  • Extracellular production of recombinant human bone morphogenetic protein-7 (rhBMP-7) was carried out through the fermentation of Bacillus subtilis. Three significant fermentation conditions and medium components were selected and optimized to enhance the rhBMP-7 production by using the response surface methodology (RSM). The optimum values of the three variables for the maximum extracellular production of rhBMP-7 were found to be 2.93 g/l starch, 5.18 g/l lactose, and a fermentation time of 34.57 h. The statistical optimization model was validated with a few fermentations of B. subtilis in shake flasks under optimized and unoptimized conditions. A 3-L jar fermenter using the shake-flask optimized conditions resulted in a higher production (413 pg/ml of culture medium) of rhBMP-7 than in a shake flask (289.1 pg/ml), which could be attributed to the pH being controlled at 6.0 and constant agitation of 400 rpm with aeration of 1 vvm.

  • Stellate ganglion block (SGB) is one of the most widely used treatment modalities for a broad range of disorders, including otolaryngologic indications such as Meniere's disease and sudden hearing loss. We present a case of a vertiginous attack following SGB for the management of Meniere's disease. A 31-year-old female, suffering from Meniere's disease, underwent repeated right side SGBs with 6 ml of 1% mepivacaine after negative aspiration tests for blood. The eleventh block was performed in the usual manner. Several seconds after injection, she showed agitation, anxiety, nystagmus, and left-sided tinnitus. Two minutes later, her tinnitus and nystagmus were resolved. Fifteen minutes after injection, she experienced acute onset of severe vertigo, nausea, and vomiting. However, her symptoms were gradually alleviated within two hours.

  • The production of red pigment from glucose by fed-batch culture of Monascus anka was investigated. In batch culture using fermentor, 200 rpm of agitation speed, 1vvm of aeration volume, and 10% (v/v) of inoculum size were optimal, respectively. The red pigment production was increased by removal of wall-attached mycelium. In an intermittent feeding fed-batch culture, dry cell weight increased to 30 g/l, adn the red pigment content reached 350 of absorbance at 495nm. In a continuous feeding fed-batch culture, dry cell weight increased to 22g/l, and the red pigment content reached 190 of absorbance at 495nm.

  • The gas-liquid mass transfer volumetric coefficients in gas-liquid agitated vessels with wire gauge impellers were measured to be compared with those in vessels with disk turbine and paddle impellers. Also mass transfer volumetric coefficients for disk turbine, paddle impeller and wire gauge impeller in cylindrical agitated vessel was measured over a wide range of Reynolds number from turbulent flow to transition regions. The effect of geometries on $k_La$ is clarified experimentally. Mass transfer volumetric coefficients $k_La$ depends only on the power consumption ($P_{gv}+P_{av}$) per unit volume.

  • Phaffia rhodozyma is the most promising microbial source of astaxanthin production, though wild-type strains are needed to increase the astaxanthin content for commercial production. To increase astaxanthin content for commercial production, a mutant strain of P. rhodozyma was selected and culture conditions of the mutant selected were optimized. P. rhodozyma was treated with mutagenic agent such as NTG, acriflavine, and UV in serial order and carotenoids hyper-producing mutant strain was selected based on the capabilities of cell growth on the agar plate containing chemical inhibitors and carotenoids production. Among the mutants tested, a mutant WS-2 was finally selected. Mutant WS-2 produced 1.26mg carotenoids/g-dry cell weight and this value was about- 4-folds higher than that of wild-type. The optimum culture conditions were $24^{\circ}C$ of temperature, 1.5vvm of aeration and 300rpm of agitation. In the optimized condition, cell and carotenoids concentrations were 7.62g/l and 14.9mg/l, respectively.

  • In this study, Bacillus licheniformis cells were immobilized by entrapment in calcium alginate beads and were used for production of alkaline protease by repeated batch process. In order to increase the stability of the beads, the immobilization procedure was optimized by statistical full factorial method, by which three factors including alginate type, calcium chloride concentration, and agitation speed were studied. Optimization of the enzyme production medium, by the Taguchi method, was also studied. The obtained results showed that optimization of the cell immobilization procedure and medium constituents significantly enhanced the production of alkaline protease. In comparison with the free-cell culture in pre-optimized medium, about 7.3-fold higher productivity was resulted after optimization of the overall procedure. Repeated batch mode of operation, using optimized conditions, resulted in continuous production of the alkaline protease for 13 batches in 19 days.

  • Rhodospirillum rubrum P17 was used to investigate the pontential for the treatment of soybean curd waste and for the utilization of the biomass produced. The maximal biomass production and COD removal from the waste water were obtained at 30C, pH 7.0 under 2,500lux production and 50 rpm of agitation. The initial COD level of the soybean curd waste water was 3,240mg/l, and after 4 days of cultivation in batch culture, 3.46g/l of cells was obtained and COD level of the waste water reduced to 150mg/l (COD removal rate 95.4%).

  • Ash melting chamber is one of the key facility of the pyrolysis-melting incineration system, and it should be designed and operated very carefully for avoiding solidification of slag. In this study, an example of numerical and experimental scale-up process of the melting chamber, in which high speed air is injected to the molten slag and generates bubbles, which enhances agitation of the slag and char combustion, is presented. Cold flow test, combustion and melting test in a lab-scale (30 kg/hr) chamber and a pilot scale (200 kg/hr) chamber. Minimum energy for maintaining molten slag is derived, and it was found that the molten slag can be maintained efficiently by concentrating heat into the bubbling slag.

  • Thermal behavior of Flow Pattern Defect (FPD) and Large Pit (LP) in Czochralski Silicon crystals was investigated by applying high temperature ($\geq$1100$^{\circ}C$) annealing and non-agitation Secco etching. For evaluation of the effect of LP upon device performance / yield, DRAM and ASIC devices were fabricated. The results indicate that high temperature annealing generates LPs whereas it decreases FPD density drastically, and LP does not have detrimental effects on the performance /

  • The Ni-SiC composite plating was performed in a Watt nickel solution and the wear resistance of the composite layer was studied on a pin-on-flat type wear tester. The volume losses and friction coefficients were measured. It was found that the quantity of SiC powder in the composite layers was affected by SiC concentration, pH, temperature, and agitation speed in the Watt nickel solution. The hardness and wear resistance of the coatings increased with SiC content. The quantity of SiC powder in the coating from a nickel sulfamate solution is larger than that of the Watt nickel solution, because the amount of nickel ions absorbed on the SiC powder in the nickel sulfamate solution is greater than that in the Watt's solution.
