Use of Molecular Detection Technique for Red Tide Warning of Cochlodinium polykrikoides (Cochlodinium polykrikoides 적조출현주의보 발령에 분자탐침기법의 활용)

  • Real-time PCR was applied to early warning of red tide. For early warning of red tide at $10cells\;mL^{-1}$, Cochlodinium polykrikoides specific real-time PCR was used as a complement of microscopy that has a lower detection sensitivity. C. polykrikoides appeared extensively in Tongyeong, Namhae waters at low densities in the end of June, and early warning of C. polyrkrikoides blooms was issued on 2 August 2015.

Fundamental Structure in Simultaneous Removal for Phytoplankton and Nutrient Salt in Lakes

  • The water quality in eutrophic lakes is affected by serious problems, such as abnormal increasing of Cyanobacteria. The purpose of this study was to investigate the possibility of a modified flotation system using a hybrid technique formed by chemical compounds and an electrostatic bridge. Therefore, experiments using the hybrid technique were performed to measure the zeta potential value on the phytoplankton surface and the removal efficiencies of phytoplankton, ammonia nitrogen, nitrate nitrogen and phosphoric acid. The results were as follows: Firstly, the zeta potential of M.aeruginosa was observed to approach charge neutralization due to adhesion of magnesium hydroxide precipitate on the phytoplankton surface in the pH range 10.5 to 11. Secondly, the concentration of chlorophyll-a decreased from about 150 to 20${\mu}g$g/L, with a maximum removal efficiency of 84% due to coagulation with pH values higher than 10. Thirdly, the N$H_4$-N concentration was observed to decrease from 0.62 to 0.54mg-N/L (13%), and the P$O_4$-P concentration, which is a limiting factor to the formation of algae blooms, decreased from 0.27 to 0.02mg-P/L (92%). These findings suggest that the modified flotation system can be applied for the purification of the raw water of numerous lakes containing high phytoplankton populations and elevated pH.

The Effect of Current and Temperature of a Reservoir by the Simulation of Dam Outflow (댐 방류조건에 따른 저수지 유속과 수온 영향)

  • Water quality in the Daecheong reservoir has been deteriorated by algal bloom due to nutrient supply from the upstream of the Daecheong reservoir after heavy rainfall. Algal bloom is propagated from eutrophicated tributary into the main body of the reservoir according to the hydrological conditions. This study is aimed to estimate the water current and temperature effect by the simulation of dam spill flow control using water quality model, CE-QUAL-W2 in 2003. Water current was resulted in nutrient transport from upstream of main reservoir and nutrients were delivered up to downstream by fast water velocity. Algal blooms occurred in stagnate zone of reservoir downstream as the current of downstream was retarded according to dam outflow control. Consequently water balance in stagnate zone triggered a rise of water temperature in summer. It affected algal bloom in the embayment of the reservoir. The simulation result by outflow control scenarios showed that spill flow augmentation induced in water body instability of stagnate zone so that water temperature declined. It could be suggested that outflow control minimize algal bloom in the downstream in the flooding season as long as water elevation level is maintained properly.

Selection of Chemicals for the Dissolved Phosphorus Control by Variations of Alkalinity and Hardness (용존인 제어를 위한 알칼리도와 경도 조절제의 선택)

  • This study was done to find chemicals adequate to control alkalinity and hardness in order to reduce dissolved phosphorus in water bodies like rivers and lakes. Five chemicals were selected for the study: calcite, lime, dolomite, magnesite, and gypsum. Data were obtained from the calculations with MINTEQ model as a function of dosage variations of each chemical. Findings are as follows: Three out of the five chemicals are found to be effective in reducing the dissolved phosphorus, i.e., calcite, lime, and dolomite. Calcite and dolomite are able to lower the phosphorus concentration up to one thousandth fold whereas lime does one hundred thousandths fold. In viewpoint of pH variation, both calcite and dolomite seem to be safe since the pH does not increase over 8.3 even in case of overdose. In the same circumstance, with lime the pH increases beyond 9 which is considered to be the highest pH level for the protection of water ecosystem. Nevertheless it is recommendable to use lime in case where there are some difficulties in water quality control due to algae blooms.

