Study on Anti-Allergic Effecst of Ganoderma lucidum Herbal Acupuncture and Ganoderma lucidum Extract (영지(靈芝) 약침(藥鍼)과 영지(靈芝) 추출액의 항알레르기 효과에 대한 연구)

  • Objectives We studied on anti-allergic effects of Ganoderma lucidum herbal acupuncture(GHA) and Ganoderma lucidum extract(GE). Methods In vivo, Animals were herbal-acupunctured GHA at both B13s three times for 5 days. Then, we investigated compound 48/80-induced active systemic anaphylatic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis using Sprague Dawley rat. In vitro, we measured cell viability, b-hexosaminidase release, IL-4 and TNF-a from RBL-2H3 cells, and nitric oxide from Raw264.7 cell after treatment of GE of various concentrations. Results In vivo, GHA pretreatments at both B13s inhibited compound 48/80-induced active systemic anaphylatic shock. Passive cutaneous anaphylaxis were inhibited by GHA10 and OP. In vitro, $0.1\;{\sim}\;2%$ GE treatments were not affect on cell viability and inhibited b-hexosaminidase release, IL-4, TNF-a and nitric oxide. Conclusions These results suggest that GHA and GE may be beneficial in the inhibition of allergic inflammatory response.

Inhibition of mast cell-mediated immediate-type allergic reactions by Bojungikgitanggamibang (보중익기탕가미방(補中益氣湯加味方)에 의한 비만(肥滿) 세포(細胞) 매개성(媒介性) 즉각형(卽刻型) 알레르기 반응(反應)의 억제(抑制))

  • Objective : Mast cells are a potent source of mediators that regulate inflammatory response in allergies and asthma. The author studied the effect of Bojungikgitanggamibang(BITB) on mast cell-mediated anaphylactic reaction. Method : When BITB was given as pre-treatment at concentrations ranging from 0.01 to 1 mg/ml, the histamine release from rat peritoneal mast cells induced by compound 48/80 was reduced in a dose-dependent manner. Result : BITB dose-dependently inhibited compound 48/80-induced systemic anaphylactic shock. BITB also inhibited passive cutaneous anaphylaxis activated by anti-dinitrophenyl IgE. In addition, BITB inhibited phorbol 12-myristate 13-acetate and A23187-induced interleukin-6 secretion from human mast cell line HMC-1 cells. Conclusion : These results indicate that BITB may be actively anti-allergic.

  • The current study evaluates the capacity of eugenol to regulate immediate allergic reaction by control of histamine release. Administrations of eugenol (1M/kg, i.p.) at 60 min b efore and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) were shown the mortality rates as 0, 44.4, and 77.8%, respectively. A 60 min before administered group revealed a significant inhibition of serum histamine release compared with those of 5 and 10 min after the compound 48/80 injection. Eugenol (6-48mM) was also showed a dose-dependent activity on the compound 48/80-induced histamine release from the highly purified population of Alcian Blue-positive peritonea] mast cells. These results indicate that in vitro treatment with exogenous eugenol inhibited the active response of mast cell populations and modulated its characteristics.

Inhibitory Effects on the Type 1 Hypersensitivity and Inflammatory Reaction of Herba Patriniae Aqua-acupuncture (패장약침(敗醬藥鍼)이 면역반응(免疫反應)에 미치는 영향)

  • Objective : Experimental studies were done to research the clinical effects of Herba Patriniae(HP) aqua-acupuncture ($BL_{13},\;BL_{17},\;BL_{13}{\cdot}BL_{17}$ and free points) on the anti-allergic inflammatory response. Methods : We measured active systemic anaphylatic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. And we measured total IgE and plasma WBC level, serum total protein, albumin, immunoglobulin and NO levels induced by egg albumin. Results : HP aqua-acupuncture pretreatments at all acupoints inhibited active systemic anaphylatic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. Total IgE and plasma WBC level inhibited by HP aqua-acupuncture pretreatment at $BL_{13}\;BL_{17}$ and free points. However, HP aqua-acupuncture didn't effect serum total protein, albumin, immunoglobulin and NO levels. Conclusion : These results suggest that HP aqua-acupuncture may be beneficial in the regulation of type Ⅰ allergic reaction, but is further required immunological studies on the allergic reaction.

The Protective Effect of Lentinus Edodes on Mast Cell-Mediated Immediate-Type Hypersensitivity (비만세포 매개 즉시형 과민반응에 대한 표고버섯 추출물의 보호 효과)

  • Mast cells are crucial as effector cells in the immediate-type allergic reaction. Lentinus edodes has been the popular edible mushroom in oriental countries and reported to have immunomodulatory, anti-tumor, anti-atherogenic, anti-viral, and anti-allergic activities. However, the roles of L. edodes in mast cell-mediated anaphylactic reaction have not been fully elucidated. In this research, we have demonstrated the effects of the methanol extract of L. edodes (MELE) on mast cell-mediated anaphylaxis-like and anaphylactic reactions. MELE suppressed systemic anaphylaxis-like reaction, plasma histamine levels, and ear swelling response in mice treated with compound 48/80. MELE also suppressed passive systemic and cutaneous anaphylaxis mediated by anti-dinitrophenyl IgE. In accordance with these findings, MELE dose-dependently decreased histamine release from RPMC evoked by compound 48/80 or the antigen-antibody reaction. To clarify the mechanism of degranulation system, intracellular cAMP levels as well as calcium influx in RPMC was evaluated. In compound 48/80-treated RPMC, MELE blocked calcium uptake into the cells. In addition, MELE elevated the intracellular cAMP content and significantly attenuated compound 48/80-induced cAMP reduction in RPMC. Taken together, we propose the clinical use of MELE in mast cell-mediated immediate-type allergic diseases.

