Analysis of Quantitative Trait Loci for Yield Component Traits in Soybean Using Recombinant Inbred Lines (콩에서 수량구성요인과 관련된 양적형질유전자좌의 분석)

  • Seed and pod numbers are the main yield components in soybean. Selection for increased yield potential is main goal of plant breeding. The objective of this study was to identify quantitative trait loci(QTLs) that control pod number per plant, seed number per plant and pod in soybean. The 117 $F_{2:10}$ recombinant inbred lines(RILs) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Two independent QTLs for pod number per plant were identified from linkage group(LG) F and L. Two QTLs for seed number per plant were located on LG F and L. Seed number per pod was related with three QTLs located on LG D1a, D1b and F. Pod and seed number per plant have two common QTLs on LG F and L.

  • The seed, the most basic component, is an important factor in increasing production and efficiency in agriculture. Seeds with superior genes can be expected to improve agricultural productivity, crop survival, and reproduction. Currently, however, screening of superior seeds depends mostly on manual work, which requires a lot of time and manpower. In this paper, we propose a system that can extract the characteristics of seed phenotypes by using computer image processing technology, so that even a small number of people and a short period of time are needed to extract the characteristics of seeds. The proposed system detects individual seeds from images containing large quantities of seeds, and extracts and stores various characteristics such as representative colors, area, perimeter and roundness for each individual seed. Due to the regularity of input images, the accuracy of individual seed extraction in the proposed system is 99.12% for soybean seeds and 99.76% for rice seeds. The extracted data will be used as basic data for various data analyses that reflect the opinions of experts in the future, and will be used as basic data to determine the expressive nature of each seed.

Multiple Genes Related to Muscle Identified through a Joint Analysis of a Two-stage Genome-wide Association Study for Racing Performance of 1,156 Thoroughbreds

  • Thoroughbred, a relatively recent horse breed, is best known for its use in horse racing. Although myostatin (MSTN) variants have been reported to be highly associated with horse racing performance, the trait is more likely to be polygenic in nature. The purpose of this study was to identify genetic variants strongly associated with racing performance by using estimated breeding value (EBV) for race time as a phenotype. We conducted a two-stage genome-wide association study to search for genetic variants associated with the EBV. In the first stage of genome-wide association study, a relatively large number of markers (~54,000 single-nucleotide polymorphisms, SNPs) were evaluated in a small number of samples (240 horses). In the second stage, a relatively small number of markers identified to have large effects (170 SNPs) were evaluated in a much larger number of samples (1,156 horses). We also validated the SNPs related to MSTN known to have large effects on racing performance and found significant associations in the stage two analysis, but not in stage one. We identified 28 significant SNPs related to 17 genes. Among these, six genes have a function related to myogenesis and five genes are involved in muscle maintenance. To our knowledge, these genes are newly reported for the genetic association with racing performance of Thoroughbreds. It complements a recent horse genome-wide association studies of racing performance that identified other SNPs and genes as the most significant variants. These results will help to expand our knowledge of the polygenic nature of racing performance in Thoroughbreds.

Qualitative PCR Detection of Stack Gene GM Rice (LS28 X Cry1Ac) Developed in Korea (국내개발 stack gene GM 벼(LS28 X Cry1Ac)에 대한 정성 PCR 분석)

  • For the development of qualitative PCR detection method of genetically modified (CM) rice, rice species-specific gene, OsCc-1 (rice cytochrome c gene), was selected as suitable far use as an endogenous gene in rice. The primer pair OsCytC-5'/3'with 111 bp amplicon was used for PCR amplification of the rice endogenous gene, OsCc-1 and no amplified product was observed from 8 different crops as templates. Qualitative PCR method was carried out with stack traits of L528$\times$CryIAc1 GM rice developed in Korea. For the qualitative PCRs, some primer pairs were designed with a construct-specific and event-specific type based on T-DNA and junction sequences of T-DNA in GM rice. Actck-5'/3' amplifying between actin promoter and OsCK1 gene introduced in LS28 gave rise to an amplicon 306 bp; also, CrLB-5'/3' from CryIAcl and CKRB-5'/3'amplifying the junction region of T-DNA and genome sequence from LS28 as event-specific primers gave rise to an amplicon 142 bp and 91 bp, respectively. These primer pairs for the detection of event-specific targets not produced PCR amplicons on non-CM rice and various crops in contrast to event lines. Therefore, in this study we verified that event-specific primers were effective to specifically detect stack trait lines and demonstrated that this method presented could be provided with the detection-method data for risk assessment analysis of GM rice to be commercialized.

