Enhancement of Hydroxylamine Reactivity of Bacteriorhodopsin at High Temperature

  • Recent denaturation experiments of bacteriorhodopsin (bR) in the dark and under illumination at high temperatures revealed that irreversible thermal bleaching occurs above ~ 70°C and the preceding reversible structural changes in the dark above 60°C are closely related to irreversible photobleaching observed in the same temperature range (Yokoyama et al. (2002). J Biochem. 131,785). In this study, structural properties of bacteriorhodopsin (bR) at high temperatures were extensively probed by hydroxylamine reactivity with the Schiff base in the dark and hydrogen-deuterium (H-D) exchange in the peptide groups. In the Arrhenius plot from kinetics measurements of the hydroxylamine reaction, a good linear relationship between the reaction time constant and the inverse of the absolute temperature was observed below 60°C, while significant increase started above 60°C, suggesting that remarkable increase in water accessibility of the Schiff base in the temperature region. FT-IR spectroscopic studies on the H-D exchange suggested increase in the deuterium exchanges rate of the peptide hydrogen in the same temperature region.

Spider Fauna of Mt. Yongmunsan, Gyeonggi-do in Korea

  • Spider fauna of Mt. Yongmunsan located in Yangpyeong-gun, Gyeonggi-do, was surveyed from April to October in 1996. A total of 75 species of 51 genera in 22 families from 996 individuals of spiders were identified. Species richness of each family, there were 17 Araneid species (22.7%), followed by each of the 7 Thomisid and Tetragnathid species (9.3%), 6 Salticid species (8.0%), and 5 Lycosid species (6.7%). Zoogeographically, spider fauna of Mt. Yongmunsan represented 1 cosmopolitan species (1.3%), 2 holarctic region species (2.6%), and 9 palearctic region species (12.0%), and 4 Korean endemic species (4.0%). Thus it is suggested that spider fauna of Mt. Yongmunsan was under northern regional influence.

  • This study explores the development process of an farmers' group in Asan City that now plays an important role in the development of organic farming of the region. While increase in income in general may be one of main purposes making farmers join or form a group, the farmers group in Asan, instead, has tried to form a cooperative of local organic farmers. In doing so, they experienced a lot of difficulties and leant by trial and error. As a result, the farmers' group has recently developed in terms of business and organisational growth. The growth is not merely due to the growth of organic food markets but also due to the strong internal ties and trust that made possible to expand into food processing as well as to do social and cultural activities fur the rural residents. It implies that trust and cooperative identity between farmers should be the most important thing to be locally embedded farmers groups in a specific region.

The Analysis of Regional Economic Feasibility of GHGs Reduction Technology in the Paddy Rice (논 물관리 방식에 따른 온실가스 감축기술의 지역별 경제성 분석)

  • This study analyzed the regional economic feasibility of greenhouse gases (GHGs) reduction technology in paddy rice. Firstly, the impact of GHGs reduction technology on productivity, emission reduction, and costs is different from region to region. Secondly, the water irrigation system contributes to productivity, GHGs reduction, and water reduction, but the profit of paddy rice will decrease because of increase in fixed costs and variable costs. Thirdly, the economic feasibility shows that water-savings plot has a 1.41 in a benefit-cost ratio.

Alternaria brassicifolii sp. nov. Isolated from Brassica rapa subsp. pekinensis in Korea

  • A new species belonging to the genus Alternaria was isolated from the necrotic leaf spots of Brassica rapa subsp. pekinensis in Yuseong district, Daejeon, Korea. It is an occasional isolate, not an etiological agent, which is morphologically similar to A. broccoli-italicae, but differs in conidial size and conidiophore shape. Phylogenetic analysis using the sequence datasets of the internal transcribed spacer (ITS) region of the rDNA, glyceraldehyde-3-phosphate dehydrogenase (gpd), and plasma membrane ATPase genes showed that it is distantly related to A. broccoli-italicae and closely related to Alternaria species in the section Pseudoalternaria, which belonged to a clade basal to the section Infectoriae. Morphologically, the species is unique because it produces solitary conidia or conidial chains (two units), unlike the four members in the section Pseudoalternaria that produce conidia as short branched chains. It exhibits weak pathogenicity in the host plant. This report includes the description and illustration of A. brassicifolii as a new species.

An Integrated Watershed Environmental Assessment and Classification of the Mid-Nakdong River Region (낙동강 중류 지역의 통합적 유역환경평가 및 유형화)

  • Many of today's environmental problems are regional in scope and their effects overlap and interact. The purpose of this paper is to developed a simple method for an integrated assessment of environmental conditions across the Mid-Nakdong River Region, by combining data on land use, impervious cover, roads, streams, riparian areas, forest patches, population, pollutant loadings, soil erosion and topography. A cluster analysis was used to identify groups of sub-watersheds with similar environmental characteristics. The mean value for each group was used to find watershed that may be more vulnerable to future environmental degradation. Watersheds in cluster I and II had high amount of forest, but the amount of riparian vegetation was low. Watersheds in cluster III, which located in the middle Geumho River and the main course of Nakdong River, had a greater proportion of their agriculture, a greater proportion of agriculture on steep slopes, and less forest adjacent to streams. Watersheds in cluster IV and V were in the most urbanized areas of the region. The principal adverse impacts for watersheds in this group were high scores for urban area, impervious cover, pollutant loadings, population density, forest fragmentation, and low amounts of forest and riparian forest cover. Notwithstanding the exploratory nature of cluster analysis, it appears to be a useful tool for grouping watersheds with similar environmental characteristics.

