Objectives: We aimed to compare the sampling performance of different flow-based impactor samplers for collecting fungal spores and bacteria and to explore the association of the level of bioaerosols with activity patterns of occupants in daycare center settings. Methods: For comparison of sampling performance, two different flow-based samplers (greater than 100 L/min or not) were selected; a low flow-based sampler (one-stage Andersen sampler) and two high flow-based samplers (DUO SAS SUPER 360 sampler, BUCK bio-culture sampler). We collected airborne mold and bacteria in 30 daycare centers with various levels of contaminated air. Three repeat samplings per each sampler were performed. Mold and bacteria were grown for 96 hours at $25{\pm}1^{\circ}C$ and 48 hours at $35{\pm}1^{\circ}C$, respectively. The Andersen and SAS samplers were used for investigating the association between the level of bioaerosols and the activity patterns of occupants in daycares. Particular matters 10($PM_{10}$), temperature, and relative humidity were monitored as well. Samplings were carried out with one-hour interval from 9 to 5 O'clock. For statistical comparisons, Kruskal-Wallis test, Wilcoxon's signed rank test, and multiple regression analysis were carried out. Results: The airborne level of molds by the low flow-based sampler were significantly higher than that of high flow-based samplers (indoor, P=0.037; outdoor, P=0.041). However, no statistical difference was observed in the airborne level of bacteria by each sampler. Also the level of bioaerosols varied by the time, particularly with different activity patterns in daycare centers. The higher level of mold and bacteria were observed in play time in indoor. Similarly, the concentrations of $PM_{10}$ were significantly associated with the level of bioaerosols (P<0.05). Conclusions: Our findings indicate that the flow rate of sampler, rather than total air volume, could be able to affect the results of sampling. Also, the level of airborne mold and bacteria vary behavior patterns of occupants in indoor of daycare settings. Therefore, different samplers with other flow rate may be selected for mold or bacteria sampling, and activity patterns should be considered for bioaerosol sampling as well.

Among different sugars, bacterial growth was favored by in the order of brown, black and white sugars, indicating that the bacterial growth might be promoted by the minor elements presented as impurities in the less purified sugars. As for nitrogen source, organic forms were better to bacterial growth than inorganic forms, that is polypeptone was best followed in order by soy sauce, soybeen milk and inoganic nitrogens. Differences in bacterial growth among different forms of inorganic nitrogen were negligible.

의료 관련 감염을 예방하기 위한 필수적 방법은 일관성이 있는 감염감시 시스템을 구축하고 효율적인 감시 통제를 수행하기 위해 신뢰할 수 있는 상황에 대해 진단을 향상시켜나가는 것이다. 또한 의료 종사자의 손과 의복 및 장비는 환자 관리 및 환경과 접촉하여 병원체가 오염되는 요인이다. 병원체를 가진 시설 표면(예: 침대 레일, 침대 옆 탁자), 음용수, 냉각탑용수, 내시경기구, 급식위생, 공기매개, 멸균검사, 내독소검사, 및 의료 장비를 포함한 의료 환경의 오염은 일반적으로 발생한다. 또한 이러한 감염원을 활동 감시를 통해 MLST, PFGE의 기법으로 역학분석을 수행한다. 따라서 HAI 예방을 위한 환경 감시배양 검사는 환자의 안전과 감염원의 차단을 향상시킬 뿐만 아니라 국가의 감염관리 시스템을 통제하여 최적의 효율적인 감염관리 예방을 가능케 하고 국가의 감염관리 시스템의 안전을 향상시킨다. 결론적으로 감시배양 검사의 표준화를 통해 실효성 있는 감염관리체계에 이바지하고 감염관리 전문인력으로서의 전문성을 확보하고자 한다. 이를 통해 표준화 마련의 일차적인 목표는 의료관련 감염을 줄이고 국가적 의료관리 체계를 향상시키는데 있다.
