H2O2로 유도된 C6 신경교세포의 세포사멸에 대한 감초 추출물과 감초 활성물질의 보호효과 (Protective Effects of Glycyrrhiza uralensis Radix Extract and Its Active Compounds on H2O2-induced Apoptosis of C6 Glial Cells)

  • Background: Glycyrrhiza uralensis Radix (GR) is a crude drugs used in Asian countries that has been reported to prevent the progression of neurodegenerative diseases such as Alzheimer's disease. The present study examined whether GR and its active compounds, glycyrrhizic acid (GA) and isoliquiritigenin (IL), exerted protective effects on $H_2O_2$-induced oxidative damage in C6 glial cells. Methods and Results: We exposed C6 glial cells to hydrogen peroxide ($H_2O_2$) for 24 h and investigated the cellular response to GR and its active compounds by evaluating cell viability, reactivie oxygen species (ROS) production, and apoptosis-related protein expression. GR successfully mitigated the reduced cell viability and ROS production induced by $H_2O_2$ in C6 glial cells, IL and GA significantly increased the cell viability and decreased ROS production. In addition, IL and GA down-regulated apoptotic Baxdependent caspase-3 activation, but each compound exerted different mechanisms, i.e., IL dose-dependently decreased ROS production and, GA up-regulated anti-apoptotic Bcl-2 expression. Conclusions: These results demonstrated that GR and its active components, IL and GA, exhibit potential for use as natural neurodegenerative agents for the modulation of apoptosis in C6 glial cells.

치매 한의진단 평가도구 적용 연구 (Study on the Application of Oriental Medical Evaluation to Dementia)

  • Objectives: Patients with dementia are increasing in Korea. So the importance of accurate diagnosis and treatment of dementia is growing. In this paper, we evaluated the result of employing the Korean medical diagnostic pattern as a tool in clinics. Methods: Patients diagnosed with Alzheimer's disease were evaluated using the Korean medical diagnostic pattern tool. Results: The number of patients with liver-kidney yin deficiency pattern/syndrome was 35. Analyzing the ratio difference between the 'liver-kidney yin deficiency' group and the 'not liver-kidney yin deficiency' group revealed that the ratio of the responses to the fourth question was less than zero. The ratio of the responses to the fifth, seventh, and eight questions were all less than 10%. The ratio of the responses to the first and second questions were greater than 30%. Conclusions: Of the six diagnostic patterns, liver-kidney yin deficiency was the greatest in the study subjects. The fourth question in the liver-kidney yin deficiency tool tests for the lack of appropriateness whereas the fifth, seventh, and eight questions test for lack of discrimination. Applying more weight to the first and second questions was an excellent choice to increase the discrimination.

Plant Phenolics as ${\beta}$-Secretase (BACE1) Inhibitors

  • Various plant phenolics were assessed for (${\beta}$-secretase (BACE1) inhibitory activity in order to screen for anti-dementia agents. Among 39 phenolics, eight compounds, 1,2,3-trigalloyl glucopyranoside, acetonyl geraniin, euphorscopin, furosine, helioscopinin A, helioscopinin B, jolkinin, and rugosin E exhibited strong inhibition of BACE1 with $IC_{50}$ values of $5.87{\times}10^{-8}-54.93{\times}10^{-6}\;M$. Among them, rugosin E was the most potent ($IC_{50}$ $5.87{\times}10^{-8}\;M$). The active compounds were shown to be non-competitive inhibitors by Dixon plot. All the phenolic BACE1 inhibitors except furosin also suppressed prolyl endopeptidase (PEP) activity. However, these phenolic compounds caused less inhibition of ${\alpha}$-secretase (tumor necrosis factor a converting enzyme; TACE) and no significant inhibition of other serine proteases such as trypsin, chymotrypsin, and elastase was seen, demonstrating that they are relatively specific to both BACE1 and PEP. No significant structure-activity relationships were found.

뇌의 MR 영상에서 번짐 현상의 명암 값 분석을 통한 백질과 회백질의 추출 및 체적 산출 (Segmentation and Volume Calculation through the Analysis of Blurred Gray Value from the Brain MRI)

