Selective Algicidal Effects of a Newly Developed GreenTD against Red Tide Harmful Alga (GreenTD 물질을 이용한 유해 적조 발생 종의 선택적 살조능 평가)

  • Harmful algal blooms (HABs) are a serious problem for public health and fisheries industries, thus there exists a need to investigate the possible ways for effective control of HABs. In the present study, we investigated the algicidal effects of a newly developed GreenTD against the HABs (Chattonella marina, Heterosigma akashiwo, Cochlodinium polykriokides, and Heterocapsa circularisquama) and non-HABs (Chaetoceros simplex, Skeletonema sp. and Tetraselmis sp.), which is focused on the different population density and concentration gradients of algicidal substances. The time series viability of target alga was assessed based on the activity of Chl. a photosynthetic efficiency in terms of $F_v/F_m$, and in vivo fluorescence (FSU). Effective control of Raphidophyta, C. marina and H. akashiwo was achieved at a GreenTD concentration of $0.5{\mu}gL^{-1}$ and $0.2{\mu}gL^{-1}$, respectively, and regrowth of both the species was not observed even after 14 days. The inhibitory ratio of the dinoflagellate, C. polykriokides was more than 80% at $0.2{\mu}gL^{-1}$ of GreenTD. H. circularisquama was constantly affected in the presence of $0.2{\mu}gL^{-1}$ of GreenTD in the high- and low-population density experimental groups. On the other hand, diatoms, C. simplex, and Skeletonema sp. were not significantly affected even in the presence of $0.2{\mu}gL^{-1}$ of GreenTD and exhibited re-growth activity with the passage of incubation time. In particular, green alga Tetraselmis sp. remained unaffected even in the presence of the highest concentration of GreenTD ($1.0{\mu}gL^{-1}$), implying that non-HABs were not greatly influenced by the algicidal substances. As a result, the algicidal activity of GreenTD on the harmful and nonharmful algae was as follows: raphidophyte>dinoflagellates>diatoms>green alga. Consequently, our results indicate that inoculation of GreenTD substances into natural blooms at a threshold concentration ($0.2{\mu}gL^{-1}$) can maximize the algicidal activity against HABs species. If we consider the dilution and diffusion rate in the field application, it is hypothesized that GreenTD will demonstrate economic efficiency, thus leading to effective control against the target HABs in the closed bay.

Early Detection of Cochlodinium polykrikoides (Dinophyceae) Blooms in Namhaedo in 2019 Using Quantitative Real-Time PCR (qPCR) (Quantitative real-time PCR (qPCR)을 이용하여 2019년 남해도 해역에서 발생한 Cochlodinium polykrikoides (Dinophyceae) 적조의 조기검출)

  • Quantitative real-time polymerase chain reaction (qPCR) was applied for the early detection of red tides in the coastal areas of South Gyeongsang in 2019. Cochlodinium polykrikoides (Dinophyceae) was detected at very low cell densities (0.0015~0.0058 cells mL-1) in early June, but its cell density increased by up to 0.163 cells mL-1 in mid-August. Higher cell densities were detected mainly in Namhaedo using both qPCR and microscopy (maximum 24 cells mL-1) in late-August. Accordingly, a red tide alert was issued on September 2 (maximum 200 cells mL-1) on this island. C. polykrikoides cell density in Namhaedo peaked on September 11 (12,000 cells mL-1). Our results indicate that C. polykrikoides was detected at very low cell density in Namhaedo prior to bloom, which occurred in the same area. Therefore, qPCR is a useful tool to detect even at very low cell densities of C. polykrikoides for early warning of blooms.

Assessment of New Algicide Thiazolidinedione (TD49) for the Control of Marine Red Tide Organisms (해양적조생물제어를 위한 살조물질 Thiazolidinedione 유도체(TD49) 평가)

  • Worldwide development of harmful algal blooms causes serious problem for public health and fisheries industries. To evaluate the algicidal impact on the harmful algae bloom species in aquatic ecosystems of coast, a new algicide thiazolidinedione derivative (TD49) were tentatively examined in the growth stages (i.e., lag, logarithmic and stationary phase) of rapidophyceae $Heterosigma$ $akashiwo$, $Chattonella$ $marina$ and $Chattonella$ sp..Three strains could easily destroy in the lag phase due to relatively weak cell walls than those of the logarithmic and stationary phase. It is thought that inoculation of TD49 substances into initial or developmental natural blooms with a threshold concentration ($2{\mu}M$) can maximize the algicidal activity. Also, bio-chemical assays revealed that the algicidal substances from all culture strains were likely to be extracellular substances because those cells have easily destroyed in cell walls. On the other hand, natural zooplankton communities were influenced within the exposure experiments of $2{\mu}M$, which is showed the maximum algcidal activity of tested organisms. These results indicate that although the TD49 substance is potential agents for the control of $H.$ $akashiwo$, $C.$ $marina$ and $Chattonella$ sp. in the enclosed eutrophic bay and coastal water, more detailed research of acute toxicity effect on high trophic organism in marine ecosystems need to be conducted.

