알로에 생장점 배양시 식물체 재분화에 미치는 Polyamine, 염류농도, 당 및 Gelling Agent의 효과 (Effect of Polyamines, Salt Strength, Sucrose, and Gelling Agents on plant Regeneration from Meristem Culture of Aloe spp.)

  • This study was carried out to investigate the effect of polyamines, salt strength. sucrose and gelling agents on the regeneration of plantlets by meristem culture of Aloe arborescens Mill. and Aloe vera L.. Shoot multiplication was more effective when 10mg/ l spermine in Aloe arborescens and 1mg/ l spermidine in Aloe vera added into MS medium than when other polyamines were treated into media. A quarter strength of MS medium was effective for rooting of shoots regenerated. Higher concentration of sucrose (45g/ l) was more effective for shoot regeneration. Addition of 4g/ l gelrite into the medium was effective for induction of multiple shoots from Aloe than that of agar or other concentrations of gelrite. When plantlets regenerated from meristem culture were transferred to pot. survival rate of plantlets was 80% on perlite and was 95% on vermiculite. respectively.

Aloe vera가 항암제의 세포독성에 미치는 영향 (Effects of Aloe vera on the Cytotoxicity of Anticancer Drugs in Vitro)

  • We investigated effects of methanol extract of Aloe vera on anticancer drugs(cisplatin, mitomycin C, 5-fluorouracil)-induced growth inhibition in p388, L1210, HCT-15, SK-HepG-1 as cancer cell lines and mouse splenocytes as a normal cell by MTT assay, respectively. We also examined the effects of aloe extract and mitomycin C on the mitogen(Con, A, LPS)-induced splenocyte proliferation. Aloe extract(0.25 mg/m , 1.25 mg/m , 2.5 mg/m , 5.0 mg/m ) showed dose-dependently selective cytotoxicity against the cancer cell lines. In contrast, Aloe extract increased the growth and proliferation of the normal mouse splenocytes. The combination of aloe extract with anticancer drugs showed an additive effect for the cytotoxicity against cancer cell lines. However, that combination reduced clealy the anticancer drugs-induced toxicity against the normal mouse splenocytes.

알로에 겔의 in vitro 생리기능 특성에 미치는 삼투탈수공정의 영향 (Effect of DIS Process on in vitro Physiological Functionality of Aloe vera Gel)

  • In vitro physiological functions such as jack bean (Canavalia ensiformis) urease inhibitory activity and retarding effect of glucose/bile acid of Aloe vera gel concentrated by the optimized DIS (Dewatering Impregnation & Soaking) process conditions were examined. Urease inhibitory activity of DIS aloes ranged from 84.6 to 94.4%, which was similar to or higher than 86.3% of fresh aloe. Also, urease inhibitory activity of DIS aloes was maintained at initial levels after heat treatment (90$^{\circ}C$, 10 min.) and drying treatment (freeze or hot air drying). Urease inhibition pattern from Lineweaver-Burk plot indicated general non-competitive inhibition, and inhibition constants ($K_{IE}$ and $K_{IES}$) of DIS aloes were 41-149 and 87-163 $\mu$L/mL, respectively. DIS(glucose) and DIS(polyethylene glycol) exhibited the highest retarding effect of glucose and bile acid. Their retarding effects were about 1.6 and 1.8 folds higher than that of fresh aloe after 0.5 and 1 hr of the dialysis, respectively. Conclusively, the above in vitro physiological functions of Aloe vera gel concentrated by DIS process suggested that aloe products treated with DIS would have the potential benefits for protection against Helicobacter pylori and reduction of blood glucose and cholesterol levels.

알로에(Aloe vera L.) 분말을 첨가한 쿠키의 품질 특성 (Quality Characteristics of Cookies with Aloe vera L. Powder)

  • Aloe is an attractive potential food source because it has functional uses. It has been widely used as a well-being food and drink. The antioxidant activity was estimated by measuring DPPH free radical scavenging activity. We investigated the quality characteristics of cookies prepared by adding various concentrations of aloe powder (0%, 1%, 3%, 5%, all w/w) as a substitute for flour. In this study, the DPPH free radical scavenging activity, moisture content, and hardness of cookies significantly increased with increasing aloe powder, while the pH, spread factor and L and b values slightly decreased. Sensory evaluation scores in terms of texture were shown to be very similar statistically, but color, flavor, taste, and overall acceptability scores were lower than the control. As a result of this study, the quality of cookies with the addition of 1% aloe powder were the most suitable in terms of taste, flavor, texture, and sensory properties.

