Effect of Lactobacillus acidophilus based probiotic product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs

  • The aim of this study was to evaluate the effect of Lactobacillus acidophilus probiotic (LAP) product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs. In total, 14 Beagle dogs with an average initial body weight of 10.19 ± 0.61 kg were randomly assigned into two dietary treatments,with and without LAP supplementation, for a 28-day feeding trial. At the end of the experiment, there was no significant (p > 0.05) difference in the concentration of serum total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), white blood cell (WBC), red blood cell (RBC), blood lymphocyte percentage, fecal hydrogen sulfide (H2S) and total mercaptans (R.SH) emission, and fecal coliforms counts. However, the serum concentrations of the triglyceride and fecal ammonia (NH3) emission of the LAP treatment were significantly (p < 0.05) decreased in the group compared with the CON dogs. Fecal total lactic acid bacteria counts were significantly (p < 0.05) increased in the LAP treatment. In conclusion, the supplementation of LAP in Beagle dog diets could decrease the blood triglyceride level and enhance the gut Lactobacillus count which may have positive effects on dogs.

  • A numerical procedure for the analysis of slender arch buckling problems for uniform dead weight is presented in this paper. Such loading changes in the arch profile. The problem is nonlinear. The numerical procedure is limited to an inextensible analysis and to elastic behavior. Based upon a numerical integration technique developed by Newmark for straight beams, a large deflection bending analysis is combined with small deflection buckling routines to formulate the numerical procedure. The numerical procedure is composed of a combination of the numerical integration and successive approximations procedure. The results obtained in this study are as follows : 1.The critical loads obtained in this study coincide with the results by Austin so that the algorithm developed in this study is verified. 2.The numerical results are converged with good precision when the half arch is divided into 10 segments in both Prime and Quadratic section. 3.The critical loads are decreased as the ratios of rise versus span are increased. 4.The critical loads are increased as the moments of inertia at the ends are increased. 5.The critical loads of Prime section are larger than that of Quadratic section under the same profile conditions.

홍삼의 자기 공명 영상 (Magnetic Resonance Imaging of Red Ginsengs)

  • MRI 시스템을 이용하여 홍삼의 MR 영상을 획득하여 내부 품질 평가 가능성을 검토해 보았다. 천삼, 내공이 존재하는 양삼 및 내백이 존재하는 양삼에 대하여 실제 단면 영상과 MR 영상과를 비교 검토해 본 결과, 비파괴적으로 홈삼 내부의 영상을 확인할 수 있었으며 내부 품질 평가에 있어서 주요한 판별인자가 될 것으로 사료된다. 또한 획득된 MR 영상의 중심부를 지나는 부분에 대하여 ID profile을 실시하여 분석해 본 결과에서 알 수 있듯이 정상 조직과 비정상 조직인 내공이나 내백 부의 구분이 용이하게 관찰됨을 관찰할 수 있었다. 즉, 홍삼의 내부 품질 평가에 있어서 비교적 용이하게 진행될 수 있는 ID profile을 적용한다면, 전체의 MR 영상을 획득하는데 있어 소요되는 시간 및 절차를 줄이고 효과적인 판별 가능성을 제시할 수 있었다.

Characterization and cDNA Cloning of a Defensin-Like Peptide, Harmoniasin, from Harmonia axyridis

  • We compared the mRNA expression profile of the Harmonia axyridis larvae that were either untreated or treated with LPS. The extracted mRNAs were subjected to ACP RT-PCR analysis using a combination of arbitrary primers and oligo (dT) primer. Among the 47 DEGs differentially expressed, we identified a cDNA showing homology with defensin-like antibacterial peptide. The cDNA showed a putative 32-residue signal sequence and a 50-residue mature peptide named harmoniasin. We also investigated the antibacterial activity of the harmoniasin analog, which exhibited potent antibacterial activities against Gramnegative and -positive bacteria strains and it also evidenced no hemolytic activity.