  • Allergic contact dermatitis (ACD) is an allergen-specific T-cell-mediated inflammatory response, albeit with unclear pathogenesis. Exosomes are nanoscale extracellular vesicles secreted by several cell types and widely distributed in various biological fluids. Exosomes affect the occurrence and development of ACD through immunoregulation among other ways. Nevertheless, the role of exosomes in ACD warrants further clarification. This review examines the progress of research into exosomes and their involvement in the pathogenesis, diagnosis, and treatment of ACD and provides ideas for exploring new diagnostic and treatment methods for this disease.

Serum allergen-specific and total IgE concentrations in dogs with allergic disorders in South Korea

  • Canine atopic dermatitis (CAD) has a genetic basis and is primarily a skin disorder characterized by allergic features. It is caused by an excessive immune response, associated with immunoglobulin E(IgE), to allergens and shows clinical signs of itching and inflammation. The present study evaluated the allergen-specific and total IgE concentration in dogs with allergic disorders. Allergen-specific IgE analysis was performed using a Multiple Allergen Simultaneous Test (MAST) assay, and total IgE analysis was conducted using an ELISA in 82 dogs with CAD. The subjects were divided into two groups: dogs aged ≤3 years and dogs aged >3 years, enabling a comparison of total IgE concentrations between the two age groups. The results showed that the major affecting environmental allergen was Alternaria alternate, and the major affecting dietary allergen was raw chestnut. The results revealed that dogs aged ≤3 years had higher total IgE concentrations than dogs aged >3 years with significant difference. The results of this study can contribute to the diagnosis and treatment of CAD in South Korea.

An Experimental Study of Sinichengpaeum(辛荑淸肺飮) on the anti-allergic effect (辛荑淸肺飮의 抗allergy效果에 關한 實驗的 硏究)

  • Sinichengpaeum has been widely used in the oriental medical treatment of nasal diseases. These studies were performed done to investigate the effect of Sinicheng- paeum on the anti-allergic action We studied the vascular permeability response induced by the histamine and serotonin injection(allergy Ⅰ type), homologous PCA provoked by the IgE-like antibody against EWA(allergy Ⅳ type), contact dermatitis induced by picryl chloride(allergy Ⅳ type), delayed type hypersensitivity response to SRBC (allergy Ⅳ type), and the amount of eosinophil and IgE. The results were as follows: 1. The effect of Sinichengpaeum on vascular permeability responses to intradermal histamine and serotonin were significant. 2. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Sinichengpaeum showed a significant effect. 3. In the delayed type hypersensitivity responses to picryl chloride, Sini-chengpaeum provoked a significant effect. 4. After reaction provoked by picryl chloride, Sinichengpaeum showed an insignficant effect on amount of eosinophil, but a significant effect in IgE amount. 5. In the delayed type hypersensitivity responses to SRBC, Sinichengpaeum provoked a significant effect. 6. After reaction provoked by SRBC, Sinichengpaeum showed a significant effect on amount of cosinophil, but an insignificant effect in IgE amount.

  • Experimental studies were done to research effects of Leejungtang and Leejungtanggamibang on the anti-allergic effects. We studied the vacular permeability response induced by histamine and serotonin, homologous PCA provoked by IgE-like antibody against EWA, contact dermatitis induced by picryl chloride and delayed type hypersensitivity response to SRBC. The results were obtained as follows: 1. In the delayed type hypersensitivity responses to picryl chloride, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 5. In the delayed type hypersensitivity responses to sheep red blood cell the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect.

Anti-pruritic Effect of Ethanol Extracts from Platycodon grandiflorum and its fermented production in Scratching Behavior Mouse Models (길경(桔梗)발효 추출물의 알레르기성 소양행동 억제효과연구)

  • Objectives : Platycodon Root is frequently used in traditional medicine to treat inflammatory diseases of the throat. The purpose of this study was to characterize the effect of the EtOH extract of fermented Platycodon grandiflorum on the ameliorative effects on the Antipruritic Effect of atopic dermatitis mouse model induced by compound 48/80 and ovalbumin (OVA)-induced allergic responses in mice. Methods : In the present study, we examined the anti allergic effect of Platycodon grandiflorum (PR) and its fermented production (FPR) in several mouse model. We measured acute ear edema in a mouse model caused by TPA and consecutively histological change of Ear tissue was observed by hematoxylin and eosin (H&E) staining. and also Scratching behaviors by compound 48/80 was investigated. The levels of allergic mediators such as immunoglobulin (Ig) E, and anti-oxidant markers such as SOD and MDA in the sera of OVA induced allergic mice were measured by enzyme-linked immunosorbent assay. Results : FPR inhibited compoud 48/80-induced scratching behavior in mice, as well as acetic acid-induced writhing in mice. The anti-scratching behavioral effect of FPR was more potent than PR. FPR extract significantly decreased the serum levels of IgE and MDA compared with those of OVA control group. Conclusions : These results indicate that Anti allergic effect of Platycodon grandiflorum is enhanced by fermentation with Saccharomyces cerevisae and FPR may be useful for protection from the itching reactions, which are IgE-mediated representative skin allergic diseases.