Estimation of Genetic Parameter for Milk Production and Linear Type Traits in Holstein Dairy Cattle in Korea (국내 Holstein 젖소의 유생산 형질과 유방 및 지제 선형심사 형질에 대한 유전모수 추정)

  • This study was conducted to estimate genetic parameters for milk production and linear type traits in Holstein dairy cattle in Korea. The data including milk yields, fat yields, protein yields, fat percent, protein percent, somatic score and 15 linear type traits for 10,218 first parity cows collected by Dairy Cattle Improvement Center, National Agricultural Cooperative, Korea, which were calving from January 2009 to April 2013. Genetic and error (co)variances between two traits selected form 19 traits were estimated using bi-trait pairwise analyses with WOMBAT package. The estimated heritabilities for milk yield(MY), fat yield(FY), protein yield(PY), fat percent(FP), protein percent(PP), somatic cell score(SCS), udder depth(UD), udder texture(UT), median suspensory(MS), fore udder attachment(FUA), front teat placement (FTP), rear attachment height(RAH), rear attachment width(RAW), rear teat placement(RTP), front teat length(FTL), foot angle(FA), heel depth(HD), bone quality(BQ), rear legs side view(RLSV), rear legs rear view(RLRV) and locomotion(LC) were 0.128, 0.144, 0.100, 0.273, 0.333, 0.090, 0.179, 0.066, 0.104, 0.109, 0.127, 0.099, 0.059, 0.069, 0.154, 0.014, 0.010, 0.052, 0.065, 0.175 and 0.031, respectively. Among the genetic correlations, UD, UT, FTP, RAW, FTL, FA and RLSV with MY were -0.334, 0.271, 0.445, 0.544, 0.076, -0.281 and -0.228, respectively, and MS, FTP, RTP, FTL, FA, BQ, RLSV, RLRV and LC with PP were -0.147, -0.182, -0.262, -0.136, 0.355, 0.311, 0.135, 0.233 and 0.143, respectively. Especially, MY had the highest positive genetic correlation with RAW (0.544), while SCS had the highest negative genetic correlation with LC (-0.603). FP had negative genetic correlation with most udder traits, whereas, FP had positive genetic correlation with leg and hoof traits (0.056 - 0.355).

Factor Analysis of Genetic Evaluations For Type Traits of Canadian Holstein Sires and Cows

  • Factor analysis was applied as a multivariate statistical technique to official genetic evaluations of type classification traits for 1,265,785 Holstein cows and 10,321 sires computed from data collected between August 1982 and June 1994 in Canada. Type traits included eighteen linear descriptive traits and eight major score card traits. Principal components of the factor analysis showed that only five factors explain the information of the genetic value of linear descriptive traits for both cows and sires. Factor 1 included traits related to mammary system, like texture, median suspensory, fore attachment, fore teat placement and rear attachment height and width. Factor 2 described stature, size, chest width and pin width. These two factors had a similar pattern for both cows and sires. In constrast, Factor 3 for cows involved only bone-quality, while in addition for sires, Factor 3 included foot angle, rear legs desirability and legs set. Factor 4 for cows related to foot angle, set of rear leg and leg desirability, while Factor 4 related to loin strenth and pin setting for sires. Finally, Factor 5 included loin strength and pin setting for cows and described only pin setting for sires. Two factors only were required to describe score card traits of cows and sires. Factor 1 related to final score, feet and legs, udder traits, mammary system and dairy character, while frame/capacity and rump were described by Factor 2. Communality estimates which determine the proportion of variance of a type trait that is shared with other type traits via the common factor variant were high, the highest ${\geq}$ 80% for final score, stature, size and chest width. Pin width and pin desirability had the lowest communality, 56% and 37%. Results indicated shifts in emphasis over the twelve-year period away from udder traits and dairy character, and towards size, scale and width traits. A new system that computes fmal score from type components has been initiated.

