Molecular and Biochemical Characterization of Opisthorchis viverrini Calreticulin

  • Calreticulin (CALR), a multifunctional protein thoroughly researched in mammals, comprises N-, P-, and C-domain and has roles in calcium homeostasis, chaperoning, clearance of apoptotic cells, cell adhesion, and also angiogenesis. In this study, the spatial and temporal expression patterns of the Opisthorchis viverrini CALR gene were analyzed, and calcium-binding and chaperoning properties of recombinant O. viverrini CALR (OvCALR) investigated. OvCALR mRNA was detected from the newly excysted juvenile to the mature parasite by RT-PCR while specific antibodies showed a wide distribution of the protein. OvCALR was localized in tegumental cell bodies, testes, ovary, eggs, Mehlis' gland, prostate gland, and vitelline cells of the mature parasite. Recombinant OvCALR showed an in vitro suppressive effect on the thermal aggregation of citrate synthase. The recombinant OvCALR C-domain showed a mobility shift in native gel electrophoresis in the presence of calcium. The results imply that OvCALR has comparable function to the mammalian homolog as a calcium-binding molecular chaperone. Inferred from the observed strong immunostaining of the reproductive tissues, OvCALR should be important for reproduction and might be an interesting target to disrupt parasite fecundity. Transacetylase activity of OvCALR as reported for calreticulin of Haemonchus contortus could not be observed.

Association Between Protein Intake From Different Animal and Plant Origins and the Risk of Non-Alcoholic Fatty Liver Disease: A Case-Control Study

  • Previous studies have frequently reviewed how different macronutrients affect liver health. Still, no study centered around protein intake and the non-alcoholic fatty liver disease (NAFLD) risk relationship. This study aimed to examine the association between the consumption of total and different sources of protein and NAFLD risk. We allocated 243 eligible subjects to the case and control groups, including 121 incidence cases of NAFLD, and 122 healthy controls. Two groups were matched in age, body mass index, and sex. We evaluated the usual food intake of participants using FFQ. Binary logistic regression was conducted to estimate the risk of NAFLD in relation to different sources of protein intake. The age of participants was 42.7 years on average, and 53.1% were male. We found Higher intake of protein in total (odds ratio [OR], 0.24; 95% confidence interval [CI], 0.11-0.52) was significantly associated with a lower risk of NAFLD, despite adjusting for multiple confounders. in detail, higher tendency to the vegetables (OR, 0.28; 95% CI, 0.13-0.59), grains (OR, 0.24; 95% CI, 0.11-0.52), and nuts (OR, 0.25; 95% CI, 0.12-0.52) as the main sources of protein, were remarkably correlated with lower NAFLD risk. In contrary, increased intake of meat protein (OR, 3.15; 95% CI, 1.46-6.81) was positively associated with a higher risk. Totally, more calorie intake from proteins was inversely associated with lower NAFLD risk. This was more likely when the protein sources were selected less from meats and more from plants. Accordingly, increasing the consumption of proteins, particularly from plants, may be a good recommendation to manage and prevent NAFLD.

  • Authors tested the grid functions with various thickness of acryl phantom, radiation field size and diameter of shielding lead. The results are as followed: 1. The characteristic values of grid are affected by phantom thickness, but free from radiation field size in the diagnostic useful range. 2. The quantity of scattered radiation was decreased according to the diameter of shielding lead under 20mm, and then the diameter the smaller the better, in accordance with proposed KS standard. 3. The quantity of primary radiation was increased a little at the 80mm diameter radiation field size, but did not have much differences. Therefore, it was thought that it is needed to limit beam size in case of absolute values in accordance with any standards, but it has no matter to use 100 mm diameter in case of relative values just to campare with.

  • This study was conducted to find out the management status of radioisotopes and radioactive waste products in Korean medical institutions during the period of three years from 1991 to 1993. The results are summarized as follows : 1. The rate of medical institution to the institutions making use of radioisotopes was decreased every year, for example, 18.11 % in the year 1991, 17.86 % in the year 1992 and 15.87 % in the year 1993. 2. The use of domestic made radioisotopes in the medical institution was increased every year, for instance, 89.68 % in the year 1991, 94.21 % in the year 1992 and 99.79 % in the year 1993. 3. 91.01 % of the half life of isotopes used in the medical Institution were two month below. 4. The rate of radioactive waste products in the medical institution to all radioactive waste products was increased every year, for example, 54.44 % in the year 1991, 75.36 % in 1992 and 78.49 % in the year 1993. 5. The rate of inflammable waste products from medical institution was 76.47 % of all inflammable waste products, and 73.99 % of whole waste products was from the medical Institution.

