Phylogenetic Analyses of Nuclear rDNA ITS Sequences of Korean Allium L. Subgenus Rhizirideum(Alliaceae)

  • Phylogenetic relationships among the Korean taxa of the genus Allium subgenus Rhizirideum and some related taxa were assessed on the basis of in sequences of nuclear ribosomal DNA. Twenty-eight accessions of the genus Allium L. consisting of subgenera Rhizirideum (19 taxa), Allium (5 taxa) and Amerallium (one taxon) were analyzed. The variation in the ITS region was informative at the levels of section except for sect. Reticulato- bulbosa which is known to be of multiple origin. The ITS 2 region was longer than the ITS 1 region, and all of the investigated Allium taxa were the same in length in the 5.8S region except for A. monanthum. Allium cyaneum var. cyaneum was the shortest (635 bp) and A. victorialis the longest (646 bp) among the investigated Korean taxa. The three morphologically similar taxa, A. thunbergii, A. sacculiferum that has been included in A. thunbergii, and A. deltoid- fistulosum, had the same ITS lengths of 641 bp, but were clearly distinguished in the phylogenetic analysis of their ITS sequences.

First report of white rot on a wild gu1ic(Allium monanthum) caused by Sclerotium cepivorum and Sclerotium sp.

  • White rot on garlic caused by Sclerotium cepivorum firstly occurred at Goheoung, Jeonnam in 1998. Thereafter, the disease rapidly spread throughout the country except Gangwon and became a major limiting factor for the cultivation of various Allium species such as garlic, onion, and welsh onion. The disease that has not been reported on a wild garlic(Allium monanthum) previously occurred severely at Seosan, Choongnam in 2003. Among cultivation areas in the region, 10.7% were infected by the disease and the ratio of diseased plant reached up to 55.0% in some heavily infected fields. Two species of Sclerotium were consistently isolated from infected samples and identified as S. cepivorum or another Sclerotium sp. Averaged size of sclerotium of the former was 455.0x562.2 urn, while the later was 374.4${\times}$347.2$\mu\textrm{m}$. Patogenicity to Allium species and mycological characteristics such as sclerotium size, growth temperature, and microconidia of the fungi were similar to those reported on other Allium species previously. Consequently, the wild garlic is a newly reported host of the two pathogenic fungi in Korea.

Effects of Physical and Chemical Treatment as the Pretreatments on Microorganisms and Quality Characteristics of Allium monanthum (전처리 방법이 달래의 품질 특성과 미생물 저감에 미치는 영향)

  • The purpose of this study was to investigate the effect of the microbial reduction and quality maintenance of the physical and chemical pretreatment of Allium monanthum. For physical treatment, handwash, bubble wash and ultrasonication were conducted at 50℃ and 60℃ for 1, 3 and 5 minutes, respectively, and for chemical treatment the sample was immersed in fumaric acid and acetic acid of 1.5% and 2% concentrations for 1, 3 and 5 minutes, respectively. As a result of the microorganism and quality analysis, 3 minutes of bubble wash was the most effective physical pretreatment in reducing fungi although the effect on reducing total viable bacterial was small. Furthermore, 5 minutes of ultrasonication at 60℃ significantly reduced microorganisms, but also resulted in the reduction of the a value of chromaticity, which cause the green color to fade. With chemical pretreatment, it was found that treating with fumaric acid was more effective in reducing the total viable bacteria and fungi than acetic acid. The result shows that 1.5% concentration of fumaric acid is the most effective with 3 minutes of treatment time. The quality of Allium monanthum were compared in the combination of the two most effective microorganism reduction pretreatments: 3 minutes of bubble wash (B3) and 3 minutes in 1.5% fumaric acid (F153). As a result of analyzing the quality characteristics over 9 days of storage at 4℃ after the treatments, it was revealed that the BF treatment is more effective in reducing fungi than the total viable bacteria. The results shows that the BF treatment is more effective in reducing total viable bacteria, whereas the F153 treatment is more effective in reducing fungi. Also, it was found that the 𝚫E value in BF was the lowest, whereas F153 treatment showed the green color faded. The maximum cohesiveness changed more significantly in the green stems than in the roots. On the 9th day of storage, the hardness of the green stem was found to be maintained at the highest level (P<0.05) after F153 treatment, whereas that of the roots decreased (P<0.05) since the 6th day after the bubble wash. Considering the reduction of microorganisms and the quality maintenance of Allium monanthum, the most effective pretreatment methods were 3 minutes in 1.5% fumaric acid for reducing microorganisms and maintaining color and maximum cohesiveness, and the combined process could also be effective if the expiration period is within 3 days.

