Species classification of the toxic dinoflagellate Alexandrium tamarense and A. catenella based on their paralytic shellfish toxin profiles

  • The annually outbreak of paralytic shellfish poisoning (PSP) were caused by toxic dinolagellate A. tamarense and A. catenella in Korea. The purpose of this study were to investigate the distribution of PSP-causative organisms, A. tamarense and A. catenella and their species classification. Sediment (Saemangeum, the south open sea) and water samples (southeastern coast) were sampled to establish clonal isolates in 2003. After isolation and purification, strains were cultured under $17^{\circ}C$, f/2 media, 14:10=L:D cycle. PST analysis and species identification were performed by HPLC-FD method and specific DNA probe, respectively. Thirty-ons strains were isolated from the Saemangeum reclamation, southeastern coast including Jinhae Bay and south open sea. PSTs were detected in all cultured strains. In eight strains from south offshore, major toxin components are GTX5, C1/2 and minors are GTX3/4, dcGTX3, neoSTX. Sixteen strains from south coastal area have GTX1/4, neoSTX, C1/2 as major toxin components and GTX2/3 as minors. Seven strains from the Saemangeum reclamation have GTX5, C1/2 as major toxin components and GTX1/2/3/4 as minors. Thus, among eight south offshore isolates, four A. tamarense have more toxic (38.31~l19.16 fmol.$cell^{-1}$) than A. catenella (3.78~13.13 fmol.$cell^{-1}$). With the previous results of different toxin composition, toxin components and toxin contents, .it is toxin profile that could used to diagnosis of regional toxic population and geographical distribution of both A. tamarense and A. catenella and their toxigenic strains.

A Versatile Method for DNA Sequencing of Unpurified PCR Products using an Automated DNA Sequencer and Tailed or Nested Primer Labeled with Near-infrared Dye: A Case Study on the Harmful Dinoflagellate Alexandrium

  • DNA sequence-based typing is considered a robust tool for the discrimination of dinoflagellate species because of the availability of extensive rDNA sequences. Here, we present a rapid, cost-effective DNA-sequencing technique for various PCR products. This sequencing strategy relies on 'nested' or 'tailed' primer labeled with near-infrared dye, and uses a minimal volume of unpurified PCR product (ca. $5{\mu}L$) as the DNA template for sequencing reactions. Reliable and accurate base identification was obtained for several hundred PCR fragments of rRNA genes. This quick, inexpensive technique is widely applicable to sequence-based typing in clinical applications, as well as to large-scale DNA sequencing of the same genomic regions from related species for studies of molecular evolution.

Dinoflagellate Cyst Assemblages in the Surface Sediments from the Northwestern East China Sea

  • Thirty-six dinoflagellate cysts, representing 15 genera were identified in the surface sediments obtained from the northwestern East China Sea. Three cyst morphotypes found in this survey have not previously been described in the East China Sea and adjacent waters: Seleno­pemphix sp. 2, Selenopemphix sp. 3 and Trinovantedinium sp. 1. In the northwestern East China Sea, Operculodinium centrocarpum, Spiniferites bulloideus and ellipsoidal cysts of Alexandrium were commonly observed. Moreover, it was recognized that the ellipsoidal cysts of Alexandrium, whose motile cells of A tamarense and/or A catenella are responsible to paralytic shellfish poisoning, distributed not only restricted to the coastal areas but also to the offshore stations far from the Changjiang River mouth.

Molecular Identification of the Toxic Alexandrium tamiyavanichii (Dinophyceae) by the Whole-cell FISH Method

  • The dinoflagellate Alexandrium tamiyavanichii Balech, a producer of toxins causing paralytic shellfish poisoning (PSP), has recently been considered as one of main organisms responsible for toxication of shellfish in Japan. In this study, A. tamiyavanichii was subjected to a molecular phylogenetic analysis inferred from 28S rDNA D1-D2 sequences and a species-specific LSU rRNA-targeted oligonucleotide DNA probe was designed to identify A. tamiyavanichii using the whole cell-FISH (fluorescence in situ hybridization). The sequences of the 28S rDNA D1-D2 region of A. tamiyavanichii showed no difference from A. cohorticular AF1746l4 (present name A. tamiyavanichii) and formed a distinct clade from the 'tamarensis species complex'. The probe, TAMID2, reacted specifically with A. tamiyavanichii cultured cells, without any cross-reaction with other species belonging to the same genus, including A. tamarense, A. catenella, A. affine, A. fraterculus, A. insuetum and A. pseudogonyaulax. In a test of cross-reactivity with a field sample, TAMID2 reacted consistently with only A. tamiyavanichii, indicating that the present protocol involving the TAMID2 probe might be useful for detecting toxic A. tamiyavanichii in a simple and rapid manner.

