황칠나무 물 추출물의 항산화, 알코올 대사 효소 및 간 보호 활성 (Antioxidant, Alcohol Metabolizing Enzyme, and Hepatoprotective Activities of Dendropanax morbifera Water Extract)

  • 본 연구에서는 황칠나무 잎과 줄기 물 추출물의 항산화 활성과 알코올 대사 효소 활성 및 간 보호 효과를 알아보았다. 황칠나무 잎과 줄기 물 추출물의 총 폴리페놀 함량은 49.56 mg TAE/g으로 나타났다. DPPH radical 소거능은 황칠나무 잎과 줄기 물 추출물의 농도 의존적으로 소거능이 증가하였으며 1,000 ㎍/ml 농도에서 84.09%로 높게 나타났다. 알코올 분해 활성을 알아보기 위해 ADH 및 ALDH 활성을 측정한 결과, 두 가지 효소 모두 황칠나무 추출물의 농도 의존적으로 증가하였으며 1,000 ㎍/ml 농도에서의 ADH 및 ALDH 활성은 각각 37.68% 및 41.67%로 나타났다. 황칠나무 잎과 줄기 물 추출물은 tacrine으로 유도된 HepG2 세포주의 세포독성에 대한 보호 활성이 나타났다. 이상의 결과를 통해 황칠나무 잎과 줄기 물 추출물은 항산화 효과와 알코올 숙취해소 효과 및 간세포 보호 효과가 높은 것으로 나타나 기능성 식품 소재로서의 활용 가능이 높은 것으로 사료된다.

  • It was studied that which components of the ginseng are related to the effects that accelerate Saccharomyces division by comparing the influences of ginseng powder, alcohol extracts of ginseng, and linoleic and stearic acids which are known as the ginseng components on the division. The addition of ginseng powder and alcohol extracts of ginseng to the glucose agar medium marked the conspicuous increases of the division of Saccharomyces: 44% increase by 0.1% powder, 53% increase by 0.05% alcohol extracts. Also, the addition of the fatty acids of less than 0.0008% marked the considerable increases of the division: 22% increase by 0.0008% linoleic acid, 31% increase by 0. 00016% linoleic acid, 12% increase by 0.0008% stearic acid. Therefore it can be concluded that a proper amount of those fatty acids contained in the ginseng has a definite effect on the acceleration of Saccharomyces division.

  • The effects of ginseng extracts on carbon dioxide generation, alcohol fermentation, and yeast cell production by Saccharomyces cerevisiae were investigated. The results are as follows. 1) In the process of fermentation, CO2 generation by yeast is faster in ginseng extracts media of 0.3%, 0. l% than in control. As the concentration of the extracts increases by 0.7% and 1.5%, CO2 generation is decreased. Among all these concentrations, CO2 generation is fastest in 0.3% of the extracts. 2) In the process of fermentation, the production of alcohol is larger in the order of 0.3%, 0.7% and 0.1% than in the control and least in 1.5%. 3) The number of yeast cell rapidly increased from 12 hours to 18 hours after cultivation and conspicuously increased in the order of 0.3%, 0.7%, 0.1%, control and 1.5%. 4) Dried yeast cell weight increased more in all the above concentration than control and among these it increased visibly in 0.3% of the extracts.

Protective Effect of Korean Medicinal Plants on Ethanol-Induced Cytotoxicity in HepG2 Cells

  • The purpose of this study is to evaluate cytoprotective effect of Korean medicinal plants on alcohol-induced cytotoxicity in liver cells. Out of the 120 plant extracts tested in this study, 53 plant extracts enhanced alcohol-induced cytotoxicity in liver cells by 50~80%, while other 11 plant extracts including Crataegus pinnatifida reduced cytotoxicity by 1~68%. The results of DPPH free radical test and LDL lipid peroxidation test on the plant extracts that sharply reduced cytotoxicity in liver cells shows that Crataegus pinnatifida and Cinnamomum cassia had antioxidative effect. This study reports that the plant extracts that enhance or reduce ethanol-induced cytotoxicity in liver cells can be research objects as cytotoxic plants or cytotoxicity-protective plants.

  • Aconitum Koreanum rhizoma has been applied in therapy from old in oriental medicine (Korean Hanbang), The action of the water and alcohol extracts of the rhizoma and root of Aconitum Koreanum R. Raymond, a common herb in Korea, on the isolated Frog heart and $LD_{50}$ were observed. The results of the studies were as follows: 1. The cardiac effect in the concentration of the extracts$(10^{-3})$ was increased and duratrin of action prolonged approximately 10 minutes, but in the extracts $(3{\times}10^{-3})blocked$. 2. The $LD_{50}$ of extracts were 722 mg/kg (water) and 151mg/kg (alcohol) intraperitoneally in mice.

