어린이의 요 중 비스페놀 A 농도에 근거한 위해성 평가와 알레르기 질환과의 관련성 (Relationship between Risk Assessment Based on Urinary Bisphenol A Concentration and Allergic Diseases in Children)

  • Objectives: The purpose of this study was to examine the relationship between urinary bisphenol A concentration and allergic diseases in children. Methods: This study was conducted in Seoul, South Korea. We collected urine samples from 231 children from a single elementary school in June 2014. Among these, 69 children with urinary creatinine levels outside the normal range were excluded. Information on allergic diseases was obtained from the parents of the children. Urinary bisphenol A was analyzed using a liquid chromatography tandem mass spectrometer. Logistic regression analysis was used to determine if allergic disease was affected by urinary bisphenol A concentration. Results: Girls had a significantly higher concentration of urinary bisphenol A than did boys (p<0.05). Children of 10-12 years old had a significantly higher concentration of urinary bisphenol A than did children 7-9 years old (p<0.01). Concentration of urinary bisphenol A was increased from underweight to overweight (p<0.05). As the concentration of urinary bisphenol A was increased by $1{\mu}g/L$ or $1{\mu}g/g$ creatinine, the risks of lifetime symptoms of atopic dermatitis in children was 1.22 times (95% CI; 1.05-1.41) or 1.08 times (95% CI; 1.01-1.15). Conclusion: Concentration of urinary bisphenol A was associated with gender, age, body mass index, and allergic disease. Particularly, urinary bisphenol A concentration was associated with lifetime symptoms of atopic dermatitis. The findings of this study could contribute to the management of health effects among sensitive groups such as children.

프로바이오틱스의 면역조절을 통한 알레르기 예방 및 치료효과 (The Preventive and Therapeutic Effects of Probiotics in Allergic Diseases Via Immune Modulation)

  • 프로바이오틱스는 인체 내에서 정상 균총의 역할을 하며 건강에 도움을 주는 미생물을 의미하며, 식품이나 미생물총에서 유래한 비교적 안전한 균주이다. 프로바이오틱스로 많이 사용되는 젖산균은 소화를 돕고 장내환경을 보호해줄 뿐 아니라 면역조절기능 또한 가지고 있다. 대표적으로 $Th_1$ 반응을 유도하여 $Th_1/Th_2$ 균형을 이루게 하는 것을 중심으로, 알레르겐에 반응하는 IgE 및 침윤된 비만세포와 호산구를 감소시키고 면역 억제 기능이 있는 Treg을 유도함으로 알레르기 반응을 완화시킨다고 알려져 있다. 하지만 프로바이오틱스를 이용한 알레르기 질환에 대한 치료 및 예방 효과는 질병 별로 다르게 나타났다. 아토피 피부염에 대해서는 예방 및 치료 효과가 검증되고 있고 알레르기 비염과 음식물 알레르기에서는 예방 효과는 미미했지만 치료효과에서 긍정적이었다. 기관지 천식의 경우 프로바이오틱스의 예방 및 치료 효과를 거의 볼 수 없었지만, 최근에는 특정 균주가 기관지 천식 환자에서 임상 증상을 향상시킨다는 보고가 있다. 균주의 장내 생존율을 높이고 재조합 유산균을 만드는 백신 기술과 함께 프로바이오틱스는 미래의 안전한 알레르기 예방 및 치료제로서 기대할 수 있을 것이며 지속적인 시장의 확대를 통해 차세대 건강기능식품으로 떠오를 것이다.

