Eletron Microscopic Observation of Calcium-Acetylated Seaweed Alginate Gel

  • Seaweed alginate was acetylated by activated carbon immobilized Pseudomonas syringae in a fluidized bed, up-flow reactor. The acetylation degree of seaweed alginate was about 30%. Calcium-acetylated seaweed alginate gel bead was made and compared to calcium-seaweed alginate gel bead by the scanning electron microscopy (SEM). Structural difference of two gel beads may results from increased viscosity and decreased affinity of acetylated seaweed alginate for calcium ion. On the basis of interior and exterior structure of calcium-acetylated seaweed alginate gels and property of acetylated seaweed alginate, it seems that acetylated seaweed alginate is used for the supporter for electrophoresis and packing materials for liquid chromatography and gel filtration.

  • 형질전환된 담배세포배양을 이용한 hGM-CSF 생산에 있어 고정화가 미치는 영향을 연구하였다. Alginate bead를 이용한 고정화 세포의 경우 hGM-CSF 생산이 6일째 $3.35\;{\mu}g/L$로써 현탁세포 $6.01\;{\mu}g/L$의 약 50%에 미쳤고 polyurethane foam 내 고정화 한 경우는 세포 생장은 낮았으나 hGM-CSF 생산량은 6일째 $6.67\;{\mu}g/L$로 가장 높았다. 높은 비 생산량을 보이는 polyurethane foam내 고농도로 세포를 포집시킨 후 세포 재사용이 용이한 장점을 이용하여 연속공정에 적용할 경우 더 높은 hGM-CSF의 생산이 기대된다.

Immobilization of Photobacterium Phosphoreum for Monitoring of Toxic Substances

  • A new sensing system based on the immobilization of luminescent batcteria, Photobacterium phosphoreum, was proposed for continuous real-time monitoring of polluants. The response curves demonstrate that Photobacterium phosphoreum immobilized on the strontium alginate was very sensitive to seven reference chemicals used. The significant inhibitory concentrations for bioluminescence emission were 5 ppm for Pb(NO3)2, NiCl2, CdCl2, 50 ppm for NaAsO2, 0.1ppm for HgCl2, 0.5ppm for pentachlorophenol and less than 5ppm for SDS, respectively. The alginate mixed-cells (AMC) retained their luminescence during experimental period (29 days) under storage condition of -8$0^{\circ}C$. The variables affecting performance of continuous flow through monitoring (CFTM) were optimized in order to ensure stability and efficiency. The flow through cell with strontium-alginate immobilized luminescent bacteria was tested with salicylate and 4-nitrophenol and a rapid response of luminescence was recorded by time drive mode in bioluminescence spectrometer after exposure to both toxicants.

Immobilization and Characterization of Tannase from a Metagenomic Library and Its Use for Removal of Tannins from Green Tea Infusion

  • Tannase (Tan410) from a soil metagenomic library was immobilized on different supports, including mesoporous silica SBA-15, chitosan, calcium alginate, and amberlite IRC 50. Entrapment in calcium alginate beads was comparatively found to be the best method and was further characterized. The optimum pH of the immobilized Tan410 was shifted toward neutrality compared with the free enzyme (from pH 6.4 to pH 7.0). The optimum temperature was determined to be $45^{\circ}C$ for the immobilized enzyme and $30^{\circ}C$ for the free enzyme, respectively. The immobilized enzyme had no loss of activity after 10 cycles, and retained more than 90% of its original activity after storage for 30 days. After immobilization, the enzyme activity was only slightly affected by $Hg^{2+}$, which completely inhibited the activity of the free enzyme. The immobilized tannase was used to remove 80% of tannins from a green tea infusion on the first treatment. The beads were used for six successive runs resulting in overall hydrolysis of 56% of the tannins.

Comparison of Immobilization Matrix for Ethanol Fermentation by Zymomonas mobilis and Saccharomyces cerevisiae

  • A continuous fermentation system employing immobilized cells of Zymomonas mobilis and Saccharomyces cerevisiae was studied for the mass production of ethanol. Ethanol production by cells immobilized with Ca-alginate was better than those by cells immobilized with K-carrageenan. Maximum ethanol production employing a continuous system by cells immobilized with Ca-alginate was 77.5 $g.l^{-1}h^{-1}$ at a dilution rate of 1.85 $h^{-1}$ with 82% conversion rate for Z. mobilis while that was 40.2 $g.l^{-1}h^{-1}$ at a dilution rate of 0.92 $h^{-1}$ with 85% conversion rate for S. cerevisiae. These results suggest that Ca-alginate is a better cell immobilization matrix than K-carrageenan and that immobilized cells of Z. mobilis are more efficient than S. cerevisiae for ethanol production.