Rapid, real-time detection of pathogenic microorganisms is an emerging and quickly evolving field of research, especially with regard to microorganisms that pose a major threat to public health. Herein, a new method that uses bioimpedance and solid culture medium for the real-time detection of microorganisms is introduced. We fabricated a new impedimetric biosensor by integrating solid media and two plane electrodes attached on two facing sides of an acryl well. During bioelectrical impedance analysis, the solid medium showed the characteristics of a homogenous conductive material. In a real-time impedance measurement, our solid-medium biosensor could monitor bacterial growth in situ with a detection time of ${\sim}4$ hrs. Our data indicate that the solid-medium biosensor is useful for detecting airborne microorganisms, thereby providing a new analytical tool for impedance microbiology.
For the purpose of acquiring microbial agents that can be utilized to billogically control the major airborne disease to apple trees, such as canker(Valsa ceratosperma), the effective microorgaisms were isolated, tested for antagonistic activity to the pathogen causing major disease to apple trees and identifed. Screening of more than 3, 000 species of microorganisms collected in nature for them antagonistic action to the pathogen, Valsa ceratosperma causing disease to apple tree resulted in selection of effective species. Out of the 11 species, one species designated as CAP141 demonstrated outstanding activity. The bacterial strain, CAP141 exerted antagonistic efficiency of 65% on Valsa ceratosperma. The CAP141 was identified as a bacterial strain to Bacillus subtilis based on morphology, culture conditions, and physio-biochemical characteristics.
Objectives: This on-site study was performed to evaluate the reduction efficiency of an air cleaner on particulate matters and biological agents in a swine facility. Materials and Methods: Particulate matter was measured using a real-time monitoring recorder and biological agents were sampled with a one-stage impactor and then analyzed based on the microbial culture method. An experimental process for the reduction effect on airborne pollutants through air cleaner operation consisted of three conditions: no treatment, wet scrapper by water spray and wet scrapper by disinfectant spray. Results: Geometric mean levels of particulate matter(TSP, $PM_{10}$, $PM_{2.5}$ and $PM_1$) were presented at $1,608{\mu}g/m^3$, $1,373.8{\mu}g/m^3$, $401.8{\mu}g/m^3$ and $144.5{\mu}g/m^3$ for no treatment; $1,503{\mu}g/m^3$, $1,017{\mu}g/m^3$, $159.4{\mu}g/m^3$ and $69.8{\mu}g/m^3$ for wet scrapper by water spray; and $1,222.17{\mu}g/m^3$, $477.17{\mu}g/m^3$, $33.2{\mu}g/m^3$ and $11.1{\mu}g/m^3$ for wet scrapper by disinfectant spray, respectively. In the case of biological agents, the geometric averaged concentrations of total airborne bacteria and fungi were as follows: $45,371cfu/m^3$ and $13,474cfu/m^3$ for no treatment, $43,286cfu/m^3$ and $8,610cfu/m^3$ for wet scrapper by water spray, and $2,440cfu/m^3$ and 1,867 cfu/ for wet scrapper by disinfectant spray, respectively. Regardless of particulate matter and biological agent, the highest concentrations were found for no treatment, while the lowest concentrations were found with wet scrapper by disinfectant spray. Conclusions: Based on the results obtained from this on-site evaluation, there was a significant reduction effect on particulate matter and biological agents through the application of an air cleaner in this study.
본 연구에서는 오이 지상부에 발생하는 두 가지 병원균 B. cinerea와 C. orbiculare에 항균활성을 가지는 유용한 방선균을 선발하고자 하였다. 토양에서 분리한 방선균 560 균주 중 두 병원균에 대해 동시 항균활성을 나타낸 7균주를 일차적으로 선발한 후, 배양여액의 항균활성 검정을 위한 대치배양과 오이 식물체 검정을 통해 최종적으로 S20-465 균주를 선발하였다. 이 균주는 염기서열 비교 분석을 통해 Streptomyces sp.로 동정하였다. S20-465 균주의 배양여액은 두 병원균의 균사 생장을 60%이상 억제하였다. 배양여액의 20배, 40배 희석액을 분무 처리할 경우 오이 잎에서의 잿빛곰팡이병에 의한 병반길이를 감소시켰을 뿐만 아니라 탄저병의 발병도도 대조구에 비해 50% 이상 감소시켜 생물검정에서도 병억제 효과가 있는 것을 확인하였다. 따라서 최종 선발한 S20-465는 앞으로 생물방제 미생물소재로써 활용할 수 있을 것으로 사료된다.
