Cytotoxicity and DNA Topoisomerase Inhibitory Activity of Constituents Isolated from the Fruits of Evodia officinalis

  • Four alkaloids (1-4), three quinolone alkaloids (5-7), and three flavanoid glucosides (8-10) were isolated from the fruits of Evodia officinalis Dode, and their structures were determined from chemical and spectral data. Compounds, 3, 8, 9 and 10 were isolated from this plant for the first time. Of these compounds, 1-3 and 5-7 exhibited moderate cytotoxicities against cultured human colon carcinoma (HT-29), human breast carcinoma (MCF-7), and human hepatoblastoma (HepG-2). Compound 8 showed strong inhibitory effects on DNA topoisomerases I and II (70 and 96% inhibition at a concentration of $20\;{\mu}M$, respectively).

전자혀를 이용한 인삼의 쓴맛 성분 특성화 (Characterization of bitterness components of ginseng using electronic tongue )

  • 인삼의 쓴맛 성분을 규명하기 위해 전자혀를 이용하여 여러 인삼성분의 쓴맛 강도를 측정한 결과, ginsenoside와 알칼로이드 성분의 쓴맛 강도가 높은 것으로 나타났다. 이 중 알칼로이드 성분은 고려인삼 내 함량이 낮아 인삼의 쓴맛에 실질적인 영향이 미비할 것으로 판단되며 함량이 높은 ginsenoside가 쓴맛에 미치는 영향이 클 것으로 사료된다. Ginsenoside는 인삼 고유의 활성성분으로 큰 폭의 ginsenoside 함량감소는 인삼의 고유 기능성에 부정적인 영향을 줄 수 있으므로, 기호성을 높여 시장성을 확대하고자 인삼의 쓴맛을 경감하기 위하여는 ginsenoside Rb1을 비롯한 인삼 내 함량이 높고 쓴맛이 강한 ginsenoside를 다른 ginsenoside로 전환시키는 가공방법을 개발하는 것이 바람직할 것으로 판단된다. 또한, 전자혀와 실제 사람의 미각과는 차이가 있을 수 있으므로 향후 사람의 미각에 대한 직접적인 추가 검증 연구도 필요하다고 사료된다.

HPLC에 의한 원지 중 알칼로이드 성분의 정량 (Analysis of alkaloids in Polygala tenuifolia by HPLC)

  • 원지(Polygala tenuifolia, Polygalaceae)에 함유되어 있는 ${\beta}$-carboline계의 알칼로이드를 HPLC를 이용하여 정량하는 방법을 검토하였다. 고정상으로는 RP-18 컬럼을 사용하였고, 이동상으로는 $CH_3CN$과 0.01M 인산완충액(pH=3.5)을 기울기 용리시켰으며, UV254nm에서 검출하였다. 알칼로이드 분획을 얻기 위한 전처리 방법으로 산, 염기 처리에 의한 용매추출법과 양이온교환수지(Amberlyst 15)를 이용한 고상추출법을 비교한 결과 양이온 교환수지를 이용한 방법에서 더 나은 크로마토그램을 얻을 수 있었으며, 정량 결과 원지 중 알칼로이드 함량은 각각 harman $2.8{\times}10^{-3}%$, norharman $1.7{\times}10^{-3}%$, 1-carbomethoxy-${\beta}$-carboline $1.3{\times}10^{-3}%$, 1-carboethoxy-${\beta}$-carboline $1.4{\times}10^{-3}%$, perlolyrine $3.3{\times}10^{-3}%$로 나타났다.

