세포 현탁배양에 의한 현호색 알칼로이드의 생산 (Studies on Production of Alkaloid by Plant Cell Culture of Corydalis remota)

  • In previous paper, we described the induced callus of Corydalis remota contains a significant amount of alkaloids. This study describes an optimal condition to maximize alkaloid production. The suspension cultures maintained alkaloid production ability after fifth subculture and a small amount of alkaloid seemed to be released out of cells. The yields of alkaloid by cultured cells was varied depending on the concentrations of NAA, carbon sources and phosphate ion and depending on the vitamin combinations and concentrations. Biosynthetic precursor and an elicitor treatment also affected the total alkaloid yield of the cultures. The optimal conditions for alkaloid production were as follows: 1) MS basal salt containing 30 g/l of glucose, 1.0 mg/l of NAA, and vitamins of LS medium should be used. 2) The culture should be treated with tyrosine 20 mg/l, and yeast extract 1.5 ml/l after the culture reached a stationary phase of growth. Five alkaloids were isolated from the cultures and they were characterized. The spectral data unambiguously revealed that the isolated compounds were dihydrosanguinarine, protopine. tetrahydropalmatine, allocyptopine and ambinine, respectively.

감초알카로이드에 관한 연구(제 4 보) -감초 알카로이드 분획의 항궤양작용에 대한 검토- (Studies on Alkaloid of Glycyrrhiza glabra L. IV. -Anti-ulcer Activity of Licorice Alkaloid Fraction-)

  • Anti-ulcer activity of licorice alkaloid fraction, separated from Glycyrrhiza glabra L. by fractionation according to general alkaloid purification procedure, were studied with various experimental gastric ulcers. In Shay ulcer, licorice alkaloid fraction inhibit significantly response rate and severity of ulcer. But there was no significant changes in volume and pH of gastric contents. A male rat was fixed on a board and immersed up to breast in water of $25^{\circ}C$ for 20 hrs. Hemorrhage and erosion develop in all the animals on the mucosa of the glandular portion of stomach. Licorice alkaloid fraction (5 mg/kg, 10 mg/kg) inhibit significantly in the severity of stress ulcer but there was no changes in the response rate. Chlorpromazine produce marked inhibition in the severity and response rate of ulcer It was suggested that at least the anti-ulcer activity of licorice alkaloid fraction was not mediated by central depression.

뽕잎의 유산발효에 의한 Piperidine Alkaloid 함량 증진 (Enhancement of Piperidine Alkaloid Contents by Lactic Acid Fermentation of Mulberry Leaves (Morus alba L.))

  • This study was carried out to investigate solid-state fermentation method using cellulolytic lactic acid bacteria Lactobacillus plantarum TO-2100 in order to increase piperidine alkaloid contents in mulberry leaves. Piperidine alkaloid, one type of which include 1-deoxynojirimycin (1-DNJ), is known to inhibit ${\alpha}$-glycosidase activities. Using this strain, the optimal solid-state fermentation conditions on mulberry leaves powder were found as the following: initial moisture content, temperature and relative humidity were 20%, $30{\sim}35^{\circ}C$ and 60 ~ 70%, respectively, and the fermentation time was 72 hrs. The piperidine alkaloid contents in the fermented mulberry leaves were 2.86% on dry powder, which is 7-fold increase from that of non-fermented mulberry leaves. The 1-deoxynojirimycin contents after applying preparative thin layer chromatography were 2.02% on dry powder, which is 8 times higher than that of non-fermented mulberry leaves. ${\alpha}$-Glycosidase activities was inhibited by 65.7 ~ 84.7% with 3 ~ 5% treatments of hot-water extracts of the fermented mulberry leaves, compared to 16.2 ~ 40.2% with 3 ~ 5% treatments of hot-water extracts of non-fermented mulberry leaves. Therefore, the results suggest that solid-state fermentation method does indeed increase of piperidine alkaloid contents on mulberry leaves.

  • From the leaves of Magnolia sieboldii a new aporphine-type alkaloid named magnoporphine was isolated. The structure of magnoporphine was all assigned by $^1H-^1H$COSY, $^1H-^{13}C$ COSY and $^1H-^{13}C$ long range NMR. In addition, costunolide, syringin, syringenin $4-O-{\beta}-cellobioside$ and echinacoside was isolated.

Two Alkaloids from Ephedra aphylla growing in Egypt

  • The aerial parts of Ephedra aphylla afforded two alkaloids belonging to two different classes. The first alkaloid, ephedradine C, belonging to the spermine alkaloid, a group characterized by its hypotensive effect. The other alkaloid, hordenine is a phenylalkylamine alkaloid isolated for the first time from Ephedra Species. The structures were elucidated by spectroscopic methods and the assignment of some carbons in ephedradine C was achieved based on 2 D-NMR experiments.