  • Benzoate produced from the degradative pathways of various aromatic chemicals is generally recognized as a pollutant compound. However, various bacterial strains isolated as benzoate degraders have exhibited certain limits to their functions, including a loss of viability and degradability when cultivated in a broth medium for a longer time. Accordingly, immobilization techniques have been utilized to overcome such problems, and the current study examined the use of alginate and polyurethane for immobilizing Klebsiella oxytoca C302 to extend its viability and degradability of benzoate. The organism was well encapsulated by both matrices and the immobilized cells showed a high stability as regards their viability and degradability of 2 mM benzoate in a MM2 broth medium during cultivation for longer than 60 h in a semicontinuous batch system.

Production of Alkaline Protease by Entrapped Bacillus licheniformis Cells in Repeated Batch Process

  • In this study, Bacillus licheniformis cells were immobilized by entrapment in calcium alginate beads and were used for production of alkaline protease by repeated batch process. In order to increase the stability of the beads, the immobilization procedure was optimized by statistical full factorial method, by which three factors including alginate type, calcium chloride concentration, and agitation speed were studied. Optimization of the enzyme production medium, by the Taguchi method, was also studied. The obtained results showed that optimization of the cell immobilization procedure and medium constituents significantly enhanced the production of alkaline protease. In comparison with the free-cell culture in pre-optimized medium, about 7.3-fold higher productivity was resulted after optimization of the overall procedure. Repeated batch mode of operation, using optimized conditions, resulted in continuous production of the alkaline protease for 13 batches in 19 days.

Photosynthetic Activity, and Lipid and Hydrocarbon Production by Alginate-Immobilized Cells of Botryococcus in Relation to Growth Phase

  • Whole-cell immobilization of the hydrocarbon rich microalgae, Botryococcus braunii and B. protuberans, in alginate beads under air-lift batch cultures resulted in a significant increase in chlorophyll, carotenoid, dry weight, and 1ipid contents at stationary and resting growth phases, as compared to free cells. Photosynthetic activity in both the species, of Botryococcus was enhanced, relative to free cells, at any growth phase of cultures. Immobilization exerted a protective influence on ageing of the cultures as reflected by higher chlorophyll and dry weight contents. Entrapment also stabilized the chlorophyll and carotenoid contents even at stationary and resting phases as compared to free cells in both the species.

Immobilization of Keratinase from Aspergillus flavus K-03 for Degradation of Feather Keratin

  • Extracellular keratinase isolated from Aspergillus flavus K-03 was immobilized on calcium alginate. The properties and reaction activities of free and immobilized keratinase with calcium alginate were characterized. The immobilized keratinase showed proteolytic activity against soluble azo-casein and azo-keratin, and insoluble feather keratin. Heat stability and pH tolerance of keratinase were greatly enhanced by immobilization. It also displayed a higher level of heat stability and an increased tolerance toward alkaline pHs compared with free keratinase. During the durability test at $40^{\circ}C$, 48% of the original enzyme activity of the immobilized keratinase was remained after 7 days of incubation. The immobilized keratinase exhibited better stability, thus increasing its potential for use in industrial application.

감 즙으로부터 에탄을 생산을 위한 Saccharomyces cerevisiae의 고정화 조건 (Immobilized Condition of Suchwowces cerevisiae for Ethanol Production from Persimmon Juice.)

  • 고정화 배양계를 이용하여 감 즙으로부터 식초생산을 효율적으로 행하기 위하여 Saccharomyces cerevisiae의 고정화 형성조건을 검토하였다. 고정화를 위한 최적 Na-alginate의 농도는 2%이었다. 담체의 농도를 1-4%로 변화시키면서 gel beads로부터 누출되는 효모 균수를 검토한 결과, 1%의 농도에서는 배양 8시간째부터 배양액의 탁도가 흐려지는 것이 관찰되어 배양 20시간째에 0.82의 흡광도를 나타내었다. 그러나 2-4%에서는 거의 변화가 없었다. 배양이 끝난 후, 전자현미경으로 gel beads내부의 균주 생육상태를 관찰한 결과 고정화한 효모는 Na-alginate의 농도에 관계없이 왕성하게 생육하였다. Gel beads의 크기는 2-3mm정도가 최적이었고, 효모의 최적 접종량은 약 33mg이었다. 효율적인 에탄을 생산을 위한 산소공급 시간을 검토한 결과, 배양초기부터 12시간까지는 면전으로 호기적 배양을 행하고, 그 이후부터는 check valve가 부착된 실리콘 plug를 사용하여 혐기적 배양을 행하는 것이 효과적이었다.