Experimental determination of design parameters for filtration trench using phosphorus removal granular materials (인 제거 입상소재를 적용한 여과수로 설계인자의 실험적 결정)

  • The algal blooms in stagnant streams and lakes have caused many problems. Excessive algae leads to disturbance of ecosystem and overload of water treatment processes. Therefore, phosphorus(P), source of algal blooms, should be controlled. In this study, a filtration trench has been developed to convert dissolved phosphorus into hydroxyapatite(HAP) so that it could be crystallized on the surface of 'phosphorus removal granular material'; and residual particulate phosphorus could be removed by additional precipitation and filtration. The front and rear parts of filtration trench consisted of 'phosphorus removal granular material contact bed' and 'limestone filtration bed', respectively. As a result of the column test using phosphorus removal granular material and limestone serially, $PO_4-P$ was removed more than 90% when EBCT(empty bed contact time) of the contact bed was over 20 minutes; and T-P represented 60% of removal efficiency when total EBCT was over 1.5 hours. The results of column tests to figure out the sedimentation characteristics showed that more than 90% of particulate phosphorus could be removed within 24 hours. It was necessary to optimize the filtration part in order to increase removal efficiency of T-P additionally. Also, it was confirmed through the simulation of Visual MINTEQ that most of particulate phosphorus in the column tests is the form of HAP. Based on the results of the study, it could be suggested that the design parameters are over 0.5 hour of EBCT for phosphorus removal granular material contact bed and over 1.5 hours of EBCT for limestone filtration bed.

Effects of Environmental Factors on Algal Communities in the Nakdong River (낙동강의 환경요인이 조류군집 구성에 미치는 영향)

  • This study was carried out to investigate algal community structures and their correlations with environmental factors on five weir areas in the Nakdong River, South Korea. Water qualities, hydrodynamics, meteorological conditions and algal species compositions were observed in studied sites from May 2010 to Dec. 2013. Results showed that average total phosphorus concentration of 2013 was decreased by 52.4% in comparing with that from 2010 to 2011. Chlorophyll.a concentrations were positive significant with water temperature, pH, total phosphorus and total nitrogen, but is not significant with turbidity and suspended solids. Seasonal successions of algae were observed that Stephanodiscus sp. was dominant species with 65.3% of dominant frequency in studied site. Large algal biomass of the low temperature-adapted diatoms were observed during temperature range of $4{\sim}9^{\circ}C$, but large cyanobacterial biomass mainly during high temperature period ranged from $22^{\circ}C$ to $32^{\circ}C$. Microcystis sp. dominated during high water temperature in summer. The yearly correlations of algal biomass with accumulated solar radiations were not significant but seasonal correlations of summer from June to August were significant with correlation coefficient 0.33 (p<0.05). There were not significant correlations between turbidities and algal biomass. Turbidity and suspended solids concentrations were not significant correlation with algal biomass. According to the results, algal communities had strong correlation with water temperature and had partially correlation with solar radiation. For an effective management of algal blooms, water managers should survey with more long-term monitoring of various environmental factors and algal communities.

Growth rates and nitrate uptake of co-occurring red-tide dinoflagellates Alexandrium affine and A. fraterculus as a function of nitrate concentration under light-dark and continuous light conditions

  • The dinoflagellate genus Alexandrium is known to often form harmful algal blooms causing human illness and large-scale mortality of marine organisms. Therefore, the population dynamics of Alexandrium species are of primary concern to scientists and aquaculture farmers. The growth rate of the Alexandrium species is the most important parameter in prediction models and nutrient conditions are critical parameters affecting the growth of phototrophic species. In Korean coastal waters, Alexandrium affine and Alexandrium fraterculus, of similar sizes, often form red-tide patches together. Thus, to understand bloom dynamics of A. affine and A. fraterculus, growth rates and nitrate uptake of each species as a function of nitrate ($NO_3$) concentration at $100{\mu}mol\;photons\;m^{-2}s^{-1}$ under 14-h light : 10-h dark and continuous light conditions were determined using a nutrient repletion method. With increasing $NO_3$ concentration, growth rates and $NO_3$ uptake of A. affine or A. fraterculus increased, but became saturated. Under light : dark conditions, the maximum growth rates of A. affine and A. fraterculus were 0.45 and $0.42d^{-1}$, respectively. However, under continuous light conditions, the maximum growth rate of A. affine slightly increased to $0.46d^{-1}$, but that of A. fraterculus largely decreased. Furthermore, the maximum nitrate uptake of A. affine and A. fraterculus under light : dark conditions were 12.9 and $30.1pM\;cell^{-1}d^{-1}$, respectively. The maximum nitrate uptake of A. affine under continuous light conditions was $16.4pM\;cell^{-1}d^{-1}$. Thus, A. affine and A. fraterculus have similar maximum growth rates at the given $NO_3$ concentration ranges, but they have different maximum nitrate uptake rates. A. affine may have a higher conversion rate of $NO_3$ to body nitrogen than A. fraterculus. Moreover, a longer exposure time to the light may confer an advantage to A. affine over A. fraterculus.

