Ethanol treatment method was attempted for the selective isolation of ethanol-tolerant fungi from two sites of rice paddy fields around Seoul area. The vertical and seasonal fluctuation of the fungal population were also investigated. The ethanol-tolerant fungi were Talaromyces stipitatus, T. flavus var. flavus, T. helicus var. major, Eupenicillium javanicum, Emericellopsis terricolor, Pseudourotium zonatum, Aspergillus flavus, Cladosporium cladosporioides, Penicillium frequentans, P. janthinellum, and P. verruculosum. The most dominant species isolated by this method was T. stipitatus. It was found that the numbers of fungal species and colony forming units(CFUs) of ethanol-tolerant fungi were higher in Ascomycota than in Deuteromycota. A particular tendency appeared the highest CFUs in autumn, but lower in spring and winter. T. stipitatus was the dominant species of ethanol tolerant microfungi. This result would suggest that membrane lipid composition of ethanol-tolerant fungi isolated from the soils may play on important role in the ethanol tolerance.
Lactic acid bacteria (LAB) are a representative group of probiotics and used in many fermented foods and beverages. Several recent studies have shown that LAB are present in makgeolli which is a traditional Korean alcoholic beverage. However, most LAB are intolerant of more than 6% (v/v) alcohol concentrations. For this reason, alcohol-tolerant LAB are isolated from kimchi, makgeolli and nuruk using alcohol containing selective media. After being cultured in MRS broth containing 13% (v/v) alcohol, the two strains which showed the highest increasing O.D values, were finally selected. As results of 16S rRNA gene sequencing and biochemical characterization using an API kit, the two species were identified as Pediococcus acidilactici K3 and S1. In addition, the identified two strains produced bacteriocins against Staphylococcus aureus. When compared with the P. acidilactici type strain, the two selected strains possessed two to three time higher growth on 12-13% (v/v) alcohol containing MRS broth. The viability of P. acidilactici K3 and S1 when inoculated in makgeolli and stored at $10^{\circ}C$ did not decrease through a period of one month indicating that the selected strains can be used for LAB containing makgeolli.
Yeast screening for effective production of alcohol from Jerusalem artichoke tubers as an alternative energy source was performed. Inulin assimilative strains with high alcohol tolera- nce were isolated from wild sources and cultured in the liquid media of Jerusalem artichoke powder varying its concentraion from 15 to 30%. As a result, four strains of 2,445 isolates showing the inulin assimilation were selected as alcohol fermentative and alcohol tolerant yeasts. These strains were assignated to be Kluyveromyces marxianus F043 and Kluyveromyces sp. F173, E040, and F334, respectively, by their cultural and physiological characteristics. The F043 strain produced ethanol of 98.1 g/l in the 25% Jerusalem artichoke medium for 3 days.
The conventional alcohol fermentation method requires a large amount of energy for cooking the starchy raw materials prior to saccharification. The aim of this study was to compare the possibility of large scale alcohol fermentation from cassava slices were compared in low and high temperature cooking systems. The same amount of saccharifying and liquefying enzymes were used for cooking at low and high temperature. At low temperature cooking, conversion of glucose consumed in fermented mash to alcohol was 0.468g alcohol per g glucose of which was higher yield than that obtained at high temperature.
An novel amylolytic yeast, Saccharomyces cerevisiae HA 27, isolated from nuruk, displayed resistance against high sugar (50% glucose) and alcohol (15%). Maximal production of amylolytic enzyme by S. cerevisiae HA 27 was achieved on 9 days of cultivation at the optimal temperature $20^{\circ}C$ and pH 6.0. The activity of amylolytic enzyme produced by S. cerevisiae HA 27 was stable, even at $70^{\circ}C$, and over a broad pH range (4.0-11.0). Also, the amylolytic enzyme of S. cerevisiae HA 27 showed optimal activity in pH 5.0 at $50^{\circ}C$. S. cerevisiae HA 27 exhibited 6.2%(v/v) alcohol fermentation ability using starch as a carbon source.

