Tea flavonoids and polyphenols are well known for their extraordinary antioxidant activity which is considered important for anti-aging processes in animals. This study evaluated the anti-wrinkle effects of three different kinds of tea (Camellia sinensis) water extracts (CSWEs) including green, white, and black teas using a photoaged hairless mouse model. Data showed that the CSWE-treatment greatly improved skin conditions of mice suffering from UVB-induced photoaging, based on the parameters including the skin erythema index, moisture capacity, and transepidermal water loss. In addition, the wrinkle measurement and image analysis of skin replicas indicated that CSWEs remarkably inhibited wrinkle formation. In histological examination, the CSWE-treated mice exhibited diminished epidermal thickness and increased collagen and elastic fiber content, key signatures for skin restoration. Furthermore, the reduced expression of MMP-3, a collagen-degradative enzyme, was observed in the skin of CSWE-treated animals. Interestingly, comparative data between green, white, and black tea indicated that the anti-wrinkle activity of white tea and black tea is equally greater than that of green tea. Taken together, these data clearly demonstrated that CSWEs could be used as an effective anti-wrinkle agent in photoaged animal skin, implying their extended uses in therapeutics.
A novel retinol derivative, polyethoxylated retinamide (Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of [$^3H$]-proline incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells (effective diffusion area: $1, 766{\;}\textrm{cm}^2$) and the excised skin of female hairless mouse aged 8 weeks were used The stabilities of retlnoids were evaluated at two different temperature ($25{\;}^{\circ}C$ and $40{\;}^{\circ}C$) and under UV in solubilized state and in OW emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.
Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 mg of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 mg/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 mg/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 mg/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.
스테로이드 호르몬인 glucocorticoid (GC)는 glucocorticoid receptor (GR)와 결합하여 염증 유전자의 발현을 억제함으로써 강력한 항염 효과를 준다. 따라서 GR을 활성화하는 GC 제제들이 개발되어 피부염의 치료제로 사용되어 왔다. 그러나 이러한 GC 제제들은 피부 장벽 기능 저하, 진피층 두께 감소 등의 부작용을 유발하여 피부를 손상시키고 피부 노화를 유발한다. 특히 GC 성분은 11 beta-hydroxysteroid dehydrogenase type 1 ($11{\beta}$-HSD1)에 의해 활성화되어 GR의 활성을 높이는 것으로 보고되어 있다. 이에 본 연구에서는 스테로이드 제제인 dexamethasone에 의해 증가된 $11{\beta}$-HSD1의 발현을 효과적으로 억제할 수 있는 천연 소재를 발굴하고자 하여, 초두구 추출물에서 유래한 alpinetin에서 유의한 효과가 있음을 확인하였다. Alpinetin은 진피 섬유아세포에서 dexamethasone에 의해 발현이 증가한 $11{\beta}$-HSD1를 억제함과 동시에 GR의 활성 및 cortisol의 생성을 감소시켰다. 또한 사람 섬유아세포 및 3D skin model을 이용한 평가에서, alpinetin은 dexamethasone에 의한 콜라겐 감소와 진피층 두께 감소를 효과적으로 회복시켰다. 따라서, 본 연구 결과로부터 alpinetin은 $11{\beta}$-HSD1의 발현 증가에 의한 피부 스트레스 및 피부 노화를 효과적으로 예방할 수 있을 것으로 사료되었다.
별불가사리(Asterina pectinifera Muller and Troschel)는 우리나라 전국 연안에서 흔히 볼 수 있는 토속종으로, 패류 양식장에 피해를 주는 불가사리류 중의 하나이다. 별불가사리 퇴치를 위해 대부분 건조하여 비료로 활용하고 있으며, 고부가가치 창출을 위한 다양한 연구가 진행되었으나 실제 활용은 미미한 실정이다. 따라서, 별불가사리로부터 피부 유용성분을 밝혀 새로운 활용 방안을 모색하고자 하였다. 성분연구를 통해 별불가사리로 부터 2종의 polyhydroxysteroid와 1종의 saponin을 분리하였으며, 이의 구조를 각각 $5{\alpha}$-cholestane-$3{\beta},6{\alpha},7{\alpha},8,15{\alpha},16{\beta},26$-heptol, $5{\alpha}$-cholestane-$3{\beta},4{\beta},6{\alpha},7{\alpha},8,15{\beta},16{\beta},2$6-octol 및 asterosaponin $P_1$으로 동정하였다. 이 물질들의 피부 효능을 확인한 결과 asterosaponin $P_1$이 표피 줄기세포의 증식을 촉진시키고, 각질형성세포에서 히알루론산을 합성하는 효소인 hyaluronan synthase-2와 hyaluronan synthase-3 유전자의 발현을 증가시킴을 확인하였다. 또한, asterosaponin $P_1$은 섬유아세포에서 진피의 주요 콜라겐인 type 1 콜라겐의 생합성을 촉진하는 효능을 보였다. 이상의 결과들로부터, 별불가사리로부터 분리한 asterosaponin $P_1$은 노화에 동반되는 피부 증상을 개선하는 항노화 화장품 소재로 활용될 수 있을 것으로 판단된다.