Effect of oyster shell powder on nitrogen releases from contaminated marine sediment

  • Nitrogen flux release from organically enriched sediments into overlying water, which may have significantly influence on water quality and increasing continuous eutrophication. The purpose of this study is to evaluate the remediation efficiency of oyster shell powder and its treated product into organically enriched sediment in terms of nitrogen flux, organic matter, chlorophyll-a, pH and dissolved oxygen (DO). The TOSP was mainly composed of CaO2. The application of TOSP into the sediment has increased the pH, DO and significantly decreased the concentrations of NH4+-N and T-N compared to other basins. On the other hand, nitrate was enriched with the addition of treated oyster powder, an oxygen releasing compound on both phases. Furthermore, chlorophyll-a was found to be increasing with time in the control basin meanwhile it dropped drastically with the addition of TOSP, which implied on the repression of algal growth owing to blockage of nitrogen source migrating from the sediment. This study has shown that the TOSP was effective to improve sediment-water quality, diminish eutrophication and control harmful algae blooms in a marine environment. Therefore, it is a good reference as an effective environmental remediation agent.

Understanding Alginate Fouling in Submerged Microfiltration Membrane System for Seawater Pretreatment (해수전처리를 위한 침지식 정밀여과 멤브레인 시스템에서 Alginate 파울링의 이해)

  • Organic fouling observed in submerged membrane filtration as a pretreatment for seawater desalination increases energy consumption for membrane operation because of requiring frequent chemical cleaning and membrane replacement. In membrane pretreatment for seawater facing with algae blooms, membrane fouling was observed in submerged microfiltration using sodium alginate model compound which is one of the main components of extracellular polymeric substances. Without aeration, aglinate fouling increased with its concentration while aeration reduced the alginate fouling effectively regardless of its concentration tested. In the absence of aeration, alingate fouling tended to be decreased with increasing calcium concentration. However, this effectiveness was reduced by increasing sodium chloride concentration. At high concentration of sodium chloride and calcium similar to the seawater conditions, aeration reduced initial fouling. However, as time progressed, the effect of increased airflow rate on fouling reduction was not significant, implying that optimum airflow rate to control alginate fouling in submerged microfiltration can exist.

Effects of Environmental Factors on Akinete Germination of Anabaena circinalis (Cyanobacteriaceae) Isolated from the North Han River, Korea (북한강 수역에서 분리한 남조류 Anabaena circinalis 휴면포자 발아에 대한 환경요인의 영향)

  • Akinete germination may be a starting point of some akinete-producing cyanobacterial blooms in eutrophic freshwater systems. This study examined germinability of akinete of a cyanobacterium Anabaena circinalis isolated from the sediment of the North Han River (Cheongpyung Dam), Korea, under several environmental factors such as temperature ($5{\sim}25^{\circ}C$), light ($0{\sim}100{\mu}mol\;photons\;m^{-2}\;s^{-1}$), nutrients (nitrogen and phosphorus) and pH (5~12). The high germination rate appeared at high temperature: >55% at $25^{\circ}C$, followed by 15% at $15^{\circ}C$, $10^{\circ}C$ and 10% at $5^{\circ}C$. Low light intensity was favorable for akinete germination. Over 45% of germination occurred at low light intensities (5, 15 and $30{\mu}mol\;photons\;m^{-2}\;s^{-1}$), while less than 10% of germination occurred at both 50 and $100{\mu}mol\;photons\;m^{-2}\;s^{-1}$. No germination occurred in the dark condition. Akinete germination rate increased with nutrient (phosphorus and nitrogen) enrichment, and nitrogen addition showed greater effect on the germination compared to phosphorus addition. Akinetes germinated well at neutral or slightly alkaline pH condition (pH 7 and pH 8: >55%), but no germination was observed at pH 11~12. The present study demonstrates some favorable ambient conditions of Anabaena circinalis germination, which could provide useful information to study the germination conditions of other Anabaena species or akinete-forming algae and predict its bloom in eutrophic freshwaters.

The Effect of $_L$=lysine on Growth Inhibition of Microcystis sp. ($_L$ =lysine에 의한 Microcystis sp.의 선택적 성장억제)

  • Various physico-chemical and biological methods have been used to remove. cyanobacteria which causes blooms and releases toxin. The purpose of the following experiment is aimed finding out which cyanobacteria are affected by $_L-lysine $ and what concentration of$_L-lysine $ inhibits cyanobacteria. The 20 samples of Microcystis sp. have been tested. To prove the growth inhibition on Microcystis sp., double-layered agar method and microplate method have been used. When the concentration of $_L-lysine $ is as heavy as 100 ${\mu}g\; ml^{-1}$~300 ${\mu}g\; ml^{-1}$, some Microcystis sp. have made halo zone. Some Microcystis sp. have shown so high activity as to be inhibited in their growth by the $_{L}$-lysine of concentration 10 ${\mu}g\; ml^{-1}$ with microplate method. These activities are various in accordance with every species. In additions, the microplate method has been proven to be an easy way which examine the lytic activity on the species of algae.e.
