Functional properties of mucilage and pigment extracted from Opuntia ficus-indica var. saboten were determined at various temperatures, pHs and alcohol concentrations. The crude mucilage extracted from pickly pear showed pH 4.2, 0.14% total acidity and 8.1% total soluble solid content(w/w, wet basis). Polysaccharide was purified from mucilage extract by isopropanol precipitation. Intrinsic viscosity of polysaccharide was 18.1dl/g and decreased with increasing KCl concentration. Relative viscosity and color stability of mucilage extract were determined with capillary viscometer and spectrophotometer at 534nm, respectively. In additions of 1~20%(v/v) ethanol, the red pigment of mucilage extract was very stable, but relative viscosity, increased gradually. For heating above 7$0^{\circ}C$, the stability of red pigment decreased drastically, but rheological property of mucilage was not changed. During storage, the red pigment was extremely unstable at above pH 8.3. At both pH 3.0 and pH 4.2, the red pigment was the most stable at 4$^{\circ}C$ for 18 days. In the case of storage at 37$^{\circ}C$, pigment of mucilage extract at pH 3.0 was destroyed more quickly than that at pH 4.2. Natural mucilage extract(pH 4.2) showed the good stability of red pigment at 3$0^{\circ}C$ for 10 days.
Transparent, photocatalytic, and self-cleaning TiO2 thin film is developed by TiO2 sol-gel coating on glass and polycarbonate (PC) substrates. Acetyl acetone (AcAc) suppresses the precipitation of TiO2 by forming a yellowish (complex) transparent sol-gel. XPS analysis confirms the presence of Ti2p and O1s in the thin films on glass and PC substrates. The TiO2-sol is prepared by stabilizing titanium (IV) isopropoxide (TTIP) with diethylamine and methyl alcohol. The addition of AcAcsilane coupling solution to the TiO2-sol instantaneously turns to yellowish color owing to the complexing of titanium with AcAc. The AcAc solution substantially improves the photocatalytic property of the TiO2 coating layer in MB solutions. The coated TiO2 film exhibits super hydrophilicity without and with light irradiation. The TiO2 thin film stabilized by adding 8.7 wt% AcAc shows the highest photo-degradation for methylene blue (MB) solution under UV light irradiation. Also, the optimum photocatalytic activity is obtained for the 8.7 wt% AcAc-stabilized TiO2 coating layer calcined at 450 ℃. The thin-films on glass exhibit fast self-cleaning from oleic acid contamination within 45 min of UV-light irradiation. The appropriate curing time at 140 ℃ improves the anti-fogging and thermal stability of the TiO2 film coated on PC substrate. The watermark-free PC substrate is particularly beneficial to combat fogging problems of transparent substrates.
In this work, $SnO_2$ modified with reduced graphene oxide (rGO) and carbon nanotubes (CNTs) separately and combined sensitized by using the co-precipitation method and their sensing behavior toward ethanol vapor at room temperature were investigated. An interdigitated electrode (IDE) gold substrate is very expensive compared to a fluorine doped tin oxide (FTO) substrate; hence, we used the latter to reduce the fabrication cost. The structure and the morphology of the studied materials were characterized by using differential thermal analyses (DTA) and thermogravimetric analysis (TGA), transmission electron microscope (TEM), X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, Brunauer-Emmett-Teller surface area and Barrett-Joyner-Halenda (BJH) pore size measurements. The studied composites were subjected to ethanol in its gas phase at concentrations from 10 to 200 ppm. The present composites showed high-performance sensitivity for many reasons: the incorporation of $SnO_2$ and CNTs which prevents the agglomeration of rGO sheets, the formation of a 3D mesopourus structure and an increase in the surface area. The decoration with rGO and CNTs led to more active sites, such as vacancies, which increased the adsorption of ethanol gas. In addition, the mesopore structure and the nano size of the $SnO_2$ particles allowed an efficient diffusion of gases to the active sites. Based on these results, the present composites should be considered as efficient and low-cost sensors for alcohol.
