홍모기(Culex pipiens pallens)의 난성숙 과정 중 alkaline phosphase의 활성 (Activity of Alkaline Phosphatase from the Mosquito, Culex pipiens pallens)

  • Culex pipiens pallens에 존재하는 Alkaline phosphatase의 연구를 위한 적정 분석조건과 우화 후, 시간 경과에 따른 Alkaline phosphatase 활성 경향에 대해서 연구하였다. C. pipiens에 존재하는 Alkaline phosphatase의 활성은 우화 직후부터 지속적으로 감소하다가, 흡혈 자극에 의해서 급격하게 증가한다. 흡혈 후 30시간이 경과했을 때, 최대의 활성도를 보이고 감소하나, 흡혈 48시간 이후에는 다시 증가하여 지속적으로 유지됨을 알 수 있다. 기관별 분석에서 첫번째 활성 증가는 중장에서 일어나고, 두번째 활성 증가는 난소에서 일어남을 알 수 있다. 그리고 흡혈 후 30시간된 성체에서는 5개의 동위효소 밴드가 보이는데, 난소에서 ALP-1와 ALP-2가 나타나고, 가슴에서는 ALP-3, ALP-4와 ALP-5가 보인다. 지방체에서는 ALP-4와 ALP-5가, 중장에서는 ALP-3, ALP-4와 ALP-5가 나타남을 알 수 있다. 그리고 흡혈 후 72시간된 성체에서, ALP-1, ALP-2가 동일하게 존재함을 알 수 있다.

호르텐스극구흡충에서 Alkaline Phosphatase 및 Acid Phosphatase의 특성 (Characteristics of Alkaline and Acid Phosphatase in Echinostoma hortense)

  • 호르텐스극구흡충 성충 (adult worm)을 재료로 하여 acid phosphatase (Acp)와 alkaline phosphatase (Alp)의 조직 세포에 대한 분포 및 동위효소의 특성을 규명하기 위하여 Gomori lead nitrate 및 Gomori calcium방법 그리고 전기영동법 등을 시행하였다. 조직 내의 Acp및 Alp의 분포는 충체의 소장 (intestine)과 난황선 (vitellaria)그리고 인두 (pharynx)에서 강한 반응을 나타냈다. 동위효소 유형은 Acp에서 179.5 및 209.4 kDa 두 개의 분획이 분리되었고, Alp는 145.9, 207.5 및 220.8 kDa의 세 개의 분획이 분리되었다. 열에 대한 안정도는 Alp가 9$0^{\circ}C$에서 12초경과 후에 완전히 불활성화 되었으며, Alp의 최적 온도 및 pH는 4$0^{\circ}C$와 pH 9이었고 Acp의 치적 pH는 5였다. 그리고 Acp및 Alp의 최대 활성도 (unit)는 Acp가 71 범위였고, Alp는 189의 범위에 있었다. 이상에서와 같이 호르텐스극구흡충은 phosphatase가 소화관과 난황선에서 주로 분포하고 있었으며 숙주 체내에서 소화관을 중심으로 대사가 활발하게 이루어지고 있음을 알 수 있었다.

세포 검사시료에서 alkaline phosphatase 염색법을 활용한 개 골육종의 감별 진단 (Application of alkaline phosphatase staining of cytology specimen for differential diagnosis of canine osteosarcoma)

  • Aspiration of lytic bone lesions is an excellent diagnostic test in the initial evaluation of primary bone tumor. However, cytologically, it can be difficult to differentiate osteosarcoma (OSA) from other bone neoplasms, including fibrosarcoma, chondrosarcoma, synovial cell sarcoma, malignant fibrous histiocytoma and malignant peripheral nerve sheath tumor. The purpose of this study is to introduce alkaline phosphatase (ALP) staining to differentiate OSA from other mesenchymal tumors. Tumors actively producing bone are specifically positive for ALP staining. Unstained, cytologic specimens were incubated for 10 minutes with nitroblue tetrazolium chloride/5-bromo-4-chloro-3-indolyl phosphate toluidine salt-phosphatase substrate. Among 20 cases of cytology specimen, 14 were positive for ALP staining and histopathology, 6 were negative for ALP staining and histopathology. ALP staining was 100% sensitive and specificity for the diagnosis of OSA. Aspirate cytology with ALP staining was a simple, fast, safe and accurate diagnostic test for the evaluation of suspected OSA lesions in dogs.