  • As conventional drug delivery system is being improved rapidly by target-based drug delivery system, finding suitable Drug Delivery System (DDS) for new drugs remains a challenge. Although there are many drug delivery vehicles in existence, a significant improvement is required to some DDS such as for local, implant-based treatments used for musculoskeletal infections. Many polymers have been considered for providing the improvement in DDS. Synthetic polymer, for example, has gained popularity for broad-spectrum physicochemical and mechanical properties. This article reviews the biomedical applications of Poly(TriMethylene Carbonate-co-Caprolactone) (PTMCC), which has attracted attention due to its biocompatibility, biodegradability and rubber-like properties. Its synthesis, physical properties, and degradation are also discussed here. Although it is relatively new in biomedical applications, it is readily usable for the fabrication of differing format of DDS of superior mechanical strength and degradation properties. The use of PTMCC is expected to increase in coming years as more is revealed about its potentials.

  • This study was conducted to find out the indoor radon concentrations from Jan. 1, to Dec. 31, 1995 in Seoul, and the following results were achieved; 1. The average concentration of indoor radon ranged from $0.51pCi/\ell$ to $0.78pCi/\ell$. 2. The correlation coefficients(r) of radon concentration and indoor meteorological conditions were as follows; 1) temperature : r=0.11 2) atmospheric pressure : r= -0.01 3) humidity : r=0.227.

  • Authors could recognize surrounding areas induding target area by the observable film density with spot radiography using Cu mask and also, radiographic images maintained high quality. Spot radiography has been utilized for a long time for the purpose of observing detail image by the control of scattered radiation. But it has been difficult to know body part in general spot radiography. Therefore it is thought that attempt to use Cu mask in spot radiography is very advantageous in a clinical examinations.

  • The purpose of this study is to find terms that can give identity to the major and occupation of clinical laboratory technologist (also known as medical technologist). The term clinical pathology includes all branches of pathology, namely anatomical pathology, chemical pathology, hematology, microbiology, and all respective subspecialties. Unfortunately, several countries exclude anatomical pathology from the term clinical pathology, a problem that gets compounded when the title is translated into languages other than English. Clinical pathology (US, UK) is a medical specialty. Similar terms are laboratory medicine (Germany, Poland), medical/clinical biology (France, Netherlands) or clinical analysis (Spain). Depending on the person questioned, medical technology is defined slightly differently by individuals, companies, and institutions. The definition also depends on the language in which the question is asked. Medical technology can be translated to define clinical laboratory technology, allied health sciences, medical equipment, biomedical engineering, and health technology. The terms 'clinical pathology technology and pathological technology' are not used in allied health sciences. The names of 'medical technology·medical technologist' can be replaced by 'biomedical laboratory science·biomedical laboratory technologist' or 'clinical laboratory analysis·clinical laboratory analyst'. In this study, it is proposed to change the name of academic and occupation to 'medical biology·medical biology technologist' that combines the term biomedical.

The Education System for Cytotechnologists and Efforts to Improve Professionalism in the United States of America (미국 세포병리사 양성 교육제도와 전문성 향상을 위한 노력)

  • The path to becoming a cytotechnologist (also known as a cytologist) in the United States of America (USA) requires a minimum of a bachelor's degree and the completion of a training program certified by the Commission on Accreditation of Allied Health Education Programs. Aspirants are recognized as cytologists after passing a test administered by the American Society for Clinical Pathology Board of Certification on completion of a recognized cytotechnology training program. Cytotechnology training programs are currently provided by 18 universities and hospitals. Six of them provide master's level education, unifying existing programs to the master's level, which is necessitated due to a shortage of pathologists and the rapid development of the health care system. The improvement and unification of the curriculum, qualification renewal system, and the effort to change the title of qualified personnel from cytotechnologist to cytologist are seen as measures that would strengthen the capabilities of cytotechnologists as well as assist in keeping pace with the rapidly changing medical system. Knowledge about the educational courses available for cytotechnologists and the efforts to strengthen their expertise in the USA would be helpful to professors who guide students preparing for overseas employment and seeking professional improvement in Korea.

Studies on the Properties of Enzymatic Hydrolysates from File-fish (말쥐치 단백의 효소 가수분해물의 특성에 관한 연구)

  • The purpose of this study was to elucidate characteristics of hydrolysates from file-fish flesh with various proteases. File-fish flesh was chopped, homogenized with water, and hydrolysed by 8 different kinds of commercially available protease. High production of peptide was observed in bromelain and neutrase treatment. On the other hand, large amount of free amino acid was observed in esp/sav and pronase treatment. Neutrase and pancreatin hydrolysate contained large amount of 5'-GMP. Organoleptic studies showed that the bromelain, esp/sav and protease hydrolysate had strong bitter taste, while pronase and esp/sav hydrolysate had strong umami taste. From these results, pronase was found to be suitable enzyme for producing file-fish hydrolysate.
