A Study on the Effect of Aging on the Xylene Toxicity in Rats (흰쥐 성장기간에 따른 Xylene의 독성에 관한 연구)

  • To evaluate an effect of aging on the xylene toxicity, 50% m-xylene in olive oil (0.25 ml/100 g body wt.) was administered to 5 week and 12 week-old rats one times intraperitoneally and sacrificed at 24 hrs afterwards. The increasing rate of urinary m-methylhippuric acid concentration was higher in 12 week-old rats than 5 week-old rats by the treatment of m-xylene. On the liver function findings, i.e., liver weigh/body weight (%), serum levels of ALT activity and hepatic malondialdehyde content, 12 week-old rats showed more severe liver injury than 5 weeks those in xylene-treated rats. And the hepatic cytochrome P-450 contents was higher in 12 weeks rats than those of in 5 week-old rats, but the increasing rate of that was lower in 12 week-old rats. Hepatic alcohol dehydrogenase activity was also higher in 12 week-old rats than in 5 week-old rats whereas the increasing rate of that was higher in 5 week-old than those in 12 week-old rats by the xylene treatment. Furthermore, the hepatic aldehyde dehydrogenase activities were no differences between the 5 and 12 week-old rats both in the control and xylene-treated group. In conclusion, age may influences upon the hepatotoxicity with xylene and it may be responsible for xylene metabolism in rats.

Effect of Hyuntogobon-hwan on Antioxidation Activity in Induced Aging Rats (현토고본환이 노화유발 백서의 항산화능에 미치는 영향)

  • Objectives : This experiment was done to evaluate the effects of Hyuntogobon-hwan(HTG) on antioxidant capability and lipidic concentration in blood, both of which are presumed to be related to aging. Methods : 12 week-old SD rats were divided into controlled group, uncontrolled group and HTG group. As controlled and HTG groups were given subcutaneous injection of D-galactose(50mg/kg/rat), at the same time HTG group was administered extract of Hyuntogobon-hwan(270mg/200g). HTG injections continued for 6 weeks. After initial injections, blood was drawn from each group and the following were measured: the activity of SOD, GSH-px, catalase in erythrocytes, TSARS value, concentration of total lipid, tryglyceride in blood plasma. Results : The activities of SOD and GSH-px in erythrocytes increased significantly in HTG group compared with controlled group. The activity of catalase seemed to increase slightly, but it was barely noticeable. The concentration of total lipid in plasma decreased significantly in HTG group compared with controlled group. The value of TBARS in plasma seemed to decrease slightly, but it was barely noticeable. Conclusions : According to the above results, Hyuntogobon-hwan has an influence on aging by virtue of activation of antioxidative enzyme systems in erythrocytes and concentrations of lipid in blood plasma.

  • 1. Objective The purpose of this study is to investigate the anti-aging and anti-oxidant effects of Bopyewon-tang(BPW) in Liver and Lung cell. 2. Methods This experiment was used the tissue of liver and lung cells of 6, 52 and 68 weeks old SD rats. Each age group was again divided into three groups. One group, as normal group, was not-treated cells, another group, as control group, was saline-treated cells, and the last group, as experimental group, was BPW -treated cells. After culture for 48 hours, each groups measured the level of SOD, GSH, MDA and NO in the tissue of liver and lung cells. 3. Results and Conclusion 1. The activity of SOD in liver of in 52 w-BPW group, 68 w-BPW group, and in lung of 68 w-BPW were significantly increased in comparison with those of the normal groups. 2. The levels of NO in liver of in 68 w-BPW group was significantly decreased in comparison with those of the normal groups. 3. The levels of MDA in lung of in 68 w-BPW group was significantly decreased in comparison with those of the normal groups.

Effect of Gamisogunjung-tang on Antioxidation Activity in Rats Induced Aging by D-Galactose (가미소건중탕(加味小建中湯)이 D-galactose로 노화(老化)를 유발(誘發)시킨 백서(白鼠)의 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objective : This experiment has been done to evaluate the effects of Gamisogunjung-tang(GST) on antioxidant capability and lipidic concentration in blood which are presumed to be related to aging. Method : In this study, we divided 14 weeks old SD rats into normal group, control group and GST group. Control and GST group were induced aging by D-galactose. At the same time GST group were administered extract of Gamisogunjung-tang for 6 weeks. After then we took blood, and measured the activities of SOD, GSH-px, catalase in erythrocytes and measured TBARS values, concentrations of total lipid, tryglyceride, total cholesterol, HDL-cholesterol in plasma. Results : The activities of SOD, GSH-px, catalase in erythrocytes increased significantly in GST group compared with control group. The value of TBARS and the concentration of total lipid, total cholesterol in plasma decreased significantly in GST group compared with control group. The concentration of HDL-cholesterol increased significantly in GST group compared with control group. The concentration of triglyceride were not noticeable. Conclusion : it is considered that Gamisogunjung-tang has an influence on control aging by activation the antioxidative enzyme systems in erythrocytes and decreasing the concentration of lipid in blood plasma.