A Phylogenetic Study of Scirpus planiculmis F. Schm. (Cyperaceae) Based on ITS1 Sequences of Nuclear Ribosomal DNA

  • This work was performed to confirm the molecular discrimination through the nrITS1 sequences among 3 taxa of Scirpus L. sensu lato (s.l.) species. S. planiculmis represented only 2 base sequence variations with S. maritimus in spite that they showed different morphological features. The nucleotide sequences of the ITS1 region from S. planiculmis were shown to have 99.1% homology with S. maritimus and 60.4% homology with S. triqueter. Although the morphology of S. planiculmis is similar with S. triqueter, molecular basis of the size and sequences on ITS1 region were shown to have distinctive differences. For divergency investigation on same sites and metapopulation, sequencing was conducted on ITS1 region with partial 5.8S and 18S regions. All plants of each species collected at the same site had identical band size pattern and sequences. Intraspecific molecular divergency was not identified in spite that these species live in different wetland sites. The ITS1 sequences described here provided a powerful genetic tool for phylogenetic studies which was difficult by morphological identification as high rate of morphological plasticity.

Phylogenetic Analysis of Phaeosphaeria Species Using Mating Type Genes and Distribution of Mating Types in Iran

  • Phaeosphaeria species are pathogenic on wheat, barley and a wide range of wild grasses. To analyze mating type loci of the Phaeosphaeria species and investigate mating type distribution in Iran, we sequenced mating type loci of 273 Phaeosphaeria isolates including 67 isolates obtained from symptomatic leaves and ears of wheat, barley, and wild grasses from two wheat growing region in Iran as well as 206 isolates from our collection from other regions in Iran which were isolated in our previous studies. Mating type genes phylogeny was successfully used to determine the species identity and relationships among isolates within the Phaeosphaeria spp. complex. In this study, we reported seven new host records for Phaeosphaeria species and the Phaeosphaeria avenaria f. sp. tritici 3 group was first reported from Iran in this study. Mating type distribution among Phaeosphaeria species was determined. Both mating types were present in all sampling regions from Iran. We observed skewed distribution of mating types in one region (Kohgiluyeh va Boyer-Ahmad) and equal distribution in the other region (Bushehr). However, when considering our entire dataset of 273 Iranian Phaeosphaeria isolates, the ratio of mating types was not deviated significantly from 1:1 suggesting possibilities for isolates of opposite mating type to interact and reproduce sexually, although the sexual cycle may infrequently occur in some regions especially when the climatic conditions are unfavorable for teleomorph development.

Analysis of Surplus Solar Energy in Venlo Type Greenhouse (벤로형 온실의 잉여 태양에너지 분석)

  • This research analyzed surplus solar energy in Venlo-type greenhouse using acquired typical meteorological year (TMY) data for designing a heat storage system for the surplus solar energy generated in the greenhouse during the day. In the case of paprika, the region-dependent heating loads for Jeju, Jinju, and Daegwanryong area were approximately 1,107.8 GJ, 1,010.0 GJ, and 3,118.5 GJ, respectively. The surplus solar energy measured in Jeju area was 1,845.4 GJ, Jinju area 1,881.8 GJ, and Daegwanryong area 2,061.8 GJ, with the Daegwanryong area showing 11.7% and 9.6% higher than the Jeju region and Jinju region respectively. In the case of chrysanthemums, regional heating loads were determined as 1,202.5 GJ for the Jeju region, 1,042.0 GJ for the Jinju region, and 3,288.6 GJ for the Daegwanryong region; the regional differences were similar to those for paprika. The recorded surplus solar energy was 1,435.2 GJ, 1,536.2 GJ, and 1,734.6 GJ for Jeju, Jinju, and Daegwanryong region, respectively. The Daegwanryong region recorded heating loads 20.9% and 12.9% higher than in the Jeju and Jinju region, respectively. From the above, it can be said that cultivating paprika, compared to cultivating chrysanthemums, requires less heating energy regardless of the region and tends to yield more surplus solar energy. Moreover, if the Daekwan Pass region is excluded, the surplus solar energy exceeds the energy required for heating. Although the required heating energy differs according to regions and crops, cucumbers were found to require the highest amount, followed by chrysanthemum and paprika. The amount of surplus solar energy was the highest in the case of paprika, followed by cucumber and chrysanthemum.
