OUTBREAK OF HARMFUL ALGAL BLOOMS RELATED WITH TEMPERATURE DISTRIBUTION DERIVED FROM IN-SITU AND REMOTE SENSING EXPERIMENTS IN THE KOREAN WATERS

  • The red tide related with Cochlodinium Polykrikoides bloom has been frequently occurred around the South Sea of Korea and caused the economic loss in the coastal breeding grounds. The outbreak scale was usually change by physical, biological and environmental condition at each years. Relatively large-scale red tide occurred in 1995, 1997, 1999, 2001, 2002 and 2003 through spatial scale, duration and maximum density. Compared the scale of red tide with physical condition around the South Sea, the lower coastal temperature on August around the South Sea corresponded with the large scale red tide. By serial oceanographic investigations on August in the South Sea and estimated wide area temperature information by satellite, SSTA around the South Sea and wide area was negative when the outbreak of red tide was large scale. From the results of temperature difference between surface and 30m layers, the occurrence of enormous red tide has a tendency when the temperature gradient around the seasonal thermocline was weakened. Larger Kuroshio volume transport in the upstream was also corresponded with the large scale red tide.

Cochlodinium Red Tide Effects on the Respiration of Abalone, Haliotis discus hannai Ino

  • Cochtodinium votykrikoides -related red tide is the most notorious tidal bloom, resulting in mass mortality to marineanimals. This study aimed to test the effect of C. polyknkoides on the lethality to Haliotis discus hannai under con-trolled conditions. The oxygen demand of C. polykrikoides increases to reach its peak duhng the night, while the oxy-gen usage by H. discus hannai was continuously decreased with a threshold of 2 mg L U. The addition of C.polykrikoides did not effect the respiration of the H. discus hannai. However, the usage of oxygen by C. polykrikoidesduhng the night may lead to anoxia in the animal. With aeration, the level of dissolved oxygen (D.O.) was between6.06 and 7.28 mg LU; 90% of abalones survived even with a high concentration of C. potykrikoides (9000 cells mL U).Without aeration (3 mg LU of D.O.), however, the H. discus hannai suffocated immediately. Once 20 hours hadelapsed, all of the abalones were dead. The density of the H. discus hannai population contributed to their mortality.Therefore, aeration during the night and maintaining lower abalone densities is the best way to promote the sur-vivorship of H. discus hannai during a C. polykrikoides red tide.

Adsorption-DAF Hybrid Process for the Simultaneous Removal of Algae and Organic Compounds (조류와 유기화합물의 동시제거를 위한 흡착 - DAF 복합공정)

  • Dissolved air flotation (DAF) is an effective solid/liquid separation process for low density floc particles such as algal, color-alum and clay-alum flocs produced from low turbidity water. The removal of taste and odor-causing organics (2-mthylisoboneol and geosmin) originating from algae in drinking water is a local and worldwide concern. Although DAF has been effectively applied for the removal of suspended solid, its application for the treatment of dissolved organic carbon is very limited. In this study, a new hybrid system consisting of adsorption and DAF processes was introduced for the simultaneous removal of algae and taste and odor-causing organics. Powdered activated carbon (PAC) was used as an adsorbent. In this proposed system, the major concern of eliminating the spent PAC from the system was also addressed. It was found that zeta potential of algae and PAC was increased with coagulant dosage, and the removal efficiency in DAF was also enhanced up to 90~95% under the given experimental conditions. Based on this study, the hybrid process was found to be a promising technology for the simultaneous removal of algae and dissolved organic pollutants.

Optimization Test of Plant-Mineral Composites to Control Nuisance Phytoplankton Aggregates in Eutrophic Reservoir (부영양 저수지의 조류제거를 위한 기능성 천연물질혼합제의 최적화 연구)

  • To optimize the natural chemical agents against nuisance phytoplankton, we examined algal removal activity (ABA) of Plant-Mineral Composite (PMC), which already developed by our teams (Kim et al., 2010), on various conditions. The PMC are consisted of extracted-mixtures with indigenous plants (Camellia sinensis, Quercusacutissima and Castanea crenata) and minerals (Loess, Quartz porphyry, and natural zeolite), and characterized by coagulation and floating of low-density suspended solids. A simple extraction process was adopted, such as drying and grinding of raw material, water-extraction by high temperature-sonication and filtering. All tests were performed in 3 L plastic chambers varying conditions; six different concentrations ($0{\sim}1.0\;mL\;L^{-1}$), six light intensities ($8{\sim}1,400\;{\mu}mol\;m^{-2}s^{-1}$), three temperatures ($10{\sim}30^{\circ}C$), four pHs (7~10), five water depths (10~50 cm), and three different waters dominated by cyanobacteria, diatom, and green algae, respectively. Results indicate that the highest ABA of PMC was seen at $0.05\;mL\;L^{-1}$ in treatment concentrations, where showed a reduction of more than 80% of control phytoplankton biomass, while $1,400\;{\mu}mol\;m^{-2}s^{-1}$ in light intensity (>90%), $20{\sim}30^{\circ}C$ temperature (>60%), 7~9 in pH (>90%), below 50 cm in water depth (>90%), and cyanobacterial dominating waters (>80%), respectively. Over the test, ABA of PMC were more obvious on the algal biomass (chlorophyll-${\alpha}$) than suspended solids, suggesting a selectivity of PMC to particle size or natures. These results suggest that PMC agents can play an important role as natural agents to remove the nuisant algal aggregates or seston of eutrophic lake, where occur cyanobacterial bloom in a shallow shore of lake during warm season.

