This study was conducted with the aim of evaluating the antibiotic effects of leaves, stems, and roots in yacon (Smallanthus sonchifolius). Antibacterial activity of leaf extract by disk diffusion method with Bacillus subtilis and Escherichia coli respectively showed 13.3 and 13.75mm diameters of clear zone. There was no significant difference between the stems and leaves. The minimum inhibitory concentration of leaves' heating and agitation extraction showed a restrain of strain at 1mg/ml, but the stems and root extract did not appear. Yacon is a functional antibacterial material, and methanol extraction is more effective than water. This research was to investigate the growth stage of collection has the most effective antibacterial effects. It has collected yacon's leaves from June to October, which is an appropriate time for collection right before reaping. Yacon leaf has antibacterial effects on Bacillus subtilis, Escherichia coli, Enterococcus faecium, and Salmonella enteritifis. There were no significant differences by the growth stage of collection. Leaves collected in July are high in phenol which helps in sulfating activity works well considering the high scavenging capability of DPPH. Leaves collected in September are high in total flavonoid.
Anodizing is a technology to generate thicker and high-quality films than natural oxide films by treating metals via electrochemical methods. It is a technique to develop metals for various uses, and extensive research on the commercial use has been performed for a long time. Aluminum anodic oxide (AAO) is generate oxide films, whose sizes and characteristics depending on the types of electrolytes, voltages, temperatures and time. Electrochemical manufacturing method of nano structure is an efficient technology in terms of cost reduction, high productivity and complicated shapes, which receives the spotlight in diverse areas. The sulfuric acid was used as an anodizing electrolyte, controlling its temperature to $10^{\circ}C$. The anode was 5083 Al alloy with dimension of $5(t){\times}20{\times}20mm$ while the cathode was the platinum. The distance between the anode and the cathode was maintained at 3 cm. Agitation was introduced by magnetic stirrer at 300 rpm to prevent localized temperature rise that hinders stable growth of oxide layer. In order to observe surface characteristics with applied current density, the electrolyte temperature, concentration was maintained at constant condition for $10^{\circ}C$, 10 vol.%, respectively. To prevent hindrance of stable growth of oxide layer due to local temperature increase during the experiment, stirring was maintained at constant rate. In addition, using galvanostatic method, it was maintained at current density of $10{\sim}30mA/cm^2$ for 40 minutes. The cavitation experiment was carried out with an ultrasonic vibratory apparatus using piezo-electric effect with modified ASTM-G32. The peak-to-peak amplitude was $30{\mu}m$ and the distance between the horn tip and specimen was 1 mm. The specimen after the experiment was cleaned in an ultrasonic, dried in a vacuum oven for more than 24 hours, and weighed with an electric balance. The surface damage morphology was observed with 3D analysis microscope. As a result of the investigation, differences were observed surface hardness and anti-cavitation characteristics depending on the development of oxide film with applied current density.
Distillery wastewater was used in a thermophilic laboratory-scale two stage anaerobic digester to test the effects of the redox potential of the first acidogenic reactor on the performance of the system. The digester consisted of first a acidogenic reactor and the an upflow anaerobic sludge blanket (UASB) reactor. The digestor was operated at a hydraulic retention time (HRT) of 48 h. Under these conditions, about 90% of the chemical oxygen demand as measured by the chromate method ($COD_{cr}$) was removed with a gas production yield of 0.4 l/g-COD removed. The redox potential of the acidogenic reactor was increased when the reactor was purged with nitrogen gas or agitation speed was increased. The increase in reduction potential was accompanied by an increase in acetate production and a decrease in butyrate formation. A similar trend was observed when a small amount of air was introduced into the acidogenic reactor. It is believed that the hydrogen partial pressure in the acidogenic reactor was decreased by the above mentioned treatments. The possible failure of anaerobic digestion processes due to over-loading could be avoided by the above mentioned treatments.
This study attempted to evaluate the process of self-forming dynamic membrane formation on mesh filter in membrane bioreactor with a two-stage method of batch (agitation) and continues (aeration) stage at different sludge concentrations. Four concentrations of activated sludge including $6{\pm}0.4$, $8{\pm}0.5$, $10{\pm}0.3$, $14{\pm}0.3g/L$ were used to demonstrate the optimal concentration of sludge for treating municipal wastewater and reducing fouling in dynamic membrane bioreactor. The formation time and effluent turbidity were decreased in the batch stage when increasing the activated sludge concentration. The minimum values of formation time and effluent turbidity were 14 min and 43 NTU for the optimum mixed liqueur suspended solids of $8{\pm}0.5g/L$, respectively. To improve operational condition and fouling reduction in the aeration stage, critical fluxes were measured for all concentrations by flux-step method. With increasing the sludge concentration, the relevant critical fluxes reduced. The optimum subcritical flux of $30L/m^2/h$ was applied as operating flux in the second stage. The maximum COD removal efficiency of 98% was achieved by the concentration of $8{\pm}0.5g/L$. Compressibility index of self-forming dynamic membrane and transmembrane pressure trend remained somewhat constant until the optimal concentration of $8{\pm}0.5g/L$ and thereafter they increased steeply.
