Effect of Persimmon Leaf Extract on Phthalic Anhydride-induced Allergic Response in Mice

  • The purpose of this study was to investigate the anti-allergy activities of persimmon leaf extract (PLE) on a phthalic anhydride (PA)-induced allergic mouse model. A human leukemic mast cell line (HMC-1) was used to examine the inhibitory activity of PLE on the histamine release by human leukemic mast cells. PLE inhibited histamine release from HMC-1 cells in response to cross-linkage of high-affinity IgE receptor-${\alpha}$ ($Fc{\varepsilon}RI{\alpha}$). Additionally, a PA-induced allergic mouse model was used to investigate the effects of PLE in vivo. Mice were orally administrated with or without PLE of single dose (250 mg/kg/day) for 31 days. Oral intake of PLE significantly inhibited passive cutaneous reactions. Oral administration of PLE to PA-induced allergic mice also led to a striking suppression of the development of contact dermatitis, ear swelling and lymph node weight. In addition, PA-specific IL-4 production of draining lymph node cells was markedly diminished by PLE oral administration, but not IFN-${\gamma}$. Furthermore, PLE treatment suppressed PA-induced thymus and activation-regulated chemokine (CCL17) and cutaneous T cell-attracting chemokine (CCL27) expressions in ear tissues. Based on these results, we suggest that PLE may have therapeutic potential as an effective material for management of irritant contact dermatitis or related inflammatory diseases.

도시형 대안학교 공간의 치유환경에 관한 연구 - 성미산 학교 공간을 중심으로 - (A Study on the Healing environment of Urban Alternative School's space - Focused on Sungmisan School's space -)

  • As de-schooling students (students who leave schools) have been produced and increased in middle and high schools every year since 1990s, urban alternative schools have founded with Seoul as the center. The objects of such urban alternative schools are de-schooling teenagers, and their educational goal is to make the students to discover their own values and grow as members of the community by accomplishing their healing and growth. Most of students in alternative schools have excessive self-centered feeling than ordinary people, and since they don't have exchanges with others, they have to receive holistic healing along with education. Here, 'healing' is a method of approaching to health through environmental, psychological, social and cultural supports unlike 'treatment' used for medical means. Therefore, holistic healing for alternative schools' students has to accomplish self-knowledge, self-control, and self-healing without repulsion through spaces of healing environments instead of heavy-handed exchanges. This study has integrated a theory of Max $L{\ddot{u}}scher$ who suggested a psychological healing theory in terms of internal character and a theory of Rudolf Steiner who suggested it in terms of practical and holistic sense and analyzed Sungmisan School, one of urban alternative schools in Seoul through the integrated theory. The analysis of the integrated theory are intended to emphasize the importance of healing environments and suggest methods in creating healing environments for urban alternative schools in the future.

온실효과에 대한 학생들의 개념 분석 (The analysis of students' ideas about the greenhouse effect)

  • The purpose of this study is to examine the concept of Greenhouse Effect as understood by middle school high school university students using a closed-form questionnaire. Based on results of the questionnaire which was administered to 619, the extent to which alternative concepts were held was quantified and compared the difference of various group based on grade level. gender, text and major. Also, subjects were divided into two groups, one is middle school students and the other is high school university students and common themes within conceptual framework of each group were identified by factor analysis. The result showed that students confused Greenhouse Effect with ozone layer depletion in stratosphere and linked familiar contamination around everyday life. acid rain. radioactive contamination, nuclear arsenal to Greenhouse Effect. These trends were more appreciable in female than male, biology major than any other major and text did not make any significant difference. In addition, the result of factor analysis showed that two groups linked familiar contamination around everyday life to Greenhouse Effect and high school university students understand the consequences of an increase in the Greenhouse Effect more systematically than middle school student, perceived the relation between the origin of an increase in the Greenhouse Effect and human activity but confused Greenhouse Effect with ozone layer depletion in stratosphere.

Alternative splicing variant of NRP/B promotes tumorigenesis of gastric cancer

  • Gastrointestinal cancer is associated with a high mortality rate. Here, we report that the splice variant of NRP/B contributes to tumorigenic activity in highly malignant gastric cancer through dissociation from the tumor repressor, HDAC5. NRP/B mRNA expression is significantly higher in the human gastric cancer tissues than in the normal tissues. Further, high levels of both the NRP/B splice variant and Lgr5, but not the full-length protein, are found in highly tumorigenic gastric tumor cells, but not in non-tumorigenic cells. The loss of NRP/B markedly inhibits cell migration and invasion, which reduces tumor formation in vivo. Importantly, the inhibition of alternative splicing increases the levels of NRP/B-1 mRNA and protein in AGS cells. The ectopic expression of full-length NRP/B exhibits tumor-suppressive activity, whereas NRP/B-2 induces the noninvasive human gastric cancer cells tumorigenesis. The splice variant NRP/B-2 which loses the capacity to interact with tumor repressors promoted oncogenic activity, suggesting that the BTB/POZ domain in the N-terminus has a crucial role in the suppression of gastric cancer. Therefore, the regulation of alternative splicing of the NRP/B gene is a potential novel target for the treatment of gastrointestinal cancer.

Antioxidant, Antimicrobial, and Cancer Cell Proliferative Inhibition Activities of Propolis

  • A commercial propolis was investigated in terms of its antioxidant, antimicrobial, and antiproliferative activities. The contents of total phenol and flavonoid of propolis were 8.3 and 6.6 mg, respectively. The reducing power of the propolis increased with concentration increasing. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was shown at 82.70% in 1,000 ${\mu}g/mL$ of the propolis. 2,2'-Azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging effect of antioxidant activity on the propolis was 35.64 g/sample. The propolis showed high antimicrobial activity against Bacillus cereus at all concentration of propolis. All of the cancer cell lines have 53-73% as effective growth inhibition. These results showed that the commercial propolis has potential antioxidant, antimicrobial, and cancer cell proliferative inhibition activities thus, propolis can be applied to the functional food, pharmaceutical, and cosmetic industry.