  • Biodiesel has very similar physical properties (density, kinematic viscosity) and has even higher cetane number compare with conventional diesel. There are no necessity to change or modify the infra-structure & engine system. It is known that fatty acid methyl ester (FAME) is oxygen-contained components increasing the combustibility, biodegradability and reduced the exhaust harmful gas. These things made the biodiesel more popular as an alternative diesel fuel. But biodiesel's sources are controversial issues about $CO_2$ reduction effect at this time because those mainly come from edible plants such as soy, palm, rapeseed already spent lot of $CO_2$ to cultivate. Whereas micro-algae is focused because they are inedible and has rapid growth rates & high carbon-dioxide adsorption rate per area. In this study, we analyze the each FAME components using $GC{\times}GC$-TOFMS in stead of GC-FID and verify the previous total FAME contents method's applicability through the micro algal biodiesel derived from Dunaliella tertiolecta.

The Community Dynamics of Microbial Food Web during Algal Bloom by Stephanodiscus spp. in Downstream of Nakdong River (낙동강 하류부에서 Stephanodiscus속에 의한 수화 발생시 미생물먹이망 군집 동태)

  • The bloom of the genus Stephanodiscus was gradually extinguished after 18 April. Counts of bacterial population were increased as the diatom bloom was disappeared. Numbers of the heterotrophic nanoflagellates and ciliates were also increased during the disappearance of the bloom. The densities of the mesozooplankton, the major predator of the diatoms, started to increase in April. However, their growth was suppressed during the bloom period of the diatoms (from January to March). During the bloom period of the diatoms, the monthly average value of the basic productivity amounted up to 11,765.7 mgC $m^{-2}day^{-1}$, which is relatively high value considering the low temperature and light during that period. The growth rate of phytoplankton in March, when the bloom was beginning to be supressed was 0.007. The growth rate of phytoplankton was negative value in April when the decreasing of the algal density was started.

Culture Condition and Growth of Larvae of the Mytilus Coruscus Gould (홍합의 사육조건과 성장)

  • The larvae of Mytilus coruscus were grown at the room temperature of approximately 15.1C under several different sulture conditions, i.e., salinity, population of the larvae, density and kind of food organisms, etc. (1) The egg of Mytilus coruscus obtained in the laboratory measured about 73.0${\mu}$ in diameter. The embryos gradually developed into larvae up to 179.0${\mu}$ shell length with the shell height of 135.9${\mu}$ even in the absence of the algal food. Beyond this size, however, the growth of larvae was considerably retarded, indicating that the better growth could be expected if the food began to be fed four days after spawning. (2) The larvae began settling upon reaching 281.4${\mu}$ to 310.9${\mu}$ in shell length or 264.3${\mu}$ to 301.9${\mu}$ in shell height. When the shell length reaches 322.6${\mu}$ to 337.1${\mu}$, the shell height also reaches about the same, i.e., 321.5${\mu}$ to 346.2${\mu}$. (3) Daily rate of food consumption was determined by the size of the larvae and the species of the algal food. Regardless of the species of food given, the rage of food consumption remained almost the same until the larva reached the straight-hinge stage, and marked variations were found as the larvae grew larger. Daily rate of food consumption was shown as follow; Chaetoceros calcitrans : Y=2.99167e$\^$0.000018243x$\^$2// Cyclotella nana : Y=3.00324e$\^$0.000015481x$\^$2// Monochrysis lutheri : Y=3.000056e$\^$0.000014485$\^$2// (4) Suitable amount of the food to be given was about five times of the consumed food by Mytilus coruscus. (5) When the numbers of the larvae was higher than ten per milliliter, the growth was significantly retarded. (6) Monochrysis lutheri and Cyclotella nana were much better than Chaetoceros calcitrans as the food of Mytilus coruscus, and even the same food organism showed some difference with the age of the organism. (7) Sea water of higher salinity showed the better result in the growth of the larvae and the water with the specific gravity of 1.020 or below was dangerous for the larvae. Application of Species-specific DNA Probe to Field Samples of Alexandrium tamarense (Lebour) Balech (자연 시료로부터 Alexandrium tamarense을 위한 종 특이적 DNA탐침의 응용)

