안지오텐신 수용체 리간드 KR-31125의 생체 내 활성에 관한 연구 (Pharmacological Profile of KR-31125, an Orally Active AT1 Receptor Antagonist)

  • 피리딜 이미다졸 시리즈의 비펩타이드 안지오텐신 수용체 리간드로 새롭게 개발된 KR-31125에 대한 생체 내활성을 동물모델에서 검증하였다. 척수장애 동물모델에서 KR-31125는 비대칭 농도의존적으로 로자탄보다 18배 이상의 경쟁적인 혈압강하 효과를 나타내었으며, 기타 수용체 촉진제들의 영향을 받지 않았다. 안지오텐신으로 유도된 정상혈압 쥐모델에서는 대조화합물인 로자탄과 비교하여 경구효과는 동등하였으나 더 빠른 초기효과가 관찰되었다. 또한 신성고혈압 쥐모델에서 KR-31125는 로자탄보다 3배 이상의 지속형 혈압강하 효과를 나타내었고, 이뇨제를 투여하여 레닌을 활성화시킨 개실험 모델에서 KR-31125는 로자탄보다 20배 이상의 지속적인 경구혈압강하 효과를 나타내었다. 이러한 KR-31125의 생체 내 활성특징은 대사물질을 통하여 효과를 발휘하는 로자탄과 달리 동일물질의 효과에 의한 것으로 고혈압 및 혈관질환과 깊은 관련이 있는 안지오텐신 조절시스템에 대한 세포영상, 비침투성 진단등의 도구물질로서 가능성이 높을 것으로 판단된다.

유산소 트레이닝과 식사조절 병행이 비만아동의 혈중지질과 아포지단백에 미치는 효과 (Effects of Aerobic Training Plus Diet on Blood Lipids and Apolipoproteins in Obese Children)

  • 비만 남자 초등학생을 대상으로 식사 조절과 병행한 유산 소운동을 HRmax의 60-70% 강도로 1일 100분, 주 5회, 9주간 실시하여 이들의 혈중지질과 아포지단백에 미치는 효과에 대해 알아보았다. 혈중지질의 TC, TG, HDL-C 농도와 TC/ HDL-C비가 개선되었는데 이는 규칙적인 유산소운동과 에너지섭취량 제한(식사제한)이 아닌 1일 영양권장량에 준하는 정상적인 에너지 섭취를 통해 비만아동의 체중감량과 더불어 CHD 위험인자인 혈중지질에 긍정적인 효과를 미칠 수 있음을 확인하였다. 또한 아포지단백의 Apo AII, B, CIII 농도가 감소하였는데 이는 유산소운동과 식사조절 병행이 성인의 경우와 유사하게 비만 남자아동들에게도 동맥경화 질환의 예방에 효과적인 방법이 될 수 있음을 증명하는 결과로 사료된다. 그렇지만 아직 운동에 따른 Apo 반응에 대한 연구들이 미흡하고 특히 Apo CII와 CIII에 대한 연구는 더욱 미흡한 상황이므로 향후 이에 관련된 후속 연구가 진행되어야할 것으로 생각된다.

유방염 우에서 원인균분리 및 sulfadimethoxine sodium 투여 후 혈청 및 유즙내 잔류량 추이 (Identification of bacterial agents causing mastitis in dairy cattle and observation of residual changes of sulfadimethoxine in serum and milk of the cattle after administration of sulfadimethoxine sodium)

