Morin, one of the major natural flavonoids has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the hepatoprotective effect of morin on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of morin (10, 20mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phospatase, and bilirubin levels. Morin also increased serum protein level and reduced the hepatic level of malondialdehyde in DMN-treated rats. (omitted)
Aspirin and aspirin-like nonsteroidal antiinflammatory drug have been the mainstay of therapy for rheumatoid arthritis. In this study, we investigated the hepatoprotective effect of aspirin on the dimethylnitrosamine (DMN)-induced liver damage in rats. Oral administration of aspirin (7.5, 15mg/kg daily for 4 weeks) into the DMN-treated rats remarkably prevented the elevation of serum alanine transaminase, aspartate transaminase and alkaline phosphatase, and bilirubin levels. Quercetin, one of the most abundant flavonoids in human diet has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the protective effect of quercetin on hepatic fibrosis induced by dimethylnitrosamine (DMN) in rats. Treatment with DMN caused a significant decrease in body and liver weight. This study was performed to investigate the effect of serum enzyme activities in rats after administration of aluminum compound. Seventy five male Sprague-Dawley rats were divided into five groups consisting of control, $250\;ppm\;AlCl_3,\;500\;ppm\;AlCl_3,\;250\;ppm\;Al_2(SO_4)_3$ and $500ppm Al_2(SO_4)_3$ groups and kept on the diet for 2 weeks. The weight gain increased from 0.53 to 3.35% in $AlCl_3$ adiministration groups but decreased from 2.82 to 6.16% in $Al_2(SO_4)_3$, administration groups as compared to control group. As compared to control group, activities of lactate dehydrogenase (LDH) and aspartate amino transaminase (AST) in serum increased 29.43 to 57.68% and 0.68 to 9.97% in $AlCl_3$ adiministration groups, and 74.60 to 29.33% and 21.04 to 24.79% in $Al_2(SO_4)_3$ adiministration groups, respectively. However, alanine amino transminase (ALT) decreased from 12.69 to 25.42% in $AlCl_3$ adiministration groups and from 24.32 to 39.62% in $Al_2(SO_4)_3$ administration groups. Cholinesterase activity increased from 28.98 to 12.73% as compared to control group by administration of $AlCl_3$ and decreased from 3.93 to 14.48% by administration of $Al_2(SO_4)_3$.

The effect of dietary capsaicin (8-methyl-N-vanillyl-6-nonenamide, CAP) on drug-metabolizing enzyme activities was investigated in mice. Male ICR mice were divided into 4 groups and fed diets containing 0, 5, 20, 100 ppm CAP for 4 seeks. Hepatic drug-metabolizing enzyme activities and serum alanine aminotransferase and aspartate transaminease activities were measured. There was no difference in hepatic alanine aminotransferse and aspartate transaminase activities among the groups. Hepatic microsomal cytochrome P450 in CAP fed groups, but p-nitrophenol hydroxylase and the cytosolic acitivity of glutathione S-transferase activities were decreased in the dietary CAP supplemetned groups compared to the control. These results suggest that the dietary CAP at a low dose differentially modulates drug-metabolizing enzyme acitvities without causing hepatic toxicity.
Artemisia, a plant widely used as traditional herbal medicine in many countries, has drawn attention of the researchers. And its extracts or compounds are known to have an efficacy of antioxidant, anti-diabete, anti-cancer, anti-inflammation and neuroprotection. Sumaeyaksuk is a variant of the Artemisia argyi and major constituents are eupatilin and jaceosidin. This study was performed to investigate the effects of the sumaeyaksuk aqueous extract on inflammatory response induced by lipopolysaccharide (LPS) in human hepatoma HepG2 cells. To examine the potential hepatoprotective properties of sumaeyaksuk extract, cell viability, as well as nitric oxide (NO), reactive oxygen species (ROS), macrophage colony-stimulating factor (M-CSF), interleukin-8 (IL-8) levels, alanine transaminase (ALT), and aspartate transaminase (AST) activities, were measured. Cytotoxic activity of extracts on HepG2 cells was measured by MTT assay. Sumaeyaksuk extract did not induce cytotoxicity at concentrations of $0{\sim}400{\mu}g/mL$. NO and ROS levels significantly decreased with increasing concentration of the extract. The secretion levels of M-CSF and IL-8 were suppressed by sumaeyaksuk extract in a dose-dependent manner. Moreover, ALT (75.4%) and AST (61.6%) levels significantly decreased in sumaeyaksuk extract-treated cells at $400{\mu}g/mL$. These results suggested that the sumaeyaksuk extract attenuates the LPS-induced hepatotoxicity resulting from regulation of inflammatory factors and could potentially be used as a hepatitis therapeutic agent.