Comparison of characteristic aroma compounds in Korean wild chive (Allium monanthum Maxim.) cultivated in open-fields or greenhouses (노지와 시설 재배 달래의 특징적인 향기 성분 비교)

  • The objective of this study was to analyze volatile and aroma-active compounds in Korean wild chive (Allium monanthum Maxim.) cultivated in open-fields or greenhouse systems using solvent-assisted flavor evaporation-gas chromatography (GC)-mass spectrometry and GC-olfactometry. Aroma-active compounds were evaluated using aroma extract dilution analysis (AEDA). Twenty-two aroma-active compounds with log2 flavor dilutions (FD) of 1-10 were detected in Korean wild chive, which was cultivated in an open-field or a greenhouse. 2-Isopropyl-3-methoxypyrazine ("earthy"), 2-sec-butyl-3-methoxypyrazine ("earthy", "musty"), and dipropyl disulfide ("sulfurous") were the most predominant aroma-active compounds with log2FD of 9-10; this was followed by dimethyl trisulfide ("onion-like") and (E)-1-propenyl propyl disulfide ("fresh onion-like"). The "sulfurous", "earthy", "pungent", and "cabbage-like" aroma notes were strong in Korean wild chive. More intense "pungent" odors were detected in Korean wild chive cultivated in an open-field, whereas more intense "cabbage-like" odors were detected in Korean wild chive cultivated in a greenhouse.

Antimicrobial Activity on Respiration Diseases Inducing Bacteria and Antioxidant Activity of Water Extracts from Wild Edible Vegetables (산채류 추출물의 항산화 활성 및 호흡기 질환을 유발하는 세균에 대한 항균활성)

  • To investigate the antimicrobial activity on bacteria causing a respiration disease and antioxidant effects of water extracts from 12 kinds of wild edible vegetables, we extracted the water extracts for 72 h in $7^{\circ}C$ using distilled water as solvent. The water extracts except Ixeris dentate and Allium monanthum had high concentrations of phenol compounds and flavonoids. Liguraia fischeri specially had the highest level on total phenol compounds and flavonoids with 205 ${\mu}g/mL$ and 98. 86 ${\mu}g/mL$, respectively. The each 0.05% extracts of Sedum sarmentosum and Liguraia fischeri had high effect on the DPPH radical scavenging activity among wild edible vegetables and the most extracts promoted antioxidant activity with increasing concentration of extract. The catalase activity of Erysimum aurantiacum and Aralia elata showed more than 150 units per g of fresh tissue. The effect of antimicrobial activity on water extracts showed characteristic activity. Only Staphylococcus aureus KCTC 1928 and Corynebacterium diptheriae KCTC 3075 were inhibited cell growth on the other hands, the remainder of bacteria was not inhibited cell growth. Nevertheless, the extracts of wild edible vegetables had specific concentration as MIC for antimicrobial activity respectively. In case of the extract of Aster scaber, Erysimum aurantiacum, and Allium monanthum had over 30% antimicrobial activity on the bacteria causing a respiration disease. In results, the wild edible vegetables include high concentrations of total phenol compounds and flavonoids that give a good antioxidant activity and antimicrobial activity. Therefore the wild edible vegetables are functional food for anti-aging and physiological activation.

Antioxidative Activities and Inhibitory Effects on Tyrosinase and Elastase by Water Extracts of Allium sp. (Allium 속 식물 물 추출물의 항산화 활성과 Tyrosinase 및 Elastase 저해 효과)

  • Three different Allium plants, including wild garlic (Allium monanthum Max; AMM), onion (Allium cepa L.; ACL) and green onion (Allium fistulosum L; AFL), have been widely used as spicy food in daily life. This study was attempted to investigate the properties of antioxidant, whitening and anti-wrinkle activities in water extracts in Allium plants. The antioxidant, whitening and anti-Wrinkle activities were evaluated by performing DPPH and ABTS radical scavenging activities, inhibitory effects on tyrosinase, and elastase, respectively. The water extract (10 mg/mL) of onion, wild garlic and green onion exhibited DPPH radical scavenging activities of 28.46%, 28.45%, and 15.91%, respectively. The inhibitory effects of tyrosinase activities by wild garlic, green onion, and onion were increased by heating the extracts at $90^{\circ}C$. Additionally, the wild garlic extract showed higher elastase inhibitory activity than those of other plant extracts. These results suggest that water extracts of Allium plants may be useful as potential sources of beauty foods due to higher antioxidant, anti-melanin and anti-wrinkle activities.