  • This study was carried out to identify causative agents from california mastitis test(CMT) positive mastitic milk, and to examine the antimicrobial susceptibility of 50 heads in Seongju and Chilgok area of Gyeongbuk province. Sulfadimethoxine sodium(SMS) was intramuscularly administered once to four mastitis Infected dairy cattle at the rate of 10mg/kg body weight. After injection of SMS, the depletion rate of serum and milk from the cattle were periodically measured for five days. The CMT positive number from 50 heads showed 46% and that of 200 quarters was 47(23.5%). From 39 quarters of 47 heads 39 different microorganisms were identified. These organisms were classified into 12 species : Staphylococcus aureus 8(20.5%), Sta hemolyticus 6(15.4%), Streptococcus bovis 4(10.3%), Sta hyicus 3(7.7%), Sta epidemidis. Sta xylosus, Sta sciuri 2(5.1%), Str agalactiae 2(5.1%), Escherichia coli(10.3%), three Enterobacter cloacae(7.7%), two Ent aerogenes(5.1%) and one Salmonella spp(2.6%). As the results of antibiotic susceptibility test, gentamicin(Gm, 11 species 27 strains, 69.5%), cephalotin(Cf, 9 species 24 strains, 61.5%), sulfamethoxazole(Stx, 8 species 14 strains, 43.6%), tetracycline(Tc, 8 species 14 strains, 35.9%), and streptomycin(Sm), lincomycin(Lm), cefoperazon(Cp) and penicillin(Pc) have a highly resistance(7.7% ~5.1%). However, carbenicillin (Cb), amikacin(Ah) have no susceptible for all drugs. The mean residual concentration of SMS in serum according to the time lapsed were showed 33.964 $\pm$ 4.435ppm at the 4 hours after intramuscularly injection(AII). It was significantly(p<.05) decreased to 6.596 $\pm$ 3.402, 0.217 $\pm$ 0.119 and 0.005 $\pm$ 0.004ppm at the 1st, 3rd and 5th day AII. The mean concentration of SMS in milk was significantly(p<.05) decreased from 0.920 $\pm$ 0.42ppm to 0.084 $\pm$ 0.016ppm between 8 hours and 1 day AII. As the results of this experiments, sulfadimethoxine was residued at the level of no less than 0.01ppm in milk on the 2nd day AII. Thus, this results would be able to be used the basic index for prevention of sulfonamides residue in milk after treatment of dairy mastitis.

Alternative Production of Avermectin Components in Streptomyces avermitilis by Gene Replacement

  • The avermectins are composed of eight compounds, which exhibit structural differences at three positions. A family of four closely-related major components, A1a, A2a, B1a and B2a, has been identified. Of these components, B1a exhibits the most potent antihelminthic activity. The coexistence of the '1' components and '2' components has been accounted for by the defective dehydratase of aveAI module 2, which appears to be responsible for C22-23 dehydration. Therefore, we have attempted to replace the dehydratase of aveAI module 2 with the functional dehydratase from the erythromycin eryAII module 4, via homologous recombination. Erythromycin polyketide synthetase should contain the sole dehydratase domain, thus generating a saturated chain at the C6-7 of erythromycin. We constructed replacement plasmids with PCR products, by using primers which had been derived from the sequences of avermectin aveAI and the erythromycin eryAII biosynthetic gene cluster. If the original dehydratase of Streptomyces avermitilis were exchanged with the corresponding erythromycin gene located on the replacement plasmid, it would be expected to result in the formation of precursors which contain alkene at C22-23, formed by the dehydratase of erythromycin module 4, and further processed by avermectin polyketide synthase. Consequently, the resulting recombinant strain JW3105, which harbors the dehydratase gene derived from erythromycin, was shown to produce only C22,23-unsaturated avermectin compounds. Our research indicates that the desired compound may be produced via polyketide gene replacement.