만손열두조충(Spirometra erinacei)에서 알칼리성과 산성 인산효소의 특성 (Characteristics of alkaline and acid phosphatase in Spirometra erinacei)

  • 자연감염된 뱀의 체내조직에서 적출한 스파르가눔(5-스파르가눔)을 흰쥐와 고양이에 인공 경구감염시켜 12주 후에 흰쥐의 피하조직에서 적출한 스파르가눔(r-스파르가눔)과 고양이의 소장에서 회수한 성충(adult)을 재료로 하여 alkaline phosphatase(Alp)와 acid phosphatase(Acp)의 충체조직내 분포 및 동위효소의 특성을 조사하기 위하여 효소조직화학적 방법과 전기영동법 등을 이용하였다 Alp와 Acp의 조직내 분포는 성충과 유충 모두 외피층과 외피하근층에 많이 분포하였으며 실질근층에서는 거의 분포하지 않았다. 이들 phosphatase는 Acp에 비해 Alp가 더 강하게 양성반응이 나타났다. Alp의 동위효소유형이 5-스파르가눔, r-스파르가눔 성충에서 각각 2개, 2개, 4개가 분리되었고. 이중 분자량 66 kDa 분획이 유충과 성충에서 공통적으로 존재하는 동위효소였다. Acp 동위효소는 s-스파르가눔, r-스파펀가눔, 성충에서 각각 2개. 2개. 3개가 분리되었다. 이 중 130 kDa 분획이 유충과 성충에 공통적으로 존재하는 동위효소이면서 유충의 주분획이었다 IEF에 의해 성충에서 등전점 5.3, 6.5, 7.7, 7.9인 동위효소가 분리되었고. 스파펄가눔에서는 등 전점 7.7, 7.9인 동위효소가 분리되었다. 열에 대한 안정도는 Alp가 $90^{\circ}C$에서 40초경과 후에 완전히 불활성화 되었다 스파르가눔과 성충에 존재하는 Alp 활성의 최적 pH는 10이나 활성범위가 pH 9~10이었고. 최적 온도 활성범위는 40~50$^{\circ}C$이며 $50^{\circ}C$ 이상에서는 활성이 급격히 감소하였다. Alp의 최대활성도(unit)는 5-스파르가눔. r-스파르가눔, 성충에서 각각 22.0, 25.0, 215.0으로 나타났으며. Alp는 유충에 비해 성충의 조직에 존재하는 동위효소들의 활성도라 월등히 높게 나타났다 만손열두조충의 스파르가눔과 성충이 숙주와 기생장소를 달리하지만 phosphatase는 외피층과 외피하근층에 주로 분포하고 있어 기생환경의 변화에 따라 Alp와 Acp의 동위효소들의 활성을 달리하딘로서 숙주환경에 적응한다는 사실을 알 수 있다.

지렁이 중장에서 분리한 Alkaline Phophatase의 분포와 특성에 관한 연구 (Distribution and Some Properties of the Alkaline Phosphatase from the Midgut of the Earthworm, Eisenia andrei)

  • The distribution and some properties of alkaline phosphatase (ALP) were investigated in the midgut of the earthworm, Eisenia andrei. The ALP activity appeared to be highly polarized toward the luminal side of epithelium, with minor ALP activity in chloragogue tissue. The epithelial and chloragogeneous tissues contained approximately 85 and 15% of total intestinal ALP activity, respectively. The optimal temperature was approximately 37$^{circ}C$ and isoelectricpoint was estimated to be 4. The treatment of neuraminidase and PtdIns-PLC failed to change the migration rate of ALPs. Also, these ALPs appeared to have a wide range of substrate specificity. The relationship between the properties and physiological significances of the midgut ALPs in Eisenia andrei was discussed.