The combination of a radiation technique with an in vitro culture system was appiled to develop submergence tolerant rice. The 3,000 $M_3$ lines with an average 80 percent of fertile grain were utilized for the selection of submergence tolerance. Salt tolerant lines were selected based on high plant height, root length and root number after submergence in plastic pots. Of the lines tested, the tolerant line (403-6) showed a dramatic difference in morphological traits under submergence compared to its original variety (Dongjinbyeo). It was suggested that genetic variations between the original variety and $M_3$-403-6 did exist. The levels of $\alpha$-amylase and alcohol dehydrogenase activities were significantly increased in the mutant line compared to its original variety. The mutant with greater tolerance showed less electrolyte leakage indicating a greater membrane integrity and better survival. Also, this line was much more resistant to a salt stress of $1.25\%$ than the original variety. The proline level of the line was significantly (p<0.01> higher than that of the original variety. The relationships between the inhibition of growth caused by stress and the physiological changes in the plant cell were discussed.
The toxicity of chlorothalonil on the growth of yeasts was investigated using several yeast strains. An alcohol tolerant yeast, Saccharomyces cerevisiae F38-1, was the most chlorothalonil-tolerant. The glutathione content and the glutathione S-transferase activity were related to the chlorothalonil-tolerant phenotype. Several thiol compounds affect the dissipation of chlorothalonil. However, there was no significant difference on the effects of chlorothalonil dissipation among the thiol compounds tested. The growth of yeast cells was arrested by chlorothalonil. It took about 13 h to dissipate 1 mg/l of chlorothalonil, and the growth was restored as the chlorothalonil content decreased. The glutathione content and glutathione S-transferase are suggested to be among the most important factors of yeast resistance to chlorothalonil.
In order to improve the productivity of ethanol by sugar-alcohol-tolerant Saccharomyces cerevisiae D1, the effect of addition of $Ca^{2+}$ on the alcohol fermentation was investigated. The addition of $Ca^{2+}$led to both the improvement of ethanol productivity and the increase of viable cell concentration. The optimal concentration of $Ca^{2+}$ was 0.8 mM. The higher was initial concentration of glucose, the greater effect of $Ca^{2+}$ was observed. Ethanol inhibition of growth, specific death rate in lethal concentration of ethanol, and extracellular final pH decreased by the addition of $Ca^{2+}$. The effect of $Ca^{2+}$ supplementation was explained by the increase of its ethanol tolerance.

Excessive alcohol consumption can have serious negative consequences on health, including addiction, liver damage, and other long-term effects. The causes of hangovers include dehydration, alcohol and alcohol metabolite toxicity, and nutrient deficiency due to absorption disorders. Additionally, alcohol consumption can slow reaction times, making it more difficult to rapidly respond to situations that require quick thinking. Exposure to a large amount of ethanol can also negatively affect a person's righting reflex and balance. In this study, we evaluated the potential of lactic acid bacteria (LAB) to alleviate alcohol-induced effects and behavioral responses. Two LAB strains isolated from kimchi, Levilactobacillus brevis WiKim0168 and Leuconostoc mesenteroides WiKim0172, were selected for their ethanol tolerance and potential to alleviate hangover symptoms. Enzyme activity assays for alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were then conducted to evaluate the role of these bacteria in alcohol metabolism. Through in vitro and in vivo studies, these strains were assessed for their ability to reduce blood alcohol concentrations and protect against alcohol-induced liver damage. The results indicated that these LAB strains possess significant ethanol tolerance and elevate ADH and ALDH activities. LAB administration remarkably reduced blood alcohol levels in rats after excessive alcohol consumption. Moreover, the LAB strains showed hepatoprotective effects and enhanced behavioral outcomes, highlighting their potential as probiotics for counteracting the adverse effects of alcohol consumption. These findings support the development of functional foods incorporating LAB strains that can mediate behavioral improvements following alcohol intake.
This study's purpose was to investigate the characteristics of a traditional Korean rice wine containing lactic acid bacteria (LAB), called makgeolli. The makgeolli was brewed with the alcohol-tolerant Pediococcus acidilactici strain K3, and was analyzed for LAB cell counts, alcoholic content, turbidity, pH, total acidity, amino nitrogen, total sugars, reducing sugars, solid contents, and organic acids. The physicochemical properties of the makgeolli were mostly maintained during fermentation (9 d) and storage (15 d). We also monitored the properties of LAB-supplemented commercial makgeollies, after adding P. acidilactici K3 at a concentration of $10^7CFU/ml$ makgeolli, for one month. Most of their properties, such as alcoholic content, turbidity, pH, total acidity, amino nitrogen, total sugars, reducing sugars, solid contents, and organic acids, were preserved during storage at $10^{\circ}C$, suggesting that makgeolli supplemented with live LAB can be produced. These results suggest that alcohol-tolerant P. acidilactici K3 can be used for makgeolli brewing either as a starter or as a supplement.
