The effects of metallothionein and cadmium ion on the hepatic aldehyde oxidase activity and brain lipid peroxidation were tested in vitro. The content of brain lipid peroxide at the condition of normal or $300{\mu}M$ Fe(II)-induced was remarkably reduced by the addition of metallothionein in the incubation mixture. The induced content of lipid peroxide by cadmium $(30{\mu}g/ml)$ ion was reduced by metallothionein $(100{\mu}g/ml)$. The activity of aldehyde oxidase was not affected by metallothionein, but cadmium ion $(8.38{\mu}g/ml)$ increased the activity of aldehyde oxidase about 80% compared to the control. The cadmium-induced activity of aldehyde oxidase was restored to the control level by metallothionein or penicillamine.
Lipid peroxidation mediated by hydroxyl radicals which are generated during myocardial ischermia has suggested as a possible mechanism of ischemic myocardial damage. Recently, it has been reported that anti-arrhythmic action of lidocaine, a local anesthetic, is attributed to its "membrane-stabilizing" properties through scavenging free radicals, thus, inhibiting lipid peroxidation. Aldehyde oxidase and xanthine oxidase which catalyze the oxidation of many purine, pyrimidine and pteridine derivatives are known as free radical generating systems. In this experiment, we studied the effect of lidocaine and procainamide on the hepatic aldehyde and xanthine oxidase activity and antioxidative activities. It was found that lidocaine and procainamide inhibited both NADPH-dependent and independent lipid peroxidation. Both of tested compounds were found to be ineffective in inhibiting xanthine oxidase. Lidocaine and procainamide, however, inhibited aldehyde oxidase activity in vitro as well as in vivo. Based on the above results, lidocaine and procainamide could be employed as a therapeutic agent for aldehyde oxidaserelated disease.d disease.
The roots of Cynanhum caudatum (Asclepiadaceae) have been used in folk medicine in Japan and China for the prevention and treatment of various geriatric diseases and also as a cardiotonic agent. Constituents of this plant have mainly been examined for glycosides: besides two steroidal alkaloids, gagaminine and gagamine which was firstly isolated by us, more than 35 polyoxypregnane glycosides and aglycones have also been identified. Gagaminine inhibits potently the hepatic aldehyde oxidase activity and lipid peroxidation in vitro. The present work deals with the comparison of antioxidative activities of gagamine, a new pregnane alkaloid, three isolated polyoxypregnanes containing a keto group at C-20 with those of gagaminine, a potent antioxidant, in order to explain the structure-activity relationships. The results of this study further prove that the cinnamoyl group of gagaminine is very important for the inhibition on the aldehyde oxidase activity while the nicotinoyl group is necessary for anti-lipid peroxidation. Besides that, the keto compounds having no ester group at C-12 were found to be more active than the others except gagaminine.
This study was done to determine the effect of lead acetate on the activities of the hepatic cytosofic xanthine oxidase and aldehyde oxidase which were well known as oxygen free radical generating enzyme in vitro. Lead ion accelerated the formation of lipid peroxide and the increment of xanthine oxidase(type O) activity and the type conversion ratio from xanthine dehydrogenase to xanthine oxidase dose-dependently. But xanthine dehydrogenase(type D) activity was decreased. Aldehyde oxidase activity was not changed by lead ion. These data suggested that lead-induced cellular to)dcity may be concerned partially with xanthine oxidase mediated lipid peroxidation.
Sex hormones not only regulate external sexual characteristics but several internal biochemical processes. It is well accepted that life-span of female is longer than that of male. Life-span is closely related with aging process in which free radicals are known to be involved. We investigated the effect of testosterone on free radical generating systems and lipid peroxidation based on the sexual difference. Lipid peroxide levels of male and female mouse were increased proportionately with age, especially in male mouse. Increase in enzyme activity of aldehyde oxidase with age was observed in male mouse, while no siginificant change in enzyme activity was found in female mouse. Enzyme activity of xanthine oxidase also showed similar results. It, however, was not significant statistically. Lipid peroxide level and xanthine oxidase type conversion ratio of male and female mouse liver homogenate incubated at $37^{\circ}C$, increased remarkably in proportion to incubation time, especially in male mouse. Lipid peroxide level and aldehyde oxidase activity were measured in normal male mouse, castrated mouse and testosterone treated-castrated mouse. Castrated mouse group showed decrease in lipid peroxide level and aldehyde oxidase activity compared with normal group. Treatment of castrated mouse with testosterone, however turned the level of lipid peroxide and aldehyde oxidase activity back to normal. From the above results, it might be concluded that testosteron could increase the activities of free radical generating enzymes which would result in the formation of lipid peroxide, consequently leading to aging.
The crude alkaloidal fraction of the root of Cynanchum caudatum Max.(Asclepiadaceae) was tested for the effects on the activities of free radical generating enzymes and the formation of lipid peroxide. Aldehyde oxidase was strongly inhibited to about 90% of the activity by treating 1.0 mg/ml of alkaloidal fraction, corresponding to competitive inhibition. Moreover, the formation of lipid peroxide which causes damage of cell membrane was reduced in proportion to the increasing alkaloid concentration. However, xanthine oxidase of which structure and function are similar to those of aldehyde oxidase was not inhibited by the alkaloidal fraction.

In vitro, levels of lipid peroxide in tissues of liver were proportionally decreased to concentration of CEHAS. They were much more decreased. when lipid peroxidation was induced with ferrous iron $(Fe^{-2})$. Also, enzyme activities of xanthine oxidase were decreased. The ratio of type conversion of xanthine oxidase was lowered, too. But, here was not special changes on enzyme activities of aldehyde oxidase. These results suggest that CEHAS decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide.

The ratio of type conversion of xanthine oxidase was lowered, two. But, it was not seen changes on enzyme activities of aldehyde oxidase. These results suggest that CEHAS decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide.
Objectives : To compare the anti-oxidant effects of Ligusticum chuanxiang and Cnidium officinale extracts and their mixture with Angelica gigas, we investigated the anti-oxidative activities using rat liver tissues. Methods : We investigated the anti-oxidative activities by analysis of lipid peroxidation, xanthine oxidase activity, aldehyde oxidase activity, hydroxyl radical scavenging activity, superoxide dismutase activity and catalase activity in rat liver tissues. Results : Both Ligusticum chuanxing and Cnidium offieinale inhibited the lipid peroxidation compared to the control, there is no significant differences between two groups. Cnidium officinale showed strong inhibitory effects on xanthine oxidase and aldehyde oxidase activities compared with that of Ligusticum chuanxing. Ligusticum chuanxing and Cnidium officinale were scavenged the hydroxy radicals, and increased SOD and catalase activities. These results suggested that Ligusticum chuanxing and Cnidium officinale were showed antioxidant activity, especially Cnidium officinale showed higher activity than Ligusticum chuanxing. Conclusions: These results suggest that Ligusticum chuanxing could be replaced by Cmdium afficinale in clinical trial.