Blood amino acids profile responding to heat stress in dairy cows

  • Objective: The objective of this experiment was to investigate the effects of heat stress on milk protein and blood amino acid profile in dairy cows. Methods: Twelve dairy cows with the similar parity, days in milk and milk yield were randomly divided into two groups with six cows raised in summer and others in autumn, respectively. Constant managerial conditions and diets were maintained during the experiment. Measurements and samples for heat stress and no heat stress were obtained according to the physical alterations of the temperature-humidity index. Results: Results showed that heat stress significantly reduced the milk protein content (p<0.05). Heat stress tended to decrease milk yield (p = 0.09). Furthermore, heat stress decreased dry matter intake, the concentration of blood glucose and insulin, and glutathione peroxidase activity, while increased levels of non-esterified fatty acid and malondialdehyde (p<0.05). Additionally, the concentrations of blood Thr involved in immune response were increased under heat stress (p<0.05). The concentration of blood Ala, Glu, Asp, and Gly, associated with gluconeogenesis, were also increased under heat stress (p<0.05). However, the concentration of blood Lys that promotes milk protein synthesis was decreased under heat stress (p<0.05). Conclusion: In conclusion, this study revealed that more amino acids were required for maintenance but not for milk protein synthesis under heat stress, and the decreased availability of amino acids for milk protein synthesis may be attributed to competition of immune response and gluconeogenesis.

Transcriptome Profiling and In Silico Analysis of the Antimicrobial Peptides of the Grasshopper Oxya chinensis sinuosa

  • Antimicrobial peptides/proteins (AMPs) are present in all types of organisms, from microbes and plants to vertebrates and invertebrates such as insects. The grasshopper Oxya chinensis sinuosa is an insect species that is widely consumed around the world for its broad medicinal value. However, the lack of available genetic information for this species is an obstacle to understanding the full potential of its AMPs. Analysis of the O. chinensis sinuosa transcriptome and expression profile is essential for extending the available genetic information resources. In this study, we determined the whole-body transcriptome of O. chinensis sinuosa and analyzed the potential AMPs induced by bacterial immunization. A high-throughput RNA-Seq approach generated 94,348 contigs and 66,555 unigenes. Of these unigenes, 36,032 (54.14%) matched known proteins in the NCBI database in a BLAST search. Functional analysis demonstrated that 38,219 unigenes were clustered into 5,499 gene ontology terms. In addition, 26 cDNAs encoding novel AMPs were identified by an in silico approach using public databases. Our transcriptome dataset and AMP profile greatly improve our understanding of O. chinensis sinuosa genetics and provide a huge number of gene sequences for further study, including genes of known importance and genes of unknown function.

  • The effect of slaughter weight and gender on fatty acid composition of homogenised rib samples of Hungarian Simmental (HS) and Holstein-Friesian (HF) cattle breeds was investigated. In the first experiment 22 Hungarian Simmental and 18 Holstein-Friesian cattle's meat, in the second experiment 15 females and 12 males Holstein-Friesian cattle's meat was analysed on fatty acid content. Saturated and mono- or polyunsaturated fatty acid profile did not seem to vary either by breed or by slaughter weight categories. The effect of gender, however, proved to be significant in influencing the quantity of polyunsaturated fatty acids, including linoleic and linolenic acids, as well as polyunsaturated fatty/saturated fatty acids (PUFA/SAFA) ratio. The amount of polyunsaturated fatty acids (PUFA) was higher in males than in females. Negative correlation was established between the quantity of PUFA and the amount of adipose tissue in rib samples.

  • In the first study of a series of experiment samples were taken from 11-13th rib of Hungarian Simmental (HS, n=22) and Holstein-Friesian (HF, n=18) cull cows. In the second one, that of females (n=15) and males (n=12) of HF breed was analysed for amino acid composition, and biological value (BV) of proteins. No significant influence of either breed or slaughter weight was established in this study. Thus, the essential amino acid content and biological value of the proteins in Hungarian Simmental breed are practically the same as in the Holstein-Friesian. On the other hand, gender proved to be a significant factor influencing the amino acid profile of beef proteins, as the quantity of essential amino acids turned out to be significantly larger in females than in males. No statistical difference could be established for the non-essential amino acids and BV between the two genders.