알로에 원액 첨가 설기떡의 저장기간에 따른 품질 특성 (Quality Characteristics of Sulgitteok Prepared with Aloe Vera Sap during Storage)

  • 알로에 베라 원액 첨가량을 달리하여(0, 3, 6, 9, 12%) 제조한 알로에 설기떡을 4일간 저장하면서 기계적, 관능적 품질특성을 평가하였다. 수분함량은 제조 직후 대조군이 33.79%였고, 알로에 원액 12% 첨가군에서는 다시 감소하는 현상을 보였다. 저장기간별로는 대조군과 첨가군 모두 저장기간이 길어질수록 수분함량은 감소하였다. 물성측정에서는 응집성, 탄력성, 점착성, 부착성, 씸힘성은 알로에 원액 첨가량이 증가할수록 감소하는 경향을 보였다. 색도에서 L값은 설기떡에 첨가한 알로에 원액의 양이 많을수록 유의적으로 감소하였고, a값의 경우 제조 직후 대조군은 -0.56이었으나, 알로에 원액 첨가량이 늘어날수록 음의 값을 나타내는 것을 볼 수 있었고, b값은 알로에 원액 첨가량에 따라 유의적으로 증가하였다. 관능검사에서는 색, 향, 맛, 부드러움, 촉촉함 및 전반적인 기호도에서 모두 9% 첨가군이 가장 높은 것으로 나타났을. 따라서 설기떡에 알로에 원액을 사용함으로써 떡의 품질특성이 향상되어 건강 기능성 상품개발이 가능할 것으로 예상된다.

알로에 베라(A. vera)와 알로에 사포나리아(A. saponaria)로 부터 효모의 분리 및 계통분석 (Selective Isolation and Phylogeny of the Yeast Species Associated with Aloe vera and Aloe saponaria)

  • 효모의 유용 기능을 탐색하기 위해서는 효모만을 스크리닝 하는 방법의 정립이 요구되는데 이를 위해서 알로에에 정착하는 효모 군집을 분석하였다. 본 연구에서는 알로에 베라(Aloe vera)와 알로에 사포나리아 (Aloe saponaria)의 잎을 파쇄하여 세균을 억제하기 위해서 항생제 chloramphenicol과 streptomycin과 곰팡이 생장을 억제하기 위해서 Triton X-100과 L-sorbose를 포함한 4가지 선택 배지(DOB with CS, GPY, YM, SCG)에 도말 하였고, A. vera에서 총 67균주, A. saponaria에서 총 42균주를 분리해 내었다. 분리된 균주를 ITS 1, 4 primer를 사용하여 sequence의 계통분석을 실시한 결과, A. vera에서는 Meyerozyma 가 56균주, Rhodotorula 가 1균주, Sporobolomyces 가 1균주, Cryptococcus 가 9균주, A. saponaria에서는 Rhodosporidium 가 41균주, Sporobolomyces가 1균주로 분포 되었다. 계통분석한 모든 분리 균주가 효모로 밝혀졌으며, 특히 대형 플레이트를 도입하여 신속하고 간편하게 대량의 효모를 발굴할 수 있음을 보였다.