우리나라 서남단의 지리산에는 천연야생에서 자생하는 한방약초의 보고라고 할 수 있다. 현재 약초로 분류되는 식물로는 1200여종이 자생 혹은 재배되고 있다. 최근들어 이들 중 현대인의 피부노화억제와 관련하여 관심을 끌고 있는 약초로 홍화에 관한 연구가 활발히 연구되고 있는데 금번연구에서는 홍화의 주성분인 폴리페놀 화합물(Polyphenol cocktail)이 30 - 40대 중년기의 여성에게 주름생성을 억제시켜 피부노화에 대한 지연효과를 줄 수 있는 천연물질임을 확인하고자 한다. 본 연구에서는 1차적으로 in vitro법으로 DPPH assay로 항산화효과를 확인하고 더불어 인체에 대한 임상시험을 통해 얼굴피부의 주름발생에 대한 억제 혹은 개선효과가 있음을 입증하고자 한다. 또한 홍화추출물을 주성분으로 하는 항노화 마스크팩 시제품의 개발로 진일보하여 기능성화장품 산업의 발전에 기여할 것임을 확신한다.
In oriental medicine, the skin of the face and the body is the mirror of the viscera and bowels. Skin aging is measured according to the elasticity and glossiness of the skin, which is perceived as a matter of fluid-humor within the realms of oriental medicine. Fluid-humor refers to normal body fluid that makes the skin moist and supple and the hair bright and glossy. If the body is lacking fluid-humor, the hair and skin will be dry and coarse. 'The improvement of fluid-humor (nutrition)' is facilitated based on the oriental physiological change theory for middle-aged women. A new study is therefore necessary to develop oriental herbal cosmetics for young women in their 20s. As yet, there has been no study on the effect of herbal cosmetics formulated for middle-aged women and used by young women in their 20s. This study aims to investigate the effect of 'the improvement of fluid-humor' for the skin of young women in their 20s within the theory of oriental medicine. This kind of study is essential for oriental skin care and the development of diverse oriental herbal cosmetics. To determine the effect of oriental herbal cosmetics on young women in their 20s based on the theory of oriental medicine, which says that a shortage of fluid-humor causes skin aging, this study has examined the skin conditions of young women in their 20s and how satisfied they are with oriental herbal cosmetics through an objective equipment-based measurement and subjective questionnaire survey.
Advancement toward aging society has presented the importance of house planning in consideration of the elderly, increasing the necessity of such planning. Therefore the importance of indoor design guideline for elderly houses has been stressed and the understanding of the aging in regard to this trend will create more mature spaces. In line with this trend the purpose of this study is to study the medical factors related with bathroom interior design guidelines of the existing elderly houses from the viewpoint of deteriorating physical functions due to aging process. In order to research the deterioration of physical functions due to aging literatures of medical and exercise physiology were used and concerning the existing bathroom design guidelines for the aged people, the guideline items of behavior facilitation, physiological maintenance and perceptual maintenance aspects out of Murtha & Lee's user benefit criteria(1976) related with physical functions were selected to research in relation with the knowledge about the deterioration phenomena of physical functions. Physical aging and deterioration aspects were classified from the viewpoint of musculoskeletal disorder, cardiovascular disorder, respiratory disorder, gastronomy disorder, urology disorder, somatosensory disorder, endocrine disorder, immune disorder, nervous disorder and skin diseases and these were utilized in interpreting total 100 items of bathroom design guideline. Because bathroom is the space where many physical movements are done, it had the closest relationship with the deterioration of musculoskeletal health in general and as bathroom is the space where people use water and feel the difference in temperature and moisture more than other spaces, deterioration of skin and somatosensory health had the next closest relationship. The result of this study revealed that regarding the deterioration of physical functions of the elderly people in aspect of their perceptibility as designers will design creatively and sincerely based on its relationship with nervous system in future, the result of this study will be used to develop better spatial designs efficiently to meet for the aging society.