Modern Dinoflagellate Cysts Distribution off the Eastern Part of Geoje Island, Korea

  • Distributional characteristics of dinoflagellate cysts in surface sediments were investigated in relation to environmental factors in the eastern part of Geoje Island, Korea. Samples were collected from 10 stations in February of 2004 and water temperature and salinity were measured in February, May, September and November of 2004. Total 30 taxa of dinoflagellate cysts were identified representing 19 genera, 28 species and 2 unidentified species. Among these dinoflagellate cysts, Brigantedinium spp. of which relative proportion in th e total dinoflagellate cysts was 23.5%, was the most abundant at all stations except St. 1, and was followed by Spiniferites bulloideus (8.6%), Lingulodinium machaerophorum (8.2%) and Diplopsalis lenticula (6.7%). In addition, ellipsoidal cysts of the genus Alexandrium (Alexandrium catenella - tamarense type) and Gymnodinium catenatum, known to be causative organisms for PSP, occurred with high concentrations. Scrippsiella trochoidea was also found; however, its cyst concentration was low. Generally, species composition in the study area was similar to these reported from Jinhae Bay and Busan Harbor and several dinoflagellate cysts reflected the eutrophic condition. Cyst distribution in th e eastern part of Geoje Island seems to be influenced by the Tsushima Warm Current flowing from the southwest. The mean water temperature was $12.0^{\circ}C$ in February, $14.7^{\circ}C$ in May, $20.9^{\circ}C$ in September and $17.2^{\circ}C$ in November, which was most favorable for Alexandrium spp. growth. The abundances of dinoflagellate cysts rang e d from 528 to 2,834 cysts/g dry sediment. Higher concentrations were recognized in sediments of west area of the Jisimdo than at other stations. The cyst composition of this area was closely related to these of Jinhae Bay and Busan Harbor from which currents flow into this area. Higher cyst concentration in the west area of Jisimdo might be due to formation of the gyre.

적토의 활성화를 통한 적조구제물질 개발에 대한 연구 (Development of Anti-red Tide Material by Activating Red-mud)

  • 적토로부터 활성화시킨 물질에 대한 적조구제제로서의 가능성에 대하여 연구하였다. 적토는 보오크사이트로부터 알루미나를 생산하기 위해 사용되는 베이어 공정의 부산물로써 적철석, 보에마이트, 방해석, 소달라이트, 석영, 저어콘, 아나타제 및 미지의 광물 등 다양한 광물로 구성되어 있다. 중금속 흡착실험결과, As ($92\%$)를 제외한 모든 중금속이 거의 $100\%$에 가까운 높은 흡착률을 보였다. 이는 중금속에 대한 철산화물의 높은 흡착능력에 기인된 것으로 사료된다. 적토에 대한 pH (pH $1\∼13$)별 용출률 측정 결과, 산성에서 As, Cu 및 Zn가 높은 값을 보임으로써, 적토를 산처리 할 때, 중금속 제거 효과가 탁월함을 지시하였다. 적토의 산처리 산물인 활성적토는 적철석 및 보에마이트 등으로 구성되어 있으며, 온도가 증가할수록 중금속의 제거 효과가 증가하였다. 활성적토는 입도가 수십 nm로써, 5종의 적조에 대한 적조구제 효율을 측정한 결과, 구제효율은 pH특히, 반응 후 pH에 반비례하였다. 이들 5종의 프랑크톤 중, Prorocentrum minimum 및 Alexandrium tamarense의 경우, 살포 즉시 $90\%$ 이상을 그리고 약 30분 후에는 거의 $100\%$에 가까운 구제효율을 보임으로써, 활성적토가 이들 적조에 대한 우수한 구제제로서의 가능성을 지시하고 있다.

한국연안에서 분리한 적조형성 미세조류 10종의 성장에 미치는 온도, 염분, 광도의 영향 (Impacts of Temperature, Salinity and Irradiance on the Growth of Ten Harmful Algal Bloom-forming Microalgae Isolated in Korean Coastal Waters)