Inhibitory Effect of Retinoids on Alkaline Phosphatase Isoenzymes Activity in Human Serum

  • Changes in the activity of alkaline phosphatase (ALP) isoenzymes and isoforms in human serum have a major diagnostic value, therefore the regulation of ALP activities is a valuable target for therapeutic interventions. To assess the pharmacological activity of retinoids, i.e., all-trans retinoic acid and 13-cis retinoic acid, their tissue-specific inhibitory effect on human serum ALP activity was elucidated by chemical inhibition methods, heat-sensitive inactivation, and wheat-germ lectin precipitation test. Retinoids showed significant inhibition of the total ALP activity in human serum at a concentration of 5 mM. All-trans retinoic acid (5 mM) and 13-cis retinoic acid (5 mM) inhibited ALP activities by up to 12% and 15%, respectively, compared to that by guanidine hydrochloride (200 mM). L-phenylalanine (100 mM) and urea (30 mM) had no further inhibitory effect on ALP activities in human serum pretreated with retinoids (5 mM). Retinoids significantly inhibited ALP activities by up to 20% compared with that of tetramisole (30 mM). The ALP activities in retinoid-pretreated serum remained unchanged after the heat inactivation process. These results suggest that retinoids are inhibitors of the intestinal ALP isoenzyme. Remarkably, retinoids revealed potent inhibitory activities against ALP in wheat-germ lectin precipitant serum, indicating that they also function as inhibitors of the bone ALP isoform. The results show that retinoids inhibit the specific tissue-derived human serum ALP activities, moreover, the inhibitory effect of retinoids against bone ALP activity suggests their clinical utility as monitoring and prevention of metastasis of bone cancer.

Radiomodifying Potential of Panax ginseng in Liver of Swiss Albino Mice against Gamma Radiation

  • Panax ginseng occupies an important role in the folk medicine of China, Korea and Japan. The present study was undertaken to determine the radioprotective efficacy of ginseng root extract in the liver of Swiss albino mice. The animals were divided into 4 groups. Group I-Only vehicle was administered. Group II-The animals received 10 mg/kg body weight ginseng root extract i.p. for 4 consecutive days. Group III-Animals were irradiated with 8Gy gamma radiation at the dose rate of 1.69 Gy/min at the distance of 80 ems. Group IV-Animals were given by ginseng root extract (10 mg/kg body weight) continuously for 4 days and on 4th day they were irradiated with 8 Gy gamma radiation after 30 min. The animals from above groups were autopsied on 1,3,7,14 and 30 days. Biochemical estimations of phosphatases (acid & alkaline), LDH (lactate dehydrogenase), LPO (lipid peroxidation) and GSH (reduced glutathione) in liver and SGOT (serum glutamate oxaloacetate transaminase), SGPT (serum glutamate pyruvate transaminase) and alkaline phosphatase in serum were done. In ginseng treated group acid phosphatase (ACP), alkaline phosphatase (ALP), LPO and LDH in liver and SGOT, SGPT and alkaline phosphatase in serum did not show any significant alteration. However, a significant increase in GSH content in liver was recorded. In irradiated group there was a significant increase in ACP, ALP and LPO content in liver and SGOT & SGPT in serum was noted. Whereas, a significant decrease was recorded in GSH and LDH activity in liver and alkaline phosphatase activity in serum. Pretreatment of ginseng with radiation significantly alters the biochemical parameters in liver and serum. A significant decline in ACP, ALP activity and LPO content in liver and SGOT and SGPT activity in serum was observed. However, a significant increase in GSH content and LDH activity in liver and ALP activity in serum was estimated. The present study suggests that pretreatment of ginseng before irradiation significantly protects the liver and maintains the enzyme activity.

Prognostic Value of Pretreatment Serum Alkaline Phosphatase in Nasopharyngeal Carcinoma

  • Background: The prognostic value of serum alkaline phosphatase (S-ALP) has not been fully validated for nasopharyngeal carcinoma (NPC). Materials and Methods: S-ALP levels were measured in 601 patients newly diagnosed with NPC before radical treatment, and possible associations of these levels with 5-year overall survival (OS) and tumor-free survival (TFS) were explored using univariate and multivariate analyses. Results: Elevated pretreatment S-ALP (>85 U/L) was significantly less frequent among patients classified as T1+2 or stage I+II than among those classified as T3+4 or stage III+IV. Multivariate analysis showed that elevated pretreatment S-ALP (>85 U/L), age, T classification and N stage were independent predictors of poor OS and TFS. Conclusions: Pretreatment S-ALP may be a reliable biomarker to evaluate the long-term prognosis of patients with NPC.

