부영양 호수에서 분리한 Acidovorax temperans AK-05의 Anabaena cylindrica 분해 특성 (Characterization of a Novel Alga-Lytic Bacterium, Acidovorax temperans AK-05, Isolated from an Eutrophic Lake for Degradation of Anabaena cylindrica)

  • 부영양 호수로부터 살조 세균을 분리하고 동정한 결과 Anabaena cylindrica NIES-19를 유릴 탄소원으로 이용하는 double layer방법으로 15종의 살조세균을 분리하였으며 높은 살조 활성을 나타내는 4종의 살조세균 AK-05, AK-07, AK-13 그리고 AK-28을 선별하여 살조 능력을 비교하였다. 이들 중에 AK-05가 가장 높은 살조 활성을 나타내었으며 이를 16S rDNA염기서열을 분석한결과 Acidovorax temperans와 99.5%의 유사성을 나타내어 Acidovorax temperans AK-05로 명명하였다. A. temperans AK-05의 배양 여액을 A. cylindrica NIES-19에 뚜렷한 살조 활성을 나타내었으며, 이것의 살조 활성 능력을 분석한 결과 살조 활성에 관여하는 주요 물질은 non-protein이며 열에 안정적이었다. 이러한 살조 활성 능력은 알칼리 조건과 25${\sim}$$30^{\circ}C$에서 가장 높게 나타냈다. 따라서 이와 같은 특성은 일반적으로 알칼리 조건을 야기하는 Cyanobacteria에 의한 water blooms이 발생하는 호수에 적용하는데 매우 유리할 것으로 여겨진다.

  • Alga와 같은 미생물은 중금속을 흡수한다. Alga(Anabaena)로 변성시킨 탄소반죽전극으로 Cd(Ⅱ)이온을 사전농축시켜서 Cd(Ⅱ)을 양극벗김 펄스차이 전압-전류법으로 정량하였다. Cd(Ⅱ)의 산화봉우리는 -0.75V vs. SCE.에서 나타났으며, 이 봉우리를 이용하여 Cd(Ⅱ) 정량의 최적조건을 조사하였다. 변성전극의 제작에 미치는 alga양의 영향과 Cd(Ⅱ)의 사전농축에 미치는 pH와 이온세기, 온도 및 사전농축시간에 대한 영향을 조사하였으며. 전극표면에 사전 농축시킨 Cd(Ⅱ)이온을 전해환원시키는 시간과 전위의 영향도 조사하였다. Cd(Ⅱ)을 정량하기 위한 검정선은 $1.0{\times}10^6\;M ~ 8.0{\times}10^6\;M$범위에서 직선성(상관계수는0.9978)이 성립하였고 검출한계는 $5.0{\times}10^{-7}\;M$이었다. $7.0{\times}10^{-6}\;M$ Cd(Ⅱ)용액에서 얻은 상대표준편차는 3.1%(n=6)이었다. Alga변성전극의 사용횟수는 0.1M HCl 용액으로 전극표면을 재생시킬 때에 10회까지 연속측정이 가능하였다.

New record of Codium lucasii (Bryopsidales, Chlorophyta) in Korea

  • A prostrate species of Codium (Bryopsidales, Chlorophyta) was collected from Daejin on the eastern coast of Korea. This alga is morphologically characterized by a prostrate, adherent or pulvinate, dark green thallus that is tightly attached to substratum. The utricles are strongly grouped and cylindrical to slightly clavate. Their apex is rounded to capitated, and it frequently has an alveolate ornament. Hair scars are found in the upper portion of the utricle. The gametangia grow on a short pedicel in the upper part of the utricle. In the phylogenetic tree based on molecular data, this alga is placed in the same clade as C. mozambiquense in UPGMA analysis, and nests in a sister clade of C. lucasii subsp. capense and C. mozambiquense in ML and NJ analyses. However, the genetic distance between the sequences of the Korean alga and the two species is 1.3-1.9%, while that between the Korean alga and C. lucasii from Japan is 1.1% within intraspecific range. The divergence value between the Korean alga and C. lucasii from the type locality (Australia) is 2.7% considered to be interspecific range. As based on this genetic divergence value, the Korean alga together with Japanese C. lucasii can be separated from genuine C. lucasii from the type locality. However, the Korean alga is identified as C. lucasii until those entities are morphologically characterized in species level. This is the first record of C. lucasii in Korea