  • 본 연구는 사람의 뇌에 대한 자기공명영상에서 백질과 회백질을 분리하고 각각의 체적을 산출하기 위한 것이다. 일반적으로 치매나 다운증후군 같은 정신질환의 경우 백질 또는 회백질의 위축으로 인해 체적이 감소하게 되므로, 사람의 뇌에 대하여 백질과 회백질의 체적 산출을 통한 크기의 변화를 추적함으로서 여러 정신질환의 진단 및 조기 발견에 유용하게 이용될 수 있다. 그러나 일정한 두께의 단면을 촬영하여 단일의 명암 값으로 표현하는 자기공명영상기기의 특성상 번짐 현상을 보이는 자기공명영상으로부터 원래의 두께 안에 존재하는 각 성분의 부분체적을 산출할 수 없음으로 인해 백질과 회백질의 체적산출이 현재까지 불가능하였다. 따라서 본 논문에서는 번짐(blurred)을 보이는 자기공명영상에서 번진 명암 값을 해석하는 새로운 알고리즘에 의해 백질과 회백질의 부분체적을 산출하고, 이를 근거로 자기공명영상에서 백질과 회백질을 분리하기 위한 판별값을 결정하였으며, 결정된 판별값에 의해 분리된 영상에서 백질과 회백질의 체적을 산출하였다. 또한 제안된 알고리즘의 검증을 위해 인위적으로 생성된 모델에 알고리즘을 적용하여 산출된 결과를 원래의 모델과 비교하여 보았다.

아위버섯(Pleurotus ferules) 추출물의 생리활성 탐색 (Studies on the Biological Activity of Pleurotus ferulea)

  • 본 연구에서는 아위버섯의 생리활성 기능규명을 목적으로 하였다 사람의 뇌세포에서의 아세틸콜린분해효소 억제효과는25∼35%의 경향을 보여 치매 예방 및 개선제로서의 가능성을 보였다. 유해산소 제거 기능은 35∼36%로 높아 항암, 노화, 심장병 등에 좋은 효과를 나타낼 수 있을 것으로 생각되어진다. 간섬유화(collagen type-I) 억 제는 3∼12%의 저해효과를 보였다. 그러나 아위버섯은 3∼8%의 포도당 흡수억제를 나타내어 당뇨와 비만억 제에 큰 효과를 보이지 않았으며, 위암과 대장암에 대한 항암효과도 거의 나타나지 않았다. 따라서 향후 아위버섯 추출물 특히, K8종에서 나타난 다양한 생리활성효과를 이용하여 고부가가치의 다양한 기능성제품이 가능할 것으로 판단된다

Sleep Promoting Effect of Luteolin in Mice via Adenosine A1 and A2A Receptors

  • Luteolin, a widespread flavonoid, has been known to have neuroprotective activity against various neurologic diseases such as epilepsy, and Alzheimer's disease. However, little information is available regarding the hypnotic effect of luteolin. In this study, we evaluated the hypnotic effect of luteolin and its underlying mechanism. In pentobarbital-induced sleeping mice model, luteolin (1, and 3 mg/kg, p.o.) decreased sleep latency and increased the total sleep time. Through electroencephalogram (EEG) and electromyogram (EMG) recording, we demonstrated that luteolin increased non-rapid eye movement (NREM) sleep time and decreased wake time. To evaluate the underlying mechanism, we examined the effects of various pharmacological antagonists on the hypnotic effect of luteolin. The hypnotic effect of 3 mg/kg of luteolin was not affected by flumazenil, a GABAA receptorbenzodiazepine (GABAAR-BDZ) binding site antagonist, and bicuculine, a GABAAR-GABA binding site antagonist. On the other hand, the hypnotic effect of 3 mg/kg of luteolin was almost completely blocked by caffeine, an antagonist for both adenosine A1 and A2A receptor (A1R and A2AR), 8-Cyclopentyl-1,3-dipropylxanthine (DPCPX), an A1R antagonist, and SCH-58261, an A2AR antagonist. From the binding affinity assay, we have found that luteolin significantly binds to not only A1R but also A2AR with $IC_{50}$ of 1.19, $0.84{\mu}g/kg$, respectively. However, luteolin did not bind to either BDZ-receptor or GABAAR. From these results, it has been suggested that luteolin has hypnotic efficacy through A1R and A2AR binding.

더덕 물 추출물과 에탄올 추출물의 인지능 개선 활성 비교 (Effect of Water and Ethanol Extracts Codonopsis lanceolata on Spatial Learning and Memory in Mice)

  • Alzheimer's disease (AD), most common form of dementia is characterized that memory deficit and loss of cognitive function. This study was evaluated cognitive enhancing effect of water and ethanol extracts of Codonopsis lanceolata and compared using Morris water maze and passive avoidance test. The water and 70% ethanol extracts (100, 300 and 500 mg/kg) were administered to mice. The neuroprotective effect on glutamate-induced cell death in HT22 cells was additionally investigated using MTT assay. Results showed 70% ethanol extract of Codonopsis lanceolata enhanced cognitive function than water extract, as shown by decrease in escape latency time in Morris water maze test. In passive avoidance test, 70% ethanol extract also increased the latency time compared to the water extract. Furthermore, 70% ethanol extract significantly protected neuronal cell against glutamate cytotoxicity and showed higher than neuroprotective effect of water extract. These results indicate that 70% ethanol extract more improve spatial cognitive ability and protected neuronal cells than water extract.

