Optimal Condition of Natural Silk 3D Matrix Production by Silkworm Spinning

  • Silk is appealing materials for many biomedical applications involving tissue engineering and implantable devices, because of its biocompatibility, environmental stability, controlled proteolytic biodegradability and morphologic flexibility. Silk matrix is required for the treatment of a wide wound area, but the present silk matrix is made by the second processing, and thus, the labor and the cost are high. In this work, we investigated the optimal production condition of natural silk 3D matrix using the silkworms and invented Automatic Silk Matrix Making Machine (ASMMM) for natural silk 3D matrix production. As a result, we determined that optimal production condition for making A4 paper size natural silk 3D matrix was used Rough aquarelle paper on surface at $25^{\circ}C$ and 30 silkworm larvae. These results are expected to provide basic data for the efficient production of the natural silk 3D matrix, and it is suggested that the produced natural silk 3D matrix is useful as a medical biomaterials.

Optimization of Culture Condition for the Hydrocinnamic Acid Production from Bacillus subtilis IJ-31 (Bacillus subtilis IJ-31에서 Hydrocinnamic Acid 생산을 위한 최적배양조건)

  • The metabolites released from cultures of rhizosphere bacteria can inhibit plant growth. Bacillus subtilis IJ-31 inhibited plant growth by the production of hydrocinnamic acid (HCA). The production of HCA by plant-growth inhibiting rhizobacterium B. subtilis IJ-31 was optimized. $90.5\;{\mu}g/ml$ of HCA was obtained under the condition of 1% rice bran as carbon source, 0.5% tryptone as nitrogen source, 0.1% $ZnCl_2$ as metal source at $37^{\circ}C$ for 60 h (pH 7.0). The optimal condition for the HCA production by B. subtilis IJ-31 in the jar fermenter was established using response surface methodology (RSM) of statistical analysis system(SAS) program. The production of HCA by B. subtilis IJ-31 in the jar fermenter culture reached $102.99\;{\mu}g/ml$ when 2.24% soil extracts was added and agitation speed was 290 rpm under the same condition. And the experimental value of HCA production is $102.5\;{\mu}g/ml$ in the same culture condition. The production of HCA by B. subtilis IJ-31 is higher as 12% than that from the flask culture.

Irrigation Water Requirements for Upland Crops Using Rainfall Data and Water Management Guidelines (강우 자료와 밭작물 물관리 지침서를 이용한 노지 밭작물의 관개 필요량 산정 연구)

  • The purpose of this study is to determine the amount of irrigation water for upland crop growth based on the 30 year of historical rainfall data and the water management guidelines as a reference. Five regions and ten crops were selected by their cultivation size. The changes of soil moisture contents were calculated using daily mean rainfall and irrigation demand. This study assumed that crops are irrigated when the soil moisture contents fell below of the field capacity for more than 5 days, which is the drought condition defined by RDA. The maximum irrigation water requirements was 167.2 mm for chinese cabbage during the growing season, which was followed by corn (112.0 mm), daikon (102.3 mm), spinach (66.1 mm), lettuce (56.7 mm), pepper (46.5 mm), potato (33.9 mm), sweet tomato (27.4 mm), peanut (11.5 mm) and bean (10.3 mm), The results of this study could contribute to providing valuable data to determine the capacity of irrigation facilities and to establish the emergency operation plans under extreme unfavorable weather condition (heat wave, etc.) for crop growth.

Analysis of Organic Matter and Nutrient Leaching Characteristics of Agricultural Land Soils in Reservoir Area (저수구역 경작지 토양의 유기물 및 영양염류 용출특성 분석)

  • Soils in agricultural lands contain large amount of organic matter and nutrients due to the injected fertilizers and manure. During heavy rain, surface water and base runoff pollutants flows into a nearby stream or lake with eroded soil from agricultural lands. On the other hands, agricultural lands near the lake are inundated due to the increase of the water level in the lake, leading to organic matter and nutrient release from the inundated soil. In this study, releasing rates of nutrient salts and organic substances were analyzed for the soil in the agricultural land, where cultivation activities has been carried out and periodically flooded, to account for the possibility of contamination from the inundated agricultural land in reservoir areas The experiment results have shown that COD was released from the soil in anaerobic conditions, and T-P was released in both anaerobic and aerobic conditions. However, in the case of T-N, it was found that the runoff by soil was not made before the rainfall occurred, and when the soil was impound due to rainfall, the elution occurred under the aerobic conditions. Through the results of this study, it was possible to account for the effect of flooded agricultural lands on the water quality in the lake, and this could be reflected in an efficient agricultural non-point pollution management policy. In order to determine the precise releasing rate for each agricultural land, it is believed that the leaching experiment for paddy fields and grasslands are needed.

  • The coagulation of Chinese cabbage juice can be accomplished by applying the combine method of the formic acid with rate of 3% and in four hours the propionic acid with rate of 1 % in the juice. The separation of coagulation into the protein paste and the brown juice completed in 6.5 hours by set up method in special storage. The protein paste can be stored safely for 30 days in anaerobic condition.