종이 공예 및 한지 산업, 문화재 보존 등에 활용을 위한 천연 바이오 접착제를 개발하기 위해 해초류 중 갈조류, 홍조류의 복합 다당류를 추출하여 이를 접착 성분으로 사용하였다. 갈조류는 감태, 다시마, 대황, 미역, 홍조류는 도박, 우뭇가사리, 풀가사리, 후노리로 갈조류와 홍조류에 포함된 비수용성 Ca 착물 형태의 다당류를 수용성의 1 가 알칼리 금속이 포함된 수용성 다당류들로 변형하여 추출하고 알코올 침전을 이용하여 다당류만을 얻고자 하였다. 갈조류는 다시마의 접착 인장 강도가 21.58 kgf, 홍조류는 도박이 32.99 kgf를 나타내어 물풀(18.45 kgf)과 딱풀(20.45 kgf)로 알려진 고체풀의 접착력에 상회하는 결과를 나타내었다. 이 추출된 다당류들은 추출 환경에 따라서 추출 수율이 결정되었지만, 접착도에는 차이를 나타나지 않고 있어서 추출 환경이 아니라 생육 환경에 의해 다당류의 형태가 결정되어지는 것을 알 수 있었다. 추출 시에 다단계의 알코올 침전법을 사용함으로 추출 과정에서 단백질 및 기타 다당류를 제외한 구성 성분들이 제거된 결과가 나타나서 곰팡이 배양이 되지 않는 안정한 결과를 보이고 있었으며, 단순 용해에 의한 접착제 제작 후에도 곰팡이의 발생이 없어서 친환경 접착 재료로서의 사용이 가능한 결과를 나타내었다.
미생물성 효소를 이용하여 인삼saponin중 조성비율이 가장 큰 ginsenoside-Rb$_1$을 약효면에서 보다 우수한 ginsenoside-Rd로 전환하고자 인삼부패균 중 Rhizopus 속의 한 균주를 선정하여 이 균주에서 얻은 효소를 ammonium sulfate 분별 침전법으로 조정제하여 사용하였다. 기질로 사용하기 위해 홍미삼 extract로부터 ginsenoside-Rb$_1$이 36.4%, ginsenoside-Rd 가 12.2%의 조성비율을 갖인 total saponin을 정제하였고 이어 ginsenoside-Rb$_1$의 함량을 증가시키기 위해 더욱 정제한 결과 ginsenoside-Rb$_1$이 54. 5%, ginsenoside-Rd가 1.1%인 ginsenoside Rb group saponin을 얻었다. 이들 기질 saponin에 본 효소를 작용시켜 본 결과 두 기질 모두 다른 ginsenoside pattern에는 변화없이 ginsenoside-Rb$_1$만이 선택적으로 감소하고 반면에 ginsenoside-Rd의 함량이 비례적으로 증가됨을 TLC 및 HPLC의 방법으로 조사하였으며 이로써 효소에 의한 인삼saponin의 선택적전환 가능성을 확인하였다.
담배거세미나방 핵다각체병바이러스에 섭식촉진물질, 다각체 침전방지제 및 전착물질과 자외선 차단제를 첨가하여 만든 3종의 바이러스 살충제를 담배거세미나방 유충에 먹여 제제별로 살충 효과를 비교하였다. 바이러스 살충제에 설탕농도 1-5% 첨가로 섭식촉진이 증대되어 살충효과가 높았으며, 바이러스의 불활화 차단효과도 있었다. 살충제 제형에 따른 바이러스 침전방지는 polyvinylalcohoI 0.5%, 식물체 전착성은 Triton X -100 0.1 %에서 효과가 좋았다. 자외선차단제의 양은 white carbon은 100 L 당 800 g, molasses는 30%첨가에서 바이러스의 불활화가 감소되었으며 white carbon-molasses 혼합제는 white carbon 500 g, molasses 10%의 비율에서는 살충효과가 약간 감소하였으나 세 제제 모두 살포 3일후 95%이상의 살충율을 보였다. White carbon 체제는 molasses 제제보다 다각체 포매가 뚜렷하여 잔류효과가 증대되었으나 유충의 섭식은 molasses제제에서 촉진되었다.
Toluene, phenyl 등의 분해균주인 Burkholderia cepacia G4로부터 tomB 유전자를 클로닝하여 얻은 재조합 균주 E. coli CNU312로부터 catechol 2,3-dioxygenase를 정제하여 효소학적 특성을 조사하였다. Catechol 2,3-dioxygenase는 native 분자량이 약 140.4 kDa이었으며 4개의 동일한 35 kDa subunit로 구성된 homotetramer로 생각된다. Catechol의 $K_(m)$값과 $V_(max)$값은 372.6 $\mu$M과 39.27 U/mg이었으며, 1.56 mM 이상의 기질 농도에서는 활성이 감소되었다. 효소 활성의 최적 pH는 8.0이었으며, pH 7.0-8.0 범위에서 안정하였다. 최적 활성온도는 $40^{\circ}C$였으며, $60^{\circ}C$이상에서 완전히 활성을 상실하였다. 또한 $Fe^(2+)$, $Fe^(3+)$ 를 비롯한 대부분의 금속 이온에 의해 활성이 감소되었으며, $Mg^(2+)$, $K^(+)$에는 영향을 받지 않았다. 효소 활성부위를 알아보기 위해 화학변형제를 처리한 결과, tryptophan과 histidine이 효소 활성부위에 존재하는 것으로 추정된다. 그리고 10%의 유기용매에 안정성을 보이지 않았으며, $H_(2)$$O_(2)$, EDTA, ο-phenanthroline에도 활성이 감소되었다. 또한 2-mercaptoethanol, dithiothreitol, 그리고 ascorbic acid와 같은 환원제에 대해서도 안정성을 보이지 않았다. 이 효소는 catechol에 대해 높은 기질 특이성을 보였으며, 3-methylcatechol, 4-methylcatechol, 그리고 4-chlorocatechol에 대해 약간의 활성을 보였다. 그러나 2,3-dihydroxybiphenyl에 대해서는 거의 활성을 보이지 않았다.