연구배경 : 황사는 우리나라에서 주로 3월에서 5월까지 봄에 집중적으로 발생하는 대기오염의 주된 원인으로 사람에게 만성 기관지염을 악화시키고 호흡기 질환을 유발하는 것으로 알려져 있다. 황사기간 중 부유분진 중의 무기이온 성분과 금속성분의 분석에 대한 연구와 천식 등 호흡기 질환과의 연관성에 대한 연구는 많으나, 아직까지 황사의 부유분진중의 미생물에 대한 연구는 부족하다. 이 연구에서는 황사 현상시 부유분진 중에 박테리아, 진균 그리고 바이러스 등의 미생물의 검출유무와 종류를 알아보고 금속 성분 및 농도에 대해 알아보고자 본 연구를 시행하였다. 방 법 : 시료의 채취는 충남 태안군 파도리에서 cascade impactor(AN-200, USA)를 설치하여 1시간 동안 얻었으며 채취는 황사현상 기간과 비황사기간중 각각 한번씩 시행하였다. Cascade impactor에 의해 채취된 부유분진중의 미생물 검사는 cascade impactor내에 있는 여과지를 그람 양성균, 그람 음성균, 진균, 그리고 바이러스의 배지(4종, influenza, parainfluenza, adenovirus, RSV)에 옮긴 후에 배양하여 분석하였다. 금속 성분의 분석은 XRF를 이용하여 분석하였다. 결 과 : 조사 대상 지역의 황사기간과 비황사 기간 중 부유분진의 농도는 각각 80.2, $40.3g/m^3$ 이었다. 부유분진 중 XRF로 분석한 금속성분은 알루미늄과 규소, 황, 칼륨과 칼슘, 철 등 대부분의 금속이 비 황사 기간에 비해 황사 기간에서 농도가 높았다. 미생물 검사 결과상 그람 양성균은 Bacillus species, Coagulase negative staphylococcus가 자랐으며 황사기간보다 비 황사 기간동안 채취한 시료에서 오히려 더 많은 단계의 여과지에서 균이 동정되었으나 두 기 간에서 자라는 균주의 종류에는 차이가 없었다. 진균 배양은 비 황사 기간동안 Mucor species, Cladosporum, Alternaria, Aspergillus, Penicillium, Alternaria등의 진균들이 자랐으며, 황사 기간동안은 Penicillium과 Alternaria, 그리고 다른 mold form fungus들이 자랐다. 바이러스와 그람 음성균은 두 기간 다 자라는 균주는 없었다. 결 론 : 이 연구는 황사 기간동안 대기분진 중에 포함된 미생물에 대한 최초의 연구로 황사 현상시 미생물의 검출 종류는 크게 차이가 없었으나 개체수의 증가가 관찰되었으며 금속 성분의 종류는 차이가 없었으나 농도는 황사 현상시에 더 높은 수치를 보여 황사현상에 따른 호흡기 질환자의 주의와 대기오염에 대한 대책이 필요할 것으로 사료된다.
For the purpose of acquiring microbial agents that can be utilized to biologically control the major airborne diseases to apple trees, such as canker(Botryosphaeria dothidea), bitter rot(Glomerella cingulata), alternaria leaf spot(Alternaria mali), root rot(rosellinia necatrix), canker(Valsa ceratosperma) and gray mold rot(Botrytis cinerea), the effective microorgaisms were isolated, tested for antagonistic activity to the pathogens causing major diseases to apple trees and identifed. Screening of more than 5,000 species of microorganisms collected in nature for them antagonistic action to the pathogens causing 5 major diseases to apple trees resulted in selection of effective species. Out of the 11 species, one species designated as CAP134 demonstrated outstanding activity. The bacterial strain, CAP134 exerted antagonistic efficiency of 57% on an isolated strain and 40% on a donated strain of Botryosphaeria dothidea., 52% on an isolated strain and 46% on a purchased strain of Alternaria mali, 60% on Valsa ceratosperma 25% on Glomerella cingulata, and 64% Rosellinia necatrix. The CAP134 was identified as a bacterial strain to Bacillus subtilis ATCC 6633 based on morephology, culture conditions, and physio-biochemical characteristics.