제주도에 서식하는 보호대상 해조류 넓미역의 생태학적 특성: 생장 및 성숙 (Ecological Characteristics of the Endangered Brown Alga, Undariopsis peterseniana (Kjellman) Miyabe et Okamura, at Jeju Island, Korea: Growth and Maturation)

  • Ecological characteristics of Undariopsis peterseniana, an endangered brown alga, were investigated from January to December 2007 at Jeju Island, Korea. The U. peterseniana population formed dense patches at depths of 12~17 m depth. Growth in length of the alga reached a maximum of $168{\pm}36cm$ at June. Sorus were observed from May to August, and a maturation peak was detected in June, when bottom seawater temperatures were around $18^{\circ}C$. After zoospore release, the alga bleached and disappeared after August. Developmental initiation of sporophytes occurred at temperature above $15^{\circ}C$, and sorus formation required approximately 236 degree-days. This is the first study to examine a U. peterseniana population throughout the entire year in a natural habitat.

New record of an economic marine alga, Ahnfeltiopsis concinna, in Korea

  • An economic marine alga, which is considered to be an important source of carrageenan, was collected from Jindo of the southern coast of Korea. This species shares the vegetative and female reproductive features of Ahnfeltiopsis and is characterized mostly by its small size (up to 8 cm), terete to subterete thalli at the lower portion, cartilaginous in texture, dichotomous branches, rarely produced proliferations, and an absence of hypha-like filaments in the medulla. It is distinguished from other Korean species within the genus by the thallus feature. In a phylogenetic tree based on the molecular data, this alga nests in the same clade with A. concinna from Japan but forms a sister clade to A. concinna from Mexico and Hawaii (type locality). However, the genetic distance among those sequences was calculated as 0.1-1.3% for rbcL and 1.1% for COI sequences, considered to be intraspecific variation within the genus. Based on the morphology and molecular analysis, this alga is identified as A. concinna originally described from Hawaii. This is the first record of the species in the Korean marine algal flora.

Ulva grossa sp. nov. (Ulvales, Chlorophyta) from Korea based on Molecular and Morphological Analyses

  • A green alga specimen was collected from the eastern coast of Korea. This species shared the typical features of genus Ulva and was characterized by irregularly shaped thalli, relatively small and thick thallus, entire undulate margins without serrations, and one or two pyrenoids per cell. In a phylogenetic tree, based on sequences of the nuclear-encoded internal transcribed spacer region, it nests as a sister clade to a few species including Ulva ohnoi, which has a relatively large thallus. This Korean algal specimen differs from the species forming the same subclades, including U. ohnoi, Ulva fasciata, Ulva reticulata, and Ulva gigantean, and has a relatively small (3-8 cm) and thick (60-100 ㎛) thallus. Of these species, U. ohnoi, originally described from Japan, is similar to the Korean alga as it had a thick thallus of 30-90 ㎛, but it has microscopic serrations on the thallus margin, unlike the Korean alga. The genetic distance between the Korean alga species and the aforementioned species was determined to be 1.8%-4.8%, indicating an inter-specific divergence level at the genus Ulva. Herein, Ulva grossa sp. nov. (Ulvales, Chlorophyta) from Korea is described based on the morphological and molecular analyses.