A Taxonomic Study on Korean Allium L. Based on the Morphological Characters (형태학적 형질에 기초한 한국산 부추속의 분류학적 연구)

  • For 20 taxa of Korean Allium, including 16 species and 5 varieties, examined were morphological characters (i.e. sexuality, structure and shape of underground part, types and growing patterns of leaf and scape, and shapes of perianth, filament and pistil). The specialization and the evolutionary trends of taxonomic characters were inferred from morphological examination. Taxonomic relationships and system of Korean Allium were also studied. The characters showing evolutionary trends were the structure and shape of underground part including rhizome and bulb, leaf, scape, inflorescens, filament and ovary. It seemed that thin and short rhizome developed from thick and long one, and the membranous simple bulb tunic evolved into fibrous reticulate one. The presence of hyaline sheath in A. monanthum of sect. Microscordum was apomorphic. Both angular leaf blade with 2-rowed vascular bundle and flat blade with 1-rowed vascular bundle were developed from terete one with 2-rowed vascular bundle. The base of filament have differentiated from entire to toothed, and 2-ovuled ovary as well as erect scape before flowering was plesiomorphic type. In addition, sexuality, structure of underground part, the presence of hyaline sheath and cross-section structure of leaf were taxonomic characters with the level of the subgenus in this genus. Shape of rhizome, bulb, leaf, scape and pedicel discriminated each section from other ones in the subgenera easily. The shape and arrangement of perianth and filaments were the diagnostic characters for species level along with shape of ovary and stigma.

Basic Research of Development for Food of Seedling in Some Plants (몇가지 식물에 있어서 어린순의 식품학적 가치개발을 위한 기초연구)

  • This experiments were carried out to develop for food in some plants seeding. Plant materials were Raphanus sativus, Perilla frutescens var. acuta, Perilla frutescens var. japonica, Scutellaria baicalensis, Angelica dahurica, Angelica gigas, Ligusticum acutilobum, Peucedanum japonicum, Platycodon grandiflorum, Codonopsis lanceolata, Allium monanthum and Fagopyrum esculentum.   • To study effects of wild garlic on components of lipid and protein, blood glucose level, and cholinesterase activity in rat's serum, Sprague-Dawley strain of 40 male rats are divided into 5 groups which are Normal, Control, A, B, and C group. Normal group is fed only Basal diet, and Control group is fed Basal diet and 0.5ml/day of 2.5% cholesterol solution, In addition to a diet of control group, A group is fed 25% wild garlic flour, B group 0.5ml/2day of concentratded wild garlic juice, C group 0.5ml/2day of concentrated ethanol extract of wild garlic, After 8 weesk the rats were fasted for 12 hours, and then decapitated to collect blood. The results of analysis of the rat's serum were summarized as follows, 1. Wild garlic diet, specially ethanol extract has an influence on decreasing the level of total cholesterol and blood glucose in rat's serum, 2. HDL-cholesterol and phospholipid content of rat's serum are increased by wild garlic diet. Therefore I think that wild garlic is good food for preventing development of atherosclerosis and diabetes.

Dietary Fiber Contents and Physical Properties of Wild Vegetables (산채류의 식이섬유 함량과 물리적 특성)

  • Nine wind vegetables were analyzed for moisture, ash, crude protein, crude lipid and dietary fiber. Wild vegetables contained 33-53% of dietary fiber on a dry weight basis. Dalle (Allium monanthum) contained 49% total dietary fiber and 22% soluble dietary fiber and dodok(Codonopsis lanceolata) contained 55% total dietary fiber and 21% soluble dietary fiber. Wild 8% more dietary fiber than cultivated one. Water holding capacities of wild vegetables were higher than commercial wheat bran and soy fiber, but lower in oil absorption. When wild dodok and dalle were wet milled by blade grinding before sieving the dietary fiber content in dodok was increased from 55 to 83 % with increasing the dietary fiber content in dalle form 49% to 69%.