한외여과 알로에 농축액의 Urease 저해 및 무기물 응집 활성 (Urease Inhibition and Flocculating Activity of Concentrated Aloe vera Gel by Using Ultrafiltration Process)

  • For physiological function of aloe concentrate by ultrafiltration (UF) process, jack bean urease inhibitory activity and bentonite flocculating activity of UF aloe concentrate was investigated and compared with fresh aloe gel. Urease inhibitory activity of UF aloe concentrate ranged from 87 to 90% in 1 mL sample. Also, urease inhibitory activity of UF aloe concentrate increased about 10% by heat treatment showing the heat stability. From Lineweaver-Burk plot for UF aloe concentrate, urease inhibition pattern indicated general non-competitive inhibition. From flocculation test of UF aloe concentrate about 1% (w/v) bentonite suspension, maximum flocclulating activity of 97% was obtained at 0.5 mL addition of UF aloe concentrate/ 5 ml bentonite suspension. However, flocculating activity of 81% was obtained at 1 mL addition of UF aloe concentrate/ 5 mL bentonite suspension, which was typical flocculating behavior of polymers with re-dispersion at overdose area. FT-IR spectra of UF aloe concentrate showed the characteristic patterns of $\beta$-binding polysaccharide and less deacetylation indicating higher level of bioactive polysaccharide content.

알로에 배라 점질물의 무기물 응집활성 (Flocculating Activity of the Mucilage Extracted from Aloe vera Linne)

  • For the purpose of screening the functionality in aloe, the mucilage from Aloe vera Linne was prepared by extraction of mucilaginous parenchyma with ethanol followed homogenation, filtration and centrifigation. Flocculability of this mucilage from Aloe vera L. was investigated and the identification fro flocculating active materials were carried out. The mucilage exhibited an excellent flocculability for 1% bentonite and 5,000 ppm kaolin suspensions. The good correlations between mucilage volume(concentration) and flocculability for 1% bentonite suspension were obtained. The flocculated volume or flocculation rate was affected depending upon the degrees of processing such as a heat treatment and purification. A major component showing the flocculaing activity was considered as a highly polymerized polysaccharide having flocculating activity was about 1,000,000 daltons.

  • Aloe vera L.(Aloe barbadensis Miller) has been used for many centuries for various medical, cosmetic and neutraceutical purposes. The gel of A. vera was reported to exert numerous biological activities including wound healing, immunomodulatory, anti-cancer, anti-inflammatory, anti-bacterial, anti-viral, anti-diabetic, and anti-psoriasis activities. Acemannan, found predominantly in the inner leaf gel of A. vera, has been identified as the main active ingredient exerting diverse biological activities. In this review, we summarized the recent findings on the immunomodulatory activities of acemannan. Studies that used purified acemannan demonstrate that acemannan exerts immune-stimulating, anti-cancer, anti-viral, and hematopoiesis-increasing activities. In addition, it is being clear that most of these activities of acemannan are mediated primarily by activating professional antigen presenting cells such as macrophages and dendritic cells.

전처리 농축 정도에 따른 Aloe Vera gel의 동결건조분말의 물성 (Physical Properties of Freeze-Dried Powder of Aloe Vera Gel with Respect to the Concentrating Degree as Pre-Treatment)

  • 농축시킨 Aloe vera gel을 동결건조하였을 때 그 농축도 차이에 의한 최종 건조분말의 물성 변화를 비교분석하였다. 분말의 최종 수분함량은 거의 차이가 없었으며. 점성 측정에서는 높은 농축 조건의 경우 전형적인 shear thinning 현상과 non-Newtonian 유체의 속성을 나타냈다. 이에 비하여 낮은 농축 조건의 경우 Newtonian 액체의 성질과 고형분이 입자가 아닌 풀린 형태로 존재할 때 나타나는 현상을 보였다. 분말이 물에 용해되는 동안 전기전도도의 변화를 실시간으로 측정한 결과 평형상태에 도달하였을 때, 가장 높은 농축 조건에서 전기전도도가 가장 낮게 나타나, 비전해질성 물질의 용해도가 가장 큰 것으로 해석되었다. 분말의 등온흡습성에서는 농축이 증가할수록 수분 흡착과 결합수의 양이 작아지는 것으로 분석되었다. 결과적으로 Aloe vera gel을 농축하여 동결건조할 때는 그 농축도에 따라 점성, 현탁시 전기전도도에 따른 용해성, 등온흡습성이 변하는 것으로 나타나, 특정 품질의 동결건조제품을 얻기 위해서는 그에 대한 농축의 최적화가 필요한 것으로 생각된다.