New record of Chondrus retortus (Gigartinales, Rhodophyta) in Korea

  • A marine red algal species was collected from Honghyeon-ri, Namhae located on the southern coast of Korea during a survey of marine algal flora. This alga shares the generic features of Chondrus, and is characterized by short and thin, canaliculated and crisped fronds as C. retortus. Tetrasporangia are developed from shallow medullary cells on both sides of branches. In a phylogenetic tree based on rbcL sequences, the Korean alga nests in the same clade as C. retortus originally described from Japan. The genetic distance between both sequences within the clade was calculated as 0.0-0.1%. Based on the morphological and molecular data, the alga is identified as Chondrus retortus. This is the first record of C. retortus in Korean marine algal flora.

New record of Callophyllis mageshimensis (Gigartinales, Kallymeniaceae) in Korea

  • A marine red algal species was collected from Pyengdae-ri, Gujwa in Jeju, Korea during a survey of marine algal flora. This alga shares the generic features of Callophyllis(Gigartinales, Kallymeniaceae), and is characterized by erect and membranous thalli, branches dichotomously or trichotomoulsy divided, margin somewhat undulate or entire and tetrasporangia scattered irregularly in the cortex. In a phylogenetic tree based on rbc L sequences, the Korean alga nests in the same clade as C. mageshimensis originally described from Japan. The genetic distance between both sequences within the clade was 0.0-0.1%. Based on the morphological and molecular data, the alga was identified as Callophyllis mageshimensis. This is the first record of C. mageshimensis in Korean marine algal flora.

남조류 분해세균 HY0210-AK1의 분리와 특성 및 Anabaena cylindrica 분해 활성 (Isolation and Characterization of Alga-Lytic Bacterium HY0210-AK1 and Its Degradability of Anabaena cylindrica)

  • 남조류의 대발생이 자주 발생하는 석촌호수와 팔당호로부터 시료를 채취하여 남조류 분해세균의 분리를 시도하였다. MDM배지 상에서 Anabaena cylindrica를 숙주로 하는 over-layer method를 이용하여 Anabaena cylindrica lawn에서 남조류 분해 세균을 분리하여 HY0210-AK1으로 명명하였다. HY0210-AK1의 형태적, 생화학적 특성은 Rhizobium속과 유사하였으며, 16S rDNA 염기서열을 분석한 결과 Sphingobium herbicido vorans IFO 16415$^{T}$ 와 99.1%의 높은 유연관계를 보여 Sphingobium herbicidovorans HY0210-AK1로 동정하였다. 잠복기, 대수성장기, 정체기에 있는 Sphingobium herbicidovorans HY0210-AK1를 Anabaena cylindrica NIES-19와 혼합 배양하였을 때, 접종 초기에는 정체기의 Sphingobium herbicidovorans HY0210-AK1를 접종한 처리구에서 가장 좋은 살조 효과를 나타냈으나, 접종 7일 이후에는 커다란 차이를 보이지 않았다 또한,Sphingobium herbicidovorans HY0210-AK1는 $1\times 10^{8}$ CFU $ml^{-1}$ 이상의 농도로 처리하였을 때, Anabaena cylindrica NIES-19가 완전히 분해되었다. Sphingo bium herbicidovorans HY0210-AK1와 Anabaena cylin dricaNIES-19를 혼합 배양한 상등액을 여과하여 접종 하였을때는 납조류 성장 저해 효과가 나타나지 않았다. 따라서 Sphingobium herbicidovorans HY0210-AK1는 Anabaena cylindrica NIES-19를 직접 공격하여 분해하는 것으로 사료된다. Sphingobium herbicidovorans HY0210-AK1는 Microcystis aeruginosa 분해하였다..

충남 고정리의 하계 해조상 (Summer Algal Flora of Gojeong-Ri, West Coast of Korea)

  • The marine benthic algae of Gojeong-Ri was investigated in a floristic aspect. As a result, 34 species, 7 blue-green, 14 red, 6 brown and 7 green algae were identified as a summer flora from the area. Among them, Sirocoleum kurzii (blue-green alga), Ralfsia clavata (brown alga), Pseudulvella consociata and Cladophora speciosa (green algae) were known as new records to Korea.