Operation and Diagnosis of DAF Water Treatment Plant at Highly Turbid Raw Water (고탁도시 DAF 정수장의 운영 및 진단)

  • DAF process has been designed considering raw water quality characteristics in Korea. Although direct filtration is usually operated, DAF is operated when the freshwater blooms occut or raw water turbidity become high. Pre-sedimentation iS prepared in case when raw water turbidity is very high by rainstorms. A main feature of this plant is that the operation mode can be changed (controlled) based on the characteristics of raw water to optimize the effluent quality and the operation costs. Treatment capacity (surface loading rate) and efficiency of DAF was found to be better than conventional sedimentation process. Moreover, low-density particles (algae and alum flocs) are easily separated while it is difficult to remove in sedimentation. One of the main concerns in adoption of DAF (Dissolved Air-Flotation) process is a high raw water turbidity problem. That is, DAF is not adequate for raw water, which is more turbid than 100NTU. In order to avoid this problem, pre-sedimentation basins are prepared in DAF plant to decrease the turbidity of DAF influent. For simulation of the actual operation, bench and full-scale tests were performed for highly turbid water conditions. Consequently, DAF process coupled with sedimentation is suggested that pre-sedimentation with optimum coagulation prior to DAF would be appropriate.

Development of a Fluorescence Measurement System Capable of Rapid Red Tide Monitoring (신속한 적조 예찰이 가능한 형광 측정시스템 개발)

  • The occurrence of harmful algae on the coast of Korea has been a cause of damage to the aquaculture industry and deterioration of the coastal ecosystem environment. A method is required to predict their outbreak in real-time at the site. Therefore, this study attempted to develop a small hybrid optical sensor and real-time monitoring system based on LiDAR that can be used in the field and laboratory and can be applied to various platforms. FMS-L specifically suggested the amount of Chlorophyll a (Chl a) in the sample by measuring and analyzing the fluorescence emitted by the irradiating light. The accuracy of FMS-L was verified by measuring the concentrations of standard Chlorophyll a substances and Margalfidinium polykirkoids. In addition, the precision was verified by comparing the measurement results of FMS-L using commercial equipment Phyto-PAM-II. This equipment is compact and easy to move. Therefore, it can be easily applied to field surveys, allows short time measurements (10 s), and can be applied at a distance of 10 m from the measurement site.

Characterization of Filamentous Cyanobacteria Encapsulated in Alginate Microcapsules (알긴산염 마이크로캡슐 내부에 동결보존된 사상체 남세균의 특성 연구)

  • Cyanobacteria are microorganisms which have important roles in the nitrogen cycle due to their ability to fix nitrogen in water and soil ecosystems. They also produce valuable materials that may be used in various industries. However, some species of cyanobacteria may limit the use of water resources by causing harmful algal blooms in water ecosystems. Many culture collection depositories provide cyanobacterial strains for research, but their systematic preservation is not well-developed in Korea. In this study, we developed a method for the cryopreservation of the cyanobacteria Trichormus variabilis (syn. Anabaena variabilis), using alginate microcapsules. Two approaches were used for the experiments and their outputs were compared. One of the methods involved the cryopreservation of cells using only a cryoprotectant and the other used the cryoprotectant within microcapsules. After cryopreservation for 35 days, cells preserved with both methods were successfully regenerated from the initial 1.0 × 105 cells/ml to a final concentration of 6.7 × 106 cells/ml and 1.1 × 107 cells/ml. Irregular T. variabilis shapes were found after 14 days of regeneration. T. variabilis internal structures were observed by transmission electron microscopy (TEM), revealing that lipid droplets were reduced after cryopreservation. The expression of the mreB gene, known to be related to cell morphology, was downregulated (54.7%) after cryopreservation. Cryopreservation using cryoprotectant alone or with microcapsules is expected to be applicable to other filamentous cyanobacteria in the future.