본 연구에서는 국내 토양에서 분리된 곤충병원성 선충 Steinernema carpocapsae Kor-A1에서 분리된 공생박테리아 X. nematophilus Kor-A1 균주를 이용하여 액체 배양을 통해 생산조건에 따른 균주의 성장과 항균활성의 영향을 검증하고 항균물질의 생산조건 최적화를 검토하고 하였다. Flask 상에서 배양시간에 따른 항균활성을 조사한 결과 growth associated form을 보여 주었으며, 공생박테리아의 성장의 정지기에 최대 활성을 타나내었다. 또한 5L jar fermentor 상에서 최적 배양 조건을 알아보기 위해 교반속도와 DO의 영향에 대한 실험을 수행한 결과 교반속도는 400rpm에서 DO는 50%일 때 X. nematophilus Kor-A1의 균체 성장 및 항균물질 분비에 최적임을 알 수 있었다.

The filamentous fungus Aspergillus tubingensis that belongs to the black Aspergillus section has the capacity to produce high-value metabolites, for instance, naphtho-gamma-pyrones (NGPs). For these fungal secondary metabolites, numerous biological properties of industrial interest have been demonstrated, such as antimicrobial, antioxidant and anti-cancer capacities. It has been observed that production of these secondary metabolites is linked with fungal sporulation. The aim of this research was to apply osmotic and oxidative environmental stresses to trigger the production of NGPs in liquid cultures with CYB (Czapek Dox Broth). In addition, numerous parameters were tested during the experiments, such as pH value, incubation time, container geometry, and static and agitation conditions. Results demonstrate that the produced amount of NGPs can be enhanced by decreasing the water activity ($a_w$) or by adding an oxidative stress factor. In conclusion, this study can contribute to our knowledge regarding A. tubingensis to present an effective method to increase NGP production, which may support the development of current industrial processes.
The objective of this study was to evaluate the effects of surfactant type and concentration upon dissolution rates of carbamazepine from an immediate-release tablet. The dissolution media used in this study were aqueous solutions containing 0.1-2% sodium lauryl sulfate, cetyltrimethylammonium bromide, or polysorbate 80. The solubility of carbamazepine in the dissolution media was determined at first. A dissolution study was then conducted by using the USP dissolution apparatus II (paddle method) with an agitation rate of 75 rpm. Aliquots of the dissolution media were taken at predetermined time intervals, and the amount of carbamazepine dissolved was measured spectrophotometrically at 285 nm. The dissolution data obtained were fitted into a biphasic exponential equation with four parameters. Excellent correlations were observed between the experimental data and the theoretical ones predicted by the equation. This equation permitted the calculation of $T_{50%}$ (the time required for dissolving 50% of carbamazepine) under various experimental conditions. Differentiation of the equation also led to the attainment of dissolution rates at dissolution time points. The addition of a surfactant to an aqueous solution led to increasing the solubility of carbamazepine by 3- to 12-folds, depending upon its type and concentration. This event also resulted in enhancing the magnitude of a sink condition during the dissolution study. As a result, the dissolution rate of carbamazepine was affected by the aqueous surfactant concentration in a proportional manner. Subsequently, $T_{50%}$ values declined rapidly, as the surfactant concentration increased. Such effects were observed in decreasing order of sodium lauryl sulfate, cetyltirmethylammonium bromide, and polysorbate 80. These results clearly demonstrated that it was possible to tailor a dissolution rate and $T_{50%}$ of carbamazepine by manipulating the type and concentration of a surfactant. Relevant information would be beneficial to setting up dissolution specifications for poorly water-soluble drug products.
Cellulose로 만들어진 다공성 미립담체를 이용하여 HeLa cell을 working volume 100mL의 spinner flask에서 배양하였으며, 세포가 완전히 자란 미립담제를 계대배양하기 위하여 담체간 세포 전이 배양 방법을 시도하였다. 부유세포의 농도는 다공성 미립담체-HeLa 시스템의 경우에 담체간 세포 전이 배양에 영향을 미치는 중요한 인자로 작용하였으며, 낮은 칼슘농도의 배지인 RPMI-1640과 빠른 교반 속도를 이용하여 활성을 유지한 많은 세포가 떨어지도록 유도하였으며, 담체간 세포 전이 배양을 효과적으로 3회 이상 실시할 수 있었다. 이렇게 배양한 세포에 재 조합 Vaccinia virus를 감염하여 그 수율을 비교한 결과 T-flask에서 떼어낸 세포로 접종한 미립담체 배양과 거의 비슷한 재조합 단백질($\beta$-galactosidase) 수율을 나타내었다. Trypsin 처리 방법에 의한 미립담체 계대 배양도 경우에 따라서는 유용한 미립담체 계대 배양 방법이 될 수 있지만 실제 생산 규모에의 적용에는 공정이 복잡해지고 정확한 제어가 필요하다는 등의 문제가 있다. 따라서, 추가적인 비용이나 공정이 필요 없는 간편한 방법인 담체간 세포 전이 배양은 동물세포 배양을 이용한 유용 단백질 및 바이러스 생산 공정의 규모 증대에 매우 유용한 수단이다.