  • 본 많은 연구에서 알레르기성 질환과 사회경제적 및 사회인구학적 변수의 상관관계가 있음을 보여주었다. 그러나 이들 변수가 한국 청소년에게 미치는 영향에 대한 연구는 거의 이루어지지 않았다. 본 연구는 한국 청소년의 알레르기성 질환에 대한 사회경제적 및 사회인구학적 위험요인을 파악하고 예방 및 관리 하기 위한 정보 제공을 목적으로 수행되었다. 2018년 한국청소년건강행태조사를 기반으로 60,040명 청소년의 자료가 사용되었다. 사회경제적 및 사회인구학적 변수는 익명으로 관리되는 온라인 설문 조사를 통해 수행되었고, 알레르기성 질환은 천식, 알레르기성 비염, 아토피 피부염이 포함되었다. 자료의 분석을 위해 SPSS 21.0 프로그램을 이용하였고, 다변량 로지스틱 회귀분석을 통한 복합표본분석이 사용되었다. 낮은 경제적 수준, 어머니와 동거, 부모의 높은 교육수준, 높은 학업성적, 성별, 비만, 음주경험 및 흡연 경험 등이 천식, 알레르기성 비염, 아토피 피부염과 관련이 있었다. 낮은 사회경제적 수준과 건강에 해로운 행동은 알레르기성 질환의 위험요인 이었다. 그러므로 지역사회의 균형적 경제성장과 함께 청소년의 해로운 건강행태를 통제하는 것은 한국 청소년의 알레르기성 질환의 유병률 감소에 도움이 될 것이다.

Effects of Panax species and their bioactive components on allergic airway diseases

  • Panax species include Panax ginseng Meyer, Panax quinquefolium L., Panax notoginseng, Panax japonicum, Panax trifolium, and Panax pseudoginseng, which contain bioactive components (BCs) such as ginsenosides and polysaccharides. Recently, growing evidence has revealed the pharmacological effects of Panax species and their BCs on allergic airway diseases (AADs), including allergic asthma (AA) and allergic rhinitis (AR). AADs are characterized by damaged epithelium, sustained acquired immune responses with enforced Th2 responses, allergenspecific IgE production, and enhanced production of histamine and leukotrienes by activated mast cells and basophils. In this review, we summarize how Panax species and their BCs modulate acquired immune responses involving interactions between dendritic cells and T cells, reduce the pro-inflammatory responses of epithelial cells, and reduce allergenic responses from basophils and mast cells in vitro. In addition, we highlight the current understanding of the alleviative effects of Panax species and their BCs against AA and AR in vivo. Moreover, we discuss the unmet needs of research and considerations for the treatment of patients to provide basic scientific knowledge for the treatment of AADs using Panax species and their BCs.

Rubus coreanus Unripe Fruits Inhibits Immediate-type Allergic Reaction and Inflammatory Cytokine Secretion

  • The immediate-type allergic reaction (anaphylaxis) is involved in many allergic diseases such as asthma, allergic rhinitis, and sinusitis. The discovery of drugs for the treatment of immediate-type allergic diseases is a very important subject in human health. In this study, we investigated the effect of Rubus coreanus Miq.(Rosaceae) unripe fruits (RCF) on mast cell-mediated allergic reaction and inflammatory cytokine secretion. RCF inhibited compound 48/80-induced systemic reactions in mice. RCF attenuated immunoglobulin (Ig) E-mediated local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells local allergic reactions. In addition, RCF dependently reduced histamine release from rat peritoneal mast cells activated by compound 48/80 or IgE. Furthermore, RCF decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated tumor necrosis factor $(TNF)-{\alpha}$ and interleukin (IL)-6 secretion in human mast cells. Our findings provide evidence that RCF inhibits mast cell-derived immediate-type allergic reactions.

Anti-allergic Effects of Schizonepeta tenuifolia on Mast Cell-Mediated Allergy Model

  • Immediate-type hypersensitivity is involved in many allergic diseases such as asthma, allergic rhinitis and anaphylaxis. The discovery of drugs for the treatment of allergic disease is an important subject in human health. Stimulation of mast cells releases inflammatory mediators, such as histamine and pro-inflammatory cytokines with immune regulatory properties. We investigated the effect of the aqueous extract of Schizonepeta tenuifolia (AEST) (Labiatae) on the immediate-type allergic reaction. AEST inhibited compound 48/80-induced systemic allergic reaction. AEST attenuated immunoglobulin E (IgE)-mediated skin allergic reaction and histamine release from human mast cell line (HMC-1) cells. In addition, AEST decreased the gene expression and secretion of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated HMC-1 cells. Our results indicate that AEST inhibits the mast cell-derived allergic reactions and involvement of histamine and pro-inflammatory cytokines in these effects.