The Effects of Korean Cucurbitaceous Plants on the Alkaline Phosphatase Activity Associated with Sonic Hedgehog Pathway

  • In order to examine the effects of Korean cucurbitaceous plants on sonic hedgehog pathway and growth of cancer cells with over-activated hedgehog pathway, we measured the sonic hedgehog conditioned medium (shh-CM) induced alkaline phosphatase (ALP) activity and cell viability of pancreatic cancer cell lines by treatment of cucurbitaceous plants. Among the tested cucurbitaceous plants, Actinostemma lobatum Maxim, Cucumis sativus L., Momordica charantia L., Schizopepon bryoniaefolius Maxim and Trichosanthes kirilowii Max, var. japonica Kitam showed the potent inhibitory effects (> 50 % at $20{\mu}g/mL$) on shh-CM induced ALP activity. We also evaluated the cell viability of pancreatic cancer cells treated with the cucurbitaceous plants. The tested cucurbitaceous plants showed the very weak effects on cancer cell proliferation but, T. kirilowii Max, var. japonica Kitam presented the inhibitory effect of 72.7 % on the proliferation of pancreatic cancer cells at $20{\mu}g/mL$. Taken together, we screened the effects of Korean cucurbitaceous plants on shh-CM induced ALP activity and cell viability of pancreatic cancers to search for the modulators of the hedgehog pathway leading to the inhibition of cancer cell proliferation. T. kirilowii Max, var. japonica Kitam, among the tested cucurbitaceous plants, showed the inhibitory effects on the shh-CM induced ALP activity and the proliferation of pancreatic cancer cells.

Nd-Fe-B 자석의 정자기장이 MC3T3-E1 세포의 alkaline phosphatase 활성도에 미치는 영향 (The effect of a static magnetic field of Nd-Fe-B magnet on alkaline phosphatase activity of MC3T3-E1 cells)

  • Nd-Fe-B 자석의 정자기장 (Static Magnetic Field)이 MC3T3-E1 세포의 alkaline phosphase (ALP) 활성도에 미치는 영향을 알아보기 위하여, MC3T3-El세포를 12 well 세포배양접시의 1열과 3열에 접종하고 1열과 3열의 첫째 칸 하방에 Nd-Fe-B 자석을 가하여 7일, 13일, 19일, 25일간 배양한 후 세포의 ALP 활성도를 측정하였으며, 100 mm 세포배양접시의 한 쪽 가장자리 하방에 Nd-Fe-B 자석을 가하여 7일, 13일, 19일, 25일간 배양한 후 ALP 염색을 하여 다음과 같은 결과를 얻었다. 1. 정자기장을 가한 13, 19, 25일에 각각 100 mT에서는 대조군에 비해 ALP활성도가 감소된 반면 4.6 mT와 0.5 mT에서는 대조군에 비해 ALP 활성도가 증가되었다 (P<0.01). 2. ALP염색에서 전체적으로는 19일까지 ALP가 증가되었다가 25일에 다소 감소되는 양상을 나타내었으며, 각 세포배양접시에서는 7일에는 뚜렷한 차이를 보이지 않았으나 13, 19, 25일에는 자석이 놓인 부위 (100 mT) 가 그 반대편(0.5 mT)에 비해 ALP가 감소되었음을 육안으로나 도립위상차현미경으로 관찰할 수 있었다. 이상의 결과 0.5 mIT와 4.6 mT의 낮은 자기장에서는 ALP 활성도가 증가되었으며 100 mT의 높은 자기장에서는 ALP활성도가 감소되어, Nd-Fe-B자석의 정자기장이 MC3T3-E1 세포의 ALP 활성도에 영향을 미쳐 골조직의 형성 및 개조에 영향을 미칠 수 있을 것으로 생각된다.