  • 한국 연안에서 출현하는 주요 적조생물의 적조발생 특성을 알아보기 위해 편모조류 10종을 대상으로 하여 수온과 염분, 광도에 따른 종의 성장률을 조사하였고, 또한 최근 13년간의 적조발생자료와 1999년도 및 2000년도 남해안연안에서 조사한 적조생물의 출현밀도를 비교, 분석하였다. 수온에 따른 적조생물의 성장은 Heterocapsa triquetra, Eutreptiella gymnastica, Alexandrium tamarense의 경우 대체로 $16{\sim}22^{\circ}C$에서 최대성장률을 보였으나, $10{\sim}16^{\circ}C$의 저수온에서도 비교적 높은 성장률을 보였고 $22^{\circ}C$이상의 고수온에서는 오히려 성장률이 급격히 감소되는 것으로 나타나 저온성종의 특성을 보였다. Prorocentrum micans와 Pyramimonas sp.는 $19^{\circ}C$ 이하의 저수온에서는 낮은 성장률을 보였으나 $22{\sim}25^{\circ}C$에서는 높은 성장률을 보여 고온종의 특성을 나타냈으며, Akashiwo sanguinea, Hetemsigma akashiwo Prorocentrum minimum, Scrippsiella tnchoidea는 $16{\sim}25^{\circ}C$의 광범위한 수온 범위에서 비교적 높은 성장률을 보여 광온성종의 특성을 보였다. 또한 이러한 결과는 이 종들이 자연상태에서 적조를 일으키는 수온 범위와도 대체로 일치하였다 염분에 따른 성장은 대부분의 종이 염분 $30{\sim}35$ psu에서 높은 성장률을 보였는데, E. gymnastica는 35 psu 이상의 고염분보다는 $10{\sim}30$ psu의 저염분에서 더 높은 성장률을 나타내 저염성을 보였으며, H. akashiwo, P. minimum, H. triquetra는 $15{\sim}40$ psu의 넓은 염분 범위에서 양호한 성장률을 보여 광염성종의 특성을 보였다. 이종들은 한국 연안에서 강우 직후 염분이 25 psu이하로 하강하는 저염분 상태에서 빈번히 적조를 일으키고 있는데, 이것은 이종들의 저염 및 광염성 특성과 밀접한 관련이 있는 것으로 판단되었다. 광도별 성장은 H. akashiwo, P. minimum, Pyramimonas sp.는 $150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$의 고광도에서 비교적 높은 성장률을 보였다. A. sanguinea, A. tamarense, H. triquetra는 $150{\sim}100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$에서 가장 높은 성장률을 보였고 $100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ 이상의 고광도에서 는 오히려 성장률이 급격히 감소되는 것으로 나타났다. 전반적으로 수온과 일조량이 높은 시기인 하계에 적조를 일으키는 고온성 종은 고광도에서 성장이 좋은 것으로 나타났고, 수온과 일조량이 낮은 동, 춘계에 적조를 일으키는 저온성 종은 저 광도에서 성장이 양호한 것으로 나타났다.

Community Dynamics and Distribution of Dinoflagellates and Their Cysts in Masan-Chinhae Bay, Korea

  • The community dynamics and distribution of dinoflagellates and their cysts were monitored monthly from June 1996 to May 1997 at six stations in Masan-Chinhae Bay, one of the famous bays for red-tide occurrence in Korea. The dinoflagellate standing crops ranged from the minimum of 306 cells $ml^{-1}$ in February to the maximum of 37,959 cells $ml^{-1}$ in May. The species causing massive blooms were Prorocentrum micans Ehrenberg and Ceratium furca (Ehrenberg) Claparede & Lachmann in July, Gymnodinium sanguineum Hirasaka in October, Alexandrium tamarense (Lebour) Balech in April and Proroeentrum minimum (Pavillard) Schiller in May. Twenty-seven taxa of dinoflagellate cysts were isolated, including 20 identified species and 7 unidentified species. The mean cyst abundance ranged from the minimum of 556 cysts $cm^{-3}$ in June to the maximum of 5,727 cysts$cm^{-3}$ in February. The spatial pattern of cyst distribution showed the gradual increase in abundance from offshore to inshore stations. The cyst genera of Protoperidinium, Gymnodinium, Serippsiella, Gyrodinium and Alexandrium were abundant throughout the year. The vertical distribution of cysts showed the surface miximum at the 0-2cm layer and the sub-surface maximum at the 2-4cm layer. Total abundance of cysts showed the most significant relationships with water temperature, and some minor relationship with dissolved oxygen, salinity, pH, phosphate and total inorganic nitrogen of surface and bottom layer in the water column. The number of dinoflagellate species decreased, while the abundance of cysts increased 4.5 times as compared with the observation of 10 years prior to the present study at the same stations of Masan-Chinhae Bay.

Mortality of Fishes and Shellfishes to Harmful Algal Blooms

  • Mortality of several species of fish and shellfish exposed to Harmful Algal Blooms (HABs) caused by Cochlodinium polykrikoides, Heterosigam akashiwo, Alexandrium tamarense, Eutreptiella gymnastica, Heterocapsa triquetra and Prorocentrum micans was studied. When fish were exposed to a cell density of 8,000 cells $mL^{-1}$ in C. polykrikoides, $35\%$ of flatfish and darkbanded rockfish died within 48 hrs. However, jacopever rockfish had mortality of higher than $85\%$. Rock bream, filefish and red sea bream showed $100\%$ mortality within 10 hrs with an exposure cell density of 8,000 cells $mL^{-1}$. The rest of HABs except for C. polykrikoides showed that there was no fish and shellfish death throughout the 48 hrs even in the maximum cell density of 100,000 cells $mL^{-1}$ These results imply that C. polykrikoides can have a serious impact on fish mortality and it is regarded as an ichthyotoxic dinoflagellate. The fish death may be attributed to anoxia caused by a combination of the production of reactive oxygen species (ROS) and polysaccharide from C. polykrikoides during blooms.