Effects of Dietary Chromium Picolinate Supplementation on Growth Performance and Immune Responses of Broilers

  • Two experiments were conducted to evaluate the effects of chromium (Cr) on the growth performance, bone trait, serum traits, and immune responses in broilers. The broilers were fed corn-soybean meal basal diet supplemented with Cr at level of 0(control), 200, 400, or 800 ppb in the form of chromium picolinate (CrPic). The broilers were fed treated diets for 6 weeks in Exp. 1, but the Cr supplement was removed for the last 3 weeks in Exp. 2. Exp. 1 showed that dietary supplement of Cr did not affect growth performance of the broiler, though improved feed efficiency (p<0.05) was observed during 0 to 3 weeks. Moreover, serum total (p<0.05) and HDL cholesterols (p<0.06) were significantly higher in pooled Cr added group at 6 weeks of age, however, the difference was not significant in Exp. 2. The pooled Cr added group in Exp.1 had significantly lower (p<0.05) alkaline phosphatase activity and higher (p<0.09) calcium at 3 weeks. Significantly lower phosphorus was also observed in Exp. 2. With continued supplement of Cr as in Exp. 1, the alkaline phosphatase activity maintained higher at 6 weeks, as opposed to significantly lower in Exp. 2, which had no further Cr supplement. Higher bone breaking strength was observed in 400 ppb Cr supplemented in Exp. 1, though not significantly different. Serum glucose and triglyceride were not affected by Cr supplement. Antibody against Infectious Bronchitis (IB) was significantly (p<0.05) higher with 400 ppb Cr supplemented, and anti-Newcastle disease (ND) antibody also tended to be higher (p<0.06) in pooled Cr added group at 6 weeks of age in Exp. 1. Peripheral blood blastogenesis activity was not different among the treatments. The results suggest that diet supplemented with 400 ppb CrPic may be beneficial to the broiler.

Investigation of Soybean Cyst Nematode Heterodera Glycines Type and Evaluation of Resistance on Soybean Varieties and Germplasms in Korea

  • Soybean cyst nematode (Heterodera glycines Ichinohe) is one of the serious soybean [Glycine max (L.) Merr.] pests in major soybean producing countries. The objective of this study was to investigate of Heterodera glycines type using the five SCN infested soybean field soils and was to evaluate resistance to the soybean cyst nematode HG 2.5.7 type on soybean varieties and germplasms. The five SCN contaminated soil samples were collected from the three provinces on November 2011 in Korea, and eggs were cultured on early spring season in 2012. For the second study, a total fifty nine soybean varieties and germplasms were tested by infestation of HG type 2.5.7 in the greenhouse. Soybean cyst nematode HG types were investigated from five locations, HG 2 (race 1) type at Donghae, HG 2.5 (race 1) type at Jeongseon and Hapcheon, HG type 2.5.7 (race 1 or 5) at Yeongwol, and HG 1.2.7 (race 5) type at Haenam locations in present study. No Korean soybean varieties and germplasms were observed with SCN resistant trait to the HG type 2.5.7. Average SCN female index were calculated with 82.7% in 59 plant materials. Our results could be provided useful information to develop a SCN resistant cultivar in Korea.

  • From development of computer science, genetic algorithm has been applied to many fields for search like non-linear problem based on various variables and optimization process. Among others, in the data mining field, there are methods to select the best input variables for model accuracy and various predict models which were merged by using the genetic algorithm. In the meantime, to improve and preserve quality of the Hanwoo (Korean cattle) which is represented the agricultural industry in our country, we need to find out outstanding economical traits of Hanwoo in having specific genotype of single nucleotide polymorphism (SNP) which is inherited to next generation. According to, This research proposed the selecting method to find genotype of SNPs marker which affects economical traits of the Hanwoo by using the genetic algorithm. And we selected the best genotypes of the principal SNPs marker by applying to real data on Hanwoo genetic.

Identification of Quantitative Trait Loci (QTLs) Associated with Oil and Protein Contents in Soybean (Glycine max L.) (콩에서 Microsatellite marker률 이용한 oil 및 단백질 함량의 양적형질 유전자좌의 분석)

  • Soybean oil and protein contents are very important as a nutritional component of food. The seed composition as oil and protein are polygenic traits. In this study, the Keunolkong${\times}$Iksan10 populations were evaluated with SSR markers to identify QTLs related to oil and protein contents. Three related independent QTLs near the marker satt100 on LG C2, satt546 on LG D1b+W and satt418 on LG L were identified oil contents. The three independent QTLs near the marker satt556 on LG B2, satt414 on LG J and satt238 on LC L were identified of protein contents. In the results of this study, common QTLs on LG L was associated with seed oil and protein contents. In the result of this study, it is believed that the seed composition material as oil and protein contents were mainly controlled by environmental stresses and they are seed size on genotypes.