알긴산이 가지는 여러가지 물성적 특성 및 기능적 특성으로 식품에 널리 이용되고 있으나 상핀 점도와 용해도 때문에 그 이용 범위가 제한적이다. 이러한 알긴산을 보다 광범위하게 식품에 이용하기 위해서 알긴산의 특성을 어느 정도 유지하면서 용해도를 높이고, 점도를 저하시킨 부분 분해 알긴산의 제조와 특성을 실험하였다. 사용된 알긴산 분해효소는 pH 8.0, 온도 $37^{\circ}C$, 0.5 M NaCl 첨가 조건에서 최대 분해 활성을 나타내었다. 기질과 반응하는 부분 정제 효소(220.4 g/ml)의 농도는 2%(v/v)가 적절하였다. 반응 알긴산 농도에 관계없이 반응시간 10분으로 알긴산이 가지고 있는 점도 80%정도가 저하되었고, 60분 이내에 90% 이상의 점도가 떨어졌다. 분해에 의한 환원당 생성량을 측정하여 본 결과, 반응 60분까지는 큰 변화가 없었으나, 그 이후에는 환원당이 크게 증가하였다. 분해 시간에 따라 얻어진 알긴산들의 가용 농도는 반응 알긴산의 농도가 1.0%와 1.5%의 경우 10분 정도의 분해만으로도 10% 농도까지 용해할 수 있었고, 반응 알긴산 농도가 2.0%인 경우는 10분간의 분해로 7%까지 녹일 수 있었다. 알코올 침전도를 알아본 결과, 2.5% 시판 알긴산은 알코올 농도 3%일 때 침전을 시작하여 알코올 농도 30%일 때 22%, 알코올 농도 50%에서 대부분의 알긴산이 침전하였다. 부분 분해 알긴산의 경우는 알코올 농도 30%일 때 침전하기 시작하여 알코올 농도 70%일 때 85% 정도의 알긴산이 침전되었다. $Ca^{2+}$이온과의 반응성은 시판 알긴산과 마찬가지로 종류에 관계없이 부분 분해 알긴산의 농도가 높아짐에 따라 반응성도 커지는 것으로 나타났다.
본 연구는 탄산칼슘 처리농도(0.1, 0.2 and 0.3%)에 따른 개량머루주의 감산 특성을 검토하였다. 탄산칼슘 처리농도에 비례하여 처리 2일째 총산함량은 큰 폭으로 감소하였으나, 이후 기간에는 비슷한 것으로 나타났다. 1차 앙금분리와 숙성 중에는 총산이 정상적으로 감소되는 반면, 발효 중에는 약 30%로 효율이 낮았다. 또한 탄산칼슘 처리농도가 많을수록 pH가 상승하기 때문에 적색도가 낮아져 와인이 보랏빛을 나타냈으며, 총 안토시아닌 함량은 다소 감소하였다. 알코올과 탄닌 함량은 처리구간 유의적인 차이가 나타나지 않았으며, 총 폴리페놀 함량은 탄산칼슘 처리에 따라 소폭 증가하는 것을 알 수 있었다. 개량머루주의 주요 유기산 중 주석산은 탄산칼슘과 반응하여 처리농도에 비례하게 감소되었으며, 그 결과 총산 함량의 감소로 이어졌다. 기호도 평가에서 와인의 색은 탄산칼슘 처리농도가 많을수록 대체적으로 낮은 선택률을 보였고, 무게감과 이미는 높은 선택률을 나타냈다. 탄산칼슘 0.1% 처리구가 신맛과 전반적인 기호성에서 가장 높은 기호도를 나타냈다. 결과적으로 탄산칼슘 처리농도가 많아질수록 개량머루주의 총산은 감소되어 신맛을 저감화시키지만 와인의 품질에는 부정적인 영향을 미치므로, 0.1% 이내의 탄산칼슘 처리가 필요할 것으로 보인다.
