Studies on the Chemical Constituents from the Seeds of Zizyphus jujuba var. inermis

  • This study analyzed the seeds of Zizyphus jujuba var. inermis commonly used as a remedy in traditional Chinese medicine, in order to determine its various biologically active compounds. Through process 3-pentadecylcatechol, ${\rho}$-menth-8-ene, and ${\gamma}$-bisabolene were isolated and identified for the first time which are urushiol, monoterpenoidal, and sesquiterpenoidal compounds, respectively. Also, found were another sesquiterpenoidal compounds, vomifoliol, and four steroidal compounds, ${\beta}$-sitosterol, stigmasterol, stigmasta-5,23-dien-$3{\beta}$-ol, and stigmast-4-en-3-one. In addition, fourteen triterpenoidal compounds were isolated and identified. These were lupeol, betulinic acid, betulinaldehyde, alphitolic acid, 3-O-cis-${\rho}$-coumaroyl-alphitolic acid, 3-O-trans-${\rho}$-coumaroyl-alphitolic acid, 2-O-cis-${\rho}$-coumaroyl-alphitolic acid, 2-O-trans-${\rho}$-coumaroyl-alphitolic acid, zizyberanalic acid, ceanothic acid, oleanolic acid, maslinic acid, 3-O-cis-${\rho}$-coumaroyl-maslinic acid, and 3-O-trans-${\rho}$-coumaroyl-maslinic acid. The structures were identified by comparing of the spectroscopic experiments, NMR and MS, and then compared that reported data, respectively. Three extracts of water, methanol, and chloroform from the seeds showed a weak anti-proliferative effect, anti-microbial activity, and anti-oxidant effect, respectively.

Isolation and Quantitative Analysis of Betulinic Acid and Alphitolic Acid from Zyziphi Fructus (대추로부터 베튜리닉 산과 알피톨릭 산의 분리 및 정량)

  • Betulinic acid and alphitolic acid, the triterpenoids of Zyziphi Fructus, were isolated with silica gel column chromatography and used as the standard substances for the analysis. The compounds were determined with HPLC and HPTLC. They were separated on reversed phase column (Nova-Pak C18) with 0.05M Na2HPO4-methanol (19:81) in HPLC and detected at 210nm. Separation on HPTLC precoated silica gel F254 plates was carried out with chloroform-methanol (6:1) and the separated compounds were reacted with p-anisaldehyde and detected at 540nm. The contents of betulinic acid and alphitolic acid in Zyziphi Fructus from four different regions in Korea were in the range of 2.9~3.8% and 3.2~3.9%, respectively.

Quantitative Determination of Triterpenoids from the Fruits of Zizyphus jujuba

  • Eleven triterpenoids, colubrinic acid (1), alphitolic acid (2), 3-O-cis-p-coumaroyl alphitolic acid (3), 3-O-trans-p-coumaroyl alphitolic acid (4), 3-O-cis-p-coumaroyl maslinic acid (5), 3-O-trans-p-coumaroyl maslinic acid (6), betulinic acid (7), oleanolic acid (8), betulonic acid (9), oleanonic acid (10), and zizyberenalic acid (11), were isolated from the fruits of Zizyphus jujuba Mill. A simple and rapid HPLC method, using a $C_{18}$ column, has been developed for the quantitative analysis of these compounds 1 (0.74%),2 (0.09%), 3 (0.19%), 4 (0.19%), 5 (0.08%),6 (0.08%), 7 (0.41%), 8 (0.05%), 9 (0.50%),10 (0.59%), and 11 (0.19%).

  • Phytochemical investigation of the MeOH extract of the leaves of Nelumbo nucifera resulted in the isolation of five norsesquiterpenes, four flavonoids, two triterpenes and one alkaloid. Their chemical structures were characterized by spectroscopic methods to be (E)-3-hydroxymegastigm-7-en-9-one (1), (3S,5R,6S,7E)- megastigma-7-ene-3,5,6,9-tetrol (2), dendranthemoside B (3), icariside $B_2$ (4), sedumoside $F_1$ (5), luteolin (6), quercetin 3-0-${\beta}$-D-glucuronide (7), quercetin 3-0-${\beta}$-D-glucoside (8), isorhamnetin 3-0-rutinoside (9), alphitolic acid (10), maslinic acid (11), and N-methylasimilobine (12). Norsesquiterpenoids (1-5) and triterpenes (10-11) were isolated for the first time from this plant. Compounds 6 and 10-12 exhibited considerable cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.