Characteristics of Cyanobacterial Occurrence and Concentration Distribution of Cyanotoxins in Hoeya Reservoir (회야호의 남조류 발생 특성과 남조류 독소의 농도분포특성)

  • Algae blooms have soared recently in the lakes across the nation due to eutrophication. Blue-green algae cause unpleasant scene, produce taste and odor problem, and hinder processes in drinking water treatment. Algae toxicity monitoring has been strengthened, because the damages of wild lives and livestocks by algal toxins have been reported. Investigation on the characteristics of cyanobacterial occurrence and concentration distribution of Cyanotoxins in Hoeya reservoir have been conducted. Physical and chemical influences of water environment on cyanobacterial occurrences have also been studied. Movements of four species of Microcystin and five species of Anatoxin-a among Cyanotoxins were observed by LC-MS/MS analysis. Microcystis spp. among the cyanobacteria have mainly dominated in the Hoeya reservoir during the investigating period. The density of cyanobacteria were positively correlated with temperature and pH of water. Highest concentrations of Microcystin-LR and Microcystin-RR were $0.424{\mu}g/L$ and $0.117{\mu}g/L$ at the sampling points. Total concentration of Cyanotoxins in water coming into the water treatment plant was $0.182{\mu}g/L$, and they were not detected in treated water.

Growth Characteristics of Blue-green Algae (Anabaena spiroides) Causing Tastes and Odors in the North-Han River, Korea (북한강 수계에서 이취미를 유발하는 남조류(Anabaena spiroides)의 증식 특성)

  • Blue-green algae blooms occurred during early winter in the North-Han River, Korea. Among blue-green algae, Anabaena spiroides were observed for approximately 33 consecutive days, between 28 November and 30 December, 2011. A. spiroides emerged from Lake Uiam to Lake Paldang, depending on the flow of the river has spread downstream. Changes of physical water environment like rising water temperature and increasing hydraulic retention time influenced the A. spiroides bloom. The A. spiroides bloom showed a very rapid increase in cell density, and a slow decrease: the cell density increased to a maximum of $11,325cells\;mL^{-1}$ in Lake Paldang (st. 5), and was completely disappeared after the water temperature dropped below $4^{\circ}C$. A decrease in water temperature was the most influential factor among all environmental parameters, for the reduction of A. spiroides cell density. The A. spiroides bloom was accompanied with the occurrence of very high concentrations of the odor metabolite geosmin. Geosmin reached the peak value of $1,640ng\;L^{-1}$ in Lake Paldang (st. 4). The geosmin concentration was very strongly correlated with cell numbers of A. spiroides.

A Study on the Application of Drone to Prevent the Spread of Green Tides in Lake Environment (호수 환경의 녹조 확산 방지를 위한 드론 적용 방안에 관한 연구)

  • Recently, water shortages have occurred due to climate change, and the need for water management of agricultural water has increased due to the occurrence of algal blooms in reservoirs. Existing algae prevention is operated by putting many people on site and misses the optimal spraying time due to movement through boats. In order to solve this problem, it is necessary to block contamination in advance and move within time to uniformly spray complex microorganisms uniformly. Control drones are used for pesticide spraying and can be applied to algae prevention work by utilizing control drones. In this paper, basic research for the establishment of a marine control system was conducted for application to the reservoir environment, and as one of the results, the characteristics of a drone nozzle, a core technology that can be used for control drones, were calculated. In particular, it was found that the existing agricultural control drones had a disadvantage that the concentration was non-uniform within the suggested spraying interval, and to compensate for this, nozzle positioning and nozzle spraying uniformity were calculated. Based on the experimental results, we develop a core algorithm for establishing an algal bloom monitoring system in the reservoir environment and propose a precision control technology that can be used for marine control work in the future.
