  • MR-387A [(2S, 3R)-2-hydroxy-3-amino-4-phenylbutanoyl-L-valyl-L-prolyl-(2, 4-trans)- L-4-hydroxy-proline] reversibly inhibits aminopeptidase N (BC 3.4.11.2) in a process that is remarkable for its unusual degree of time dependence. The time required to inactivate the enzyme by 50$%$ ($t_{1/2}$) for establishing steady-state levels of $EI^*$complex was approximately 5 minutes. This indicates that the inhibition is a slow-binding process. In dissociation experiments of $EI^*$ complex, enzymic activity was regained slowly in a quadratic equation, indicating that the inhibition of aminopeptidase N by MR-387A is tight-binding and reversible. Thus, the binding of MR-387A by aminopeptidase N is slow and tight, with $K_{i}$ (for initial collision complex, EI) and $K_i{^*}$ (for final tightened complex, $EI^*$) of $2.2\times10^{-8}$ M (from Lineweaver-Burk plot) and $4.4\times10^{-10}$ M (from rate constants), respectively. These data indicate that MR-387A and aminopeptidase N are bound approximately 200-fold more tightly in the final $EI^*$complex than in the initial collision EI complex.

  • Rice (Oryza sativa L.) is an important feeding crop in Asia, and utilization of genetically modified (GM) rice is highly demanding. For co-existence of GM rice and non-GM rice, the proper confinement measures should be provided. Thus, we surveyed gene flow from herbicide resistant GM rice to the conventional rice cultivars in the field tests. Gene flow frequency decreased with increasing distance between the pollen donor and recipients and did not exceed more than 1% even at the nearest distance. In single recipient model plot, a maximum gene flow frequency was observed at the shortest distance and hybrid was detected up to 12 m from the pollen donor. The direction of gene was coincided with the dominant wind direction. Gene flow assessment to multiple recipient plots was conducted under the high raining season by chance, and abrupt decline of gene flow frequency and maximum distance were resulted. According to the survey results, current regulation for isolation distance is reasonable for environmental safety or for general crop production. However, we suggest an alternative measure for GM rice cultivation that should be supplemented to overcome the out of estimation and in the environment asking higher security levels.

  • This study was carried out to investigate the effect of application dosage and methods of selenium (Se) on mineral contents in garlic. The content of Se was higher in upland than paddy fields. Application of higher dosage of Se salt increased higher content of Se in leaf sheath and bulb of garlic. Se was detected also in the non-treated plot but its amount was negligible. Absorption of Se in garlic was higher when Se was applied in the form of sodium selenate. Foliar application of sodium selenate enriched Se content in garlic as compared with the soil dressing. However, Se content in garlic fluctuated depending on growing conditions. Se content of garlic grown on the artificial soil (vermiculite and commercial soil) in the plastic house was higher than the garlic grown in the paddy field. The growth of garlic was not affected by types, dosage and application methods of selenium. Mineral contents in garlic such as Mg, S and K were higher in the selenium treatment than control. However, this trend was not evident according to dosages of selenium fertilizers.

  • The increasing area of abandoned rice fields could provide new opportunities for wetland restoration in Asia. However, it is unknown how quickly or completely abandoned rice fields will recover from agricultural disturbances. We assessed water quality and plant community succession in abandoned rice fields with different hydrology in a mountain valley to understand the effects of hydrological regime on recovery. Water level, soil redox potential, water quality, plant composition, and primary production were measured. The sites, coded as D6, N13, and N16, had been recovering for 6, 13, and 16 years by 2006. N13 and N16 have been recovering naturally whereas D6 has been drained with a nearby dike and was tilled in 2001. The typical hydroperiods of D6, N13, and N16 were no surface water, permanently flooded, and seasonally flooded, respectively. The major change in vegetation structure of both D6 and N13 was the replacement of herbaceous species by woody species. Drawdown accelerated this change because Salix koreensis grew better in damp conditions than in flooded conditions. Phragmites japonica reduced plot-level plant species richness. The removal efficiency of $NH_4-N$, $NO_3-N$, and $PO_4-P$ from water varied seasonally, ranging between -78.8 to 44.3%, 0 to 97.5%, and -26.0 to 44.4%, respectively. In summary, abandoned rice fields quickly became suitable habitat for native wetland plant species and improved regional water quality. Variation among our sites indicates that it is likely possible to manage abandoned rice fields, mostly through controlling hydrology, to achieve site-specific restoration goals.

  • An antagonistic bacterial strain KLF01 was isolated from rhizosphere of tomato and identified to be Lactobacillus sp. by biochemical and genetic analysis. This strain showed antagonism against the used plant pathogenic bacteria like Ralstonia solanacearum, (bacterial wilt), Xanthomonas axonopodis pv. citri, (Citrus canker), Xanthomonas campestris pv. vesicatoria (Bacterial spot), Eriwinia pyrifoliae (Shoot-blight) and Eriwinia carotovora subsp. carotovora group (Potato scab) through agar well diffusion method. In planta test done by drench application of strain KLF01 $(4{\times}10^8 cfu/ml)$ into the experimental plot containing tomato (Solanum lycopersicum L.) cultivar 'Lokkusanmaru' and red pepper (Capsicum annuum L.) cultivar 'Buja' plants, in pot test post-inoculated with the plant pathogenic bacteria, R. solanacearum significantly reduced the disease severity, compared to the non-treated plants.