[ $\beta$ ]-Secretase (BACE1) Inhibitors from Pomegranate (Punica granatum) Husk

  • In the course of screening for anti-dementia agents from natural products, two $\beta$-secretase (BACE1) inhibitors were isolated from the husk of pomegranate (Punica granatum) by activity-guided purification. They were identified as ellagic acid and punicalagin with $IC_{50}$ values of 3.9 $\times$$10^{-6}$ and 4.1$\times$$10^{-7}$ M and Ki values of 2.4$\times$$10^{-5}$and 5.9$\times$$10^{-7}$ M, respectively. The compounds were non-competitive inhibitors with a substrate in the Dixon plot. Ellagic acid and punicalagin were less inhibitory to $\alpha$-secretase (TACE) and other serine proteases such as chymotrypsin, trypsin, and elastase, thus indicating that they were relatively specific inhibitors of BACE1.

Analysis of behavioral management for dental treatment in patients with dementia using the Korean National Health Insurance data

  • Background: The global population is aging rapidly, and accordingly, the number of patients with dementia is increasing every year. Although the need for dental treatment increases for various reasons in patients with dementia, they cannot cooperate during dental treatment. Therefore, behavioral management, including sedation (SED) or general anesthesia (GA), is required for patients with dementia. Thus, this study aimed to investigate the trends and effects of SED or GA in patients with dementia undergoing dental treatment in South Korea based on the Korean National Health Insurance claims data. Methods: This study utilized customized health information data provided by the Health Insurance Review and Assessment Service. Among patients with records of using sedative drugs during dental treatment from January 2007 to September 2019, patients with the International Classification of Diseases-10 code for dementia (F00, F01, F02, F03, and G30) were selected. We then analyzed the full insurance claims data for dental care. Age, sex, sedative use, and dental treatment of patients were analyzed yearly. In addition, the number of cases of GA or SED per year was analyzed, and changes in behavioral management methods with increasing age were investigated. Results: Between January 2007 and September 2019, a total of 4,383 (male, 1,454; female, 2,929) patients with dementia received dental treatment under SED or GA. The total number of SED and GA cases were 1,515 (male, 528 ; female, 987 ) and 3,396 (male, 1,119 ; female, 2,277) cases, respectively. The total number of cases of dental treatment for 4,383 patients with dementia was 153,051 cases, of which 2.22% were under GA and 0.98% were under SED. Midazolam was the most commonly used drug for SED. Conclusion: Although gingivitis and pulpitis were the most common reasons for patients with dementia to visit the dentist, GA or SED for patients with dementia was frequently used in oral and maxillofacial or periodontal surgery.

Chromenone Derivatives as Monoamine Oxidase Inhibitors from Marine-Derived MAR4 Clade Streptomyces sp. CNQ-031

  • Three compounds were isolated from marine-derived Streptomyces sp. CNQ-031, and their inhibitory activities against monoamine oxidases (MAOs), acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and β-secretase (BACE-1) were evaluated. Compound 1 (5,7-dihydroxy-2-isopropyl-4H-chromen-4-one) was a potent and selective inhibitor of MAO-A, with a 50% inhibitory concentration (IC50) of 2.70 µM and a selectivity index (SI) of 10.0 versus MAO-B. Compound 2 [5,7-dihydroxy-2-(1-methylpropyl)-4H-chromen-4-one] was a potent and low-selective inhibitor of MAO-B, with an IC50 of 3.42 µM and an SI value of 2.02 versus MAO-A. Compound 3 (1-methoxyphenazine) did not inhibit MAO-A or MAO-B. All three compounds showed little inhibitory activity against AChE, BChE, and BACE-1. The Ki value of compound 1 for MAO-A was 0.94 ± 0.28 µM, and the Ki values of compound 2 for MAO-A and MAO-B were 3.57 ± 0.60 and 1.89 ± 0.014 µM, respectively, with competitive inhibition. The 1-methylpropyl group in compound 2 increased the MAO-B inhibitory activity compared with the isopropyl group in compound 1. Inhibition of MAO-A and MAO-B by compounds 1 and 2 was recovered by dialysis experiments. These results suggest that compounds 1 and 2 are reversible, competitive inhibitors of MAOs and can be considered potential therapies for neurological disorders such as depression and Alzheimer's disease.
