiroi tincture inhibited acetylcholinesterase (AchE) activity. That study identified AChE inhibitor in the C. loureiroi extract. Furthermore, the C. loureiroi extract enhanced memory in a trimethyltin (TMT)-induced model of cognitive dysfunction, as assessed via two behavioral tests. Rosa laevigata extract protected against oxidative stress-induced cytotoxicity. Administrating R. laevigata extracts to mice significantly reversed $A{\beta}$-induced learning and memory impairment, as shown in behavioral tests. Methods and Results: We conducted behavioral to examine the synergistic effects of C. loureiroi and R. laevigata extracts in inhibiting AChE and counteracting TMT-induced learning and memory losses. We also performed biochemical assays. The biochemical results showed a relationship between increased oxidative stress and cholinergic neurons damage in TMT-treated mice. Conclusions: A diet containing C. loureiroi and R. laevigata extracts ameliorated learning and memory impairments in the Y-maze and passive avoidance tests, and exerted synergistic inhibitory effect against AChE and lipid peroxidation.

곤달비 잎과 뿌리의 생물 활성 (Biological Effects of the Leaves and Roots of Ligularia stenocephala)

  • 국내에서 식품으로 사용되고 있는 국화과 식물인 곤달비(Ligularia stenocephala) 잎과 뿌리의 생물활성을 과학적으로 검증하기 위하여 각 추출물에 대하여 항산화 활성 및 세포독성을 in vitro에서, 간보호 효과, 알코올 해독작용 및 기억증진 효능 등을 in vivo에서 평가하였으며 잎의 독특한 향기성분을 GC-MS로 분석하였다. 지질과산화 억제효과는 잎(20.4% 억제율)이 뿌리 보다 좋았으며, 유해 라디칼의 일종인 superoxide anion은 뿌리에서 생성 억제효과가 더 좋았고, DPPH 소거활성은 잎과 뿌리 모두 77~79%로 매우 뛰어났다. 사염화탄소로 유발된 급성 간독성 개선효과를 AST와 ALT 효소활성도를 지표로 확인한 결과, 잎의 ALT 억제활성이 대조군에 비해 약 78% 정도 감소하였으며 알코올을 투여한 mouse의 혈중 알코올 농도는 잎추출물 투여시 약 60% 가량 유의성 있게 감소되어 효과를 인정할 수 있었다. 세포독성은 뿌리에서 비교적 강하게 나타났는데, 흑색종의 경우, $IC_{50}=40.14mg/ml$이었으며, 잎의 세포독성은 비교적 약하였다. 기억증진 효과를 동물모델을 이용한 수동회피시험법으로 평가한 결과, 잎과 뿌리 모두 scopolamine에 의해 유도된 기억력 감소를 80% 이상 향상시킨 것으로 조사되었다. 곤달비 잎의 n-헥산 추출물을 GC-MS로 분석한 결과, 독특한 향기는 주로 terpene 화합물에서 유래되는 것으로 추정되었다.

Inhibitory Effect of Ginsenosides on NMDA Receptor-mediated Signals in Rat Hippocampal Neurons

  • Ginseng is the best known and most popular herbal medicine used worldwide. Ameliorating effects of ginseng were observed on the models of scopolamine-induced, aged or hippocampal lesioned learning and memory deficits. Further beneficial effects of ginseng were observed on neuronal cell death associated with ischemia or glutamate toxicity. In spite of these beneficial effects of ginseng on the CNS, little scientific evidence shows at the cellular level. In the present study, we have employed cultures of rat hippocampal neurons and examined the direct modulation of ginseng on NMDA receptor-induced changes in $[Ca^{2+}]_i$ and -gated currents using fura-2-based digital imaging and perforated whole-cell patch-clamp techniques, respectively. We found that ginseng total saponins inhibited NMDA-induced but less effectively glutamate-induced increase in $[Ca^{2+}]_i$ Ginseng total saponins also modulated $Ca^{2+}$ transients evoked by depolarization with 50 mM KCI along with its own effects on $[Ca^{2+}]_i$. Among ginsenosides tested, ginsenoside $Rg_3$ was found to be the most potent component for ginseng actions on NMDA receptors. Furthermore, we examined the inhibitory effects ofbiotransformants of ginsenosides on NMDA receptor using purified stereoisomers of ginsenosides. 20(S)-ginsenoside $Rg_3$ and its metabolite, 20(S)-ginsenoside $Rh_3$, produced the strongest inhibition while 20(S)-ginsenoside $Rh_1$ and Compound K produced the moderate inhibition on NMDA-induced increase in $[Ca^{2+}]_i$. The data obtained suggest that the inhibition of NMDA receptors by ginseng, in particular by 20(S)-ginsenoside $Rg_3$ and its metabolite, 20(S)-ginsenoside $Rh_2$, could be one of mechanisms for ginsengmediated neuroprotective actions.