Constipation anti-aging effects by dairy-based lactic acid bacteria

  • Constipation, which refers to difficulties in defecation and infrequent bowel movement in emptying the gastrointestinal system that ultimately produces hardened fecal matters, is a health concern in livestock and aging animals. The present study aimed to evaluate the potential effects of dairy-isolated lactic acid bacteria (LAB) strains to alleviate constipation as an alternative therapeutic intervention for constipation treatment in the aging model. Rats were aged via daily subcutaneous injection of D-galactose (600 mg/body weight [kg]), prior to induction of constipation via oral administration of loperamide hydrochloride (5 mg/body weight [kg]). LAB strains (L. fermentum USM 4189 or L. plantarum USM 4187) were administered daily via oral gavage (1 × 10 Log CFU/day) while the control group received sterile saline. Aged rats as shown with shorter telomere lengths exhibited increased fecal bulk and soften fecal upon administration of LAB strains amid constipation as observed using the Bristol Stool Chart, accompanied by a higher fecal moisture content as compared to the control (p < 0.05). Fecal water-soluble metabolite profiles showed a reduced concentration of threonine upon administration of LAB strains compared to the control (p < 0.05). Histopathological analysis also showed that the administration of LAB strains contributed to a higher colonic goblet cell count as compared to the control (p < 0.05). The present study illustrates the potential of dairy-sourced LAB strains as probiotics to ameliorate the adverse effect of constipation amid aging, and as a potential dietary intervention strategy for dairy foods including yogurt and cheese.

Acute Toxicity Study of Yan-Sheng Health Liquid in Rats and Mice (랫드와 마우스에서 연생호보액의 급성 경구 독성 시험에 관한 연구)

  • The acute toxicity of orally administered Yan-Sheng health liquid (YSHL), water extracts from twelve Chinese drugs (Cervi Cornu, Lonicerae Flos, Foeniculi Fructus, Glycyrrhizae Radix, Liriopis Tuber, Raphani Semen, Bombyx, Ginseng Radix alba, Cinnamomi loureirii Cortex, Epimedii Herba, Zingiberis Rhizoma, Lycii Fructus) was evaluated in male and female Sprague-Dawley rats and ICR mice. Rats and mice aging 5 weeks were gavaged with 0, 2.0, 3.0, 4.4, 6.7, 10.0, 66.7, or 100.0 ml/kg of YSHL. No animal died by oral treatment and no toxic symptom was observed in the treated animals during 5 days. The body weight of the treated animals was not significantly different from the controls. The results of macroscopic examination on the organs of the treated animals revealed no abnormal findings. Therefore, it was concluded that YSHL was practically non-toxic when it was orally administered to rats and mice, and its LD50 was suspected to be greater than 100 ml/kg in rats and mice.

Effects of Dietary Fiber Sources on the Composition Intestinal Microflora in Aged Rate (노화 흰쥐의 장내 균총 조성에 대한 식이섬유질원의 영향)

  • This study was performed to investigate the influence of vadrious dietary fiber sources such as mugwort, apple, sea mustard, butterbur on the intestinal in aged rats. Nine groups of aged rats (12months after weanling) were fed each experimental diet containing 5% and 15% dietary fibers for 4 weeks. The contents in large intestine of aged rats used for the measurement of intestinal microflora. Total viable counts and the numbers of Bifidobacterium, Lactobacillus, Clostridium, E. coli and Staphylococcus were determined by nonselective medium and various selective media. The intake of dietary fiber sources employed did not show any differece in the number of total microorganisms, Lactobacillus and Bifidobacterium. It was remarkable that Clostridium perfringens appeared in the colon of aged rats. The prpulations of Peptococcus and Clostridium were significantly larger in the apple 5% group than in other dietary fiber groups while the number of Staphylococcus was significantly smaller in the dietary fiber groups than in cellulose group. And the Streptococcus populations decreased in the mugwort 15% group. Accordingly, the mugwort 15% group is effective in improving intestinal flora of aged rats.

  • The acute toxicity of recombinant human epidermal growth factor, DWP-401 was evaluated in SD rats. Male and female rats aging 6 weeks were administered orally or subcutaneously with 0, 0.125, 0.25, 0.5, 1 and 2 mg/kg of DWP-401. No deaths and no toxic symptoms related to the DWP-401 were observed. The body weights of treated animals were not significantly different from the controls. The results of necropsy revealed no abnormal gross findings of the organs in treated animals. LD50 values of DWP-401 for male and female rats were estimated to be over 2 mg/kg, which is approximately 2, 000 times of expected clinical dose.

A Quantitative Comparison of Fibroblasts, Collagen and Elastic Fiber Densities in the Young and Aged Rat Skin

  • Computerized image processing and analysis system was used for quantitative assessment of skin tissue components in color histological sections. The purpose of this study was to determine that the changes in the collagen fiber density and elastic fiber density in dermis in the rat skin as aging. And also to determine that the correlation between fibroblast density and collagen fiber density and elastic fiber density in the aged rat skin. Ten weeks old ($130{\sim}150g$) eight and fifty-four weeks old ($300{\sim}350g$) eight female Sprague-Dawley rats were used. The full-thickness skin biopsy specimens were prepared serial sections and stained with hematoxylin and eosin, Masson's trichrome and Verhoeff-van Gieson. The collagen fiber and the elastic fiber were identified using the image analysis processing system and then calculated the collagen fiber density rate and the elastic fiber density rate in the dermis. It also identified fibroblast and calculated fibroblast density in the dermis. By using a Student's t-test, a decrease in the collagen fiber density rate (t=-4.650, P<0.001) and the elastic fiber density rate (t=-6.494, P<0.001) of dermis can be observed in aged rats as compared with the young rats. A Student's t-test showed a significantly less fibroblast density in the aged rats than the young rats (t=-4.497, P<0.001). There were significantly positive correlation between the fibroblast density and the collagen fiber density rate (r=.69, P<0.001) and the elastic fiber density rate (r=.91, P<0.001). These results indicate that the aging may decrease the collagen fiber density and elastic fiber density due to reduced the proliferative and synthetic activity of fibroblast in the dermis.