알코올 및 사염화탄소에 의해 유발된 급만성 간손상에 대한 지구자 열수추출물의 보호효과 (Hepatoprotective Effects of Hovenia dulcis Extract on Acute and Chronic Liver Injuries induced by Alcohol and Carbon Tetrachloride)

  • Objectives : The purpose of this study was to evaluate the hepatoprotective effect of Hovenia dulcis extract on acute and chronic liver injuries induced by alcohol and $CCl_4$ in mice and rats. Methods : In acute alcohol-induced liver injury, mice were administered Hovenia dulcis extracts (60 and 200 mg/kg) orally before and after alcohol administration. In chronic alcohol-induced liver injury, mice were administered alcohol containing liquid diet for 4 weeks. The mice were administered H. dulcis extracts (60 and 200 mg/kg) mixed with the liquid diet. In acute $CCl_4$-induced liver injury, rats received a single dose of $CCl_4$ (2 mL/kg in olive oil, intraperitoneally). Rats were administered H. dulcis extracts (30, 100 and 300 mg/kg) before and after $CCl_4$ administrations. After the ends of the administrations, the serum levels of AST and ALT were measured using chemical analyzer, and ${\gamma}$-GTP levels were measured using spectrophotometer. Results : In acute alcohol-induced liver injury, H. dulcis extracts treated group showed significant reduction in ALT levels compared to those of control group. In chronic alcohol-induced liver injury, it inhibited weight-loss compared to normal group and showed significant reduction in AST, ALT and ${\gamma}$-GTP levels compared to control group. In acute $CCl_4$-induced liver injury, it also showed significant reduction in AST, ALT levels compared to control group. Conclusions : The results show that H. dulcis extract has hepatoprotective effect in acute and chronic alcohol-induced liver injury and acute $CCl_4$-induced liver injury. These findings suggest that H. dulcis could be a potent hepatoprotective agent.

In vitro Anti-obesity Effect of 4-hydroxybenzyl Alcohol from Cudrania tricuspidata

  • The present study was investigated on in vitro anti-obesity effect of 4-hydroxybenzyl alcohol from Cudrania tricuspidata. We isolated various compounds from Cudrania tricuspidata. Among these compounds, anti-obesity effects of 4-hydroxybenzyl alcohol was examined by lipase activity assay, cyclic adenosine monophosphate (cAMP)-specific phosphodiesterase type IV (PDE4) activity assay, and citrate synthase activity assay. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts inhibited the enzymatic activities of lipase, PDE4, and citrate synthase. Lipase is known to mediate the hydrolysis of triacylglycerol in adipose tissue and cholesterol esters in other tissue or cells. Also, PDE4 hydrolyses cAMP, a crucial secondary messenger for in metabolic pathways including glucose and lipid metabolism, lipolysis, and thermogenic function. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts induced the inhibitory effect against each enzymatic activity on several specific substrates as observed by detection at 405 or 412 nm. These findings might be attributable to the inhibition of adipogenesis, and partial prevention of obesity. In conclusion, these results show that 4-hydroxybenzyl alcohol and Cudrania tricuspidata may be a critical candidate as a natural anti-obesity source.

은복과 한약재 복합물이 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향 (Effects of Water Extracts from Lagocephalus wheeleri with Several Herbs on Hyperlipemia and Liver Damage Induced by Alcohol)

  • Objectives : This study was performed to investigate the effect of water extracts from Lagocephalus wheeled with several herbs(LW) on hyperlipemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : At first, we observed effects of LW on hyperlipemia induced by alcohol. LW group didn't show significant increase of total cholesterol in comparison with those of the control group. LW group didn't show significant increase of HDL(High-Density lipoprotein) cholesterol in comparison with those of the control group. LW group showed significant decrease of triglyceride in comparison with those of the control group. LW group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. LW group showed significant increase of liver weight in comparison with those of the control group. At second, we observed effects of LW on liver damage induced by alcohol. LW group showed significant decrease of ALP, GOT, GPT and LDH in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from Lagocephalus wheeleri with several herbs have pharmaceutical efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of this.

채소의 용매분획 추출물들이 Saccharomyces cerevisiae의 alcohol dehydrogenase 활성에 미치는 영향 (Effects of Vegetable Extracts by Solvent Separation on Alcohol Dehydrogenase Activity from Saccharomyces cerevisiae)

  • 용매분획에 의한 콩나물, 미나리, 무의 추출물들이 Saccharomyces cerevisiae의 ADH의 활성에 미치는 영향을 in vitro에서 조사하였다. 이 추출물들은 알코올로 추출한 것을 용매분획에 의해 수용성 분획물과 유기성인 염기성, 산성, 중성, 페놀성 분획물들을 얻었다. 수용성 분획물들은 유기성 분획물들 보다 ADH 활성을 훨씬 높게 촉진시켰다. 수용성 분획물인 콩나물, 미나리, 무의 촉진율은 각각 125.75%, 104.94%, 87.63%를 나타냈다. 염기성, 산성, 중성의 분획물들에서 염기성 분획물이 약 40%로 가장 높았고 다른 분획물들은 25% 이하로 나타났으며 큰 차이를 보이지 않았다. 페놀성 분획물들 역시 ADH 활성에 큰 영향을 나타내지 못했다. 따라서, 이 분획물들을 ADH 활성 촉진에 이용할 때는 무기물, 아미노산 등의 상승효과를 얻을 수 있는 수용성 분획물들을 이용해야 할 것으로 사료된다.
