The antifungal effect of pine needle extract prepared by a distinguishable extraction method and the dry distillation method, was examined. The effect of this extract itself was insignificant. The chemical components of pine needle extract were then investigated by gas chromatographic analysis, and four, chemical components, acetol, furfural, 5-methyl furfural, and terpine4-ol, were identified. The antifungal effects of those four chemical components against Alternaria mali (A. mali), an agent of Alternaria blotch of apple, were then examined. It was observed that the minimum inhibitory concentrations (MICs) were 6.25, 0.78, 0.78, and 12.5 (mg/ml) of acetol, furfural, 5-methyl furfural, and terpine-4-ol, respectively. MICs of furfural and 5-methyl furfural had the same order of magnitude as that of an antifungal agrochemical, chlorothalonil. Although furfural itself can not be completely substituted for an antifungal agrochemical, a partial mixture of furfural and antifungal agrochemical may be used as a substitute. The use of agrochemicals for the prevention of plant disease caused by pathogenic fungus such as A. mali could be partially reduced by the application of this mixture.
Changes in the numbers of air-borne spores of Alternaria mali causing the Alternaria leaf spot of apple were investigated in apple orchards, by monitoring their AM- toxin(host-specific toxin) producing ability. Most of the air-borne Alternaria in apple orchards were not AM-toxin producers. In culture, only about 2% of Alternaria isolates from a commercially managed apple orchards produced the toxin that was highly toxic only to susceptible leaves.
In the 82 apple varieties tested, the distribution of varieties grouped on the damage degree to Alternaria leaf spot were 15 varieties(l8.3%), which damaged above 50%, and 51 varieties(62.2%) damaged below 5%. Susceptible varieties to Alternaria leaf spot were Raritan, Indo, Winesap and Starkrimson, moderately resistant varieties Sekaiichi and Starking, and resistant Jonathan, Tsugaru, Molliies, Jonagold, Gala etc. The leaf hair density on the backside of leaf were rarer inresistant varieties than susceptible one. In inoculation test, penetration and infection of Alternaria mali were easier on the backside than surface of apple leaf at the same varity, and infection rate with Alternaria mali were higher at removal treatment than non-removal one of leaf hairs, so we can guess that leaf hair are related to resistance of variety to Alternaria mali.
Isoates with changed pathogenicity were selected among iprodione-resistant Alternaria mali to investigate any relationship between their pectolytie enzyme activity and pathogenicity. In artificial medium, strongly pathogenic isolates $S_1$, and $R_3$ showed higher enzyme activity than weakly pathogenic isolate $R_8$.Activity of endo-polymethylgalacturonase and end-o-polygalacturonase was more than 3 times. But activity of pectin methylesterase and pectin Iyase by isolaste $S_1$ was higher than those by $R_3$ and $R_8$ isolates. In apple medium dialyzed against distilled water, activity of enzyme by each isolate was increased but growth of each isolate was reduced. When iprodione was added to the medium, enzyme activity and growth of isolate $S_1$, were reduced but strongly pathogenic isolate .$R_3$ among iprodione-resistant ones showed increased enzyme activity except for exo-polygalacturonase in dialyzed apple medium.
In early June 2014, leaf spot symptoms were observed on black chokeberry (Aronia melanocarpa) in Yesan-gun and Geumsan-gun, Chungcheongnam-do in Korea. The initial symptoms on leave surfaces were brown small-circular spots with a yellow halo lesion, and gradually the small spots were fused, all of infected leaves dropped eventually. A fungus were isolated from the initial lesion, and cultured on potato dextrose agar. Colony color on upper surface of plate varied from olive gray to charcoal gray. Size of conidia mostly extend to $19-50{\times}5-9{\mu}m$ in nature and $20-59{\times}8-13{\mu}m$ in culture, with 3-8 transverse septa and usually no longitudinal septum or only 1 longitudinal septum in 1-3 of the transverse compartments, and also have a short or long beak. Pathogenicity was investigated using wounded or unwounded black chokeberry and apple leaves. After 7 days of inoculation, leaf spots were similar to the symptoms naturally occurred in the field. On the basis of mycological characteristics, pathogenicity, and ITS rDNA sequence analysis, this fungus was identified as Alternaria mali. This is the first report of Alternaria leaf spot on black chokeberry caused by A. mali in Korea.