흰쥐에 있어서 Symphytum officinale L. 추출액의 독성에 관한 연구 (Toxicological Study on the Water Extract of Symphytum officinale L. in Rats)

  • This study was performed to investigate the toxic effect of pyrrolizidine alkaloids from symphytum officinale i n rat. For this experiment, 120 male and female rats of Sprague-Dawley strain were used. The experimental groups were divided into five: Group CM and CF served as normal control with its gender. Group EM1 and EF1 were fed a 1% Symphytum officinal extract diet for 8 weeks. Group EM2 and EF2 fed a diet containing 2% extract diet. 4% extract diet into group EM3 and EF3 and 8% extract diet into group EM4 and EF4 were given. The results were as follows: 1. The major alkaloids of Symphytum officinale extract were symphytine, echmidine, and lasiocarpine. The amounts of total alkaloid were 168 $\mu\textrm{g}$ PAs/$m\ell$ extract. And contents of Pas in leaves were 0.05% wt.. 2. Total serum bilirubin concentrations increased significantly in group EM2, EM3, and EM4. Group EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 3. Aspartate transaminase activities were increased significantly in group EM3 and EM4 (p<0.05). Aspartate transaminase activities of EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 4. Alanine transaminase activities increased significantly in group EM3, EM4 (p<0.05). Alanine transaminase activities of EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 5. Alkaline phosphatase activities increased significantly in group EM2, EM3, and EM4 (p<0.05). Alkaline phosphatase activities of EF1, FE@, EF3, and EF4 showed statistical sigmificance for the group CF (p<0.05). 6. istopathological analysis of liver specimens from group EM3 and EM4 showed focal necrosis, periportal necrosis and apoptpsis. Hepatocytes obtained from group EM2 showed fatty change and hydropic degeneration in group EM3 and EM4. Chromatolysis and chromatin margination was shown in group EF2 and EF3. With the above results, it was demonstrated that the Symphytum officinal extract could induce functional change of liver, and histopathological change of liver in rats fed a diet containing extract. In conclusion, because of the risk of intoxication or adverse effect, the composition, dosage and mode of administration of herbal products should be monitored strictly. And this study serves as a reminder that herbal as well as orthodox medications may have serious side effects.

Protective effect of Macleaya cordata isoquinoline alkaloids on lipopolysaccharide-induced liver injury in broilers

  • Objective: This experiment aimed to explore the protective action of dietary supplementation with isoquinoline alkaloids (IA) from Macleaya cordata on lipopolysaccharide (LPS)-induced liver injury in broilers. Methods: Total 216 healthy broilers were selected in a 21-d trial and assigned randomly to the following 3 treatments: control (CON) group, LPS group, and LPS+IA group. The CON and LPS groups were provided with a basal diet, whereas the LPS+IA group received the basal diet supplemented with 0.6 mg/kg Macleaya cordata IA. Broilers in LPS and LPS+IA groups were intraperitoneally injected with LPS (1 mg/kg body weight) at 17, 19, and 21 days of age, while those in CON group were injected with equivalent amount of saline solution. Results: Results showed LPS injection caused systemic and liver inflammation in broilers, inhibited immune function, and ultimately lead to liver injury. By contrast, supplementation of IA ameliorated LPS-induced adverse change in serum parameters, boosted immunity in LPS+IA group. Furthermore, IA suppressed the elevation of hepatic inflammatory cytokines and caspases levels induced by LPS, as well as the expressions of genes related to the toll-like receptor 4 (TLR4)/myeloid differentiation primary response 88 (MyD88)/nuclear factor-kappa B (NF-κB) pathway. Conclusion: Dietary inclusion of 0.6 mg/kg Macleaya cordata IA could enhance immune function of body and inhibit liver damage via inactivating TLR4/MyD88/NF-κB signaling pathway in broilers.

  • By bioassay guided fractionation followed by chromatographic separation of the MeOH extract from the fruit of Evodia rutaecarpa (Juss.) Benth. (Rutaceae), a new cyclooxygenase-2 inhibitor was isolated and identified as an alkaloid, rutaecarpine. Other alkaloids such as evodiamine and dehydroevodiamine together with limonoids were also isolated and characterized.

  • Dihaploid lines were derived from the anthers of the F, of single cross, "Va 115" x"SC 72" (Nicotiana tabacum L.). The path-analysis, correlation coefficient, and variation of characteristics among dihaploid families were evaluated. Dihaploid lines displayed much greater variation than their parents. The genetic correlation coefficient between yield and total alkaloids of dihaploids was significantly lower than that of conventional varieties and lines. In path-analysis which relates yield components to yield, number of leaves harvested and leaf width had a direct effect on yield. a direct effect on yield.