쇠뜨기와 화살나무의 항암 효능에 관한 연구 (Studies on the Effects of Seddugi and Hwasalnamoo on the Anti-Cancer in Mice)

  • This study was designed to find out the anti-cancer effects of two herbs using a folk remedy in Korea-The two herbs were A: Equisetum arvense L. (Seddugi), B: Eunymus alatus Siebold. (Hwasalnamoo) The experiment for the anti-cancer effects were conducted in mice and experimental groups were divided into normal(NO), standard (ST) and two other(A,B) groups for each herb and each of the two experimental groups consisted of three sub-groups(I, II, III) according to three different quantity doses of a folk remedy using five times and ten times the quantity of a commonly used in a folk remedy. The water soluble extracts of two herbs were orally administered to mice during six weeks followed by inoculation of the sarcoma-180 tumor cells into the peritoneal cavity. During experimental periods, the contents of ascites, abdominal organs were observed histopathologically. The results of effects on anticancer were as follows : 1. The survival time was significantly prolonged by the two herbs, especially in groups AII & BII.2. Body weight, circumference of the abdomen and contents of ascites at the time of sacrifice were noted and significantly decreased in groups AI, AII & BII. 3. Anti-cancer effects evaluated by the invasiveness and thickness of the tumor cells were noted in groups AIII & BII.

Purification and Comparison of Properties of the C-Terminus Truncated Agarase of Pseudomonas sp. W7

  • Three plasmids derived from the ${\beta}-agarase$ gene (PjaA) of Pseudomonas sp. W7 were expressed in Escherichia coli AD494(DE3) pLysS with lactose as an inducer. These products corresponded to the complete (PjaA) and the two C-terminal truncated (PjaAI and PjaAII) forms of ${\beta}-agarase$. The PjaAI and the PjaAII were originated from exonuclease L treatment from PjaA by deleting 127 and 182 amino acid residues-encoded nucleic acids at 3' region, respectively. The molecular weights of the purified proteins were 71 kDa, 58 kDa, and 50 kDa on SDS-PAGE, respectively. The $K_m$ value of PjaAI was lower than that of the PjaA, and the catalytic efficiency ($k_{cat}/K_m$) of PjaAI was increased to 5 times. The enzyme of PjaAI retained more than 90% activity at $50^{\circ}C$. In contrast to the PjaAI, the remaining activity of the PjaA was only 20% at the same temperature.

Effect of age on endothelial function in rat aorta

  • The Influence of age on the endothelial modulation of angiotensin II (AII)-induced contractile response was investigated in isolated aortic rings of rats ranging in age from 0.7 to 20 months. Hemoglobin and L-NAME were used to examine whether age-related changes in the EDRF-releasing system were involved in endothelial modulation of All-induced contraction in rat aorta. In all five age groups (0.7, 1.5, 3, 6, 20 months), hemoglobin (10 ${\mu}$M) significantly enhanced All-induced contractile response only in aorta with endothelium intact. L-NAME (10 ${\mu}$M) Produced a significant enhancement in All responses in endothelium-intact aortas from rats aged 0.7 and 1.5 months, but it had no effect in aortas from older rats aged 6 and 20 months. Indomethacin (10 ${\mu}$M) did not affect All-induced contractile responses in both endothelium intact and removed aortas from rats at the age of 0.7 to 20 months. Hemoglobin (10 ${\mu}$M) abolished acetylcholine-induced relaxation response in aortas from young and old rats. L-NAME completely abolished the relaxation in aortas from young (0.7 and 1.5 months), but incompletely in aortas from older (6 and 20 months) rats. The sensitivity of endothelium-dependent relaxation to A23187 increased with age between ages of 0.7 and 6 months, with no further increase noted up to 20 months of age. These results suggest that endothelial modulation of AII-induced contraction in rat aorta might involve age-related alteration in EDRF-releasing system, probably via post-receptor mechanism.

AII-Bond - Fourth Generation Dentin Bonding System

  • The All-Bond system is a unique development in the field of adhesive dentistry. It is a universal bonding system that will bond composite to all dental-related surfaces: dentin, enamel, metal alloy (precious and nonprecious), amalgam, porcelain, and composite. It is also the only system that allows use of both the conservative and all-etch techniques. The main purpose of this paper is to explain the features of the All-Bond system. Its chemistry and a working hypothesis are shown as well.

The neurons in CVO are considered to playa pivotal role in blood-brain communication.