  • Purpose : To investigate the effect of alkaloid fraction from Korean ginseng on radiation-induced DNA double strand breaks (dsb) formation and repair in murine lymphocytes Materials and Methods : We used the neutral filter elution technique to assay $^{60}Co\;{\gamma}$ ray-induced DNA double strand breaks formation and repair in C57BL/6 mouse spleen lymphocytes for evaluating the dose-response relationship in the presence of alkaloid fraction as a radioprotective agent. The lymphocytes were stimulated with Phytohemagglutinin (PHA, 2 u g/ml) to label $^3[H]-thymidine.$ Isotope-labelled lymphocytes in suspension were exposed to 100 Gy at $0^{\cdot}C$ in the alkaloid fraction-treated group and elution procedure was performed at PH 9.6. The extents of formation of radiation-induced DNA double strand breaks and repair were compared respectively via strand scission factor (SSF) and relative strand scission factor (RSSF). Results: Alkaloid fraction reduced the formation of double strand breaks with dose modification factor of 2 15, compared to control group Rejoining of DNA dsb appeared to take place via two components. The first fast component was completed within 20.4 minutes, but the second slow component was not completed until 220.2 minutes after irradiation. About $30\%$ of dsb formed by irradiation was ultimately unrejoined despite the administration of alkaloid fraction. The administration of alkaloid fraction had a great effect on the second slow component of repair; the half-time of fast component repair was not changed, but that of slow component was 621.8 minutes. Conclusion: Neutral filter elution assay Proved to be a very effective method to quantitate the extents of DNA dsb formation and its repair. By using this technique, we were able to evaluate the efficiency of alkaloid fraction from Korean ginseng as a valuable radioprotector. Alkaloid fraction can be used prophylactically to prevent or ameliorate the severe radiation damages in workers and neighbors around the atomic power plants. For more refined study, however, more advanced purification of alkaloid fraction wil be needed in the near future.

미치광이풀 모상근 배양에서 적정 sucrose 농도 및 hydroxyapatite 첨가에 의한 tropane alkaloid 생산성 향상 (Improvement of Tropane Alkaloid Productivity by Optimization of Sucrose Concentration and Addition of Hydroxyapatite in Hairy root Cultures of Scopolia parviflora)

  • 미치광이풀(Socopolia parviflora Nakai) 모상근 배양에서 모상근 생장과 tropane alkaloid 생산에 대한 sucrose농도와 일부 흡착제의 효과를 조사하였다. 모상근 클론 SP 11은 5% sucrose 농도를 함유한 1/2 B5 배지에서 최적인 생장과 tropane alkaloid 생산을 보였다. 모상근에서 tropane alkaloids 생성양상은 모식물에서와 약간의 차이를 보였고 그 중 식물체에서 검출되지 않는 littorine의 생성이 모상근에서 두드러졌다. 조사된 흡착제(0.01% 1%) 중에서, charcoal 0.01%와 XAD-II l%에서 약간의 생장촉진효과를 보인 것을 제외하고는 그 외 농도나 흡착제(amberlita chitosan)에서는 억제효과 또는 유의적인 변화를 보이지 않았다. 반면에 hydroxyapatite는 미치광이풀 모상근의 생장과 tropane alkaloid 생산에 있어 촉진효과가 있음이 확인되었다.

독말풀(Datura stramonium var. tatula Torr.) 모상근의 성장과 tropane alkaloid 생성에 미치는 pH, 서당 및 비타민의 영향 (Effects of pH, Sucrose and Vitamins on the Growth and Tropane Alkaloid Production of Hairy Roots of Datura stramonium var. tatula Torr.)

  • 모상근의 성장 및 tropane alkaloids의 생성에 미치는 pH, 서당, 비타민의 영향을 구명하기 위하여, 독말풀(Datura stramonium var. tatula Torr.)의 잎에 Agrobacterium tumefaciens $A_4$T를 접종하여 모상근을 유도하였다. 유도된 모상근 중 성장률이 양호한 clone (DTLA9)을 선발하고, 이를 pH, 서당, 비타민을 각각 여러 농도로 처리한 SH(Schenk and Hildebrandt, 1972) 기본배지에 배양하였다. 모상근의 성장에 있어 최적 pH는 6.3이었으며, 최적 서당농도는 3.0%이었다. Tropane alkaloids의 함량에 있어 최적 PH는 6.5이었으며, 최적 서당농도는 2.8%이었다. 한편, 비타민이 제거된 SH기본배지에 ascorbic acid, D-pantothenate, nicotinic acid, pyridoxine, riboflavin. 그리고 thiamine을 각각 농도별로 첨가하여 배양한 경우에, 모상근의 성장에 있어 비타민의 최적 농도는 각각 0.1, 0.003, 0.07, 0.002, 0.025, 0.01 mM이었다. Tropane alkaloids의 함량은 0.1 mM의 ascorbic acid 단독처리구에서 대조구(vitamin-free구) 및 SH기본배지에서의 경우에 비하여 가장 높게 나타났다.

  • An alkaloid was isolated from the roots of Glycyrrhiza uralensis Fisch. It was identified as 3-methyl-6, 7, 8-trihydro-pyrrolo[1, 2-alpyrimidin-2-one(I) by spectral analysis. The compounds is first found in the plant kingdom.