  • Allergic disorders such as atopic dermatitis and asthma are common hyper-immune disorders in industrialized countries. Along with genetic association, environmental factors and gut microbiota have been suggested as major triggering factors for the development of atopic dermatitis. Numerous studies support the association of hygiene hypothesis in allergic immune disorders that a lack of early childhood exposure to diverse microorganism increases susceptibility to allergic diseases. Among the symbiotic microorganisms (e.g. gut flora or probiotics), probiotics confer health benefits through multiple action mechanisms including modification of immune response in gut associated lymphoid tissue (GALT). Although many human clinical trials and mouse studies demonstrated the beneficial effects of probiotics in diverse immune disorders, this effect is strain specific and needs to apply specific probiotics for specific allergic diseases. Herein, we briefly review the diverse functions and regulation mechanisms of probiotics in diverse disorders.

Effects of Ascaris suum Extract and Sulfamethoxazole on Allergic Airway Inflammation

  • Allergic asthma is complex inflammatory airway disorder caused by genetic and environmental factors. Sulfamethoxazole, a sulfonamide, is the cause of drug rash with eosinophilia and systemic symptoms syndrome. Parasites infection also related with eosinophilia and allergic diseases. In the present study, we investigated the modulating effects of parasitic derivative and sulfamethoxazole (SMX) on allergic airway inflammation in the ovalbumin (OVA)-induced murine asthma model. Histopathological changes, cytokine secretion, and total and allergen-specific IgE were investigated. BALB/c mice were treated with Ascaris suum extract or SMX for 4 weeks before sensitized and challenged to ovalbumin. Pre-treatment of Ascaris suum extract decreased allergic inflammation in lung tissue and IL-4, total IgE, and OVA-specific IgE levels in bronchoalveolar lavage fluid. However, pre-treatment of SMX did not show any effects on allergic airway inflammation. These results indicate that parasitic infection has protective effects on allergic asthma, but the sulfamamides may not relate with allergic asthma.

In this study, we investigated the effect of small black soybean (Glycine max Merr.) (Leguminosae) on mast cell-mediated allergic reaction and pro-inflammatory cytokine secretion. Small black soybean (SBS) inhibited compound 48/80-induced systemic reaction. SBS attenuated immunoglobulin (Ig) E-mediated local allergic reaction. In addition, SBS decreased the phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated tumor necrosis factor $(TNF)-{\alpha}$ and interleukin (IL)-8 secretion in human mast cells. These results indicate that SBS may be beneficial in the treatment of mast cell-mediated immediate-type allergic reactions.

Fermentation-Mediated Enhancement of Ginseng's Anti-Allergic Activity against IgE-Mediated Passive Cutaneous Anaphylaxis In Vivo and In Vitro

  • Ginseng (the root of Panax ginseng Meyer) fermented by Lactobacillus plantarum has been found to attenuate allergic responses in in vitro and in vivo experimental models. Ginseng has been reported to also possess various biological functions including anti-inflammatory activity. The present study was aimed at comparing the anti-allergic effect of ginseng and fermented ginseng extracts on IgE-mediated passive cutaneous anaphylaxis in vitro in a murine cell line and in vivo in mice. Fermented ginseng extract (FPG) showed higher inhibitory effect against in vitro and in vivo allergic responses when compared with ginseng extract (PG). The secretion of ${\beta}$-hexosaminidase and interleukin (IL)-4 from the IgE-DNP-stimulated RBH-2H3 mast cells were significantly (p < 0.05) inhibited by FPG treatment, and this effect was concentration-dependent. Further, MKK4 activation and subsequent JNK phosphorylation were attenuated by FPG treatment. The inhibitory effect of FPG on the in vitro allergic response was verified in vivo against IgE-DNP-induced passive cutaneous anaphylaxis in a mouse model. These data indicated that the fermentation of ginseng with L. plantarum enhanced its anti-allergic effects both in vitro and in vivo. We predict that compositional changes in the ginsenosides caused by the fermentation may contribute to the change in the anti-allergic effects of ginseng. The results of our study highlight the potential of the use of FPG as a potential anti-allergic agent.