판 구조론 학습에 사용되는 시각적 표상이 판구조론 개념에 대한 고등학생들의 응답에 미치는 영향 (The Effect of Visual Representation in Plate Tectonics Topics on High School Students' Conceptions on Plate Tectonics)

  • This study aimed to investigate the high school students' conceptions about the plate tectonics through visual representation. For this purpose, the subjects were 67 students in 11th-grade high schools in Chungbuk. In order to in-depth understand the students' conceptions about plate tectonics, so the investigator conducted a semi-structured interview. The conclusions were as in the following. After learning the plate tectonics, the students had the alternative conceptions associated with terminology, colors' meanings, plate-related melting, plate's movement, plates' boundaries, mantle's physical conditions, driving forces for plate movement, and they had the organic relations about colors' meanings, mantle's physical conditions, and driving forces of plate movement. Also, the visual representation used to teach plate tectonics influenced on the students' responses about terminology, plates' boundaries, plate-related melting and the mantle's physical features, also this study found the factors of visual representation causing the learners to create alternative conceptions. These results implicated the importance of teacher's role in identifying the students' interpretation process on visual representation, and it needed to improve the factors creating students' alternative conceptions about visual representation and to study the factors further.

대구 지역 고등학생의 아침식사 이용실태 및 아침식사 대용식 인식조사 (Study on Breakfast Status and Perception of Substitution Foods for Breakfast in High School Students in Daegu Area)

  • This study investigated eating behaviors of high school students (209 males and 230 females) in Daegu region by gender. As a result of analyzing breakfast intake frequency, 'Not rarely eat' showed the highest (25.1%), followed by '5 times a week' (24.4%), '2 times a week' (17.3%), '3 times a week' (15.0%), '1 time a week' (11.6%), and '4 times a week' (6.6%). Regarding reasons for skipping breakfast, 'lack of time' showed the highest percentages. For their breakfast, 53.5% of students ate boiled rice with side dishes. Regarding reasons for skipping breakfast, 'buy and eat snack' showed the highest percentages. Regarding favorite breakfast menu, 'rice roll, rice ball, rice burger' showed the highest percentage (55.4%), followed by 'bread & cereal' (28.7%) and 'fruits & vegetable' (7.7%). Regarding purchase of breakfast alternatives, 'Yes' was highest. Considering actors for choosing a breakfast alternative, there is a need for 'convenience of food consumption', 'spending less time', 'easy cooking', 'favorite menu' and 'taste' of the breakfast alternative. When asked how much they like breakfast alternative menus, they responded that they liked 'rice roll, rice ball, rice burger' (3.91), 'bread & cereal' (3.67), 'fruit & vegetable' (3.66), 'noodle' (3.39), 'porridge' (3.18) and 'rice cake' (3.07). This result shows that breakfast menus should be developed according to high school students preferences.

Analyses of alternative polyadenylation: from old school biochemistry to high-throughput technologies

  • Alternations in usage of polyadenylation sites during transcription termination yield transcript isoforms from a gene. Recent findings of transcriptome-wide alternative polyadenylation (APA) as a molecular response to changes in biology position APA not only as a molecular event of early transcriptional termination but also as a cellular regulatory step affecting various biological pathways. With the development of high-throughput profiling technologies at a single nucleotide level and their applications targeted to the 3'-end of mRNAs, dynamics in the landscape of mRNA 3'-end is measureable at a global scale. In this review, methods and technologies that have been adopted to study APA events are discussed. In addition, various bioinformatics algorithms for APA isoform analysis using publicly available RNA-seq datasets are introduced.

Students' Alternative Conceptions of Plate Boundaries and Their Conception Revision According to Their Reasoning Patterns

  • This study investigated students' alternative conceptions of plate boundaries and their conception revision according to the pattern of students' reasoning. Participants were twenty-two 10th grade high school students. All participants were asked to draw the three types of plate boundaries and to explain their drawings. Nine students participated in the reasoning activity. To this end, a semi-structured interview was conducted during which key questions were asked for the students to individually answer. The key questions used in the reasoning activity were created, by utilizing questions used in the previous studies. The findings revealed that the alternative conceptions of plate boundaries were classified into three levels based on established criteria. Students who attempted a variety of reasoning strategies such as causal reasoning, using an analogy, abductive reasoning, data reconstruction and concept combination, revised their alternative conception to a scientific conception after the reasoning activity. On the other hand, some students could not revise their alternative conceptions because they only conducted an incomplete reasoning strategy. The study also found that they were unable to use other reasoning strategies, either.

Antioxidant, Antimicrobial, and Antiproliferative Activities of Olive (Olea europaea L.) Leaf Extracts

  • Total phenol, total flavonoid, reducing powder, electron donating activity, ascorbic acid equivalent antioxidant capacity, antimicrobial and antiproliferative activities of olive leaf extracts were investigated. The contents of total phenol and flavonoid were 257.48 and 92.33 mg in 100 g of olive leaf extract, respectively. The reducing power of the olive leaf extract increased with concentration increasing. Electron donating activity was high in 100 ${\mu}g/mL$ treated olive leaf extract as 95.20%. The ascorbic acid equivalent antioxidant capacity of the olive leaf extract was 68.93 mg/g olive leaf extract. The olive leaf extracts showed relatively high antimicrobial activity against Escherichia coli, Salmonella typhimurium, Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, and Pseudomonas aeruginosa. All of the cancer cell lines including MKN45, HCT116, NCI-H460, and MCF7 have 70-81% as effective growth inhibition.