  • The objectives of this research were to investigate the system of rice intensification (SRI) on early growth, grain yield, and yield components under Chinese milk vetch residue-mulched no-tillage cropping systems at silt loam soil. The field was prepared as a split-plot design with three replications, main plots consisted of Dongjinbyeo, and Sobibyeo as a cultivar, and subplots consisted of $10{\times}10$ cm, $20{\times}20$ cm, and $30{\times}30$ cm as a planting density. Weed infestation during rice growing season was more severe in wider planting density $30{\times}30$ at 35 days after transplanting (DAT), and $20{\times}20$ cm at 95 DAT in both Sobibyeo and Dongjinbyeo. The maximum plant height was recorded in Sobibyeo compared with Dongjinbyeo, $10{\times}10$ cm and $20{\times}20$ cm planting density compared with $30{\times}30$ cm from 20 DAT until 60 DAT. Among the three planting densities, SPAD values were significantly greater in planting density of $20{\times}20$ cm both in Sobibyeo and Dongjinbyeo followed by $30{\times}30$ cm compared with closer planting density of $10{\times}10$ cm. The lowest grain yield was observed in wider planting density of $30{\times}30$ in both Sobibyeo and Dongjinbyeo due to lower number of panicle per unit area. Our findings suggest that optimum planting density for SRI in no-tillage paddy was $20{\times}20$ cm and it should be useful the systems to small-scale rice farmers in Korea as a sustainable farming system.

  • In this study, we investigated the effects of the eco-friendly chemical bio-sulfur, on the citrus melanose-causing pathogen, Diaporthe citri, and on the pest, Panonychus citri. In an open field experiment with a plot-scale application of the chemicals: Mancozeb, lime sulfur, lime sulfur + machine oil, and bio-sulfur, the control group showed 70.6% disease severity compared with 10.3% for the Mancozeb-treated group. Among the eco-friendly treatments, disease severity was the lowest for the group treated with lime sulfur + machine oil (32.2%) and was 53.9%, 58.8%, and 58.1% following treatment with lime sulfur, and bio-sulfur diluted 500 and 1000 times, respectively. The proportion of diseased fruit showed similar results, suggesting that bio-sulfur is an effective alternative to lime sulfur. Three days after treatment acaricidal effects on P. citri showed a 197.6% control survival rate whereas the machine oil, and bio-sulfur diluted 500 and 1000 times treatments showed rates of 2.9%, 5.8%, and 9.0%, respectively. After three days, the control value for bio-sulfur diluted 1000 times was 73.2% compared with the values for the machine oil (96.4%) and bio-sulfur diluted 500 times (94.6%) treatments. Therefore, we suggest that additional research is needed on the combined application of bio-sulfur and oils to enhance the additive control effect on citrus melanose and Panonychus citri.

  • The experiment was implemented to introduce the drainage electrode irrigation system as early as possible after transplanting in order to save the nutrient solution in a convenient way. Drainage electrode irrigation method was introduced 15, 19 or 22 days after transplanting after irrigation was firstly controlled by time clock. Time clock method was also treated as a control plot. Drainage electrode method could be adopted from 15 days after transplanting, 15 days earlier than the present introducing time. The growth and yield was better in treatments with drainage electrode method. Water and fertilizer use efficiency were the highest in the treatment of 15 days, the lowest in time clock treatment.

  • This experiment was performed to analyze variation of growth traits of soybean to determine the information of less sensitive traits under different environmental conditions. Sowing was carried out on June 2 each year and the experimental plot was laid out in three replicates by randomized complete block design with thirty soybean varieties which consisted of 19 cultivars for beancurd and soypaste, 8 cultivars for soy-sprout including 1 landrace, and 3 cultivars for cooking with rice including 1 landrace during the period 2014-2015. The weather conditions during the experimental period were quite different with extremely low precipitation and longer sunshine duration in 2015 than 2014. The variation of characteristics related to growth period such as days of growth, days of maturity, days of flowering and the 100-seed weight was less in spite of different environmental conditions. While the variation of the number of pods per plant was high. Considering growth and seed characteristics like the number of pods per plant, the number of seeds per pod and 100-seed weight which are linked directly to the yield, cultivars such as Shingi, Daewonkong, Danbaegkong, and Daepung for beancurd and soypaste, Pungwon, Haepum and Shingang for soy-sprout and Seoritae for cooking with rice were more stable and could be expected to have high yield in Suwon, the south-central part of South Korea. These results could be useful for the selection of breeding resources to develop cultivars with high stability under changeable weather condition.

  • In this study, we attempted to find alternative materials to rice bran (Japonica in Korea) such as rice bran pellet (Indica in Pakistan), corn distillers dried grains with solubles, sesame oil meal, and kapok meal for bottle cultivation of Pleurotus eryngii. Among all treatments, the total carbon content of the mixed substrate was the highest in T1 and the total nitrogen content was the highest in T4. The C/N ratio of T2 was comparable to that of the control and was higher than that of other treatments. The diameter of the pileus was the largest in T1 and T2, and the length of the stipe was the largest in T1 and T2. The yield and bio-efficiency of the fruit body were higher in T2 than in the other treatments, and were similar to that of the control plot. Therefore, the findings of our study suggest that rice bran pellet (Indica in Pakistan) could be a suitable alternative to rice bran (Japonica in Korea) for the cultivation of P. eryngii.