Streptococcus zooepidemicus 유래의 세포외 고분자물질인 히알루론산(hyaluronic acid) (HA)을 대량 생산하기 위해, 균주 개량, 생산배지 및 배양공정 개발에 관한 연구를 수행하였다. HA 고생산성 변이주를 선별하기 위해 약 99%의 사멸률을 보이는 ethylmethane sulfonate (EMS) 처리조건을 적용해서, 지속적인 random screening 방법으로 고생산성, 고안정성의 변이주들을 선별할 수 있었다. HA를 고농도로 생산하기 위해서는, 이 균주의 생화학 및 배양생리적 특성에 기반한 최적 배지개발이 필수적이라고 판단하여, one-factor-at-a-time (OFAT), full factorial design (FFD), steepest ascent method (SAM) 및 response surface method (RSM) (반응표면분석법)을 순차적으로 적용하여 통계적 배지 최적화 실험을 수행하였다. 최적 배지조성에서 플라스크 배양에 의한 HA 생산성은 5.38 g/l로서, 이전 배지(3.54 g/l)에 비해 약 52% 향상된 생산량을 얻을 수 있었다. 또한 선별된 우량균주와 최적화된 생산배지를 이용하여 5 L 발효조에서 배양공정 최적화 연구를 수행하였다. 이 균주의 생리학적 특성을 고려할 때, HA 생산성을 높이기 위해서는 (배양 중 HA 축적으로 인해 고점도를 띠는) 배양액으로의 충분한 용존산소 공급이 매우 중요한 요인인 것으로 판단되었다. 따라서 용존산소 공급과 밀접하게 관련있는 발효조의 교반시스템(교반 날개 종류, 크기 및 배치 등) 및 교반속도에 대한 최적화 연구를 수행하였다. 그 결과, 교반축 하부에는 Rushton turbine-type, 상부에는 marine-type의 확장된 교반날개(기존 대비 직경 1.3배 확장)가 설치된 경우, 450 rpm에서 강화된 혼합력과 충분한 용존산소 공급으로 인해 HA 생산성이 기존 플라스크 배양 대비 약 1.8배(9.79 vs. 5.38 g/l) 더 높은 것으로 확인되었다. 최종적으로 HA 배양공정의 scale-up 가능성을 확인하기 위해, pilot 규모의 50 L 발효조 배양을 최대 300 rpm의 교반속도에서 수행하였다. 처음으로 시도한 50 L 배양임에도 불구하고, HA 최대 생산성 면에서 볼 때, 5 L 발효조 결과와 거의 동일한 수준(98.5%) (9.11 vs 9.25 g/l)의 생산량을 얻을 수 있었다. 반면 지수기 성장단계인 배양 15시간까지의 50 L 배양의 HA 평균생산속도(rp)는 0.46 g/l/hr로서 0.62 g/l/hr인 5 L 배양 대비 약 74% 정도에 머무는 것으로 나타났다. 따라서 생산 발효조의 scale-up 시, 생산균주의 전단응력 민감성(shear damage)을 함께 고려하면서, 산소전달계수(kLa)를 기반으로 하는 교반시스템에 대한 체계적인 연구가 진행된다면, HA 생산속도도 증가될 수 있는 긍정적인 결과를 얻을 수 있을 것으로 기대된다.
In this work, mass production of Bacillus licheniformis SCD121067 through medium optimization by statistical experimental method was studied. First, galactose, yeast extract and potassium phosphate dibasic were selected as carbon, nitrogen and phosphate sources for mass production of B. licheniformis SCD121067 by using one factor at a time method. Second, according to the result of Plackett-Burman experimental design, key factors was yeast extract and $K_2HPO$. Finally, the response surface methodology was performed to obtain the optimum concentrations of two selected variables. The optimized medium composition consisted of 20 g/L galactose, 36 g/L yeast extract, 0.41 g/L $K_2HPO4$, 0.25 g/L $Na_2CO_3$, 0.4g/L $MgSO_4$ and 0.01g/L $CaCl_2$. Dry cell weight (15.4 g/L) by optimum production medium were increased 10 times, as compared to that determined with basic production medium (1.5 g/L). Fermentation conditions were examined for the mass production of B. licheniformis. The effect of temperature, agitation speed, pH and aeration rate on the mass production of B. licheniformis were also studied in a batch fermenter which was carried out in a 2.5 L bioreactor with a working volume of 1.5 L containing optimized production medium. As a result, dry cell weight of batch culture was 30.7 g/L at $42^{\circ}C$, 300 rpm, pH 8.0 and 2 vvm.