  • Fluorescent species-specific DNA probe (AT1) of toxic dinoflagellate Arexandrium tamarense was tested on several other species, on comparison of binding activity at different preservatives for fixation of the cells, at different culture age and estimation of cell density by light microscope or epifluorescent microscope using whole cell hybridization. Th AT1 probe specifically bound to Alexandrium tamarense, whereas it did not bind to other phytoplankton, in particular Alexandrium catenella, morphologically similar to Alexandrium tamarense, could not react to AT1 probe. When cells were fixed with all three preservatives, labeling cells of Alexandrium tamarense emitted strong fluorescent signal intensity. In addition, regardless culture days, binding activity with AT1 probe was strong. The tell densities estimated by epifluorescent microscope were than those estimated by light microscope. The enumeration and identifying of Arexandriurn tamarense using DNA probe method will be contributed to a new biotoxin monitoring and prediction system in field.

Combined Effects of Filter-feeding Bivalve and Zooplankton on the Growth Inhibition of Cyanobacterium Microcystis aeruginosa (남세균 제어를 위한 동물플랑크톤(Daphnia magna)과 패류(Unio douglasiae)의 단독 및 혼합적용)

  • Single - and combined effects of a domestic freshwater bivalve Unio douglasiae (7.6~8.6 cm in shell length) and zooplankton Daphnia magna (1~2 mm in body size) were examined to understand whether they inhibit the growth of harmful cyanobacterial bloom (i.e. Microcystis aeruginosa) in a eutrophic lake. The experiments were triplicated with twelve glass aquaria (40 L in volume); three aquaria without mussel and zooplankton, served as a control, three zooplankton aquaria (Z, density=40 indiv. $L^{-1}$), three mussel aquaria (M, density=0.5 indiv. $L^{-1}$), and three mussel plus zooplankton aquarium (ZM, density=40 indiv.Z $L^{-1}$ plus 0.5 indiv.M/L), respectively. Algal growth inhibition (%) calculated as a difference in the concentration of chlorophyll-a (Chl-a) before and after treatment. Chl-a in all aquaria decreased with the time, while a greatest algal inhibition was seen in the ZM aquaria. After 24 hrs of incubation, Chl-a concentration at the mid-depth (ca. 15 cm) in ZM aquaria reduced by 90.8% of the control, while 63.2% and 79.8% in Z and M aquaria, respectively. Interestingly, during the same period, the surface Chl-a was diminished by 51.9% and 65.4% relative to the control in Z and ZM aquaria, while 27.4% of initial concentration decreased in M aquarium, respectively. These results suggest that 1) this domestic freshwater filter-feeding bivalve plays a significant role in the control of cyanobacterial bloom (M. aeruginosa), and 2) the combination with zooplankton and mussel has a synergistic effect to diminish them, compared to the single treatment of zooplankton and mussel.

  • The digestive tract of the rockfish, Sebastes schlegeli composed of pharynx, esophagus, stomach, intestine, anus and ten or eleven pyloric caeca. Pyloric caeca is blind sac of banana shape, and that is originated from pyloric portion of the stomach. The relative length of gut(RLG), that is length of digestive tract to standard length, is about 1.56(n=10). Esophageal muscularis consists of thin outer layer of longitudinal muscle and thick inner layer of circular muscle. Mucosal epithelium consists of columnar epithelium with short microvilli and contains numerous mucous secretory cell. The mucosal folds of the stomach are regular, and the muscularis consists of longitudinal, oblique and circular muscle layer. The chief cell of the gastric gland have a tubular mitochondria, endoplasmic reticula and numerous secretory granules in electron-dense. However, parietal cell contains small mitochondria, endoplasmic reticula and vacuoles in low electron density. Mucosal epithelium of the pyloric caeca and intestine composed of columnar epithelium, goblet cell, rodlet cell and dark cell. Columnar absorptive cell in the pyloric caeca and intestine contains well developed mitochondria, endoplasmic reticula, vesiculated granules in high electron density, pinocytotic vesicles and multivesicular body. Rodlet cell have a well developed cytoplasmic capsule and the endoplasmic reticula in the cytoplasm. Dark cell showing a high electron density in the cytoplasm and contains well developed mitochondria. Columnar epithelium of the intestine have a well developed intercellular junction and the microvilli which contains actin filament originated from the cytoplasm. Mucosal epithelium of the intestine have a longer microvilli and more abundant goblet cells than in the pyloric caeca.