The fifty percent effective concentration (EC50) and the minimum inhibitory concentration (MIC) of iprodione on the resistant isolates of Alternaria mali obtained from infected leaves in Kyeongsan region ranged $550\~1,310{\mu}g/ml$ and $33,800-39,800{\mu}g/ml$, respectively. Conidia germination of the resistant isolates reached $70\%$ on PSA added with iprodione, $500{\mu}g/ml$ When treated with $500{\mu}g/ml$ iprodione soon after the inoculation the diameters of lesions on apple fruits 7days after inoculation with the resistant isolates were 1.6-14.6mm but no lesions appeared on those inoculated with the sensitive isolates. A similar trend of responses with the sensitive isolates was shown to benomyl, chlorothalonil, garbenda, thiophanatemethyl and triademefon with the resistant isolates. One isolate among the resistant ones was cross resistance to captan and folpet that were effective to the sensitive isolates.
This studies was carried out to find the effect of inorganic matters of Apple Leaves on the occurrence of Alternaria leaf spot and calicium compounds on mycelial growth of Alternaria mali. Diseased leaf rate of Alternaria leaf spot of susceptible apple varieties was 84.8%, moderate resistant 8.1% and resistant 0.3% respectively and in No. of lesion per leaf, the tendency was same. CaO contents of apple leaves was contained much in the resistant varieties than susceptible one. It was estimated that calcium is one of factors on resistance to apple leaf spot by the results of relations among CaO contents and diseased leaf rate from July to August and No. of lesion per leaf was correlated significantly as r=-0.551, r=-0.585, -0.485 but in T-N, $P_2O_5,\;K_2O,\;MgO,\;Na_2O$, there was no difference among varieties. Then, growth of Alternaria mali was possible in the media of which pH ranged up to $11{\sim}13$, and calcium has effect of inhibition on growth of Alternaria mali.
In the survey from 1992 to 2000, twenty-eight parasitic diseases were observed in major apple producing areas in Korea. The predominant apple diseases were white rot(Botryosphaeria dothidea), Marssonina blotch(Marssonina mali), Valsa canker(Valsa ceratosperma), Alternaria leaf spot(Alternaria mali), and bitter rot(Collectotrichum gloeosporioides and C. acutatum). Apple scab that reappeared in 1990 after disappearance for 15 years was disappeared again since 1997. A viroid disease(caused by apple scar skin viroid) was newly found in this survey. The five diseases, fire blight(Erwinia amylovora), black rot(Botryosphaeria obtusa), scab(Cladosporium carpophilum), Monochaetia twig blight(Monochaetia sp.), and brown leaf spot(Hendersonia mali), which had once described in 1928 but no further reports on their occurrence, were not found in this survey. However, blossom blight(Monilinia mali), brown rot(Monilinia fructigena), and pink rot(Trichothecium roseum), which did not occur on apple after mid 1970s, were found in this survey.
Of the 1,200 single spore isolates of Alternaria mali causing apple leaf spot and fruit decay, the mycelial colonies showed 76, 96 and 15% growth at 100,000 ${\mu}g/ml$ of captafol, chlorothalonil and folpet, respectively, and five and three percent growth at 10,000 ${\mu}g/ml$ of iprodione and polyoxin. These isolates showed cross-tolerance of mycelial growth, spore formation, or spore germination to the above five fungicides plus captan, garbenda, and thiophanate methyl. Captafol, mancozeb, polydong, and propineb did not show any cross-tolerance of spore germination since no spore was germinated at 100 ${\mu}g/ml$ which was much lower than the concentration used by apple growers. Benomyl, fenarimol, oxidong, and triademefon showed medium degree of cross-tolerance of the fungal growth, sporulation, and spore germination.
RAPD analysis was performed from four host-specific toxin (HST) producing Alternaria, i.e., A. kikuchiana, A. mali, a. longipes and A. Longipes and A. alternata f. sp. lycopersici, nonpathogenic A. alternata and A. brassicicola to assess their phylogenetic relationship. DNA polymorphism was detected among species (pathotypes) of HST producing Alternaria by PCR amplification and differentiation of the species was recognized by RAPD analysis. Primer OPA-02 was the most profitable among 7 notificated primers for differentiation of the HST producing Alternaria species. UPGMA analysis of the RAPD bands from alternaria spp. revealed that HST producing Alternaria and nonpathogenic a. alternata are closely related.