This study was performed to investigate the toxic effect of pyrrolizidine alkaloids from symphytum officinale i n rat. For this experiment, 120 male and female rats of Sprague-Dawley strain were used. The experimental groups were divided into five: Group CM and CF served as normal control with its gender. Group EM1 and EF1 were fed a 1% Symphytum officinal extract diet for 8 weeks. Group EM2 and EF2 fed a diet containing 2% extract diet. 4% extract diet into group EM3 and EF3 and 8% extract diet into group EM4 and EF4 were given. The results were as follows: 1. The major alkaloids of Symphytum officinale extract were symphytine, echmidine, and lasiocarpine. The amounts of total alkaloid were 168 $\mu\textrm{g}$ PAs/$m\ell$ extract. And contents of Pas in leaves were 0.05% wt.. 2. Total serum bilirubin concentrations increased significantly in group EM2, EM3, and EM4. Group EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 3. Aspartate transaminase activities were increased significantly in group EM3 and EM4 (p<0.05). Aspartate transaminase activities of EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 4. Alanine transaminase activities increased significantly in group EM3, EM4 (p<0.05). Alanine transaminase activities of EF1, EF2, EF3, and EF4 showed statistical significance for the group CF (p<0.05). 5. Alkaline phosphatase activities increased significantly in group EM2, EM3, and EM4 (p<0.05). Alkaline phosphatase activities of EF1, FE@, EF3, and EF4 showed statistical sigmificance for the group CF (p<0.05). 6. istopathological analysis of liver specimens from group EM3 and EM4 showed focal necrosis, periportal necrosis and apoptpsis. Hepatocytes obtained from group EM2 showed fatty change and hydropic degeneration in group EM3 and EM4. Chromatolysis and chromatin margination was shown in group EF2 and EF3. With the above results, it was demonstrated that the Symphytum officinal extract could induce functional change of liver, and histopathological change of liver in rats fed a diet containing extract. In conclusion, because of the risk of intoxication or adverse effect, the composition, dosage and mode of administration of herbal products should be monitored strictly. And this study serves as a reminder that herbal as well as orthodox medications may have serious side effects.
Objectives: The purpose of this study is to investigate safety of postpartum herbal medicine by assessing the effect of taking herbal medicine of postpartum period on liver function. Methods: A retrospective chart review was conducted on 167 mothers who underwent liver function tests (LFT) within 3 months before and after childbirth among mothers who gave birth at ○○ Hospital between January 1, 2016 and May 31, 2018. Mothers with abnormally elevated LFT during pregnancy were excluded. Among 167 women, 6 women are herbal-medicine-group took herbal medicine for 5-6 weeks during postpartum period, and 161 patients are general -group who did not take herbal medicine. LFT Variation of Subjects before and after childbirth were compared between the two groups. And subjects who had elevated liver levels above the normal range after delivery were classified separately, the characteristics and causes of changes in liver levels were analyzed, and the presence or absence of drug-induced liver damage was confirmed. Results: Among a total of 167 subjects, there were 5 women in the herbal-medicine-group and 150 women in the general-group who had changes in liver values within the normal range after childbirth. Aspartate transaminase (AST) change before and after childbirth in the herbal-medicine-group was 3.40±1.82, and AST change in the general-group was 2.92±8.59, showing no significant difference between the two groups (p=0.901). Increase of Alanine transaminase (ALT) before and after childbirth in the herbal-medicine-group was 5.60±3.65, and ALT change in the general-group was 8.01±11.81, showing no significant difference between the two groups (p=0.651). There were 12 subjects who had elevated AST, ALT above the normal range after delivery, including 1 in the herbal-medicine-group and 11 in the normal mothers group. Valuation of 1 Subject of the herbal-medicine-group before and after delivery was 17 IU/L of AST and 52 IU/L of ALT. Because results of AST, ALT is under the standard to diagnose to liver damage, she was observed without any treatment. However the cause of AST, ALT elevation was not found in the chart, she was receiving treatment for diabetes and hyperlipidemia. The general-group had an average increase of AST 35.64±22.67 IU/L and ALT 53.00±26.80 IU/L. As a result of analyzing the cause, there were direct causes such as autoimmune hepatitis, chronic hepatitis B, and acute pyelonephritis. Abnormal elevations in liver levels were also found in mothers with hypothyroidism, diabetes, and fever of unknown cause, although they were not direct causes. Conclusions: To investigate the safety of taking herbal medicines, we assess the variation in AST and ALT within 3 months before and after delivery in the herbal-medicine-group and general-group. There was no significant difference between two groups.
The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid,monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.
Objectives: Although mercury (Hg) exposure is known to be neurotoxic in humans, its effects on liver function have been less often reported. The aim of this study was to investigate whether total Hg exposure in Korean adults was associated with elevated serum levels of the liver enzymes aspartate aminotransferase (AST), alanine transaminase (ALT), and gamma-glutamyltransferase (GGT). Methods: We repeatedly examined the levels of total Hg and liver enzymes in the blood of 508 adults during 2010-2011 and 2014-2015. Cross-sectional associations between levels of blood Hg and liver enzymes were analyzed using a generalized linear model, and nonlinear relationships were analyzed using a generalized additive mixed model. Generalized estimating equations were applied to examine longitudinal associations, considering the correlations of individuals measured repeatedly. Results: GGT increased by 11.0% (95% confidence interval [CI], 4.5 to 18.0%) in women and 8.1% (95% CI, -0.5 to 17.4%) in men per doubling of Hg levels, but AST and ALT were not significantly associated with Hg in either men or women. In women who drank more than 2 or 3 times per week, AST, ALT, and GGT levels increased by 10.6% (95% CI, 4.2 to 17.5%), 7.7% (95% CI, 1.1 to 14.7%), and 37.5% (95% CI,15.2 to 64.3%) per doubling of Hg levels, respectively, showing an interaction between blood Hg levels and drinking. Conclusions: Hg exposure was associated with an elevated serum concentration of GGT. Especially in women who were frequent drinkers, AST, ALT, and GGT showed a significant increase, with a significant synergistic effect of Hg and alcohol consumption.
